Degradation Of Di (2-Ethylhexyl) Phthalate By Marine Sediment Bacteria And Identification Of Key Degraders And Degradative Genes, Ritu Ningthoujam
Chulalongkorn University Theses and Dissertations (Chula ETD)
Di (2-ethylhexyl) phthalate (DEHP) is a toxic phthalate ester (PAE) plasticizer that is predominantly detected in marine sediment and biota. DEHP degradation by bacteria from several environments has been studied, but very little is known about marine sediment bacteria that can degrade DEHP and other PAEs. Therefore, in this study, we enriched a bacterial consortium C10 that can degrade four PAEs of varying alkyl chain lengths (DEHP, dibutyl phthalate, diethyl phthalate, and dimethyl phthalate ; separately and as a mixture) from marine sediment. The major bacterial genera in C10 during the degradation of the PAEs were Glutamicibacter, Ochrobactrum, Pseudomonas, Bacillus, …

Characterization And Property Investigation Of Novel Human Glucose 6-Phosphate Dehydrogenase Inhibitors In Lung Cancer Cell Lines, Makamas Chanda
Chulalongkorn University Theses and Dissertations (Chula ETD)
G6PD plays fundamental roles in many cellular processes, including redox balance and lipid and nucleotide synthesis. Overexpression of G6PD is required to promote the proliferation and survival of cancer cells, including lung cancer. Targeting G6PD is a prominent strategy to inhibit cancer cell progression. Finding novel natural compounds with anticancer properties to inhibit G6PD activity needs to be investigated. Our study indicated that SJ006, which is 1,2-naphthoquinone, promoted cytotoxic activity against NSCLC cell lines. Interestingly, SJ006 exhibited a direct inhibitory effect on G6PD activity without interfering with mRNA or protein levels, which were strongly observed in both A549 and H292. …

Role Of Jak/Stat Signaling Pathway During Viral Infection In Black Tiger Shrimp Penaeus Monodon, Pasunee Laohawutthichai
Role Of Jak/Stat Signaling Pathway During Viral Infection In Black Tiger Shrimp Penaeus Monodon, Pasunee Laohawutthichai
Chulalongkorn University Theses and Dissertations (Chula ETD)
Janus Kinase/signal transducers and activators of transcription (JAK/STAT) signaling pathway plays an important role for antiviral immunity. To stimulate the pathway, the cytokines, interferons, or other stimulators activate the receptor, which is then dimerized and activate JAKs, causing phosphorylation of the tyrosine residues of the receptor. Consequently, STATs are phosphorylated, dimerized and localized into the nucleus to regulate their target gene transcription. This study aims to better understand the role of JAK/STAT signaling pathway in Penaeus monodon during viral infection. From tissue distribution, PmDOME, PmJAK and PmSTAT transcripts were expressed in all tested tissues and up-regulated about 2-fold upon WSSV …
Macrophage-Cancer Cell Interaction In Three Dimension Liver Cancer Model, Pornlapat Keawvilai
Macrophage-Cancer Cell Interaction In Three Dimension Liver Cancer Model, Pornlapat Keawvilai
Chulalongkorn University Theses and Dissertations (Chula ETD)
Hepatocellular carcinoma (HCC) is the one of the leading causes of cancer-related mortality. Liver cancer progression is significantly influenced by immune cells which infiltrate tumors and their interaction with liver cancer cells in the tumor microenvironment (TME). Circulating monocytes are recruited to tumor site by TME and differentiated into tumor-associated macrophages (TAMs) which are roughly divided into pro-inflammatory macrophages (M1) or pro-tumoral macrophages (M2). In order to study the interaction between macrophages and liver cancer cells. In in vitro, an effective liver cancer model which closely mimicks this crosstalk is necessary. This study aimed to fabricate a three-dimension (3D) liver …

Development Of Electrochemical Detection For Cortisol And Hepatitis C Virus Core Antigen, Pannaporn Pusomjit
Chulalongkorn University Theses and Dissertations (Chula ETD)
This dissertation focused on the development of electrochemical biosensors for the determination of essential biomarkers by using a label-free assay. This dissertation can be divided into two parts. In the first part, a non-invasive aptamer-based electrochemical biosensor using disposable screen-printed graphene electrodes (SPGEs) was developed for simple, rapid, and sensitive determination of cortisol. The specific recognition of the cortisol DNA aptamer (CApt) was modified with streptavidin magnetic beads (MBs) before simple immobilization onto the electrode surface using a neodymium magnet. The electrochemical behavior of the aptamer-based biosensor was assessed by using electrochemical impedance spectroscopy (EIS) and cyclic voltammetry (CV). Furthermore, …

Photocatalytic Films For Efficient Gas Phase Co2 Reduction Into Value-Added Chemicals, Mohammad Fereidooni
Chulalongkorn University Theses and Dissertations (Chula ETD)
A viable method to lower the atmospheric CO2 concentration is artificial photosynthesis, which converts CO2 and H2O into products with added value. Herein, the catalytic activity of thin photocatalytic films was examined in water vapor, batch, and flow reactor configurations for CO2 reduction. In addition to the experimental study, a theoretical analysis of the reaction mechanism over silica-modified amorphous TiO2 (AM-TiO2-SiO2) and indium tin oxide (ITO) photocatalysts is conducted. In the case of AM-TiO2-SiO2, the research shows that surfaces with low loading of SiO2 groups have a higher affinity for certain target molecules (T-M). This affinity accelerates adsorption and reaction …

