Focusing Of High-Amplitude Sound Waves Using The Time Reversal Process, Brian D. Patchett Dec 2022

Focusing Of High-Amplitude Sound Waves Using The Time Reversal Process, Brian D. Patchett

Theses and Dissertations

Time reversal is a method often used to focus sound to a desired location, and works best in a reverberant environment. The effect of focus location within a reverberant environment is presented first, revealing that proximity to reflecting surfaces has a significant effect on the amplitude of the focus both experimentally and when using a modal summation model. These effects are a primary component to creating focus signals at high amplitudes. High-amplitude focusing experiments show that when multiple sources are used simultaneously to generate a focus, a peak amplitude pressure spike of 200 dB can be achieved in air. A …

NK cell deficiencies (NKD) are a type of primary immune deficiency in which the major immunologic abnormality affects NK cell number, maturity, or function. Since NK cells contribute to immune defense against virally infected cells, patients with NKD experience higher susceptibility to chronic, recurrent, and fatal viral infections. An individual with recurrent viral infections and mild hypogammaglobulinemia was identified to have an X-linked damaging variant in the transcription factor gene ELF4. The variant does not decrease expression but disrupts ELF4 protein interactions and DNA binding, reducing transcriptional activation of target genes and selectively impairing ELF4 function. Corroborating previous murine models …

The year 2021 saw the start of the UN Decade of Ocean Science for Sustainable Development. Ocean and coastal ecosystems play crucial roles in supporting our planet and our lives, from regulating climate and protecting shorelines, to providing food and employment. The UN estimates that billions of people rely on the ocean for their livelihoods. The First Global Integrated Marine Assessment (World Ocean Assessment I), published by the United Nations in 2016 (UN 2016), flagged that much of the ocean was severely degraded. The second (WOAII), released last year, suggests that the situation has not improved (UN 2021). The UN …

Ecological Niche Modeling Of The Macedonian Mouse, Mus Macedonicus (Mammalia, Rodentia), Under Climate Change Conditions, S Dermitas, Ne Gunduz, David T. Bilton

School of Biological and Marine Sciences

Mus macedonicus Petrov & Ružic, 1983, also known as the Macedonian mouse or Balkan short-tailed mouse, lives in the southern Balkans and the Middle East. While this species is common in Mediterranean ecosystems and is listed as “least concern” by the IUCN, little is known about how its distribution may shift with climate change. This study explores the ‘species’ potential distribution in three different periods: during the Last Glacial Maximum (LGM) and under current and future climate scenarios, using Maximum Entropy modelling. Modelling was based on 137 georeferenced occurrence records from Macedonia, Bulgaria, Greece, Turkey, Syria, Lebanon, Israel, Iran, Georgia, …

Oncogenic FOXO1 gene fusions drive a subset of rhabdomyosarcoma (RMS) with poor survival; to date, these cancer drivers are therapeutically intractable. To identify new therapies for this disease, we undertook an isogenic CRISPR-interference screen to define PAX3-FOXO1-specific genetic dependencies and identified genes in the GATOR2 complex. GATOR2 loss in RMS abrogated aa-induced lysosomal localization of mTORC1 and consequent downstream signaling, slowing G1-S cell cycle transition. In vivo suppression of GATOR2 impaired the growth of tumor xenografts and favored the outgrowth of cells lacking PAX3-FOXO1. Loss of a subset of GATOR2 members can be compensated by direct genetic activation of mTORC1. …

The Protein Phosphatase 2a Holoenzyme Is A Key Regulator Of Starch Metabolism And Bradyzoite Differentiation In Toxoplasma Gondii, Jin-Lei Wang, Ting-Ting Li, Hany M Elsheikha, Qin-Li Liang, Zhi-Wei Zhang, Meng Wang, L David Sibley, Xing-Quan Zhu

2020-Current year OA Pubs

Phenotypic switching between tachyzoite and bradyzoite is the fundamental mechanism underpinning the pathogenicity and adaptability of the protozoan parasite Toxoplasma gondii. Although accumulation of cytoplasmic starch granules is a hallmark of the quiescent bradyzoite stage, the regulatory factors and mechanisms contributing to amylopectin storage in bradyzoites are incompletely known. Here, we show that T. gondii protein phosphatase 2A (PP2A) holoenzyme is composed of a catalytic subunit PP2A-C, a scaffold subunit PP2A-A and a regulatory subunit PP2A-B. Disruption of any of these subunits increased starch accumulation and blocked the tachyzoite-to-bradyzoite differentiation. PP2A contributes to the regulation of amylopectin metabolism via dephosphorylation …

