Getting By With A Little Help From Friends: A Qualitative Case Study Of Students’ Strategies For Coping With Failure In An Undergraduate Biology Laboratory Course, Soo Won Shim, Nancy Pelaez
Purdue University Libraries Open Access Publishing Fund
Recent emphasis on research competencies in undergraduate biology education means that more students are doing course-based research. Professional research scientists learn from failed research, but undergraduate students who encounter failure in their biology lab research may not always respond in ways that advance their learning. There is a need to examine individual students’ responses to failed research as they conduct investigations in an undergraduate lab course. Here, we report a qualitative research case study based on data from interviews and course work to examine five undergraduate students’ emotional responses, coping strategies, and perceptions of learning as they confronted failure in …

Major depressive disorder (MDD) is a progressive deteriorating mental state with a feeling of worthlessness and frequent mood swings. Several studies reported the favorable effects of natural drug substances on MMD associated oxidative stress and neuroinflammation. The present study is attempted to examine whether carveol could affect lipopolysaccharide- (LPS-) induced depression, and if so, how nuclear factor E2-related factor (Nrf2) contributed to the neuroprotective effects of carveol mechanistically. Two experimental cohorts were used using the SD rats: first to evaluate the promising dose of carveol (whether 20 mg/kg or 50 mg/kg) and secondly to determine the effect of carveol on …

Wildlife translocations alter animal movement behavior, so identifying common movement patterns post-translocation will help set expectations about animal behavior in subsequent efforts. American and Eurasian beavers (Castor canadensis; Castor fiber) are frequently translocated for reintroductions, to mitigate human-wildlife conflict, and as an ecosystem restoration tool. However, little is known about movement behavior of translocated beavers post-release, especially in desert rivers with patchy and dynamic resources. We identified space-use patterns of beaver movement behavior after translocation. We translocated and monitored nuisance American beavers in desert river restoration sites on the Price and San Rafael Rivers, Utah, USA, and compared …

Multidrug resistance (MDR) is one of the major therapeutic challenges that limits the efficacy of chemotherapeutic response resulting in poor prognosis of ovarian cancer (OC). The multidrug resistance protein 1 (MRP1) is a membrane-bound ABC transporter involved in cross resistance to many structurally and functionally diverse classes of anticancer drugs including doxorubicin, taxane, and platinum. In this study, we utilize homology modelling and molecular docking analysis to determine the binding affinity and the potential interaction sites of MRP1 with Carboplatin, Gemcitabine, Doxorubicin, Paclitaxel, and Topotecan. We used AutoDock Vina scores to compare the binding affinities of the anticancer drugs against …

An Overlooked Epidemic: Pediatric Oral Health Concerns On The Rise In Rural Communities, Emma Verbrugge
Honors Program: Senior Projects (Public)
Pediatric oral health is a concern for many dentists and parents as they lay the foundation for long-term proper hygiene in children. Early childhood caries (ECC) is a disease of chronic tooth decay marked as the most common chronic disease of childhood in the world. Children living in rural areas are even more susceptible to this disease and thus significant attention is needed to curve these numbers. The consequences of ECC are much more than just cosmetic, but include heightened risk of gingivitis, periodontal disease, diabetes, cardiovascular disease, and much more. A foundational cause of poor dental hygiene lies with …

A ratiometric genetically encoded voltage indicator (GEVI) would be desirable for tracking transmembrane voltage changes in the presence of sample motion. We performed combinatorial multi-site mutagenesis on a cyan-excitable red fluorescent protein to create the bright and monomeric mCyRFP3, which proved to be uniquely non-perturbing when fused to the GEVI ASAP3. The green/red ratio from ASAP3-mCyRFP3 (ASAP3-R3) reported voltage while correcting for motion artifacts, allowing the visualization of membrane voltage changes in contracting cardiomyocytes and throughout the cell cycle of motile cells.

Mucins are components of the mucus layer overlying the intestinal epithelial cells, which maintains physiological homeostasis. Altered mucin expression is associated with disease progression. Expression of MUC4 decreases in colorectal cancer (CRC); however, its functional role and implications in the intestinal pathology in CRC are not studied well. Therefore, we generated a genetically engineered Muc4 knockout (Muc4-/-) CRC mouse model by crossing with Muc4-/- and Apcflox/flox mice in the presence of colon-specific inducible Cre. We observed that deficiency of Muc4 results in an increased number of macroscopic tumors in the colon and rectal region and leads to poor survival. …
Aptamer Functionalisation Of Back-Gated Graphene Field Effect Transistors For Pb2+ Sensing, S Awan
Aptamer Functionalisation Of Back-Gated Graphene Field Effect Transistors For Pb2+ Sensing, S Awan
School of Engineering, Computing and Mathematics
We discuss the development of aptamer functionalized, back-gated, graphene field effect transistor (GFET) biosensors for the targeted detection of Lead (Pb2+) ions. The widespread existence of the heavy metal Pb2+ in the environment is a severe threat to the health of humans. It is a neurotoxin that accumulates over time in the body restricting the cognitive, behavioral and psychological development of children along with causing irreversible harm to the human fetus. New biosensors which allow for the rapid, sensitive and low-cost detection of Pb2+ are required to monitor this toxicant in water sources worldwide. The GFET devices were fabricated using …

