Targeting a tumor’s metabolic dependencies is a clinically actionable therapeutic approach; however, identifying subtypes of tumors likely to respond remains difficult. The use of lipids as a nutrient source is of particular importance, especially in breast cancer. Imaging techniques offer the opportunity to quantify nutrient use in preclinical tumor models to guide development of new drugs that restrict uptake or utilization of these nutrients. We describe a fast and dynamic approach to image fatty acid uptake in vivo and demonstrate its relevance to study both tumor metabolic reprogramming directly, as well as the effectiveness of drugs targeting lipid metabolism. Specifically, …

Assessing The Behavioural Responses Of Small Cetaceans To Unmanned Aerial Vehicles, J. Castro, F. O. Borges, A. Cid, M. I. Laborde, R. Rosa, Heidi C. Pearson

Faculty, Staff, and Students

Unmanned Aerial Vehicles (UAVs), or drones, have recently emerged as a relatively affordable and accessible method for studying wildlife. Vertical Take-off and Landing (VTOL) UAVs are appropriate for morphometric, behavioural, abundance and demographic studies of marine mammals, providing a stable, nonintrusive and highly manoeuvrable platform. Previous studies using VTOL UAVs have been conducted on various marine mammal species, but specific studies regarding behavioural responses to these devices are limited and scarce. The aim of this study was to evaluate the immediate behavioural responses of common (Delphinus delphis) and bottlenose (Tursiops truncatus) dolphins to a VTOL UAV flown at different altitudes. …

Biological Threats Are National Security Risks: Why Covid-19 Should Be A Wake Up Call For Policy Makers, Rep. Eric M. Swalwell, R. Kyle Alagood

Washington and Lee Law Review Online

A national security strategy is the “nation’s plan for the coordinated use of all the instruments of state power—nonmilitary as well as military—to pursue objectives that defend and advance its national interest.” Perhaps the most straightforward national security objective is to protect the country from foreign invasion, but national security involves other objectives that aim to protect people in the United States as well as their values. For example, protecting U.S. elections from foreign interference is a security objective that advances the nation’s interest in democratic governance. The outbreak of a highly contagious disease like COVID‑19 strikes at the core …

And We're Rollin' On A River...: The Poetics And Musical Dimensions Of Breaking With Eurocentric Knowledge And Decolonizing Humanity, Hannah Renee Shelton

Electronic Theses and Dissertations Archive

American society to date is wearing a mask. Although claiming to secure rights and liberites for all, American society hides under Eurocentrism's guise which overvalorizes the white maleand undervalorizes minority bodies through reason, space, and place. Therefore, I assert there must be an epistemology which breaks from Eurocentricism. Utilizing poetics as a revalorizing logic, I will ask: How can poetics, as a legitimate source of knowledge, decolonize humanity? Keeping within the American scope, I employ critical race theory and theorists to demonstrate the dangers of employing hegemonic knowledge. I will then utilize poetry, literary devices, jazz, and blues to demonstrate …

Clinical Utility Of Molecular Profiling In Recurrent Glioblastoma Multiforme, Rebecca P. Bystrom, Pavalan Selvam, Jens Rueter, Honey Reddi, Christine Lu-Emerson

Journal of Maine Medical Center

Introduction: Glioblastoma multiforme (GBM) is the most common and aggressive primary malignant brain tumor found in adults. GBM has limited therapeutic options. Initial tumor sampling establishes the histopathologic diagnosis, identifies prognostic and therapeutic biomarkers, and provides an opportunity for molecular profiling. By contrast, the utility of repeat tumor sampling and molecular profiling in recurrent GBM is not well established.

Clinical Findings: We present a 69-year-old woman with GBM whose tumor recurred after standard treatment with temozolomide (TMZ) and concurrent radiation, followed by adjuvant TMZ. This patient had a methylated O6-methylguanine-DNA methyltransferase (MGMT) promoter, which ordinarily predicts a favorable response to …

Ovarian Dynamics And Fecundity Regulation In Blueback Herring, Alosa Aestivalis, From The Connecticut River, Us, Foivos Alexandros Mouchlianitis, Eric T. Schultz, Thassya C. Dos Santos Schmidt, Justin P. Davis, Kostas Ganias Jan 2021

Ovarian Dynamics And Fecundity Regulation In Blueback Herring, Alosa Aestivalis, From The Connecticut River, Us, Foivos Alexandros Mouchlianitis, Eric T. Schultz, Thassya C. Dos Santos Schmidt, Justin P. Davis, Kostas Ganias

EEB Articles

We analyzed ovarian dynamics of anadromous blueback herring, Alosa aestivalis, in Connecticut River with the principal aim of exploring oocyte recruitment and how it shapes the fecundity pattern. We examined the oocyte release strategy and analyzed spawning cyclicity by linking oocyte growth to the degeneration of postovulatory follicles. Females were accordingly classified as pre-spawners, early and late active spawners, and oocyte recruitment intensity was compared among the different spawning phases. Oocyte recruitment occurred continuously and in parallel with spawning activity, a pattern which is diagnostic of indeterminate fecundity. However, both fecundity and oocyte recruitment intensity progressively decreased (tapered) throughout …

