Inflammatory monocytes (iMOs) and B cells are the main targets of the poxvirus ectromelia virus (ECTV) in the lymph nodes of mice and play distinct roles in surviving the infection. Infected and bystander iMOs control ECTV's systemic spread, preventing early death, while B cells make antibodies that eliminate ECTV. Our work demonstrates that within an infected animal that survives ECTV infection, intrinsic and bystander infection of iMOs and B cells differentially control the transcription of genes important for immune cell function and, perhaps, cell identity. Bystander cells upregulate metabolism, antigen presentation, and interferon-stimulated genes. Infected cells downregulate many cell-type-specific genes …

The Development Of A Genomic Toolbox For Studying The Evolutionary Genetics Of Reptilian Lungs Using The Chicken Model, Logan Thomas Edvalson
Theses and Dissertations
There is a vast diversity in tetrapod lung branching morphology. Phylogenetically, much of the pulmonary diversity among vertebrates appears to arise from the way epithelial tubes branch or form saccular (cyst) structures. Fgf10 activity has been shown to play a critical role in regulating branch versus cyst morphology. We hypothesize that the species-specific differences in lung morphology may be primarily due to species-specific differences in Fgf10 expression. To test this hypothesis, we have performed bioinformatic analyses on the Fgf10 locus and have identified a conserved 11 kb noncoding region that potentially contains the Fgf10 lung enhancer. We are taking a …

Alcoholic hepatitis (AH) is the most severe form of alcoholic liver disease for which there is no efficacious treatment aiding most patients. AH manifests differently in individuals, with some patients showing debilitating symptoms more so than others. Previous studies showed significant metabolic dysregulation associated with AH. Therefore, we sought to analyze how the activity of metabolic pathways differed in the liver of patients with varying degrees of AH severity. We utilized a genome-scale metabolic modeling approach that allowed for integration of a generic human cellular metabolic model with specific RNA-seq data corresponding to healthy and multiple liver disease states to …

BACKGROUND: Endometriosis is an estrogen-dependent inflammatory reproductive disease. Therefore, systematic estrogen depletion and anti-inflammatory drugs are the current treatment for endometriosis. However, current endometriosis treatments have low efficacy and cause adverse effects in endometriosis patients. Consequently, alternative endometriosis treatments targeting endometriosis-specific factors are in demand. In this context, ERβ was selected as a druggable target for endometriosis due to its critical role in progression. Therefore, selective targeting of ERβ without inhibiting ERα activity would be a new paradigm for endometriosis treatment to overcome the low efficacy and adverse effects of hormonal endometriosis therapy.
METHODS: Cell-based ERβ and ERα activity assay …

Von Willebrand Factor (VWF) plays a critical role in thrombus formation, stabilization, and propagation. Previous studies have demonstrated that targeted inhibition of VWF induces thrombolysis when administered in vivo in animal models of ischemic stroke. The study objective was to quantify dose-dependent inhibition of VWF-platelet function and its relationship with thrombolysis using BB-031, an aptamer that binds VWF and inhibits its function. VWF:Ac, VWF:RCo, T-TAS, and ristocetin-induced impedance aggregometry were used to assess BB-031-mediated inhibition of VWF. Reductions in original thrombus surface area and new deposition during administration of treatment were measured in a microfluidic model of arterial thrombolysis. Rotational …

Transition To Kindergarten: Parental Efficacy And Experiences During Covid-19, Samira Amirazizi, Emily Edelman, Matthew Quirk, Erin Dowdy
Perspectives on Early Childhood Psychology and Education
Previous research has highlighted the effectiveness of supporting parents in fostering children’s academic and/or social-emotional development in preparation for kindergarten. Considering the emergence of COVID-19 as a risk factor to which all children and parents have been exposed, it is essential to understand parental mental health as it relates to early childhood development. This study utilized mixed methods to examine the experiences and perspectives of parents (N = 82) whose children were transitioning into kindergarten during the fall of 2020 amidst the COVID-19 pandemic. Results indicate that parents’ knowledge/confidence in managing their child’s transition to kindergarten was inversely associated with …

