Tumor interactions predominantly dominate cancer progression and its tumor microenvironment. Such interactions include, tumors generating signals that cause dysfunction and death to immune cells [1]. Inflammatory and immune cells present to eliminate cancer cells fail and lose their overall function [1]. These cells lose the overall ability to aid and are co-opted to promote tumor growth. Purpose: To explore and assess awareness of and attitudes toward "undetectable equals untransmittable" (U=U) and associations with social-ecological factors among minority women. Population: Ethnic minority women 18 years and older residing in the El Paso, TX, US - Juarez, Mexico border region. Methods: A cross-sectional convergent parallel mixed methods study using in-depth interviews and a survey instrument was conducted among a purposive sample. Thematic analysis was done on qualitative data, and descriptive, bivariate and binary logistic regression analyses were done on quantitative data. Caliche or pedogenic calcium carbonate (CaCO3) layers are a common feature of arid soils, but the environmental controls on their distribution are not fully understood. Caliche layers are thought to play an important role in shallow subsurface water storage and movement, due to their water retention capacity and ability to impede vertical water transport (Hennessy et al., 1983). Therefore, caliche distribution and stability play a role in controlling water distribution in arid lands. However, due to its largely subsurface nature, caliche can be difficult to investigate and map without significant effort. Aedes aegypti is a medically important mosquito species that transmits multiple arboviruses, including dengue, chikungunya, Zika, and yellow fever. This mosquito species has expanded its geographical range into expanded into the Northern Chihuahuan Desert to further increase the risk of infection by these viruses in naïve human populations. This work presents an in-depth numerical investigation into a hypothesized two-layer central pattern generator (CPG) that controls mammalian walking and how different parameter choices might affect the stepping of a simulated neuromechanical model. Particular attention is paid to the functional role of features that have not received a great deal of attention in previous work: the weak cross-excitatory connectivity within the rhythm generator and the synapse strength between the two layers. Sensitivity evaluations of deafferented CPG models and the combined neuromechanical model are performed. Transformer neural networks have made a significant impact on natural language processing. The Transformer network self-attention mechanism effectively addresses the vanishing gradient problem that limits a network learning capability, especially when the time series gets longer or the size of the network gets deeper. This dissertation examines the usage of the Transformer model for time-series forecasting and customizes it for a simultaneous multistep-ahead prediction model in a surrogate model predictive control (MPC) application. To act effectively, humans store event schemas and use them to predict the near future. How are schemas learned and represented in memory, and used in online comprehension? One means to answer these questions is modeling event comprehension. What are, then, computational principles of event comprehension? We proposed three candidate properties: 1) abstract representation of visual features, 2) predictive mechanism and prediction error as feedback, and 3) contextual cues to guide prediction, and adapted a computational model embodying these properties. Background: Respiratory syncytial virus (RSV) is known to cause bronchiolitis and other upper respiratory infections (URIs) in children. However, given the COVID-19 pandemic and the resultant lockdowns, circulation of other respiratory viruses was uncharacteristically low. Restrictions are quickly falling and with increased interaction there has been an uncharacteristic jump in cases.
Purpose: To summarize the pathogenesis of RSV and the mechanisms the virus uses to evade immune responses. Additional discussion of the increased rates of RSV hospitalizations as well as the role of vaccinations in controlling infection.

Light scattering experiments were performed on bovine βH crystallin, an important protein component of the mammalian eye lens. Light scattering results are compared to simplified models for the free energy of mixing of β crystallins with water. The static light scattering data are well-represented by hard convex body equations of state having dimensionless non-sphericity coefficients of close to 2, compatible with prolate spherocylinders, and weight-average molecular weights ranging between 2−5×105 g/mole. Additionally, quasi-elastic light scattering data was obtained which shows that βH has a hydrodynamic radius near 7.3 nm. These experiments extend light scattering work on β crystallins well into …

