Efforts to efficiently target brain tumors are constrained by the dearth of appropriate models to study tumor behavior toward treatment approaches as well as potential side effects to the surrounding normal tissue. We established a reproducible cerebral organoid model of brain tumorigenesis in an autologous setting by overexpressing
Ground-Based Modeling Of Deep-Space Risks To The Spleen And Intestine In Mice, Edith Nathalie Pineda Sotomayor
Ground-Based Modeling Of Deep-Space Risks To The Spleen And Intestine In Mice, Edith Nathalie Pineda Sotomayor
Theses and Dissertations
Deep-space missions, such as those planned for the cis-lunar space and Mars, pose unique challenges to astronaut health due to prolonged microgravity (MG) exposure and low-dose chronic irradiation (CIR). Understanding the impact of these stressors on secondary lymphoid tissues, such as the spleen and intestine, is crucial for ensuring crewmembers' well-being, given the impracticality of conducting this research with human samples. Using a ground-based facility, we exposed C57BL/6 female and male mice to CIR (1.74 Gy at a dose rate of 0.0029 Gy/h) and/or simulated MG (SMG) via hind limb unloading (HLU) for one month. Spleen and intestinal tissues were …

Comparative Pollen Morphology Of The Genus Rhaponticoides Vaill. (Asteraceae) In Türkiye: Implications For Taxonomy And Phylogeny, Foroogh Tavakoli, Gürkan Semi̇z, Batikan Günal, Meryem Bozkurt, Tuna Uysal
Turkish Journal of Botany
The main focus of this research is the palynological investigation of Rhaponticoides Vaill. species naturally occurring in Türkiye. Light microscopy and scanning electron microscopy (SEM) were employed to examine the pollen morphology of nine Rhaponticoides species in detail. Hierarchical cluster analysis (HCA) was also conducted to identify closely related species and distinguish the investigated taxa based on palynological traits. The study found that the pollen grains were tricolporate, isopolar, monad, and radially symmetric, with an oblate-spheroidal shape as indicated by the polar/equatorial (P/E) ratio. The pollen measurements derived from the P/E ratio ranged between 0.85 and 1, values associated with …

Introduction: Ischemia-induced retinopathies, such as retinopathy of prematurity and proliferative diabetic retinopathy, are major causes of vision loss. Both pathological neovascularization and neuronal damage contribute to vision impairment in these conditions, yet current therapies primarily target neovascularization and may further compromise neuronal health.
Objectives: Cyclic AMP (cAMP) is a widely recognized second messenger regulating diverse physiological and pathological processes. Exchange protein directly activated by cAMP (Epac) is a recently identified effector of cAMP signaling. This study aimed to investigate the role of Epac1 in ischemia-induced retinopathy and elucidate its underlying mechanisms using a mouse model of oxygen-induced retinopathy (OIR).

The global reliance on fossil fuels and natural gas has largely dominated the energy production field, but due to finite resource depletion and escalating greenhouse gas emissions, the immediate exploration of sustainable energy alternatives to mitigate climate change and ensure resource security has been a major concern. There has been extensive research into other more renewable methods of energy production, such as wind and hydropower. Of these current energy generation types, there are many areas of untapped potential from the mechanical movements generated ambiently not only in the large scale of power generation but also on a smaller scale. Two Essays On Health And Health Behavior In China, Jiamin Tan
Lingnan Theses (MPhil & PhD)
This thesis consists of two empirical studies on elderly mental health and on health behaviors. The first study investigates the long-term impact of childhood hunger on mental health in later life, using data from two nationally representative surveys in China. The study finds that childhood hunger exposure significantly increases the risks of depression and anxiety in later life. These effects are most pronounced among individuals exposed after age 5, suggesting that autobiographical memory formation plays an important role beyond socioeconomic and health pathways. The findings further indicate these impacts can be mitigated through economic interventions, such as welfare programs, and …

Fetal congenital anomalies and genetic disorders complicate 3%-5% of pregnancies and can have a significant impact on pregnancy outcomes. Precise and individualized prenatal diagnosis is crucial for effective counseling and management. The identification of new biomarkers holds promise for enhancing prenatal screening, diagnosis, and prognostic counseling in affected pregnancies. Recently, metabolomics has emerged as a potential adjunct in the interpretation of genetic variants identified through genome-wide sequencing for rare genetic conditions. To assess the potential of metabolomic profiling as a functional assay capable of providing deeper insights into the pathological processes and genetic findings associated with prenatal congenital anomalies, we …

