Pediatrics Research and Scholarship
Somatic variants in TET2 and DNMT3A are founding mutations in hematological malignancies that affect the epigenetic regulation of DNA methylation. Mutations in both genes often co-occur with activating mutations in genes encoding oncogenic tyrosine kinases such as FLT3ITD, BCR-ABL1, JAK2V617F , and MPLW515L , or with mutations affecting related signaling pathways such as NRASG12D and CALRdel52 . Here, we show that TET2 and DNMT3A mutations exert divergent roles in regulating DNA repair activities in leukemia cells expressing these oncogenes. Malignant TET2-deficient cells displayed downregulation of BRCA1 and LIG4, resulting in reduced activity of BRCA1/2-mediated homologous recombination (HR) and DNA-PK-mediated non-homologous …

In plants, the reactive oxygen species (ROS) formed during normal conditions are essential in regulating several processes, like stomatal physiology, pathogen immunity and developmental signaling. However, biotic and abiotic stresses can cause ROS over-accumulation leading to oxidative stress. Therefore, a suitable equilibrium is vital for redox homeostasis in plants, and there have been major advances in this research arena. Salicylic acid (SA) is known as a chief regulator of ROS; however, the underlying mechanisms remain largely unexplored. Women entering science, technology, engineering, and math (STEM) occupations increased by 19% between 1970 and 2019, but at 74% of the STEM workforce, men still dominate the field (Martinez & Christnacht, 2021). In the nuclear industry, this disparity is most apparent with few women holding leadership roles within nuclear organizations (Kovaleski, 2014; Gaspar & Dubertrand, 2019). While research suggests that companies with women leaders in the manager level through the board level excel in organizational performance (Catalyst, 2007), a problem exists for women when trying to promote to leadership positions, especially to the C-suite in STEM fields (Warner, 2014).

Structural And Kinetic Analyses Of Holothurian Sulfated Glycans Suggest Potential Treatment For Sars-Cov-2 Infection, Rohini Dwivedi, Priyanka Samanta, Poonam Sharma, Fuming Zhang

Certain sulfated glycans, including those from marine sources, can show potential effects against SARS-CoV-2. Here, a new fucosylated chondroitin sulfate (FucCS) from the sea cucumber Pentacta pygmaea (PpFucCS) (MW ∼10.60 kDa) was isolated and structurally characterized by NMR. PpFucCS is composed of {→3)-β-GalNAcX-(1→4)-β-GlcA-[(3→1)Y]- (1→}, where X = 4S (80%), 6S (10%) or nonsulfated (10%), Y = α- Fuc2,4S (40%), α-Fuc2,4S-(1→4)-α-Fuc (30%), or α-Fuc4S (30%), and S = SO3-. The anti-SARS-CoV-2 activity of PpFucCS and those of the FucCS and sulfated fucan isolated from Isostichopus badionotus (IbFucCS and IbSF) were compared with that of heparin. Wastewater treatment systems collect and treat sewage that includes microplastics (MPs). However, we are not aware of any studies on the occurrence and distribution of MPs in wastewater stabilization ponds (WSPs), which serve small communities worldwide. Here, we characterized MPs (~45 µm–5 mm) in an aerated WSP serving ~500 houses and an adjacent lake. Putative MPs were most abundant in duckweed (Lemna minor) and sludge (75 ± 22 and 12.8 ± 3.1 particles/g, respectively: ±1 standard deviation (SD), n = 6, dry weight). Cattle are the main reservoir of Enterohemorrhagic Escherichia coli (EHEC), with O157:H7 the distinctive serotype. EHEC is the main causative agent of a severe systemic disease, Hemolytic Uremic Syndrome (HUS). Argentina has the highest pediatric HUS incidence worldwide with 12–14 cases per 100,000 children. Herein, we assessed the genomes of EHEC O157:H7 isolates recovered from cattle in the humid Pampas of Argentina. According to phylogenetic studies, EHEC O157 can be divided into clades. Clade 8 strains that were classified as hypervirulent. Most of the strains of this clade have a Shiga toxin stx2a-stx2c genotype. To better understand the molecular bases …