Lipoprotein(a) (Lp(a)) represents a unique subclass of circulating lipoprotein particles and consists of an apolipoprotein(a) (apo(a)) molecule covalently bound to apolipoprotein B-100. The metabolism of Lp(a) particles is distinct from that of low-density lipoprotein (LDL) cholesterol, and currently approved lipid-lowering drugs do not provide substantial reductions in Lp(a), a causal risk factor for cardiovascular disease. Somatic genome editing has the potential to be a one-time therapy for individuals with extremely high Lp(a). We generated an LPA transgenic mouse model expressing apo(a) of physiologically relevant size. Adeno-associated virus (AAV) vector delivery of CRISPR-Cas9 was used to disrupt the LPA transgene in …

Development Of A 3d-Printed Microfluidic Droplet-On-Demand System For The Deterministic Encapsulation And Processing Of Biological Materials, Chandler A. Warr

Theses and Dissertations

The growing threat of antimicrobial resistance is among the largest concerns in the world today. One method under development to combat this issue is the encapsulation of microbes in microfluidic droplets for single-cell testing. This method may be able to circumvent the need for a traditional positive cell culture which consumes the majority of the testing time using current diagnostic methods. This dissertation presents a method by which to deterministically encapsulate microbes using an artificial intelligence object detection algorithm and a Droplet-On-Demand microfluidic device. To accomplish this, the Droplet-On-Demand microfluidic device was first developed using a unique 3D-printing manufacturing method. …

Worldwide Distribution Of Cave-Dwelling Chelodesmidae (Diplopoda, Polydesmida), Rodrigo Salvador Bouzan, Jackson C. Means, Kaloyan Ivanov, Rodrigo L. Ferreira, Antonio Domingos Brescovit, Luiz Felipe Moretti Iniesta

International Journal of Speleology

Chelodesmidae is one of the most species rich families within the Myriapoda. However, little is known regarding their association with caves. We provide a list of all Chelodesmidae taxa reported from caves, map their worldwide distribution, and discuss the troglomorphic features of the group. A total of 25 species and subspecies from 20 genera and 2 subfamilies have been recorded from 59 caves and cave systems in 11 countries. These numbers represent a surprisingly modest proportion (~3%) of the approximately 800 described species in the family. Records of cave-dwelling chelodesmids appear to be geographically biased with most taxa reported from …

Simulated Problem-Based Learning: Teaching International Law In Exciting Way, Nik Nor Suhaida Ali

Indonesian Journal of International Law

This research aimed to discuss the methods for teaching International Law to non-law students. The subject was offered to second and third-year students of the International Affairs Management program at the School of International Studies, Universiti Utara Malaysia. Since the subject required higher-order thinking skills, it was introduced with Simulated Problem-Based Learning (SPBL), which combines role-playing simulation and problem-based learning (PBL). In western universities, simulations, PBL, and role-playing were popular methods in teaching International Law and International Relations. The methods were shown to improve learning skills but were rarely used in teaching International Law in International Relations programs in Malaysia. …

Herein we report the first molecular assessment of intra-species genetic variation and interrelationships within the Rio Grande Chirping frog, Eleutherodactylus campi. We analyzed 548 base pairs of 16S rRNA gene for 71 ingroup individuals belonging to the genus Eleutherodactylus (including 42 E. campi sampled from 15 localities in the United States and Mexico) and four outgroup samples. By unveiling two highly divergent and geographically structured clades within E. campi this study provides a novel phylogenetic placement of E. campi populations north and south of the Rio Grande Valley as sister groups to each other. The observed level of genetic …

Pre-Hospital Conditions Affecting The Hospitalization Risk In Older Adults At The Emergency Department, Karin Erwander, Kjell Ivarsson, Mona Landin-Olsson, Björn Agvall Dec 2022

Pre-Hospital Conditions Affecting The Hospitalization Risk In Older Adults At The Emergency Department, Karin Erwander, Kjell Ivarsson, Mona Landin-Olsson, Björn Agvall

Journal of Geriatric Emergency Medicine

BACKGROUND: The Emergency Department (ED) is a common route to hospitalization for critically ill and older adults. Older patients are admitted to hospital at a higher rate and have longer length of stay (LOS) when hospitalized. To be able to confront an increasing aging population, meet their medical needs and influence rising costs of health care, there is a need to focus on the older population. In Scandinavia, few studies are made that focus on the geriatric population at the ED. It is essential to early identify risk factors for hospitalization at the ED to improve the medical care for …