This study aimed to understand the essential nature of media literacy, evaluate pre-developed higher education classes for existing media literacy context, and recommend best practices for incorporating media literacy into an undergraduate curriculum. This mixed-methods study of media literacy in undergraduate college courses explored the presence and absence of media literacy lessons within core classes by auditing 15 online course shells accessed through the university’s Learning Management System (LMS). Specifically, all the courses surveyed included the first skill of media literacy, Access; 33% of the classes included Analyze; 27% included Creation; 20% included Reflection; and 20% included Action. This phenomenological study was designed to evaluate the experiences of African American Christian adult clients within traditional psychotherapy and faith-based counseling settings. Research has shown various therapist/counselor identities affect Christian African American clients, and that Christians as well as African Americans have historically been less likely to pursue traditional psychotherapy for a variety of reasons. Participants were six African American Christian adult participants (three traditional psychotherapy participants and three faith-based counseling participants). All participants were asked open-ended questions about their experiences in traditional psychotherapy or faith-based counseling. AIM: To describe adults with (non-dialysis) chronic kidney disease (CKD) in nine public renal practice sites in the Australian state of Queensland.
METHODS: 7,060 persons were recruited to a CKD Registry in May 2011 and until start of kidney replacement therapy (KRT), death without KRT or June 2018, for a median period of 3.4 years.
RESULTS: The cohort comprised 7,060 persons, 52% males, with a median age of 68 yr; 85% had CKD stages 3A to 5, 45.4% were diabetic, 24.6% had diabetic nephropathy, and 51.7% were obese. Younger persons mostly had glomerulonephritis or genetic renal disease, while older persons …

HSP90 inhibitors destabilize oncoproteins associated with cell cycle, angiogenesis, RAS-MAPK activity, histone modification, kinases and growth factors. We evaluated the HSP90-inhibitor ganetespib in combination with standard chemotherapy in patients with high-risk early-stage breast cancer. I-SPY2 is a multicenter, phase II adaptively randomized neoadjuvant (NAC) clinical trial enrolling patients with stage II-III breast cancer with tumors 2.5 cm or larger on the basis of hormone receptors (HR), HER2 and Mammaprint status. Multiple novel investigational agents plus standard chemotherapy are evaluated in parallel for the primary endpoint of pathologic complete response (pCR). The Orphanet Rare Disease Ontology (ORDO) provides a structured vocabulary encapsulating rare diseases. Downstream applications of ORDO depend on its accuracy to effectively perform their tasks. In this paper, we implement an automated quality assurance pipeline to identify missing is-a relations in ORDO. We first obtain lexical features from concept names. Then we generate related and unrelated feature sharing concept-pairs, where a feature sharing concept-pair can further generate derived term-pairs. If an unrelated and related feature sharing concept-pair generate the same derived term-pair, then we suggest a potential missing is-a relation between the unrelated feature sharing concept-pair. Higher-order chromatin structure is important for the regulation of genes by distal regulatory sequences. Structural variants (SVs) that alter three-dimensional (3D) genome organization can lead to enhancer-promoter rewiring and human disease, particularly in the context of cancer3. However, only a small minority of SVs are associated with altered gene expression4,5, and it remains unclear why certain SVs lead to changes in distal gene expression and others do not. Activation-induced cytidine deaminase, AICDA or AID, is a driver of somatic hypermutation and class-switch recombination in immunoglobulins. In addition, this deaminase belonging to the APOBEC family may have off-target effects genome-wide, but its effects at pan-cancer level are not well elucidated. Here, we used different pan-cancer datasets, totaling more than 50,000 samples analyzed by whole-genome, whole-exome, or targeted sequencing. AID mutations are present at pan-cancer level with higher frequency in hematological cancers and higher presence at transcriptionally active TAD domains. AID synergizes initial hotspot mutations by a second composite mutation. AID mutational load was found to be independently associated with …

Metastatic involvement of the spine is a common complication of systemic cancer progression. Surgery and external beam radiotherapy are palliative treatment modalities aiming to preserve neurological function, control pain and maintain functional status. More recently, with development of image guidance and stereotactic delivery of high doses of conformal radiation, local tumor control has improved; however recurrent or radiation refractory disease remains a significant clinical problem with limited treatment options. Immune checkpoint inhibitors (ICIs) have yielded remarkable responses but often lead to immune-related adverse events (irAEs). Although germline causes for irAEs have been hypothesized, no individual variant associated with developing irAEs has been identified. We carried out a genome-wide association study of 1,751 patients on ICIs across 12 cancer types. We investigated two irAE phenotypes: (1) high-grade (3-5) and (2) all-grade events. We identified 3 genome-wide significant associations (P < 5 × 10-8) in the discovery cohort associated with all-grade irAEs: rs16906115 near IL7 (combined P = 3.6 × 10-11; hazard ratio (HR) = 2.1); rs75824728 near IL22RA1 (combined P = 3.5 × 10-8; HR = 1.8); and rs113861051 on 4p15 (combined P = 1.2 × 10-8, HR = 2.0); rs16906115 was replicated in 3 independent studies. The association near IL7 colocalized with the gain of a new cryptic exon for IL7, a critical regulator of lymphocyte homeostasis. Patients carrying the IL7 germline variant exhibited significantly increased lymphocyte stability after ICI initiation, which was itself predictive of downstream irAEs and improved survival.