Analysis Of Genetic Diversity In A Novel Vaccine Candidate, The Glutamic Acid-Rich Protein Of Plasmodium Falciparum (Pfgarp), Rattanaporn Rojrung
Chulalongkorn University Theses and Dissertations (Chula ETD)
Malaria vaccine development confronts high genetic polymorphism in some antigenic components of the vaccine candidates, which could potentially limit the vaccine efficacy. An antigen of Plasmodium falciparum, designated glutamic acid-rich protein (PfGARP), has recently been considered as a strong vaccine target against asexual-blood-stages parasite. It has been shown that naturally acquired anti-PfGARP antibodies elicited clinical protection against severe malaria among people living in hyperendemic areas. Furthermore, PfGARP has been involved in binding to human erythrocyte band 3, which induces erythrocyte aggregation and might bring about severe malaria. However, the extent of genetic variation in the pfgarp locus among field isolates …

Composition And Diversity Of Meibum Microbiota In Meibomian Gland Dysfunction And Its Correlation With Tear Cytokines Levels, Ubonwan Rasaruck
Chulalongkorn University Theses and Dissertations (Chula ETD)
Meibomian gland dysfunction (MGD) is commonly caused by obstruction of the terminal meibomian gland duct, which is associated with alterations in the quantity and quality of the meibum. This may affect the composition of meibum microbiota, causing aberrant cytokine production, epithelial hyperkeratinization, and meibomian gland blockage. This cross-sectional study included 44 patients with moderate to severe MGD and 44 healthy controls, to determine the meibum microbiota by next-generation sequencing (NGS) and its association with tear cytokines levels. We observed reduced bacterial diversity in the meibum microbiota of patients with severe MGD. Significantly higher abundance of Bacteroides and Novosphingobium, and substantially …

Checkpoint blockade immunotherapy has transformed the course of cancer treatments. The use of monoclonal antibodies (mAbs) specific against immune checkpoints has displayed satisfactory clinical benefits. However, its effectiveness is limited to just a small percentage of cancer patients. In this work, a recombinant DVD-AT2C bispecific antibody (bsAb) was developed for dual inhibition of PD-1/PD-L1 and CTLA-4 axes and produced in plants. Plant system was utilized as an alternative protein production platform. Transient expression of DVD-AT2C targeting PD-L1 and CTLA-4 in Nicotiana benthamiana was determined highest after 4 days of infiltration. The size, assembly, and protein properties of the purified bsAb were …

Development Of Plant-Produced Anti-Il-6r Mab, Namthip Kaewbandit
Chulalongkorn University Theses and Dissertations (Chula ETD)
After the body is invaded by pathogens, it responds by secreting cytokines, for example, interleukin 6 (IL-6), which activates an intracellular signaling pathway involving JAKs/STAT3 through phosphorylation. However, overexpression of pSTAT-3 can lead to a positive feedback loop of IL-6, causing the release of more cytokines and potentially leading to cytokine storm. Thus, inhibition of IL-6 signaling is another important target for Cytokine storm treatment. Tocilizumab, a humanized IL-6R antibody has been approved for RA treatment. There are several concerns about mammalian cell protein expression systems, such as higher production costs and pathogen contamination risks. Recently, plants have become increasingly …

Surface-Enhanced Raman Scattering Technique For Nanoplastics Detection, Boonphop Chaisrikhwun
Surface-Enhanced Raman Scattering Technique For Nanoplastics Detection, Boonphop Chaisrikhwun
Chulalongkorn University Theses and Dissertations (Chula ETD)
The size-dependent effect of nanoplastics (NPLs) restricts the quantification of NPLs by surface-enhanced Raman scattering (SERS) technique. In this study, we successfully established an innovative preparation method to eliminate this effect. This developed SERS method allowed us to quantify 100-, 300-, 600-, and 800-nm polystyrene nanospheres (PSNSs) in diverse aqueous conditions at a low concentration. The SERS substrate was easily fabricated and used as NPLs signal enhancement with sputtering gold particles onto a glass cover slide. By preconcentrating and dissolving PSNSs in toluene, SERS technique can be successfully quantify NPLs at extremely low concentrations with a limit of detection (LOD) …
Enzyme-Free Hydrogen Peroxide Sensor From Dual-Shaped Plasmonic Silver Nanoparticles, Charles Oliver Avenido
Enzyme-Free Hydrogen Peroxide Sensor From Dual-Shaped Plasmonic Silver Nanoparticles, Charles Oliver Avenido
Chulalongkorn University Theses and Dissertations (Chula ETD)
A novel growth-based strategy for the determination of hydrogen peroxide has been developed using a mixture of silver nanospheres (AgNSs) and citrate-capped silver nanoprisms (AgNPrs). The unique ability of hydrogen peroxide to oxidize silver nanoparticles and reduce silver metal consequently was exploited to produce a highly sensitive and selective sensor (AgNSs/NPrs). Citrate-capping of the AgNPrs only allows the lateral growth that resulted in a red shift and intensity enhancement of the in-plane dipole plasmon resonance (IPDPR) band. Consequently, distinct hues from yellow, orange, red, purple to blue were produced from the diluted sensors. Ratiometric analysis was employed to quantify H2O2 …

Chulalongkorn University Theses and Dissertations (Chula ETD)
Vanillic acid is an important compound used in food, beverage, fragrance, and pharmaceutical industries. Vanillic acid can be produced by using either natural plant, native, and engineered microorganisms. However, the industrial scale production of vanillic acid compound still difficult due to low yielding production which can be improved by metabolic engineering of bacterial cells. Hence, the efficient biocatalyst, engineering, and biotechnology process can be a promising strategy to enhance vanillic acid production from lignin derivatives and strengthen the large-scale production for industrial approach. Among several lignin-derived compounds, ferulic acid is a widely used substrate for vanillic acid production. Recently, the …
