Accelerating Multiparametric Mri For Adaptive Radiotherapy, Shraddha Pandey Oct 2022

Accelerating Multiparametric Mri For Adaptive Radiotherapy, Shraddha Pandey

USF Tampa Graduate Theses and Dissertations

MR guided Radiotherapy (MRgRT) marks an important paradigm shift in the field of radiotherapy. Superior tissue contrast of MRI offers better visualization of the abnormal lesions, as a result precise radiation dose delivery is possible. In case of online treatment planning, MRgRT offers better control of intratumoral motion and quick adaptation to changes in the gross tumor volume. Nonetheless, the MRgRT process flow does suffer from some challenges that limit its clinical usability. The primary aspects of MRgRT workflow are MRI acquisition, tumor delineation, dose map prediction and administering treatment. It is estimated that the acquisition of MRI takes around …

Enhancing Production Of Functional Proteins In Plant Cell Culture Through Cellular Engineering And Rational Protein Design, Uddhab Karki Oct 2022

Enhancing Production Of Functional Proteins In Plant Cell Culture Through Cellular Engineering And Rational Protein Design, Uddhab Karki

Student Theses and Dissertations

Plant cell culture is emerging as a promising alternative bioproduction platform for valuable proteins due to its low cost of production, safety (free from animal pathogen contamination) and ease of scale-up. However, there are common challenges for commercialization, including low protein yields, difficulty in cryopreservation, and non-human glycosylation, etc. This research project addresses the overarching research hypothesis that strategically modifying the plant cell genome using CRISPR/Cas9 technology and rationally designing protein molecules by fusing with a functional module enhance the production of protein in plant cell culture and improve protein function. There are two specific aims addressed in this project: …

The Flaws Of Magic Bullet Theory: Retraining Unconscionability To Discretely Target Different Contexts Of Unfairness In Contracts, Marcus Moore Dr. Oct 2022

The Flaws Of Magic Bullet Theory: Retraining Unconscionability To Discretely Target Different Contexts Of Unfairness In Contracts, Marcus Moore Dr.

Dalhousie Law Journal

Unconscionability has long been a troublesome area in Canadian jurisprudence. This is of significant concern given unconscionability’s pre-eminence as a protection of contractual fairness. This article elaborates a much-needed reorganization and rationalization of unconscionability in Canada. Under current law, a single doctrine hopelessly targets two divergent purposes. I set out here a proposed redevelopment rather of separate common law doctrines, each fit-for-purpose: (1) An English-style unconscionable bargains doctrine for avoiding bargains that exploited disability, and (2) an American-style unconscionable clauses doctrine to control unfair terms in standard form contracts. Extensive Canadian precedent supports this solution, assuring its feasibility and legitimacy. …

Transnational Corporations And Climate Governance: A Case Study Of Amazon.Com’S Net-Zero Climate Pledge, Jason Maclean Oct 2022

Transnational Corporations And Climate Governance: A Case Study Of Amazon.Com’S Net-Zero Climate Pledge, Jason Maclean

Dalhousie Law Journal

“Net zero” has become the predominant way of framing global, national, and nonstate climate change commitments. Hundreds of countries and thousands of corporations promise to achieve net-zero emissions by 2050 or earlier. Hopeful as this may seem, early evidence suggests the need to carefully scrutinize corporations’ climate promises. Specifically, there is an urgent need to critically assess the claim that strategic collaboration and compromise at the science-business-society interface can deliver the transformative social, economic, and political change required to address climate change.

Analyzing Amazon.com’s net-zero pledge as a case study, this article argues that strategic conflict with—and within—transnational corporations is …

Successes with anti-CD20 antibodies in chronic lymphocytic leukemia (CLL) and enhanced activity of Fc-engineered vs unmodified antibody therapy suggest a potentially impactful role for natural killer (NK) cells and other innate immune cells in controlling this disease. Stimulated NK cells have shown promise as a cellular therapy, but their application has been constrained by limited expansion capacity and low cytotoxic activity against CLL cells. Here, we demonstrate that both healthy donor-derived and CLL patient-derived NK cells expand rapidly when stimulated with feeder cells expressing membrane-bound interleukin-21 (mbIL-21) and have potent cytotoxic activity against allogeneic or autologous CLL cells. Combination with …