The Ecosystem Of Racial Inequalities In Discipline In Early Childhood Education, Alexandra Aylward, Rachel Garver, Catherine Kramarczuk Voulgarides, Christine Hodgson
Perspectives on Early Childhood Psychology and Education
Preschool children are suspended and expelled at a rate greater than school-aged youth, and exclusionary discipline practices are further inequitable across racial and ethnic groups. Denied the documented benefits of early childhood education, Black students are disproportionately excluded from US educational institutions beginning in early childhood, effectively preserving and reproducing racial inequities. Black students, especially boys, are the most likely to lose access to schooling due to exclusionary discipline. The disparities are dehumanizing and detrimental to students’ opportunities to learn, as early educational experiences greatly influence development and future outcomes. Although there is a plethora of evidence concerning the significant …

Humboldt Journal of Social Relations
This essay explores some of the gendered dimensions of contemporary cannabis politics and culture from the perspective of a long-time sexuality and gender scholar recently migrated into the world of drug policy reform. My essay suggests that inattention to gendered stereotypes and inequalities creates obstacles to women’s full participation in drug policy reform and complicates efforts to end marijuana prohibition in the U.S.
Small-Scale Marijuana Growing: Deviant Careers As Serious Leisure, Craig Boylstein, Scott R. Maggard
Small-Scale Marijuana Growing: Deviant Careers As Serious Leisure, Craig Boylstein, Scott R. Maggard
Humboldt Journal of Social Relations
Previous research on marijuana growers in the United States has primarily focused on those who have been incarcerated for the activity (Weisheit, 1990; 1998). These growers tend to produce large amounts of the product. It may be that large scale marijuana growers who have been arrested may differ in their reasons for growing, their style of growing and distributing and in how they view marijuana growing as an activity from different types of growers; namely small-scale, indoor hydroponic growers who grow primarily for personal use and enjoyment. Our study analyzes one social network of marijuana growers in central and northern …
Should Per Se Limits Be Imposed For Cannabis? Equating Cannabinoid Blood Concentrations With Actual Driver Impairment: Practical Limitations And Concerns, Paul Armentano
Humboldt Journal of Social Relations
Fourteen US states have amended their longstanding, effect-based DUI drug laws to per se or zero tolerant per se statutes in regard to cannabis. Other states are considering enacting similar legislation. Under these amended traffic safety laws, it is a criminal violation for one to operate a motor vehicle with trace levels of cannabinoids or their metabolites in his or her blood or urine. Opponents of per se cannabinoid limits argue that neither the presence of cannabinoids nor their metabolites are appropriate or consistent predictors of behavioral or psychomotor impairment. They further argue that the imposition of such per se …
Designing “‘Queer’ Across Cultures:” Disrupting The Consumption Of Diversity, Kim Berry
Designing “‘Queer’ Across Cultures:” Disrupting The Consumption Of Diversity, Kim Berry
Humboldt Journal of Social Relations
This article charts the underlying logic and structure of a course titled “‘Queer’ Across Cultures,” created to provide a transnational focus for the groundbreaking minor in Multicultural Queer Studies, spearheaded by Eric Rofes. Concerned that students would too easily revert to a practice of consuming diversity, I have attempted to weave together readings which complicate a colonial gaze by challenging hegemonic constructions of essential difference. My primary strategy in crafting this course has been to focus on the term “across” in the title. In other words to foreground and theorize the transnational within the production of both similarities and differences …

Racial discrimination and inequality have perpetuated within the U.S. since its inception. In 2016, Colin Kaepernick initiated the national anthem protests to oppose the oppression of people of color in America. This study was developed in 2018 to identify social determinants of health underlying discriminatory beliefs and behaviors. The objective was to investigate the impacts of college students’ race, gender, political ideology, socio-economic status [SES], NFL interest, patriotism, and general protest support on support for the national anthem protests. We administered paper-and-pencil surveys across locations on the James Madison University campus using a convenience sample. There were 408 participants included, …