Light-gated ion channels from protists (channelrhodopsins or ChRs) are optogenetic tools widely used for controlling neurons and cardiomyocytes. Multiplex optogenetic applications require spectrally separated molecules, which are difficult to engineer without disrupting channel function. Scanning numerous sequence databases, we identified three naturally blue-shifted ChRs from ancyromonads. They form a separate branch on the phylogenetic tree and contain residue motifs characteristic of anion ChRs (ACRs). However, only two conduct chloride, whereas the closely related Nutomonas longa homolog generates inward cation currents in mammalian cells under physiological conditions, significantly exceeding those by previously known tools with similar spectral maxima (peak absorption at …
A Computational Framework For Multi-Scale Analysis Of Mouse Cerebral Blood Flow Regulation, Hadi Esfandi
A Computational Framework For Multi-Scale Analysis Of Mouse Cerebral Blood Flow Regulation, Hadi Esfandi
Electronic Theses and Dissertations
Cerebral hemodynamic imaging is increasingly used as a non-invasive technique to study brain function and dysfunction in both research and clinical settings. Recent advances in imaging technologies have enabled high-speed, high-resolution, whole-brain measurements of blood flow and oxygenation. Improving the interpretation of these cerebral hemodynamic signals through a mechanistic understanding of their cellular drivers would enable more accurate characterization of brain function and pathology. However, no existing biophysical model fully captures the fine-grained dynamics observed in these signals. This dissertation introduces a biophysically grounded computational framework for multi-scale analysis of the mouse cerebral blood flow regulatory system, which captures mechanistic …

The number of reliable and non-invasive techniques to characterize ocular blood flow is limited. Here, we present a protocol for identifying a soft tissue window in the orbital aperture using pulsed-Doppler ultrasonography to offer real-time, non-invasive blood flow velocity monitoring in the mouse ophthalmic artery. We describe steps for preparing a mouse, positioning the ultrasound probe, and acquiring and recording signals. We then detail procedures for analyzing results using a workstation equipped with Doppler flow velocity software. For complete details on the use and execution of this protocol, please refer to Sharina et al.
Investigating The Role Of Chaperones And Sequestrases In Misfolded Protein Sequestration, Sarah Rose Rolli
Investigating The Role Of Chaperones And Sequestrases In Misfolded Protein Sequestration, Sarah Rose Rolli
Dissertations (1934 -)
Protein homeostasis, or proteostasis, is responsible for a folded and functional proteome and is essential for cell survival. Proteins can misfold during stress or because of mutation. The accumulation of misfolded proteins is observed in many diseases, including Huntington’s disease. Misfolded proteins will clump together into inclusions, which can be sequestered to membrane-less compartments by proteostasis mechanisms. Several proteins, including chaperones, co-chaperones, and other proteins, have been identified as sequestrases that target inclusions and misfolded proteins to different protein quality control (PQC) compartments. The juxtanuclear quality control (JUNQ) compartment forms in the nucleus-vacuole junction in yeast cells, and several soluble …

Spatial And Temporal Distribution Of Large (1–5 Mm) Microplastics On The Strandline Of A Macrotidal Sandy Beach (Polzeath, Southwest England) And Their Association With Beach-Cast Seaweed, Catherine Beale, Andrew Turner
School of Geography, Earth and Environmental Sciences
Microplastics (MPs) are ubiquitous and persistent contaminants of the marine environment, but a clear understanding of their cycling, fate, and impacts in coastal zones is lacking. In this study, large MPs (1–5 mm) were sampled spatially and temporally from the strandline of a macrotidal, sandy beach (Polzeath) in southwest England. MPs encompassing a diversity of sources were categorised by morphology (foams, nurdles, biobeads, fragments, fibres, films) and quantified by number and mass, with a selection analysed for polymer type. A total of about 17,600 particles of around 350 g in mass were retrieved from 30 samples over a period of …

A series of unsymmetrically substituted BODIPY dyes featuring fused benzo- or naphtho-fragments on one pyrrolic unit were synthesized from the corresponding pyrrolic precursors. The synthetic route was optimized using a modular approach based on the condensation of formylpyrroles with alkylpyrroles, enabling the identification of precursor combinations that minimize byproduct formation and improve preparative yields. The resulting benzo- and naphtho-fused BODIPYs display intense fluorescence in the red region, with emission maxima spanning 590–680 nm and fluorescence quantum yields ranging from 0.27 to 0.84. Their two-photon absorption (2PA) properties were studied both experimentally and computationally. An increase in the two-photon absorption cross-section …

MraY is an essential bacterial enzyme for peptidoglycan synthesis in cell walls and serves as a promising but unrealized target for developing effective antibacterial drugs. Nature has provided a remarkable array of nucleoside inhibitors of MraY, and researchers have skillfully refined these structures to develop inhibitors that effectively mimic natural products. Yet, both natural products and their synthetic variants often face challenges regarding inadequate

BACKGROUND: Diagnosis of autism spectrum disorder (ASD) significantly improves quality of life, yet current diagnostic practices rely on subjective behavioural assessments. While machine learning models using functional Magnetic Resonance Imaging (fMRI) show promise for objective diagnosis, existing approaches have critical limitations. High-accuracy models often neglect interpretability, increasing clinical distrust, while interpretable approaches frequently suffer from low accuracy and lack neuroscientific validation of identified biomarkers. Furthermore, no studies have systematically benchmarked which interpretability methods are most effective for fMRI data, hindering the translation of Artificial intelligence (AI) findings into clinical practice.
METHODS: In this observational study, we used the Autism Brain …

