This work combines microbiology, molecular biology, Next-Generation Sequencing and system biology approaches to explore the mechanism of antibiotic persistence: a multi-drug tolerant, non-dividing, and metabolically altered state present in a subpopulation of cells due to phenotypic diversity rather than genetic variation (i.e. mutations). Persister can survive lethal antibiotic state and resuscitate after the treatment period is over. They are considered as the major contributing factor behind recurring infections. They also have a high mutation rate, which increases the chances of bacteria gaining antibiotic resistance. Alternative ingredients and co-products from ethanol and biofuel industries represent an opportunity for utilization in swine diets. In this context, understanding nutritional value to practical swine feeding begins with product characterization and determination of nutrient digestibility. The objective of this research was to test the hypothesis: yeast-based high-protein DDGS (HP-GDDY) has comparable AA and metabolizable energy (ME) digestibility to feedstuffs commonly used in swine diets and that it could be included as an alternative protein source in nursery diets without negative impact on growth performance. Human dental pulp stem/stromal cells (hDPSCs) derived from the permanent secondary dentition are recognised to possess certain advantageous traits, which support their potential use as a viable source of mesenchymal stem/stromal cells (MSCs) for regenerative medicine-based applications. However, the well-established heterogeneous nature of hDPSC subpopulations, coupled with their limited numbers within dental pulp tissues, has impeded our understanding of hDPSC biology and the translation of sufficient quantities of these cells from laboratory research, through successful therapy development and clinical applications. Human diets containing greater resistant starch (RS) are associated with superior glycemic control. Although high amylose rice has higher RS (29 g/kg to 44 g/kg) than lower amylose content varieties, sensory and processing properties associated with RS have not been evaluated. This study used variants of Waxy and starch synthase II a (SSIIa) genes to divide high amylose (256 g/kg to 284 g/kg) varieties into three haplotypes to examine their effects on RS, RVA parameters, and 14 cooked rice texture properties. Do addictions share common traits of an “addictive personality” or do different addictions have distinct personality profiles? This narrative review examines the differences in the associations between substance use disorder (SUD) and compulsive sexual behavior disorder (CSBD), on the one hand, and personality traits, attachment dispositions, and temperament, on the other hand. We found that both people with a SUD and people with CSBD tended to be more spontaneous, careless, and less reliable, to place self-interest above getting along with others, to show emotional instability and experience negative emotions such as anger, anxiety, and/or depression, to be less able to …

Using a mixed-methods convergent design within an Improvement Science framework, this Improvement Science Dissertation in Practice analyzed the effects of embedding self-regulation breathing techniques into the phonemic segmentation lesson versus conducting the techniques prior to the lesson or not at all. The project was conducted across two elementary schools, using six kindergarten educators and classes. Three studies in this dissertation examined a topic centered around delayed flight progress in aviation pilot training. Study one explored the impact of nonconcurrent flight laboratory training on the academic outcomes of collegiate aviation students, while studies two and three explored virtual reality and artificial intelligence as potential solutions to help alleviate the strain of delayed flight progress on the flight training organization. A fully solid-state, software-defined, one-handed, handle-type control device built around a machine-learning (ML) model that provides intuitive and simultaneous control in position and orientation each in a full three degrees-of-freedom (DOF) is proposed in this paper. The device, referred to as the “Smart Handle”, and it is compact, lightweight, and only reliant on low-cost and readily available sensors and materials for construction. The activation of relatively inert C-H bonds has remained a challenge in organic chemistry for over a century. Metal catalysts have been deployed to overcome the labile reactivity of these bonds and through gas-phase studies, detailed mechanisms can be elucidated for this important class of chemistry. The powerful combination of ion-molecule reactions, collisional activation of ions, and density functional theory calculations allows for structural elucidation, intermediate determination, and as shown throughout this work, a complete picture of catalytic processes. As plastic pollution increases, top marine predators such as marine mammals are becoming increasingly susceptible to plastic particles and their additives. Plastic particles have been found in gastrointestinal tracts and scat of many marine mammals, and quantifying plastic pollution in those that are pelagic can provide insight into plastic pollution in mesopelagic ecosystems that are just beginning to be analyzed. Adapting well-developed laboratory techniques for microplastic (MP) isolation (i.e. density separation and chemical digestion), I isolated MPs from the scat of the deepest diving pinniped, the northern elephant seal (NES), and found that100% of scat samples (n=11) contained high counts …

This project describes the development of a new antitumor therapeutic platform that combines the benefits of small-molecule drug conjugates (SMDCs) and antibody drug conjugates (ADCs). Valine-citrulline (VCit) dipeptide linkers are popular cathepsin B cleavable ADC linkers. Due to its instability in mouse serum, translating efficacy data from mouse to human is more difficult. It has been reported that replacing the VCit linker with glutamic acid-valine-citrulline (EVCit) improves ADC stability in mouse serum. This research explores the intersections between Indigenous-led tourism and resurgence among Indigenous communities. Resurgence is understood as practices promoting connections with culture, Country (i.e. Indigenous lands and waterways), and community. The research utilises a comparative case study approach to specifically analyse the experience of Karajarri Traditional Owners (TOs) of the saltwater Kimberley region of northwestern Australia. Comparative examples are drawn from Indigenous-led tourism development by other coastal Kimberley Aboriginal tourism leaders, as well as from the author’s own Tribal Nation, the Cherokee Nation of Oklahoma, United States.

This research program investigated the use of acupuncture intervention as an adjunct for hypertrophic scar (HTS) management. Mechanistic evidence from ex vivo and animal studies suggests that acupuncture has the potential to modulate neurogenic inflammation to influence pain and itch associated with HTS. Further, acupuncture may mediate non-neuronal cells involved in the proliferation and remodelling stage of healing to promote scar maturation. Previous studies using acupuncture for scar management demonstrated positive outcomes, however low-quality clinical trials and case studies provide limited evidence for treatment benefit.

Recent filmic representations of the fairy tale villain can offer powerful counter-narratives to archetypal fairy tale stories. This thesis examines how certain contemporary representations of fairy tales have reinterpreted the traditional ‘villain’ archetype, revealing a greater diversity of perspectives. Using John Stephens’ and Peter Stockwell’s cognitive poetics theory on schema and scripts as a theoretical framework, this thesis closely analyses the known Disney fairy tale villain, Sleeping Beauty’s (1959) Maleficent, as she is portrayed in the live-action films Maleficent (2014) and Maleficent: Mistress of Evil (2019), as well as Disney’s Descendants (2015, 2017, 2019) films. Cattle ranching specifically, a principal land use in the Blackfoot watershed, has faced increasing socioeconomic challenges due to global competition, falling cattle prices, amenity migration, and climate change. Patterns of cultural mountain adaptations are recorded throughout the Western Cordillera of North America. They demonstrate prolonged and complex uses of these ecosystems over the Holocene. What behaviors or circumstances in the Early Holocene transitioned mountains from inhospitable to reliable territory? This research addresses this question in the Beartooth Mountains of the Greater Yellowstone Ecosystem (GYE).

Foraging behaviors dictate hunter-gatherer mobility and habitation patterning. These behaviors are reflected in the spatial arrangement of archaeological activity areas and lithic tools in mountainous ecosystems.