Anthropocene Fiction: Empathy, Kinship, And The Troubled Waters At The End Of The World, Megan Mandell Stowe
Anthropocene Fiction: Empathy, Kinship, And The Troubled Waters At The End Of The World, Megan Mandell Stowe
USF Tampa Graduate Theses and Dissertations
Anthropocene literature features characters and environments grappling with the devastation of climate change. Donna Haraway’s term Chthulucene narrows this category further by exposing the tentacular connections between overpopulation, dependence on material goods, species loss, and other facets of global warming—such as water as a resource. Three novels of the Chthulucene exhibit these connections, centering on the importance of water to both ecologies and personal identity. As seen in the novels, place attachment brings ecology and identity together by forging a stewardship between person and location, often resulting in efforts to defend and preserve a place. In Octavia Butler’s Parable of …

An electrical device that transforms the electricity into the waves of radio and vice versa is termed the antenna. Its main deployment is in the transmitter and receiver of the antenna. While transmission, the transmitter of radio at the extremities of the antenna furnishes the electricity which oscillates at the frequency of radio wave and energy is released as current as em waves. Some of the voltage is formed from the em wave that is invaded at the point of receiving to amplify the receiver. This study focuses on the analysis of the satellite system to aid in mobile antenna …

Natives And Nativars: Understanding Pollinator Preference For Native Plants And Their Cultivated Counterparts In The Pacific Northwest, Jen Hayes, Gail Langellotto
Urban Ecosystem Research Consortium of Portland/Vancouver
Native cultivars present a paradox for ecologically-minded gardeners: growing interest in native plants is largely related to their benefits to pollinators, yet many native plants found at nurseries may be significantly altered, compared to wild genotypes. The goals of our research are to identify potential differences in bee preference for wild type natives or cultivars, and to understand what floral traits plant breeding impacts. In this presentation, we focus on native bee visitation to 7 genera of Oregon native plants and one to three associated cultivars, documented over two field seasons (2020 and 2021). This study is ongoing, and takes …

Urban Ecosystem Research Consortium of Portland/Vancouver
Invasive species pose a serious statewide threat to Oregon. In 2017, the Oregon Invasive Species Council (OISC) published the Statewide Strategic Plan for Invasive Species. Development of a web-based hub that facilitates learning and collaboration on the prevention, early detection and management of invasive species is a priority in the Statewide Strategic Plan. The hub transformed the former Oregon’s Worst Invaders List into a searchable and sortable web-based database. The hub has different metrics which can be queried, such as species status in Oregon, introduction or dispersal pathways of concern, suitable habitat, timely alerts, or combinations of multiple metrics. Through …

The Future Of Portland’S Forests: Conifer Regeneration And Barriers To Recruitment In Urban And Rural Forests, Mila Pruiett
Urban Ecosystem Research Consortium of Portland/Vancouver
Some reports suggest that Portland’s urban forests have low conifer regeneration rates. If so, this could be due either to a shortage of viable seeds or to a shortage of viable habitat. Nurse logs, in particular, have been identified as a substrate that increases conifer germination and survival rates. My study aims to determine 1) if there is less conifer regeneration in urban forests compared to rural forests, and, if so, 2) do urban forests produce less conifer seeds than rural forests, and 3) do urban forests have less nurse log surface area than rural forests. I sampled five of …

Enhancer of zeste homolog 2 (EZH2), a core component of polycomb repressive complex 2, plays an important role in cancer development. As both oncogenic and tumor suppressive functions of EZH2 have been documented in the literature, the objective of this study is to determine the impact of Ezh2 deletion on the development and progression of endometrial cancer induced by inactivation of phosphatase and tensin homolog (PTEN), a tumor suppressor gene frequently dysregulated in endometrial cancer patients. To this end, we created mice harboring uterine deletion of both Ezh2 and Pten using Cre recombinase driven by the progesterone receptor …

Faculty, Staff and Students Publications
Determinants of length of life are not well understood, and therefore increasing lifespan is a challenge. Cardinal theories of aging suggest that oxidative stress (OxS) and mitochondrial dysfunction contribute to the aging process, but it is unclear if they could also impact lifespan. Glutathione (GSH), the most abundant intracellular antioxidant, protects cells from OxS and is necessary for maintaining mitochondrial health, but GSH levels decline with aging. Based on published human studies where we found that supplementing glycine and N-acetylcysteine (GlyNAC) improved/corrected GSH deficiency, OxS and mitochondrial dysfunction, we hypothesized that GlyNAC supplementation could increase longevity. We tested our hypothesis …

Oral squamous cell carcinoma (OSCC) is a top-ranked cancer in the Pakistani population, and patient survival has remained unchanged at ∼50% for several decades. Recent advances have claimed that a subset of tumour cells, called cancer stem cells (CSCs), are responsible for tumour progression, treatment resistance, and metastasis, which leads to a poor prognosis. This study investigated the impact of CSC markers expression on overall survival (OS) and disease-free survival (DFS) of OSCC patients. Materials and Methods. Immunohistochemistry was used to evaluate CD44, CD133, L1CAM, and SOX2 expression in a well-characterized cohort of 100 Pakistani patients with primary treatment …

Faculty and Staff Scholarship
Life history and metabolism covary, but the mechanisms and individual traits responsible for these linkages remain unresolved. Dispersal capability is a critical component of life histories that is constrained by metabolic capacities for energy production. Conflicting relationships between metabolism and life histories may be explained by accounting for variation in dispersal and maximal metabolic rates. We used female wing-polymorphic sand field crickets, Gryllus firmus, selected either for long wings (LW) and flight-capability or short wings (SW) and high early lifetime fecundity to test the hypothesis that selection on dispersal capability drives the evolution of metabolic capacities. While resting metabolic …