Thin Safety Margin charts the history of SEFOR, a twenty-megawatt reactor that operated for three years in the rural Ozark Mountains of Arkansas as part of an internationally sponsored program designed to demonstrate the Doppler effect in plutonium-oxide-fueled fast reactors. Authors Jerry Havens and Collis Geren draw upon this history to assess the accidental explosion risk inherent in using fast reactors to reduce the energy industry’s carbon dioxide emissions.

There is a longstanding and widely held view, often associated with Catholicism, that intrinsically nonprocreative human sex acts are intrinsically immoral. Some philoso- phers who hold this view, such as Edward Feser, claim that they can defend the view on purely philosophical grounds by relying on the perverted faculty argument. This paper argues that Feser’s defense of the perverted faculty argument does not work because Feser fails to recognize the full implications of the species-dependence of natural goodness. Neural networks -- specifically, deep neural networks -- are, at present, the most effective machine learning techniques. There are reasonable explanations of why deep neural networks work better than traditional "shallow" ones, but the question remains: why neural networks in the first place? why not networks consisting of non-linear functions from some other family of functions? In this paper, we provide a possible theoretical answer to this question: namely, we show that of all families with the smallest possible number of parameters, families corresponding to neurons are indeed optimal -- for all optimality criteria that satisfy some reasonable requirements: : …

Complex organic matter is the most abundant form of organic carbon in the fossil record on Earth and in geological materials delivered from Space. It is unclear how complex organic matter preserved in fossils and meteorites forms in different terrestrial and extraterrestrial geological settings. The molecular make-up of decay-resistant organic polymers in fossils and meteorites has the potential to preserve biological signals and record geological conditions in deepest time. Oscillatory phosphorylation/dephosphorylation can be commonly found in a biological system as a means of signal transduction though its pivotal presence in the workings of circadian clocks has drawn significant interest: for example in a significant portion of the physiology of Synechococcus elongatus PCC 7942. The biological oscillatory reaction in the cyanobacterial circadian clock can be visualized through its reconstitution in a test tube by mixing three proteins—KaiA, KaiB and KaiC—with adenosine triphosphate and magnesium ions. Surprisingly, the oscillatory phosphorylation/dephosphorylation of the hexameric KaiC takes place spontaneously and almost indefinitely in a test tube as long as ATP is present. This …

The TU Dublin IMPACT project was a National Forum and HEA-funded project under the Strategic Alignment of Teaching and Learning Enhancement (SATLE)) fund which materialised over/during the 2020/21 Academic Year. The aim of the SATLE fund was to develop a strong foundation for future enhancement of teaching and learning (T&L) within an institution and in line with local and national priorities. TU Dublin received the largest funding allocation of any Irish HEI; a total of €602,000. This significant funding was used to develop strategically aligned, sustainable processes and infrastructure to maximise the impact of TU Dublin’s T&L practices on the …

Background: Head and neck cancer treatment modalities can significantly impact functional outcomes of patients, especially oral intake (Brizel, et al N Engl J Med 1998; Kamal, et al Support Care Cancer 2019). Radiation therapy in particular has been associated with posttreatment xerostomia and dysphagia (Adelstein, et al J Clin Oncol 2003; Hutcheson, et al Cancer 2013) which can affect quality of life and impair weight gain, contributing to worse long-term outcomes (Payakachat, et al Head Neck, 2013). Artist Martin Puryear’s commitment to seeing objects as connected to history and culture resonates with a moment when Gettysburg College reflected on the significance of its own historical place and time. As the College approached the sesquicentennial anniversary of its founding, it decided to mark the occasion not with a ball or parade, but with “an intelligent artifact,” a permanent marker that both recognizes its maker and offers its own history.
