A cure for human immunodeficiency virus (HIV-1) is restricted by the continued presence of a latent reservoir of memory CD4+ T cells with proviruses integrated into their DNA despite suppressive antiretroviral therapy (ART). A predominant strategy currently pursued in HIV-1 cure-related research is the “kick and kill” approach, where latency reversal agents (LRAs) are used to reactivate transcription from integrated proviruses. The premise of this approach is that “kicking” latent virus out of hiding allows the host immune system to recognize and kill infected cells. Simple and low-cost fabrication of microfluidic devices has attracted considerable attention among researchers. The traditional soft lithography fabrication method requires expensive equipment like a UV exposure system and mask fabrication facility. In this work, an alternative and low-cost UV exposure system was introduced along with an alternative mask fabrication system. A previously reported passive microfluidic mixer was fabricated successfully using this modified soft lithography method. Challenges were presented during this modified fabrication method. Another emerging potential alternative for the fabrication of microfluidic mixers is 3D printing. It was also used in this experiment to fabricate a passive micromixer. This method …

With the rapid development of new data collection and acquisition techniques, high-dimensional data have emerged from various fields. Consequentially, new variable selection methods especially in ultra-high dimensional problems are demanding.
The first part of this dissertation focuses on developing a new Bayesian variable selection method for a differential expression analysis using raw NanoString nCounter data. The medium-throughput mRNA abundance platform NanoString nCounter has gained great popularity in the past decade, due to its high sensitivity and technical reproducibility as well as remarkable applicability to ubiquitous formalin fixed paraffin embedded (FFPE) tissue samples. The paper inquires into public art produced by artists and government authorities. It interprets collaboration as a site of negotiating various interests; holding possibilities for both disruption and complicity; antagonism and amelioration. The research considers two art projects in the Philippine context: first; the Paoay sand dunes sculpture park made between 2012 and 2015 in the Northern Ilocos region; and second; the ongoing Pasig River Art for Urban Change; beginning with Bakawan in 2015 in the Philippine capital Metro Manila. Toxoplasmosis gondii exposure has been linked to increased impulsivity and risky behaviors, which has implications for eating behavior. Impulsivity and risk tolerance is known to be related with worse diets and a higher chance of obesity. There is little known, however, about the independent link between Toxoplasma gondii (T. gondii) exposure and diet-related outcomes. Using linear and quantile regression, we estimated the relationship between T. gondii exposure and BMI, total energy intake (kcal), and diet quality as measured by the Health Eating Index-2015 (HEI) among 9,853 adults from the 2009–2014 National Health and Nutrition Examination Survey. …

In order to better understand the students who enroll in Freshman Inquiry (FRINQ) courses and identify areas where they may need support, University Studies (UNST) began collecting and compiling data on these students. This report presents a profile of FRINQ students from Fall 2021. It presents an overview of all students enrolled in FRINQ during Fall 2021 and then summarizes information from the Prior Learning Survey. It is organized around three areas that contribute to student success: academic preparation and concern; financial and physical wellbeing; and connectedness to campus.

Bacterial tyrosine kinases (BY-kinases) and shikimate kinases (SKs) comprise two structurally divergent P-loop containing enzyme families that share similar catalytic site geometries, most notably with respect to their Walker-A, Walker-B, and DxD motifs. We had previously demonstrated that in BY-kinases, a specific interaction between the Walker-A and Walker-B motifs, driven by the conserved “catalytic” lysine housed on the former, leads to a conformation that is unable to efficiently coordinate Mg2+•ATP and is therefore incapable of chemistry. Here, using enhanced sampling molecular dynamics simulations, we demonstrate that structurally similar interactions between the Walker-A and Walker-B motifs, also mediated by …

Sparked by the police killings of George Floyd and Breonna Taylor, the 2020 uprisings accelerated a momentum of abolitionist organizing that demands the defunding and dismantling of policing infrastructures. Although a growing body of legal scholarship recognizes abolitionist frameworks when examining conventional proposals for reform, critics mistakenly continue to disregard police abolition as an unrealistic solution. This Essay helps dispel this myth of “impracticality” and illustrates the pragmatism of abolition by identifying a community-driven effort that achieved a meaningful reduction in policing we now take for granted. We present a new algorithm for learning unknown governing equations from trajectory data, using and ensemble of neural networks. Given samples of solutions x(t) to an unknown dynamical system x˙(t) = f(t, x(t)), we approximate the function f using an ensemble of neural networks. We express the equation in integral form and use Euler method to predict the solution at every successive time step using at each iteration a different neural network as a prior for f. This procedure yields M-1 time-independent networks, where M is the number of time steps at which x(t) is observed. Finally, we obtain a …

The novel Coronavirus is not only exposing old patterns of racism and systemic inequalities, but deepening them as well. The notion of blindspotting, as described in the film by the same name, is used to understand how the COVID-19 pandemic impacts the “spiritual emergency” or crisis of racism in America. "Blindspotting" is an image or situation that can be interpreted in two ways but is understood by some in only one way, thereby producing a blind spot. Life history theory (LHT) is the conceptualization of tradeoffs in resources allocation. Plasticity, the ability of phenotypes to vary in response to environmental cues, is integral to LHT. Prior to completed maturation, resources are distributed towards maintenance or somatic maturation, a combination of growth and development (G&D). This dissertation presents an examination of intragroup variation in the G&D of the dental and skeletal systems within a sample of 20th century White females to consider whether they provide evidence for tradeoffs within the subadult interval. Veratrum spp. grow throughout the world and are especially prevalent in high mountain meadows of North America. All parts of Veratrum plants have been used for the treatment of ailments including injuries, hypertension, and rheumatic pain since as far back as the 1600s. Of the 17–45 Veratrum spp., Veratrum californicum alkaloids have been proven to possess favorable medicinal properties associated with inhibition of hedgehog (Hh) pathway signaling. Aberrant Hh signaling leads to proliferation of over 20 cancers, including basal cell carcinoma, prostate and colon among others. Education, Antidomination, And The Republican Guarantee, Kip M. Hustace