Excess Extracellular K+ Causes Inner Hair Cell Ribbon Synapse Degeneration, Hong-Bo Zhao, Yan Zhu, Li-Man Liu Jan 2021

Excess Extracellular K+ Causes Inner Hair Cell Ribbon Synapse Degeneration, Hong-Bo Zhao, Yan Zhu, Li-Man Liu

Otolaryngology--Head & Neck Surgery Faculty Publications

Inner hair cell (IHC) ribbon synapses are the first synapse in the auditory system and can be degenerated by noise and aging, thereby leading to hidden hearing loss (HHL) and other hearing disorders. However, the mechanism underlying this cochlear synaptopathy remains unclear. Here, we report that elevation of extracellular K+, which is a consequence of noise exposure, could cause IHC ribbon synapse degeneration and swelling. Like intensity dependence in noise-induced cochlear synaptopathy, the K+-induced degeneration was dose-dependent, and could be attenuated by BK channel blockers. However, application of glutamate receptor (GluR) agonists caused ribbon swelling but …

Metastatic Breast Cancer, Nigora E. Atakhanova, Farrukh Sh. Fayziev, Dilbar M. Almuradova, Dilorom S. Shodmonova

Central Asian Journal of Medicine

Breast cancer is the most common cause of death among women from malignant tumors. Metastatic breast cancer is not currently a curable disease. According to most authors, especially in developing countries, from 50 to 70% of patients already have advanced forms of the disease upon admission (locally common forms, in the form of lesions of regional lymph nodes and distant metastases to various organs). The nature and frequency of distant breast cancer metastases is very diverse. In 40-80% of cases, breast cancer metastasizes to bone. However, it can metastasize other organs and systems, for example, to the liver, lungs, ovaries, …

Cyclin-Dependent Kinase 1 And Aurora Kinase Choreograph Mitotic Storage And Redistribution Of A Growth Factor Receptor, C. D. Cota, M. S. Dreier , '20, W. Colgan, A. Cha, Twan Sia , '21, Bradley Justin Davidson , '91

Biology Faculty Works

Endosomal trafficking of receptors and associated proteins plays a critical role in signal processing. Until recently, it was thought that trafficking was shut down during cell division. Thus, remarkably, the regulation of trafficking during division remains poorly characterized. Here we delineate the role of mitotic kinases in receptor trafficking during asymmetric division. Targeted perturbations reveal that Cyclin-dependent Kinase 1 (CDK1) and Aurora Kinase promote storage of Fibroblast Growth Factor Receptors (FGFRs) by suppressing endosomal degradation and recycling pathways. As cells progress through metaphase, loss of CDK1 activity permits differential degradation and targeted recycling of stored receptors, leading to asymmetric induction. …

Development Of A Novel Orthotopic Gastric Cancer Mouse Model., Wonyoung Kang, Leigh Maher, Michael Michaud, Seong-Woo Bae, Seongyeong Kim, Hye Seung Lee, Seock-Ah Im, Han-Kwang Yang, Charles Lee

Faculty Research 2021

BACKGROUND: Gastric cancer metastasis is a highly fatal disease with a five-year survival rate of less than 5%. One major obstacle in studying gastric cancer metastasis is the lack of faithful models available. The cancer xenograft mouse models are widely used to elucidate the mechanisms of cancer development and progression. Current procedures for creating cancer xenografts include both heterotopic (i.e., subcutaneous) and orthotopic transplantation methods. Compared to the heterotopic model, the orthotopic model has been shown to be the more clinically relevant design as it enables the development of cancer metastasis. Although there are several methods in use to develop …

Single-Cell Fluidic Force Microscopy Reveals Stress- Dependent Molecular Interactions In Yeast Mating, Marion Mathelié-Guinlet, Felipe Viela, JéRôMe Dehullu, Sviatlana Filimonava, Jason M. Rauceo, Peter N. Lipke, Yves F. DufrêNe

Publications and Research

Sexual agglutinins of the budding yeast Saccharomyces cerevisiae are proteins mediating cell aggregation during mating. Complementary agglutinins expressed by cells of opposite mating types “a” and “α” bind together to promote agglutination and facilitate fusion of haploid cells. By means of an innovative single-cell manipulation assay combining fluidic force microscopy with force spectroscopy, we unravel the strength of single specific bonds between a- and α-agglutinins (~100 pN) which require pheromone induction. Prolonged cell–cell contact strongly increases adhesion between mating cells, likely resulting from an increased expression of agglutinins. In addition, we highlight the critical role of disulfide bonds of the …