Reactive oxygen species (ROS), a by-product of aerobic life, are highly reactive molecules with unpaired electrons. The excess of ROS leads to oxidative stress, instigating the peroxidation of polyunsaturated fatty acids (PUFA) in the lipid membrane through a free radical chain reaction and the formation of the most bioactive aldehyde, known as 4-hydroxynonenal (4-HNE). 4-HNE functions as a signaling molecule and toxic product and acts mainly by forming covalent adducts with nucleophilic functional groups in proteins, nucleic acids, and lipids. The mitochondria have been implicated as a site for 4-HNE generation and adduction. Several studies clarified how 4-HNE affects the …

Cancer cachexia is a lethal metabolic syndrome featuring muscle wasting with preferential loss of fast-twitching muscle mass through an undefined mechanism. Here, we show that cancer induces muscle wasting by selectively degrading myosin heavy chain (MHC) subtypes IIb and IIx through E3 ligase UBR2-mediated ubiquitylation. Induction of MHC loss and atrophy in C2C12 myotubes and mouse tibialis anterior (TA) by murine cancer cells required UBR2 up-regulation by cancer. Genetic gain or loss of UBR2 function inversely altered MHC level and muscle mass in TA of tumor-free mice. UBR2 selectively interacted with and ubiquitylated MHC-IIb and MHC-IIx through its substrate recognition …

A Cop1-Gata2 Axis Suppresses Ar Signaling And Prostate Cancer, Tao Shen, Bingning Dong, Yanling Meng, David D Moore, Feng Yang

Faculty, Staff and Students Publications

Androgen receptor (AR) signaling is crucial for driving prostate cancer (PCa), the most diagnosed and the second leading cause of death in male patients with cancer in the United States. Androgen deprivation therapy is initially effective in most instances of AR-positive advanced or metastatic PCa. However, patients inevitably develop lethal castration-resistant PCa (CRPC), which is also resistant to the next-generation AR signaling inhibitors. Most CRPCs maintain AR expression, and blocking AR signaling remains a main therapeutic approach. GATA2 is a pioneer transcription factor emerging as a key therapeutic target for PCa because it promotes AR expression and activation. While directly …

Neural crest cells (NCCs) are multipotent stem cells that can differentiate into multiple cell types, including the osteoblasts and chondrocytes, and constitute the majority of the craniofacial skeleton. Here, we show through in vitro and in vivo studies that the transcriptional regulators Yap and Taz have redundant functions as key determinants of the specification and differentiation of NCCs into osteoblasts or chondrocytes. Primary and cultured NCCs deficient in Yap and Taz switched from osteogenesis to chondrogenesis, and NCC-specific deficiency for Yap and Taz resulted in bone loss and ectopic cartilage in mice. Yap bound to the regulatory elements of key …

Mice with a functional human immune system serve as an invaluable tool to study the development and function of the human immune system in vivo. A major technological limitation of all current humanized mouse models is the lack of mature and functional human neutrophils in circulation and tissues. To overcome this, we generated a humanized mouse model named MISTRGGR, in which the mouse granulocyte colony-stimulating factor (G-CSF) was replaced with human G-CSF and the mouse G-CSF receptor gene was deleted in existing MISTRG mice. By targeting the G-CSF cytokine-receptor axis, we dramatically improved the reconstitution of mature circulating and tissue-infiltrating …

Waste Management Practices Of Small Hotels In Dehradun, Arakhita Behera, Surendra Singh Jatav

Journal of Tourism Insights

Waste has emerged as a major issue in the modern society. Due to urbanization, the volume of waste is speedily rising. If the origin of waste creation makes no effort, then all wastes are either disposed of or burnt which results in waste of land, air, water, and energy as well as the growth of undesirable animals, plants and microorganism. Through the associate and involvement of the local population, waste may be managed locally in a sustainable manner. Waste management may be an expensive endeavor if not carried out appropriately. In most of the developing nations, 20-50 percent of urban …

Multi‐Scale Effects Of Land Cover, Weather, And Fire On Columbian Sharp‐Tailed Grouse, Bryan S. Stevens, Courtney J. Conway, Jeffrey M. Knetter, Shane B. Roberts, Patrick Donnelly Oct 2022

Multi‐Scale Effects Of Land Cover, Weather, And Fire On Columbian Sharp‐Tailed Grouse, Bryan S. Stevens, Courtney J. Conway, Jeffrey M. Knetter, Shane B. Roberts, Patrick Donnelly