Free Radical-Based Sequencing For Native Top-Down Mass Spectrometry, Carolina Rojas Ramírez, Rayan Murtada, Jinshan Gao, Brandon T. Ruotolo Dec 2022

Free Radical-Based Sequencing For Native Top-Down Mass Spectrometry, Carolina Rojas Ramírez, Rayan Murtada, Jinshan Gao, Brandon T. Ruotolo

Department of Chemistry and Biochemistry Faculty Scholarship and Creative Works

Native top-down proteomics allows for both proteoform identification and high-order structure characterization for cellular protein complexes. Unfortunately, tandem MS-based fragmentation efficiencies for such targets are low due to an increase in analyte ion mass and the low ion charge states that characterize native MS data. Multiple fragmentation methods can be integrated in order to increase protein complex sequence coverage, but this typically requires use of specialized hardware and software. Free-radical-initiated peptide sequencing (FRIPS) enables access to charge-remote and electron-based fragmentation channels within the context of conventional CID experiments. Here, we optimize FRIPS labeling for native top-down sequencing experiments. Our labeling …

Pseudo-Embryology And Personhood: How Embryological Pseudoscience Helps Structure The American Abortion Debate, Scott F. Gilbert Dec 2022

Pseudo-Embryology And Personhood: How Embryological Pseudoscience Helps Structure The American Abortion Debate, Scott F. Gilbert

Biology Faculty Works

There is a pseudo-embryology flourishing today, well nourished by popular science, religious ideologies, and the public media. Just as eugenics was a pseudoscience that influenced (and still influences) American popular culture and which was responsible for racist anti-immigration laws (such as the Immigration Restriction Act of 1924), pseudo-embryology is also influencing popular culture and legislation. This new pseudoscience promotes the belief that science supports current zygotic and fetal personhood movements as well as anti-abortion legislation. However, what often passes for science are actually ideological myths, often grounded in and supporting male superiority. Indeed, the first myth of pseudo-embryology is that …

Computing Individual Area Per Head Group Reveals Lipid Bilayer Dynamics, Michael L. Greenfield, Lenore M. Martin, Faramarz Joodaki Dec 2022

Computing Individual Area Per Head Group Reveals Lipid Bilayer Dynamics, Michael L. Greenfield, Lenore M. Martin, Faramarz Joodaki

Chemical, Biomolecular, and Materials Engineering Faculty Publications

Lipid bilayers express a range of phases from solid-like to gel-like to liquid-like as a function of temperature and lipid surface concentration. The area occupied per lipid head group serves as one useful indicator of the bilayer phase, in conjunction with the two-dimensional radial distribution function (i.e., structure factor) within the bilayer. Typically, the area per head group is determined by dividing the bilayer area equally among all head groups. Such an approach is less satisfactory for a multicomponent set of diverse lipids. In this work, area determination is performed on a lipid-by-lipid basis by attributing to a lipid the …

Computer Simulation Of The Light Absorption Band Of The Jumping Spider Isorhodopsin, Noah Zoldak Dec 2022

Computer Simulation Of The Light Absorption Band Of The Jumping Spider Isorhodopsin, Noah Zoldak

Honors Projects

In order to simulate the photoisomerization of the 9-cis Jumping Spider Isorhodopsin (JSiR-1) it is necessary to first simulate its light-absorption band. Here we report on the absorption band simulated using protein models constructed using the advanced Automatic Rhodopsin Modeling (a-ARM) program. A population of S0 models was created and the corresponding S0 to S1 transitions were determined for each member of the resulting population. The calculation resulted in a Gaussian plot showing that the wavelength of the absorption maximum of 560 nm (a violet color) that is consistent, but red-shifted, with respect the experimentally observed value.

Il-21 Signaling Promotes The Establishment Of Kshv Infection In Human Tonsil Lymphocytes By Increasing Differentiation And Targeting Of Plasma Cells, Nedaa Alomari, Jennifer Totonchy

Pharmacy Faculty Articles and Research

Introduction: Factors influencing Kaposi’s sarcoma-associated herpesvirus (KSHV) transmission and the early stages of KSHV infection in the human immune system remain poorly characterized. KSHV is known to extensively manipulate the host immune system and the cytokine milieu, and cytokines are known to influence the progression of KSHV-associated diseases. Our previous work identified the early targeting of plasma cells for KSHV infection. In this study, we examine whether IL-21, a cytokine known to profoundly influence plasma cell fate, influences the early stages of KSHV infection in B lymphocytes.

Methods: Using our unique model of ex vivo KSHV infection in tonsil lymphocytes, …
