“Glass Ceilings” And “Sticky Floors:” Multiple Identities Of Female Superintendents And Intersectionality, Lindsey Brooke Mcbride Jan 2021

“Glass Ceilings” And “Sticky Floors:” Multiple Identities Of Female Superintendents And Intersectionality, Lindsey Brooke Mcbride

Theses and Dissertations

According to AASA, “men are four times more likely to serve in the most powerful position in education” (Robinson et al, 2017). That position being the superintendent in public school setting. The School Superintendents Association (AASA) found that less than 25% of superintendents were women (Superville, 2016). The purpose of this study was to examine the influence, if any, that is created by a female superintendent choosing certain career pathways or having certain identities such as the identity of a female, spouse/mother, and leader and how those identities shape her professional identity making her a better fit in the organization …

Drug resistance is the main obstacle to achieving cures with both conventional and targeted anticancer drugs. The emergence of acquired drug resistance is initially mediated by non-genetic transcriptional changes, which occur at a much higher frequency than mutations and may involve population-scale transcriptomic adaptation. CDK8/19 kinases, through association with transcriptional Mediator complex, regulate transcriptional reprogramming by co-operating with different signal-responsive transcription factors. Here we tested if CDK8/19 inhibition could prevent adaptation to drugs acting on epidermal growth factor receptor (EGFR/ERBB1/HER1). The development of resistance was analyzed following long-term exposure of BT474 and SKBR3 breast cancer cells to EGFR-targeting small molecules …

Cerebral autosomal dominant arteriopathy with subcortical infarct and leukoencephalopathy (CADASIL) is a Notch3 mutation-induced cerebral small vessel disease, leading to recurrent ischemic stroke and vascular dementia. There is currently no treatment that can stop or delay CADASIL progression. We have demonstrated the efficacy of treatment with combined stem cell factor (SCF) and granulocyte colony-stimulating factor (G-CSF) (SCF+G-CSF) in reducing cerebral small vessel thrombosis in a TgNotch3R90C mouse model of CADASIL. However, it remains unknown whether SCF+G-CSF treatment protects neurons from microvascular thrombosis-induced ischemic damage. Using bone marrow transplantation to track thrombosis, we observed that capillary thrombosis was widely distributed in …

Saudi Early Childhood Educators' Perceptions Of Gender Roles In Children's Dramatic Play, Dalal Alanazi Jan 2021

Saudi Early Childhood Educators' Perceptions Of Gender Roles In Children's Dramatic Play, Dalal Alanazi

USF Tampa Graduate Theses and Dissertations

This study was conducted in a unique time of cultural and social change in Saudi Arabia. The new Saudi Vision 2030 promotes gender equality, and it is within this context that I investigated four Saudi preschool teachers’ perceptions of children’s gender roles in dramatic play. I focused on how the teachers have responded to the current move to think differently about gender roles in the Saudi context and the relevant policy changes. Rogoff’s sociocultural theory, which highlights the significance of cultural contexts on peoples’ perceptions, framed the study. I conducted semi-structured interviews with the four Saudi teachers. As a result …

Terrestrial ecosystems are increasingly enriched with resources such as atmospheric CO2that limit ecosystem processes. The consequences for ecosystem carbon cycling depend on the feedbacks from other limiting resources and plant community change, which remain poorly understood for soil CO2efflux, JCO2, a primary carbon flux from the biosphere to the atmosphere. We applied a unique CO2enrichment gradient (250 to 500 μL L-1) for eight years to grassland plant communities on soils from different landscape positions. We identified the trajectory of JCO2responses and feedbacks from other resources, plant diversity [effective species richness, exp(H)], and community change (plant species turnover). We found linear …

