At the interface between host and external environment, the airway epithelium serves as a major protective barrier. In the present study we show that protein kinase D (PKD) plays an important role in the formation and integrity of the airway epithelial barrier. Either inhibition of PKD activity or silencing of PKD increased transepithelial electrical resistance (TEER), resulting in a tighter epithelial barrier. Among the three PKD isoforms, PKD3 knockdown was the most efficient one to increase TEER in polarized airway epithelial monolayers. In contrast, overexpression of PKD3 wild type, but not PKD3 kinase-inactive mutant, disrupted the formation of apical intercellular …

Neuropilin-1 (NRP-1), a member of a family of signaling proteins, was shown to serve as an entry factor and potentiate SARS Coronavirus 2 (SARS-CoV-2) infectivity in vitro. This cell surface receptor with its disseminated expression is important in angiogenesis, tumor progression, viral entry, axonal guidance, and immune function. NRP-1 is implicated in several aspects of a SARS-CoV-2 infection including possible spread through the olfactory bulb and into the central nervous system and increased NRP-1 RNA expression in lungs of severe Coronavirus Disease 2019 (COVID-19). Up-regulation of NRP-1 protein in diabetic kidney cells hint at its importance in a population at …

Effect Of Age And Sex On The Discrimination Of Monozygotic Twins Using Cg18562578 Dna Methylation In Saliva, Carla Pierre Louis
Student Theses
DNA methylation is a type of epigenetic modification that impacts gene expression without disturbing the genetic nucleotide sequence. DNA methylation has the potential to be included in the daily forensic analysis because among other uses, it has the potential to be applied to discrimination of monozygotic (MZ) twins that currently cannot be differentiated via forensic DNA profiling. In previous research, our group showed that DNA methylation at the cg18562578 site in chromosome 3 could be used to discriminate MZ twins. Because studies have shown that DNA methylation measures can be influenced by demographic factors such as sex and age; here, …

Pre-mRNA splicing is an essential step for the regulation of gene expression. In order to specifically capture splicing variants in plants for genome-wide association studies (GWAS), we developed a software tool to quantify and visualise Variations of Splicing in Population (VASP). VASP can quantify splicing variants from short-read RNA-seq datasets and discover genotype-specific splicing (GSS) events, which can be used to prioritise causal pre-mRNA splicing events in GWAS. We applied our method to an RNA-seq dataset with 328 samples from 82 genotypes from a rice diversity panel exposed to optimal and saline growing conditions. In total, 764 significant GSS events …

Heme oxygenase 1 (HO-1) and the cytochromes P450 (P450s) are endoplasmic reticulum–bound enzymes that rely on the same protein, NADPH-cytochrome P450 reductase (POR), to provide the electrons necessary for substrate metabolism. Although the HO-1 and P450 systems are interconnected owing to their common electron donor, they generally have been studied separately. As the expressions of both HO-1 and P450s are affected by xenobiotic exposure, changes in HO-1 expression can potentially affect P450 function and, conversely, changes in P450 expression can influence HO-1. The goal of this study was to examine interactions between the P450 and HO-1 systems. Using bioluminescence resonance …

Contest Powwow: Sport And Native American Culture, Steven J. Aicinena Dr, Sebahattin Ziyanak
The Qualitative Report
The Native American powwow has served to maintain the culture of North America’s Indigenous peoples since before the arrival of European colonialists. In traditional forms of the powwow, competition is not common whereas contest powwows are characterized by the primacy of competition. We assess similarities and differences between the contest powwow and competitive sport found within the broader American society through in-depth semi-structured interviews with 14 competitive contest powwow dancers. We address a number of questions through the qualitative research process, such as what does the powwow mean to you? What are your primary goals for competing? We analyzed the …

Emotional Dimensions Of Teaching In Elementary Education Preparation, Kathleen M. Crawford, Juliann Sergi Mcbrayer, Katherine Fallon
The Qualitative Report
Student teaching is a pivotal event in teacher education preparation programs, and there is a need to investigate emotions in teaching. This study examined how one elementary program navigated the emotional dimensions of teaching. Findings revealed that the emotional dimensions of student teachers were influenced by individualized factors unique to the teacher; certain emotions were perceived as more acceptable to express; and supervisors needed to support student teachers to manage and respond to the emotional dimensions of teaching. The recommendation is to go beyond the technical and academic aspects of teaching and address the emotional dimensions to best prepare the …

Alkenones are biomarkers produced solely by algae in the order Isochrysidales that have been used to reconstruct sea surface temperature (SST) since the 1980s. However, alkenone-based SST reconstructions in the northern high latitude oceans show significant bias towards warmer temperatures in core-tops, diverge from other SST proxies in down core records, and are often accompanied by anomalously high relative abundance of the C37 tetra-unsaturated methyl alkenone (%C37:4). Elevated %C37:4 is widely interpreted as an indicator of low sea surface salinity from polar water masses, but its biological source has thus far remained elusive. Here we identify a lineage of Isochrysidales …

Communicating Scientific Uncertainty In An Age Of Covid-19: An Investigation Into The Use Of Preprints By Digital Media Outlets, Alice Fleerackers, Michelle Riedlinger, Laura Moorhead, Rukhsana Ahmed, Juan Pablo Alperin
Communication Faculty Scholarship
In this article, we investigate the surge in use of COVID-19-related preprints by media outlets. Journalists are a main source of reliable public health information during crises and, until recently, journalists have been reluctant to cover preprints because of the associated scientific uncertainty. Yet, uploads of COVID-19 preprints and their uptake by online media have outstripped that of preprints about any other topic. Using an innovative approach combining altmetrics methods with content analysis, we identified a diversity of outlets covering COVID-19-related preprints during the early months of the pandemic, including specialist medical news outlets, traditional news media outlets, and aggregators. …

Localizing The Green Energy Revolution, Hannah J. Wiseman
Localizing The Green Energy Revolution, Hannah J. Wiseman
Emory Law Journal Online
The United States is on the verge of a new industrial revolution. Renewable energy could replace more than 60% of our current energy generation infrastructure in fifteen years. This change is critical, yet it risks failure. The renewable generation already built in the United States consists primarily of large-scale projects connected to transmission lines in rural areas. The expansive new generation needed to reduce carbon emissions must also be predominantly large-scale, and rural, for reasons of efficiency. But a revolution that focuses nearly exclusively on “big energy” is likely to encounter obstacles, and it has downsides that could be mitigated …

Numerical Analysis Of Coupled Systems Of Odes And Applications Toenzymatic Competitive Inhibition By Product, Vinh Quang Mai, Thái Ahn Nhan
Mathematics and Computer Science
Enzymatic inhibition is one of the key regulatory mechanisms in cellular metabolism, especially the enzymatic competitive inhibition by product. This inhibition process helps the cell regulate enzymatic activities. In this paper, we derive a mathematical model describing the enzymatic competitive inhibition by product. The model consists of a coupled system of nonlinear ordinary di erential equations for the species of interest. Using nondimensionalization analysis, a formula for product formation rate for this mechanism is obtained in a transparent manner. Further analysis for this formula yields qualitative insights into the maximal reaction velocity and apparent Michaelis-Menten constant. Integrating the model numerically, …
