The U.S. government is one of the largest polluters on the planet. With over 700 domestic military bases and countless more federal facilities and vessels operating within state borders, there exists an enormous potential for spills and discharges of pollutants into state waters. The regulatory burden for enforcing environmental laws against the federal government falls on the Environmental Protection Agency and state regulators. But enforcing laws and regulations against the federal government and its progeny is a daunting regulatory task.

Background: The daily cycling of plant physiological processes is speculated to arise from the coordinated rhythms of gene expression. However, the dynamics of diurnal 3D genome architecture and their potential functions underlying the rhythmic gene expression remain unclear. Results: Here, we reveal the genome-wide rhythmic occupancy of RNA polymerase II (RNAPII), which precedes mRNA accumulation by approximately 2 h. Rhythmic RNAPII binding dynamically correlates with RNAPII-mediated chromatin architecture remodeling at the genomic level of chromatin interactions, spatial clusters, and chromatin connectivity maps, which are associated with the circadian rhythm of gene expression. Thermal Inactivation Kinetics Of Salmonella In Milk Powder As Impacted By Water Activity And Powder Type, Erika Litta Kadas

Historically, low-water activity (aw) foods (aw <0.65) were considered to be microbiologically safe for consumption. However, these foods have been implicated in outbreaks of pathogens such as Salmonella enterica, at a frequency that surely challenges this assumption of safety. Although usually implicated in outbreaks involving poultry and egg products, Salmonella spp. have frequently been the culprit in several outbreaks and recalls associated with low-water activity foods (LWAF) due to contamination resulting from the environment, animals, or even the employees during pre-or post-processing. One such LWAF that has been associated with Salmonella spp. outbreaks is milk powder. Milk powders are used in a variety of products ranging from infant formula to confectionary goods. With their widespread use, improving their safety is imperative. A key step in improving their safety is to thoroughly investigate preventive controls related to milk powder processing. The Food Safety Modernization Act (FSMA) established the Preventive Controls for Human Foods Rule, which mandates that processors design and establish a food safety plan outlining any potential hazards and the steps they will take to ensure those hazards do not compromise the safety of the product. Presently, the dairy industry does not have an established kill-step for milk powders post-spray drying. Spray drying may reduce some microbial populations, however, desiccation(e.g., spray drying) is not considered a kill-step. The work outlined in this thesis aims to assist our dairy industry partners in establishing their own thermal process for milk powders post-spray drying. Moisture sorption isotherms of two milk powders (nonfat dry milk: NFDM and a milk protein concentrate with 85% protein content: MPC-85) were determined and showcased that the temperatures at which the isotherms were determined (23, 40, or 60°C) significantly affected both the adsorption and desorption isotherms (p<0.0001) for each powder. Secondly, the thermal inactivation kinetics of Salmonella spp. in both powders were evaluated at two different water activities (0.20 and 0.30) and three temperatures (75, 80, and 85°C) to determine the effect of water activity on the thermal resistance of a cocktail of Salmonella spp. The D-values of Salmonella were heavily influenced by the thermal treatment temperature (p< 0.0001), but not the water activity (p >0.05). The overall findings contained within this thesis provide valuable information about the thermal inactivation kinetics of Salmonella spp. to our partners in the dairy industry so that they may implement an appropriate thermal process for their milk powders.

3D bioprinting of biological scaffolds requires control of the physicochemical properties of each unique structures. A promising material for control of properties is hydrogels, which can help create biomimetic scaffolds with controlled spatial arrangement of materials by integrating biological materials directly into layers during the bioprinting process. Nanocellulose offers a unique combination of properties including mechanical, biomimetic, and biocompatibility. These properties offer flexibility over the types, shapes, and applications of their printed hydrogel scaffolds, (i.e., tissue, drug, encapsulation). Introduction: Genome-Wide Association Studies (GWAS) identify tagging variants in the genome that are statistically associated with the phenotype because of their linkage disequilibrium (LD) relationship with the causative mutation (CM). When both low-density genotyped accession panels with phenotypes and resequenced data accession panels are available, tagging variants can assist with post-GWAS challenges in CM discovery.