A functional association is uncovered between the ribosome-associated trigger factor (TF) chaperone and the ClpXP degradation complex. Bioinformatic analyses demonstrate conservation of the close proximity of tig, the gene coding for TF, and genes coding for ClpXP, suggesting a functional interaction. The effect of TF on ClpXP-dependent degradation varies based on the nature of substrate. While degradation of some substrates are slowed down or are unaffected by TF, surprisingly, TF increases the degradation rate of a third class of substrates. These include λ phage replication protein λO, master regulator of stationary phase RpoS, and SsrA-tagged proteins. Globally, TF acts to …

While genome recoding using quadruplet codons to incorporate non-proteinogenic amino acids is attractive for biotechnology and bioengineering purposes, the mechanism through which such codons are translated is poorly understood. Here we investigate translation of quadruplet codons by a +1-frameshifting tRNA, SufB2, that contains an extra nucleotide in its anticodon loop. Natural post-transcriptional modification of SufB2 in cells prevents it from frameshifting using a quadruplet-pairing mechanism such that it preferentially employs a triplet-slippage mechanism. We show that SufB2 uses triplet anticodon-codon pairing in the 0-frame to initially decode the quadruplet codon, but subsequently shifts to the +1-frame during tRNA-mRNA translocation. SufB2 …

Children are potentially exposed to products that contain nanoparticles (NPs). In particular, silver NPs are commonly present both in products used by and around children, primarily due to their antibacterial properties. However, very few data are available regarding the ability of silver NPs to penetrate through the oral mucosa in children. In the present work, we used baby porcine buccal mucosa mounted on vertical Franz diffusion cells, as an in vitro model to investigate penetration of silver NPs (19 ± 5 nm). Permeability experiments were performed using pristine physiologically-relevant saline solution in the receiver chamber and known concentrations of NPs …

Hepatocellular carcinoma (HCC) is a highly malignant tumor. In this study, we sought to identify a novel biomarker for HCC by analyzing transcriptome and clinical data. The R software was used to analyze the differentially expressed genes (DEGs) in the datasets GSE74656 and GSE84598 downloaded from the Gene Expression Omnibus database, followed by a functional annotation. A total of 138 shared DEGs were screened from two datasets. They were mainly enriched in the “Metabolic pathways” pathway (Padj = 8.21E-08) and involved in the carboxylic acid metabolic process (Padj = 0.0004). The top 10 hub genes were found by protein-protein interaction …

Human-induced ecological boundaries, or anthropogenic ecotones, may arise where administrative boundaries meet on undeveloped lands. Landscape-level ecological processes related to factors such as fire, invasive species, grazing, resource extraction, wildlife, and water may be affected due to unique management strategies adopted by each administrative unit. Over time, different management can result in discernible ecological differences (e.g., species composition or soil characteristics). Thus, fragmentation in the management landscape can correspond to ecological fragmentation. Different ecological patterns may emerge due to an increase in the number of management units in a region, or due to an increase in the number of different …

Chemistry Faculty Research & Creative Works

The flavin reductase (FRED) and isobutyl amine N-hydroxylase (IBAH) from Streptomyces viridifaciens constitute a two-component, flavin-dependent monooxygenase system that catalyzes the first step in valanimycin biosynthesis. FRED is an oxidoreductase that provides the reduced flavin to IBAH, which then catalyzes the hydroxylation of isobutyl amine (IBA) to isobutyl hydroxylamine (IBHA). In this work, we used several complementary methods to investigate FAD binding, steady-state and rapid reaction kinetics, and enzyme-enzyme interactions in the FRED: IBAH system. The affinity of FRED for FADox is higher than its affinity for FADred, consistent with its function as a flavin reductase. Conversely, IBAH binds FADred …

Single-Locus Species Delimitation And Ecological Niche Modeling Provide Insights Into The Evolution, Historical Distribution, And Taxonomy Of The Pacific Chorus Frogs, Robert C. Jadin, Sarah A. Orlofske, Tereza Jezkova, Christopher Blair