OBJECTIVE: Optical imaging studies of oral premalignant lesions have shown that optical markers, including loss of autofluorescence and altered morphology of epithelial cell nuclei, are predictive of high-grade pathology. While these optical markers are consistently positive in lesions with moderate/severe dysplasia or cancer, they are positive only in a subset of lesions with mild dysplasia. This study compared the gene expression profiles of lesions with mild dysplasia (stratified by optical marker status) to lesions with severe dysplasia and without dysplasia.

The oldest form of radiation therapy, brachytherapy, has been investigated and reported in the scientific and medical literature for well over a century. Known by many names over the years, radium-based, empirical practices evolved over decades to contemporary practice. This includes treatment at various dose rates using multiple radionuclides or even electrically generated photon sources. Predictions or prognostications of what may happen in the future enjoy a history that spans centuries, e.g. those by Nostradamus in the 1500s. BACKGROUND: Biobehavioral factors such as social isolation and depression have been associated with disease progression in ovarian and other cancers. Here, the authors developed a noninvasive, exosomal RNA profile for predicting ovarian cancer disease progression and subsequently tested whether it increased in association with biobehavioral risk factors.
METHODS: Exosomes were isolated from plasma samples from 100 women taken before primary surgical resection or neoadjuvant (NACT) treatment of ovarian carcinoma and 6 and 12 months later. Lymphoma risk is elevated for relatives with common non-Hodgkin lymphoma (NHL) subtypes, suggesting shared genetic susceptibility across subtypes. To evaluate the extent of mutual heritability among NHL subtypes and discover novel loci shared among subtypes, we analyzed data from eight genome-wide association studies within the InterLymph Consortium, including 10,629 cases and 9,505 controls. We utilized Association analysis based on SubSETs (ASSET) to discover loci for subsets of NHL subtypes and evaluated shared heritability across the genome using Genome-wide Complex Trait Analysis (GCTA) and polygenic risk scores. BACKGROUND & AIMS: Populations consuming soy have reduced risk for breast cancer, but the mechanisms are unclear. We tested the hypothesis that soy isoflavones, which have ovarian hormone-like effects, can reduce fibroglandular breast tissue (FGBT, 'breast density'), a strong risk marker for breast cancer.

Skeletal muscle atrophy is a prevalent complication in multiple chronic diseases and disuse conditions. Fibroblast growth factor-inducible 14 (Fn14) is a member of the TNF receptor superfamily and a bona fide receptor of the TWEAK cytokine. Accumulating evidence suggests that Fn14 levels are increased in catabolic conditions as well as during exercise. However, the role of Fn14 in the regulation of skeletal muscle mass and function remains poorly understood. Dis/misinformation was a major concern in the 2016 U.S. presidential election and has only worsened in recent years. Even though domestic actors often spread dis/misinformation, actors abroad can use it to spread confusion and push their agenda to the detriment of American citizens. Even though this report focuses on actors outside the United States, the methods they use are universal and can be adapted to work against domestic agents. A solid understanding of these methods is the first step in combating foreign dis/misinformation campaigns and creating a new information literacy paradigm.

While statistics tend to focus on the difficulties facing tribal education, this article endeavors to look at the matter with fresh eyes. The federal administrative paradigm governing tribal schools has gone from a tool of cultural genocide to a mechanism for empowerment. A survey of recent governmental reforms demonstrates an embrace of the diversity of Indigenous communities, an interest in empowering students through learning, and an acknowledgement of a history of active disenfranchisement. Attention-deficit/hyperactivity disorder (ADHD) is a highly prevalent, inheritable, and heterogeneous neurodevelopmental disorder. Children with a family history of ADHD are at elevated risk of having ADHD and persisting its symptoms into adulthood. The objective of this study was to investigate the influence of having or not having positive family risk factor in the neuroanatomy of the brain in children with ADHD. Produced water (PW) is the effluent generated during oil mining and extraction. On average, for every barrel of oil, 4-5 barrels of PW are generated worldwide. The presence of various contaminants in PW makes it toxic. Disposal of untreated PW into oceans and water bodies can cause adverse effects on human health and the environment. Taking into account the large volumes of it being generated, and its effects on the environment, proper treatment is required before reuse or disposal.