Injury To Soybean Seedlings From Alfalfa Weevil (Coleoptera: Curculionidae) Larvae, Robert L. Koch, M. Lourdes Chamorro
The Great Lakes Entomologist
Alfalfa weevil, Hypera postica (Gyllenhal) (Coleoptera: Curculionidae), is primarily a pest of alfalfa, but it has been reported feeding on other plants, including soybean. The relatively limited literature on H. postica in soybean indicates that adults and larvae can feed on soybean, but it is not suitable for larval development and survival. There have been instances of H. postica adults moving from alfalfa to nearby soybean fields where they have inflicted significant injury to the soybean plants. Here we report an instance of H. postica larvae causing severe feeding injury to soybean. This infestation of soybean was the result of …

Sampling approaches are commonly adapted to reflect the study objectives in biodiversity monitoring projects. This approach optimizes findings to be locally relevant but comes at the cost of generalizability of findings. Here, we detail a comparison study directly examining how researchers’ choice of arthropod trap and level of specimen identification affects observations made in small-scale arthropod biodiversity studies. Four arthropod traps (pitfall traps, yellow ramp traps, yellow sticky cards, and novel jar ramp traps) were compared with respect to an array of biodiversity metrics calculated at two levels of identification. The arthropod community captured varied by trap type. Pitfalls and …

Pollen Loads Of Insects Caught In Cultivated Fields Of Oilseed Pennycress, Frank Forcella, Ellinor Sheik, Michael Zavada, Mark Bernards
The Great Lakes Entomologist
Although many insects are known to be associated with the floral canopies of oilseed pennycress (Thlaspi arvense), those that carry the crop’s pollen are unknown. During early spring of 2024 insects associated with pennycress flowers in Illinois and Minnesota were captured in sweep nets, frozen, sorted by species, and sonicated to remove pollen. Pollen grains were counted, and pollen loads per individual insect were calculated. Members of the orders Diptera, Hemiptera, and Hymenoptera accounted for 66%, 16%, and 14% of the 850 insects examined, but these same orders were responsible, respectively, for 18%, 3%, and 78% of the …

The conservation of bees is crucial for maintaining the essential ecological services they provide. Understanding bee species' risk of extinction is a key part of conservation efforts, and can include assessment of dietary specialization, which has been linked to higher extinction risk. Dietary specialization in bees, called oligolecty, occurs when females select pollen only from closely related plants to feed their larvae. While the solitary bee, Megachile (Chelostomoides) campanulae (Robertson 1903), has been associated with flowers in the genus Campanula L. (Campanulaceae: Asterales), evidence from pollen analysis is limited, and dietary classification is reported inconsistently. This study examined dietary specialization …

Nocturnal Flight Periodicity Of Caddisflies (Trichoptera) Along The Lake Huron Shoreline Of Drummond Island, Michigan (Usa), Robert A. Haack, David Houghton
The Great Lakes Entomologist
Over 7000 caddisfly specimens, representing 33 species in 10 families, were collected between sunset and sunrise on 12 nights: three nights each during early July and early August in both 2023 and 2024. Collections were made on the southwestern side of Drummond Island along the shoreline of Lake Huron. Sampling occurred for five minutes at 15-minute intervals from near sunset (21:00) to 01:00 and then once again at 03:00 and 05:00. Nocturnal flight periodicity was broadly similar between years and months, with mean caddisfly abundance and species richness peaking at about one hour after sunset. Mean species accumulation varied between …

Managers of northern hardwood forests in the Great Lakes region create gaps in the forest canopy of various sizes during harvesting to achieve different silvicultural objectives. Single-tree harvesting favors recruitment of shade-tolerant tree species by creating small gaps in the canopy, whereas group selection creates larger gaps that can favor shade-intolerant tree species. We evaluated changes in carabid beetle abundance and diversity in harvest-created gaps of five sizes (gap diameters of 6, 10, 20, 30, and 46 m) and uncut forests in northern Wisconsin where sugar maple (Acer saccharum) was the dominant tree species. Gaps were cut during …

A Day In The Life Of An Orbital And Oculoplastic Surgeon, Alexis Duffy, Ba
inSIGHT
Have you ever heard of an oculoplastic surgeon before? And if so, ever wonder what they do or how they differ from plastic surgeons? Treating the periocular region requires an in-depth understanding not only of the eye, but also its complex interplay with adjacent orbital, sinus, and craniofacial anatomy; there is much more than meets the eye! The most common pathway to become an oculoplastic surgeon includes completion of a four-year ophthalmology residency, followed by one to two years of subspecialized oculoplastic fellowship. Oculoplastic fellowships provide surgical and medical experience managing pathology of the eyelid and orbit, and different programs …

Research Colloquium
Aim: Repurposing of FDA-approved antifungal drugs to combat prostate cancer.
Background: Prostate cancer is one of the leading causes of cancer death among men in the United States, second only to non-melanoma skin cancer. The American Cancer Society estimates about 313,780 new cases and 35,770 deaths from prostate cancer in 2025. Despite various treatments such as radiation therapy (both external beam and internal), chemotherapy, hormone therapy, cryotherapy, biological therapy, and high-intensity focused ultrasound, men are monitored for side effects over many years. Therefore, new drugs and treatment strategies are urgently needed to address this challenge.
Methods: In this study, we …