United States Fish and Wildlife Service: Publications

Columbian sharp‐tailed grouse (Tympanuchus phasianellus columbianus) are endemic to grassland and shrub‐steppe ecosystems of western North America, yet their distribution has contracted to <10% of their historical range. Primary threats to Columbian sharp‐tailed grouse include loss of native habitat and conversion to agriculture, reductions in habitat once provided by the Conservation Reserve Program (CRP), wildfire, and drought conditions, yet population‐level consequences of these threats and their spatio‐temporal scales of effect are poorly understood. We evaluated multi‐scale effects of land cover, weather, and fire histories on patterns of abundance and productivity for Columbian sharp‐tailed grouse populations during 1995–2020 in Idaho, USA, using mixedeffects generalized regression and remotely sensed data. We demonstrated negative effects of fire, tree encroachment, and bare ground, positive effects of spring and summer precipitation and cover of shrubs and perennial forbs and grasses, and positive effects of CRP on grouse abundance that changed in magnitude with cover of perennials and shrubs near leks (i.e., strongest effects when average cover of shrubs and perennial forbs and grasses were less abundant). We also demonstrated per capita recruitment of Columbian sharp‐tailed grouse is positively associated with late‐summer greenness. Our results show that several suspected threats have measurable, population‐level impacts to Columbian sharp‐tailed grouse within Idaho. Moreover, our results suggest ongoing changes occurring within the core range of Columbian sharp‐tailed grouse, including loss of CRP cover to tilled agriculture and changes to wildfire and precipitation dynamics are likely to have negative effects on populations.

Simulation Modeling Of A High-Throughput Oyster Cryopreservation Pathway, Sarah Bodenstein, Isabelina Nahmens, Terrence R. Tiersch

Faculty Publications

The genetic resources of oysters in Louisiana and the Gulf of Mexico are threatened due to high unexplained oyster mortality. Germplasm repositories are collections of cryopreserved genetic material stored alongside associated information that are used to protect genetics resources and facilitate breeding programs in agricultural industries. Therefore, there is great need for oyster repositories. Development of repositories has been slow despite research on high-Throughput cryopreservation protocols because of logistical complexities. The goal of this study was to begin to address the gap between cryopreservation research and repository development in oyster aquaculture by modeling a cryopreservation protocol to understand and improve …

Otolith organs of the inner ear are innervated by two parallel afferent projections to the brainstem and cerebellum. These innervations were proposed to segregate across the line of polarity reversal (LPR) within each otolith organ, which divides the organ into two regions of hair cells (HC) with opposite stereociliary orientation. The relationship and functional significance of these anatomical features are not known. Here, we show regional expression of Emx2 in otolith organs, which establishes LPR, mediates the neuronal segregation across LPR and constitutes the bidirectional sensitivity function. Conditional knockout (cKO) of Emx2 in HCs lacks LPR. Tmie cKO, in which mechanotransduction …

Development Of Combinatorial Antibody Therapies For Diffuse Large B Cell Lymphoma., Eric S. Geanes, Stacey A. Krepel, Rebecca Mclennan, Stephen Pierce, Santosh Khanal, Todd Bradley

Manuscripts, Articles, Book Chapters and Other Papers

Diffuse large B-cell lymphoma (DLBCL), the most common form of lymphoma, is typically treated with chemotherapy combined with the immunotherapy rituximab, an antibody targeting the B cell receptor, CD20. Despite the success of this treatment regimen, approximately a third of DLBCL patients experience either relapse or have refractory disease that is resistant to rituximab, indicating the need for alternative therapeutic strategies. Here, we identified that CD74 and IL4R are expressed on the cell surface of both CD20 positive and CD20 negative B cell populations. Moreover, genes encoding CD74 and IL4R are expressed in lymphoma biopsies isolated from all stages of …

Re-Connecting Children To Nature In Cities: The Role Of Urban Green Infrastructure Smouha Neighborhood, Alexandria, Egypt Case., Marwa N. Charkas

Mansoura Engineering Journal

Sustainability, by its definition, is an investment in the future. This interprets the importance of linking sustainability studies to children as they are 'the future'. Achieving and practicing sustainability is more than a rigid roadmap to be followed, it is rather beliefs and faiths that have to be deeply rooted and considered truthful. So, embedding environmentally conscious attitudes and right behaviors about nature and its norms is crucial to children. This paper investigates the potential that the green infrastructure could present to reconnect children to the natural environment in cities. It develops a model that correlates all the spatial aspects: …
