Heat stress occurring during rice (Oryza sativa) grain development reduces grain quality, which often manifests as increased grain chalkiness. Although the impact of heat stress on grain yield is well-studied, the genetic basis of rice grain quality under heat stress is less explored as quantifying grain quality is less tractable than grain yield. To address this, we used an image-based colorimetric assay (Red, R; and Green, G) for genome-wide association analysis to identify genetic loci underlying the phenotypic variation in rice grains exposed to heat stress. We found the R to G pixel ratio (RG) derived from mature …

Innate immune recognition of bacterial pathogens is a key determinant of the ensuing systemic response, and host or pathogen heterogeneity in this early interaction can impact the course of infection. To gain insight into host response heterogeneity, we investigate macrophage inflammatory dynamics using primary human macrophages infected with Group B Streptococcus. Transcriptomic analysis reveals discrete cellular states within responding macrophages, one of which consists of four sub-states, reflecting inflammatory activation. Infection with six additional bacterial species—Staphylococcus aureus, Listeria monocytogenes, Enterococcus faecalis, Yersinia pseudotuberculosis, Shigella flexneri, and Salmonella enterica—recapitulates these states, though at …

Blood lipids are heritable modifiable causal factors for coronary artery disease. Despite well-described monogenic and polygenic bases of dyslipidemia, limitations remain in discovery of lipid-associated alleles using whole genome sequencing (WGS), partly due to limited sample sizes, ancestral diversity, and interpretation of clinical significance. Among 66,329 ancestrally diverse (56% non-European) participants, we associate 428M variants from deep-coverage WGS with lipid levels; ~400M variants were not assessed in prior lipids genetic analyses. We find multiple lipid-related genes strongly associated with blood lipids through analysis of common and rare coding variants. We discover several associated rare non-coding variants, largely at Mendelian lipid …

The presence of insoluble aggregates of amyloid β (Aβ) in the form of neuritic plaques (NPs) is one of the main features that define Alzheimer's disease. Studies have suggested that the accumulation of these peptides in the brain significantly contributes to extensive neuronal loss. Furthermore, the content and distribution of cholesterol in the membrane have been shown to have an important effect on the production and subsequent accumulation of Aβ peptides in the plasma membrane, contributing to dysfunction and neuronal death. The monomeric forms of these membrane-bound peptides undergo several conformational changes, ranging from oligomeric forms to beta-sheet structures, each …

In live cells, phase separation is thought to organize macromolecules into membraneless structures known as biomolecular condensates. Here, we reconstituted transcription in condensates from purified mitochondrial components using optimized in vitro reaction conditions to probe the structure-function relationships of biomolecular condensates. We find that the core components of the mt-transcription machinery form multiphasic, viscoelastic condensates in vitro. Strikingly, the rates of condensate-mediated transcription are substantially lower than in solution. The condensate-mediated decrease in transcriptional rates is associated with the formation of vesicle-like structures that are driven by the production and accumulation of RNA during transcription. The generation of RNA alters …

Remitted waves are used for sensing and imaging in diverse diffusive media from the Earth's crust to the human brain. Separating the source and detector increases the penetration depth of light, but the signal strength decreases rapidly, leading to a poor signal-to-noise ratio. Here, we show, experimentally and numerically, that wavefront shaping a laser beam incident on a diffusive sample enables an enhancement of remission by an order of magnitude at depths of up to 10 transport mean free paths. We develop a theoretical model which predicts the maximal remission enhancement. Our analysis reveals a significant improvement in the sensitivity …

The birth of a child from an unregistered polygamous marriage according to the prevailing laws is a juridical phenomenon that cannot be denied. As the part of legal events, unregistered polygamous marriages can bring further legal consequences to children born out of wedlock, which in law children born out of wedlock have a lower position than children born from legal marriages. Because of these differences, the law provides a solution for children born out of wedlock to have the same legal status as children born from legal marriages, namely through child legalization. This research will be continued by focusing on …

Personality And Psychopathology Correlates Of Institutional Misconduct Among Juvenile Offenders, Paula Floyd
Dissertations
Juvenile delinquency is a national concern, as delinquent behavior is associated with a host of poor psychosocial outcomes during later adolescence and adulthood. To address delinquency, it is important to understand psychological adjustment among adolescents who have already made contact with the juvenile justice system. One way to explore adjustment within this population involves examining personality and psychopathology correlates of institutional misconduct (i.e., behavioral infractions while incarcerated). While there is a robust body of literature regarding personality and psychopathology correlates of misconduct in adult inmate samples, there has been far less work devoted to these relationships among justice-involved youth. Furthermore, …