This Article offers a new interpretation of the United States Constitution’s republican guarantee and theorizes its protection of a fundamental right to education. Courts and education law scholars have identified the republican guarantee as a plausible source of educational rights but have not detailed how. Drawing on recent work by legal scholars, historians, political scientists, and philosophers, this Article reinterprets the guarantee as the federal government’s obligation to secure freedom as nondomination, and it argues that excellent, equitable public education is necessary to fulfilling this duty. Nondomination, a robust conception of freedom, is freedom from subjection to the will of …

Multiple lines of evidence show that modern humans interbred with archaic Denisovans. Here, we report an account of shared demographic history between Australasians and Denisovans distinctively in Island Southeast Asia. Our analyses are based on ∼2.3 million genotypes from 118 ethnic groups of the Philippines, including 25 diverse self-identified Negrito populations, along with high-coverage genomes of Australopapuans and Ayta Magbukon Negritos. We show that Ayta Magbukon possess the highest level of Denisovan ancestry in the world—∼30%–40% greater than that of Australians and Papuans—consistent with an independent admixture event into Negritos from Denisovans. Regulation of the mammalian embryo involves cell-signaling molecules produced by the maternal oviduct and endometrium. Here, datasets on the transcriptome of the gestational Days 5 and 6 bovine morula and Day 5 maternal endometrium were examined to identify receptor genes expressed by the morula and expression of the corresponding ligand-related genes in the endometrium. A total of 175 receptor genes were identified in the morula, including 48 encoding for growth factors or WNT signaling molecules, 25 for cytokines and chemokines, 35 involved in juxtacrine and matricellular signaling and 25 encoding for receptors for small molecules. From smart work scheduling to optimal drug timing, there is enormous potential in translating circadian rhythms research results for precision medicine in the real world. However, the pursuit of such effort requires the ability to accurately estimate circadian phase outside of the laboratory. One approach is to predict circadian phase noninvasively using light and activity measurements and mathematical models of the human circadian clock. Most mathematical models take light as an input and predict the effect of light on the human circadian system. OBJECTIVE: The aim of this study was to determine which initial surgical treatment results in the lowest rate of death or neurodevelopmental impairment (NDI) in premature infants with necrotizing enterocolitis (NEC) or isolated intestinal perforation (IP).
SUMMARY BACKGROUND DATA: The impact of initial laparotomy versus peritoneal drainage for NEC or IP on the rate of death or NDI in extremely low birth weight infants is unknown.
METHODS: We conducted the largest feasible randomized trial in 20 US centers, comparing initial laparotomy versus peritoneal drainage. The primary outcome was a composite of death or NDI at 18 to 22 months corrected …

The purpose of this transcendental phenomenological study was to understand the shared lived experiences of Central Pennsylvanian high school administrators’ efforts to address the barriers to parental investment for non-nuclear families. The theory guiding this study was the path-goal theory, which builds on motivational theory to select the proper leadership style to achieve the desired outcome. The central research question was intended to discover the essence of Central Pennsylvania school administrators’ shared lived experiences addressing parental investment barriers for non-nuclear families. The purpose of this transcendental phenomenological study was to describe Black millennial teachers’ self-perceived factors of resilience in Virginia public elementary schools. The central question of this study was: “What are the lived experiences that Black millennial teachers ascribe to their resilience in Virginia public school elementary settings?” The theory guiding this study was resilience theory by Masten (2014) which described positive human adaptation through the interplay of individual and contextual risk and protective factors that contributed to resilience. Currently, there is no standardized screening and counseling model that enables Navy Chaplains to counsel those with post-traumatic stress injury, moral injury, and loss of purpose. The shame associated with these three conditions is a significant contributor to Service Member suicide, mental health problems, and substance abuse. Chaplains are often the first line of care Service Member’s access because of the chaplain’s accessibility and legally protected complete confidentiality. Hypertension is a global chronic medical condition that is life threatening and eventually fatal if left untreated. Elevated blood pressure will disrupt the function of the cardiovascular system and impact the function of major organs, resulting in devastating complications for the patient while increasing cost for the healthcare system. The purpose of this embedded, explanatory multiple case study was to explain how internal and external factors shape the instructional spelling practices of third and fourth grade educators in Delaware. Internal factors were personal characteristics, such as content knowledge and pedagogy, that have been developed by past experiences and trainings. External factors were environmental and behavioral elements that vary between schools. Public water systems are an integral part of community infrastructure. Drinking water contamination or service disruptions have the potential to cause economic losses, limit fire suppression capability, and result in human illnesses. Until 2016, the United States federal government had not issued a disaster declaration due to contaminated water. The first federal drinking water disaster declaration due to contaminated water serves as a sentinel event demonstrating the need to increase focus on public water systems during all phases of emergency management: mitigation, preparation, response, and recovery.
