In this work, we put forward a hypothesis about the decisive role of multivalent nonspecific interactions in the early stages of PML body formation. Our analysis of the PML isoform sequences showed that some of the PML isoforms, primarily PML-II, are prone to phase separation due to their polyampholytic properties and the disordered structure of their C-terminal domains. The similarity of the charge properties of the C-terminal domains of PML-II and PML-VI isoforms made it possible for the first time to detect migration of PML-VI from PML bodies to the periphery of the cell nucleus, similar to the migration of …

Background: Cancer stem cells’ (CSCs) self-maintenance is regulated via the pluripotency pathways promoting the most aggressive tumor phenotype. This study aimed to use the activity of these pathways for the CSCs’ subpopulation enrichment and separating cells characterized by the OCT4 and SOX2 expression. Methods: To select and analyze CSCs, we used the SORE6x lentiviral reporter plasmid for viral transduction of colon adenocarcinoma cells. Additionally, we assessed cell chemoresistance, clonogenic, invasive and migratory activity and the data of mRNA-seq and intrinsic disorder predisposition protein analysis (IDPPA). The basic helix–loop–helix/Per-ARNT-SIM (bHLH-PAS) proteins are a family of transcription factors regulating expression of a wide range of genes involved in different functions, ranging from differentiation and development control by oxygen and toxins sensing to circadian clock setting. In addition to the well-preserved DNA-binding bHLH and PAS domains, bHLH-PAS proteins contain long intrinsically disordered C-terminal regions, responsible for regulation of their activity. Our aim was to analyze the potential connection between disordered regions of the bHLH-PAS transcription factors, post-transcriptional modifications and liquid-liquid phase separation, in the context of disease-associated missense mutations. Recently, two cases of complete remission of classical Hodgkin lymphoma (cHL) and follicular lymphoma (FL) after SARS-CoV-2 infection were reported. However, the precise molecular mechanism of this rare event is yet to be understood. Here, we hypothesize a potential anti-tumor immune response of SARS-CoV-2 and based on a computational approach show that: (i) SARS-CoV-2 Spike-RBD may bind to the extracellular domains of CD15, CD27, CD45, and CD152 receptors of cHL or FL and may directly inhibit cell proliferation. Proteins are exposed to fluctuating environmental conditions in their cellular context and during their biotechnological production. Disordered regions are susceptible to these fluctuations and may experience solvent-dependent conformational switches that affect their local dynamism and activity. In a recent study, we modeled the influence of pH in the conformational state of IDPs by exploiting a charge–hydrophobicity diagram that considered the effect of solution pH on both variables. However, it was not possible to predict context-dependent transitions for multiple sequences, precluding proteome-wide analysis or the screening of collections of mutants. Recently, it was reported that near-sourced COVID-19 convalescent plasma (CP) is more efficient than distantly sourced CP. What was left behind in this analysis is the investigation of the possible causes of mortality associated with the CP transfusion itself. Knowing this information is important for determining whether not receiving CP of near source is the main cause of high rate of death in the group of patients who received distantly sourced CP. We argue that the thrombotic and thromboembolic events may act as risk factors for adverse complications and death associated with CP transfusion. Therefore, they have to be considered …

Analysis by attenuated total reflection–Fourier transform infrared spectroscopy shows that each coexisting phase in aqueous two-phase systems has a different arrangement of hydrogen bonds. Specific arrangements vary for systems formed by different solutes. The hydrogen bond arrangement is shown to correlate with differences in hydrophobic and electrostatic properties of the different phases of five specific systems, four formed by two polymers and one by a single polymer and salt. The results presented here suggest that the arrangement of hydrogen bonds may be an important factor in phase separation.

The nucleocapsid protein N from SARS-CoV-2 is one of the most highly expressed proteins by the virus and plays a number of important roles in the transcription and assembly of the virion within the infected host cell. It is expected to be characterized by a highly dynamic and heterogeneous structure as can be inferred by bioinformatics analyses as well as from the data available for the homologous protein from SARS-CoV. Previous studies found that individuals with Autism Spectrum Disorder (ASD) tend to demonstrate poor health behaviors, including unhealthy nutrition habits and low physical activity (PA) levels. Growing research suggests the PA can improve ASD-related symptoms and attenuate the mental and physical adverse effects of the COVID-19 pandemic. Unfortunately, the stay-at-home order was put in place because of COVID-19, which negatively affected health behaviors in many individuals with ASD, including an increase in sedentary behavior and screen time and less PA participation. Pediatric and neonatal patients are especially vulnerable to suffering from distress. This literature review identifies research which applies passive music listening to distressed neonatal and pediatric acute care patients. Databases searched to find relevant studies include CINAHL plus with full text, MEDLINE, Alt HealthWatch, APA PsycArticles, and APA PsycInfo from EBSCOhost. Six studies were identified to meet search criteria. The studies that supported music listening with the neonatal and pediatric populations had a positive effect in reducing distress levels. This study examined the relationship between exposure to COVID-19 fake news, right-wing authoritarianism (RWA), sociodemographic factors (i.e., race, biological sex), and xenophobia along with anti-Asian sentiment during the pandemic. Participants included 133 female and male college students. Participants were randomly assigned to one of three conditions: (1) exposed to COVID-19 fake news, (2) exposed to real news related to COVID-19 news, and (3) no news exposure. All participants from three groups were then asked to complete a series of measures regarding their attitudes toward Asian Americans and xenophobia. AIM To provide a detailed gene and protein expression analysis related to mitochondrial biogenesis and assess mitochondrial content in skeletal muscle of children with cerebral palsy (CP). METHOD Biceps brachii muscle samples were collected from 19 children with CP (mean [SD] age 15y 4mo [2y 6mo], range 9–18y, 16 males, three females) and 10 typically developing comparison children (mean [SD] age 15y [4y], range 7–21y, eight males, two females). Seed shatter is an important weediness trait on which the efficacy of harvest weed seed control (HWSC) depends. The level of seed shatter in a species is likely influenced by agroecological and environmental factors. In 2016 and 2017, we assessed seed shatter of eight economically important grass weed species in soybean [Glycine max (L.) Merr.] from crop physiological maturity to 4 wk after maturity at multiple sites spread across 11 states in the southern, northern, and mid-Atlantic United States. Soil amendments with high carbon (C) content can be effective in semi-arid regions where soils are characterized by low C. A field study was conducted in 2016–2018 to evaluate the effect of char on soil chemical properties and irrigated maize (Zea mays L.) yields in sandy loam fertilized with urea or composted manure. Carbon-rich char used was a product of coal combustion residue from a local factory in western Nebraska. In this report, we describe a case of atypical neuroleptic malignant syndrome (NMS) presenting with an isolated lesion in the splenium of the corpus callosum (ILSCC). There is a paucity of information regarding this topic within the literature and only 7 previous case reports have been published at the time of writing. To our knowledge, this case report is also the first to describe an atypical NMS variant in the context of an ILSCC. ‘As One’, a popular modern chamber opera by composer Laura Kaminsky, presents the transition of transgender woman, Hannah. Hannah, however, is casted and composed as two separate voices, a baritone and mezzo-soprano, thus creating “Hannah before” and “Hannah after”. In doing so, a dichotomy of thought surrounding the attributes of a binary based practice of vocalization and character representation is created. Through the analysis of gender theory, voiced gender theory, and the ‘As One’ chamber opera, storytelling of complex gender-based characters may be solidified within biased gender compositional and musical tools.