Publications and Research

The Pacific chorus frogs are a complex of three wide-ranging species (i.e. Hyliola hypochondriaca, Hyliola regilla, Hyliola sierra) whose current taxonomy remains unresolved. We conducted species delimitation analyses of these taxa using fragments of the cytochrome b and 12S–16S mtDNA genes to assess the species diversity. Importantly, we included samples from new locations throughout the range to better understand species distributions and identify potential contact zones among clades. Our analyses revealed three slightly parapatric but distinct species-level clades. Molecular dating revealed that these species began diverging in the Pleistocene c. 1.4 Mya with H. hypochondriaca and …

Resistance To Aflatoxin Accumulation In Maize Mediated By Host-Induced Gene Silencing Of Aspergillus Flavus Alkaline Protease And O-Methyltransferase-A Genes, Olanike Omotola Omolehin

LSU Doctoral Dissertations

Aspergillus flavus is a soil-borne fungal pathogen that infects maize and produces aflatoxins. In the current study, portions of the alkaline protease (alk) and the O-methyl transferase (omtA) genes, which are key in A. flavus virulence and aflatoxin biosynthesis, respectively, were targeted for suppression through an RNAi (RNA interference) approach known as Host-Induced Gene Silencing (HIGS). Separate RNAi vectors were designed to carry regions of the alk and omtA gene fragments (Alk-RNAi and OmtA-RNAi) and introduced into B104 maize zygotic embryos. Eight and six transformation events were positive for the alk and omtA transgene, respectively. …

Mental Health Workers’ Perceptions Of Risk Factors For Human Trafficking In Nairobi, Kenya: A Preliminary Qualitative Investigation, Stacey Diane A. Litam, Dakota King-White, Kathryn C. Maccluskie, Julia C. Phillips

Journal of Counseling Sexology & Sexual Wellness: Research, Practice, and Education

The researchers of this pilot study conducted three in-depth semi-structured interviews with four mental health workers in Nairobi to obtain a deeper understanding of their perceptions of human trafficking in Kenya. Four themes that increased vulnerability for entrance into the human trafficking trade were identified. Individuals were at increased risk for forced labor exploitation due to socioeconomic factors, traditional African practices, cultural beliefs, and political risk factors. This article provides implications for practice and support for community mental health workers, counselors, and educators working with survivors of human trafficking. Implications for future research and practice are discussed.

The Influence Of Cyber-Sexual Assault On The Mental Health Outcomes Of Survivors, Kelley R. Holladay, W. Bryce Hagedorn, David Boote

Journal of Counseling Sexology & Sexual Wellness: Research, Practice, and Education

Researchers examined data collected from the only national sample of cyber-sexual assault (CBSA) survivors (N = 97; 3.75\% response rate to the online survey) using three valid instruments that measure symptomology of sexual assault. We found that participants scored high across each of the inventories, indicating CBSA survivors respond in ways that are similar to sexual assault survivors concerning emotional dysregulation, depression, and post-traumatic stress symptomology, particularly if they had a history of sexual assault. Implications for counselors are reviewed, and clinical recommendations are offered.

Between 2015 and 2019, the list of Lepidoptera from “cave” habitats (i.e., proper caves, rock shelters and artificial subterranean structures) in Austria grew from 17 to 62 species, although the effort of data collection remained nearly constant from the late 1970s onwards. The newly recorded moths and butterflies were resting in caves during daytime in the the warm season, three species were also overwintering there. We observed Catocala elocata at 28 cave inspections, followed by Mormo maura (18), Catocala nupta (7), Peribatodes rhomboidaria, and Euplagia quadripunctaria (6). More than half of the species have been repeatedly observed in caves …

Understanding diagnostic tests and examining important features of novel coronavirus (COVID-19) infection are essential steps for controlling the current pandemic of 2020. In this paper, we study the relationship between clinical diagnosis and analytical features of patient blood panels from the US, Mexico, and Brazil. Our analysis confirms that among adults, the risk of severe illness from COVID-19 increases with pre-existing conditions such as diabetes and immunosuppression. Although more than eight months into pandemic, more data have become available to indicate that more young adults were getting infected. In addition, we expand on the definition of COVID-19 test and discuss …