Late Cretaceous (late Campanian) diatom assemblages from the Campbell Plateau (Zealandia), southwest Pacific Ocean, obtained from Deep Sea Drilling Project (DSDP) Leg 29 Site 275, contain well-preserved specimens of two enigmatic diatom species currently assigned to the genus Ktenodiscus; Micrampulla parvula originally described from the Maastrichtian-age Moreno Shale, California, and Pterotheca cretacea from DSDP Site 275. In general, the two species share a number of common features with modern Corethron (domed valves, probable heterovalvate frustules, T-shaped serrated articulated spines, marginal sockets), but differ in the location of the sockets (i.e. vertically at the base of the valve dome and …

Context: Yield gap (Yg) analyses using farmer-reported yield and management data have been performed for a number of annual grain crops, but it lacks for perennial forages. The U.S. accounts for 21 % of the global alfalfa production with a large rainfed area located in the central Great Plains, serving as an interesting case-study for Yg in perennial forages. Most existing alfalfa Yg analyses quantified the magnitude of the Yg but failed to identify associated management practices to reduce it. Challenging this analysis, a systematic benchmark for alfalfa water productivity [WP, kg dry matter per mm evapotranspiration (ETc)] that …

Weeds compete with crops for soil moisture, along with other resources, which can impact the germination, growth, and seed production of weeds; however, this impact has not been systematically recorded and synthesized across diverse studies. To address this knowledge gap, a global meta-analysis was conducted using 1,196 paired observations from 86 published articles assessing the effect of water stress on weed germination, growth characteristics, and seed production. These studies were conducted and published during 1970 through 2020 across four continents (Asia, Australia, Europe, and North America). Imposed water stress was expressed as solution osmotic potential (ψsolution), soil water potential (ψsoil), …

Excessive gestational weight gain (GWG) is associated with adverse pregnancy outcomes. This metabolome-wide association study aimed to identify metabolomic markers for GWG. This longitudinal study included 39 Black and White pregnant women with a prepregnancy body mass index (BMI) of ≥ 25 kg/m2. Untargeted metabolomic profiling was performed using fasting plasma samples collected at baseline (mean: 12.1 weeks) and 32 weeks of gestation. The associations of metabolites at each time point and changes between the two time points with GWG were examined by linear and least absolute shrinkage and selection operator (LASSO) regression analyses. Pearson correlations between the …

Peripheral T-cell lymphomas (PTCLs) are a heterogeneous group of aggressive non-Hodgkin lymphomas, the majority of which have high relapse rates following standard therapy. Despite use of consolidative stem cell transplant (SCT) following frontline therapy, there remains no consensus on its utility. The double-blind randomized phase 3 ECHELON-2 study (#NCT01777152; clinicaltrials.gov) demonstrated improved progression-free survival (PFS) and overall survival with frontline brentuximab vedotin plus cyclophosphamide, doxorubicin, and prednisone (A+CHP). Herein, we conducted an exploratory subgroups analysis of the impact of consolidative SCT on PFS in patients with previously untreated CD30+ PTCL (ALK- anaplastic large cell lymphoma [ALCL] and non-ALCL) …

The Qualitative Report
Many special-needs children, including students with learning disabilities, are enrolled in public schools. The current state of education is transitioning from online to face-to-face learning due to the improving situation after the COVID-19 pandemic. Hybrid learning is one of the alternative methods applied during the transition period. Therefore, this study aims to explore the experiences of teachers in implementing hybrid learning in students with disabilities in Indonesia using a qualitative description. Data was collected through interviews with one class teacher, two students with learning disabilities and their parents, and one school principal. Furthermore, observations were made on the learning process …

To assess the levels of infection across communities during the coronavirus disease 2019 pandemic, researchers have measured severe acute respiratory syndrome coronavirus 2 RNA in feces dissolved in sewer water. This activity is colloquially known as sewer monitoring and is referred to as wastewater-based epidemiology in academic settings. Although global ethical principles have been described, sewer monitoring is unregulated for health privacy protection when used for public health surveillance in the United States. This study used Qualtrics XM, a national research panel provider, to recruit participants to answer an online survey. Respondents (N = 3,083) answered questions about their knowledge, …

Analyzing antigen-specific T cell responses at scale has been challenging. Here, we analyze three types of T cell receptor (TCR) repertoire data (antigen-specific TCRs, TCR-repertoire, and single-cell RNA + TCRαβ-sequencing data) from 515 patients with primary or metastatic melanoma and compare it to 783 healthy controls. Although melanoma-associated antigen (MAA) -specific TCRs are restricted to individuals, they share sequence similarities that allow us to build classifiers for predicting anti-MAA T cells. The frequency of anti-MAA T cells distinguishes melanoma patients from healthy and predicts metastatic recurrence from primary melanoma. Anti-MAA T cells have stem-like properties and frequent interactions with regulatory …