Objectives: Our objective was to identify additional GWAS evaluation criteria to assess correspondence between genomic variants and phenotypes, as well as enable deeper analysis of the localized landscape of association.

Since its introduction, the amine-thiol cosolvent system has been successfully utilized for the deposition of various thin-film devices, but its mechanism of action is still uncertain. Herein, we have attempted to dissect some of the chemical characteristics of amine-thiol cosolvents, with a special interest taken towards a mixture of ethylenediamine (en) and mercaptoethanol (ME). Conductivity was measured for multiple amine-thiol combinations at different ratios to determine extent of ionization in solution, with en-ME having one of the highest solution conductivities. Type-1 diabetes mellitus (T1DM) has been repeatedly linked to executive functioning (EF) deficits in children and adolescents but the mechanisms underlying this association are not well understood. Chronic hyperglycemia — frequently measured as glycosylated hemoglobin A1c (HbA1c) levels — and increased glycemic variability have both been proposed as risk factors for EF difficulties in this population, but it is unclear whether they each uniquely reflect T1DM-related risk for EF deficits. As universities contend with high rates of student attrition from intended STEM majors, due to many students’ difficulty in passing entry-level mathematics courses, they must examine the systems they have in place and determine how best to support these students. Historically, graduate teaching assistants (GTAs) are assigned to introductory calculus classes, which serve as undergraduate-level gatekeeping courses. Epitope mapping a protein that enables pathogenesis is crucial for the development of therapies and prophylactics that can inhibit the pathogen’s function and its transmission of disease. The lipoprotein OspA enables Lyme Disease etiologic pathogen, Borrelia burgdorferi, to inhabit the tick midgut until transmission occurs. Anti-OspA mAbs and their smaller VHH counterparts are highly specific and tailored to bind proteins such as OspA, reproducibly, at established binding sites or epitopes. The Eocene-Oligocene Transition (EOT) at ∼34 Ma marked a climatic shift from greenhouse to icehouse conditions, toward long-lasting lower global temperatures and a continental ice sheet in the Antarctic. We report on sedimentological and inorganic geochemical results across the EOT at Ocean Drilling Program (ODP) Site 696 in the Weddell Sea, within the Antarctic limb of the Atlantic circulation. The geochemical composition of detrital, authigenic and biogenic marine sediment components, and sortable silt proxies demonstrate the impact of ice growth on high latitude water masses. Optogenetics has been widely applied to cardiovascular research using different models. Among them, Drosophila melanogaster (fruit fly) outstands for its similarity of human genes for disease modeling and short life cycle for rapid screening to analyze genetic mechanisms of heart disease. However, most of the current models only allow either activation or inhibition of the heartbeat, which is not sufficient to model complex arrhythmia. Many poor health outcomes later into adulthood have been triggered by adverse life events that have occurred during childhood. Most of the general population has experienced some sort of adverse childhood event that has negatively impacted their future health. Adverse childhood events include any traumatic event that has occurred during childhood including maltreatment (sexual abuse, mental abuse, and physical abuse), neglect, parental loss, and peer victimization.

Early pregnancy loss continues to be problematic in the cattle industry. A large proportion of the pregnancy loss occurs during the maternal recognition of pregnancy (MRP) phase of gestation. During the MRP, a developing conceptus must secrete sufficient interferon tau to inhibit release of the uterine- derived and luteolytic mediator, prostaglandin F2 alpha (PGF2��). If successful, the progesterone- secreting corpus luteum (CL) will be maintained, which is necessary for establishment and maintenance of pregnancy. Berberine (BBR) is a plant-derived alkaloid popularly used in a complementary context to treat splenomegaly and chronic disease states such as rheumatoid arthritis, diabetes, and cancer. Recent research has examined BBR as a regulator of lipid and glucose metabolism in association with the AMPK pathway; but little is known concerning its immunological impacts. The prevalence of prediabetes continues to rise in the United States and without effective intervention, a significant portion of those with prediabetes will go on to develop diabetes. Prediabetes affects approximately one-third of the population (96 million American adults) and 80% of those people are unaware of their prediabetic status. Proper screening for prediabetes could provide patients with access to necessary interventions, such as intensive lifestyle modifications or pharmacologic treatments, that could prevent or delay the onset of diabetes.