Mesopelagic organisms play a crucial role in marine food webs, channelling energy across the predator-prey network and connecting depth strata through their diel vertical migrations. The information available to assess mesopelagic feeding interactions and energy transfer has increased substantially in recent years, owing to the growing interest and research activity in the mesopelagic realm. However, such data have not been systematically collated and are difficult to access, hampering estimation of the contribution of mesopelagic organisms to marine ecosystems. Here we present MesopTroph, a georeferenced database of diet, trophic markers, and energy content of mesopelagic and other marine taxa compiled from …
Synthetic Amphiphiles As Antibiotic Potentiators, Helena Spikes
Synthetic Amphiphiles As Antibiotic Potentiators, Helena Spikes
Dissertations
Antibiotic resistance has become a massive threat to modern medicine. Bacteria acquire resistance either through genetic mutations or mobile genetic elements, such as plasmids. The growing resistance crisis is exacerbated by over-prescription of antibiotics and improper use. As antimicrobial resistance becomes more widespread, superbugs (bacteria resistant to more than one class of drug) have evolved. Since few new drugs reach clinical trials and even fewer are approved by the FDA, we must find a way to make existing drugs more potent. One technique to accomplish this is by using combination therapy. By administering two or more drugs at a time, …

Species Substitution And Mislabeling Of Ceviche, Poke, And Sushi Dishes Sold In Orange County, California, Courtney J. Kitch, Amanda M. Tabb, Grace E. Marquis, Rosalee S. Hellberg
Food Science Faculty Articles and Research
Raw, ready-to-eat (RTE) seafood products have become increasingly popular globally, but they are vulnerable to species substitution and mislabeling. DNA barcoding allows for fish species identification by extracting, amplifying, and sequencing a standardized gene target. A wide variety of fish products have been studied with DNA barcoding, but little investigation of ceviche and poke has occurred in the United States. Sushi is known to be a target of mislabeling but has not been extensively studied in Orange County, CA. The objective of this study was to investigate species substitution and mislabeling of sushi, poke, and ceviche dishes sold at restaurants …

Humans are social animals who engage in a variety of collective activities requiring coordinated action. Among these, music is a defining and ancient aspect of human sociality. Human social interaction has largely been addressed in dyadic paradigms, and it is yet to be determined whether the ensuing conclusions generalize to larger groups. Studied more extensively in non-human animal behavior, the presence of multiple agents engaged in the same task space creates different constraints and possibilities than in simpler dyadic interactions. We addressed whether collective dynamics play a role in human circle drumming. The task was to synchronize in a group …
Aldosterone Treatment Protects Connexin 30 And 43 Expression For Age-Related Hearing Loss, Jennifer A. Pineros Valencia
Aldosterone Treatment Protects Connexin 30 And 43 Expression For Age-Related Hearing Loss, Jennifer A. Pineros Valencia
USF Tampa Graduate Theses and Dissertations
Age-related hearing loss (ARHL), also known as presbycusis, is the number one communication disorder affecting our aged population. Connexin proteins are essential for intercellular communication throughout the human body. Mutations in connexin genes have been linked to human syndromic and nonsyndromic deafness; thus, we hypothesize changes in connexin gene and protein expression with age may be involved in the development of ARHL. In our current study, connexin mRNA gene expression for CBA/CaJ mice at different age points were examined and correlations were analyzed between the changes in gene expression and functional hearing measures, such as ABRs and DPOAEs. Moreover, we …

Alzheimer's disease biomarkers are becoming increasingly important for characterizing the longitudinal course of disease, predicting the timing of clinical and cognitive symptoms, and for recruitment and treatment monitoring in clinical trials. In this work, we develop and evaluate three methods for modelling the longitudinal course of amyloid accumulation in three cohorts using amyloid PET imaging. We then use these novel approaches to investigate factors that influence the timing of amyloid onset and the timing from amyloid onset to impairment onset in the Alzheimer's disease continuum. Data were acquired from the Alzheimer's Disease Neuroimaging Initiative (ADNI), the Baltimore Longitudinal Study of …

Quantifying Environmental Sensitivity Of Marine Resources To Oil Well Blowouts In The Gulf Of Mexico, Emily Chancellor
USF Tampa Graduate Theses and Dissertations
This dissertation focused on oil spill risk assessment techniques historically utilized for coastal resources (e.g., environmental sensitivity indices (ESIs), vulnerability assessments, species-specific vulnerability frameworks) and their application and expansion to offshore resources found in the Gulf of Mexico (GoM). Additional included techniques (e.g., multi-sector trade-off analysis, marine spatial planning (MSP) software) provide support in decision making processes regarding the potential siting of oil production sites and/or the withdrawal of areas from oil and natural gas production. Chapter 1 included an overview and justification for this study. Chapter 2 demonstrated an initial methodology for the creation of offshore ESIs and how …
