Management Of The Phosphorus-Cladophora Dynamic At A Site On Lake Ontario Using A Multi-Module Bioavailable P Model, Martin Auer, Cory P. Mcdonald, Anika Kuczynski, Chenfu Huang

Michigan Tech Publications, Part 1

The filamentous green alga Cladophora grows to nuisance proportions in Lake Ontario. Stimulated by high phosphorus concentrations, nuisance growth results in the degradation of beaches and clogging of industrial water intakes with attendant loss of beneficial uses. We develop a multi-module bioavailable phosphorus model to examine the efficacy of phosphorus management strategies in mitigating nuisance algal growth. The model platform includes modules simulating hydrodynamics (FVCOM), phosphorus-phytoplankton dynamics (GEM) and Cladophora growth (GLCMv3). The model is applied along a 25 km stretch of the Lake Ontario nearshore, extending east from Toronto, ON and receiving effluent from three wastewater treatment plants. Simulation …

Improved Prognostic Stratification Using Circulating Tumor Cell Clusters In Patients With Metastatic Castration-Resistant Prostate Cancer., Chun Wang, Zhenchao Zhang, Weelic Chong, Rui Luo, Ronald E Myers, Jian Gu, Jianqing Lin, Qiang Wei, Bingshan Li, Timothy R Rebbeck, Grace Lu-Yao, William K Kelly, Hushan Yang Jan 2021

Department of Medical Oncology Faculty Papers

Liquid biopsy-based biomarkers have advantages in monitoring the dynamics of metastatic castration-resistant prostate cancer (mCRPC), a bone-predominant metastatic disease. Previous studies have demonstrated associations between circulating tumor cells (CTCs) and clinical outcomes of mCRPC patients, but little is known about the prognostic value of CTC-clusters. In 227 longitudinally collected blood samples from 64 mCRPC patients, CTCs and CTC-clusters were enumerated using the CellSearch platform. The associations of CTC and CTC-cluster counts with progression-free survival (PFS) and overall survival (OS), individually and jointly, were evaluated by Cox models. CTCs and CTC-clusters were detected in 24 (37.5%) and 8 (12.5%) of 64 …

Humans Express Seven Heparan Sulfate (HS) 3-O-Sulfotransferases that Differ in Substrate Specificity and Tissue Expression. Although Genetic Studies Have Indicated that 3-O-Sulfated HS Modulates Many Biological Processes, Ligand Requirements for Proteins Engaging with HS Modified by 3-O-Sulfate (3-OS) Have Been Difficult to Determine. in Particular, the Context in Which the 3-OS Group Needs to Be Presented for Binding is Largely Unknown. We Describe Herein a Modular Synthetic Approach that Can Provide Structurally Diverse HS Oligosaccharides with and Without 3-OS. the Methodology Was Employed to Prepare 27 Hexasaccharides that Were Printed as a Glycan Microarray to Examine Ligand Requirements of a …

A Microscale Approach To Investigate Breast Cancer Cellular Communication And Migration, Sharif Mohammad Mizanur Rahman Jan 2021

A Microscale Approach To Investigate Breast Cancer Cellular Communication And Migration, Sharif Mohammad Mizanur Rahman

LSU Doctoral Dissertations

Metastatic breast cancer significantly decreases patient survival, with the majority of deaths caused by secondary metastasis. Cancer metastasis is a multi-step process that starts with the detachment of a cancer cell from the primary tumor and ends with establishing a secondary tumor at a distal location. This work's focus was to use an interdisciplinary microscale approach to study how cancer cells respond to neighboring cells and how they migrate in response to chemical gradients. Two distinct devices, a microfluidic device, and a 3D-printed plate insert, were developed to perform co-culture studies to elucidate the impact of ASCs on cancer cell …

Compared to flat devices based on rigid substrates, cable-shaped dye-sensitized solar cells hold advantages of smaller size, light weight, facile fabrication, flexibility, and low cost, thus a promising direction for applications such as wearable electronic devices. However, most reported fiber-shaped dye-sensitized solar cells use Pt wires as counter electrodes, which are high in cost. Herein, a flexible Pt-free counter electrode is fabricated via depositing ternary nickel cobalt selenide (Ni–Co–Se) particles on the surface of carbon fibers. Scanning electron microscopy and X-ray diffraction are used to characterize the counter electrode and alloy material. Results from bare and modified carbon fiber counter …

Despite the established roles of the epigenetic factor UHRF1 in oncogenesis, no UHRF1-targeting therapeutics have been reported to date. In this study, we use fragment-based ligand discovery to identify novel scaffolds for targeting the isolated UHRF1 tandem Tudor domain (TTD), which recognizes the heterochromatin-associated histone mark H3K9me3 and supports intramolecular contacts with other regions of UHRF1. Using both binding-based and function-based screens of a ~ 2300-fragment library in parallel, we identified 2,4-lutidine as a hit for follow-up NMR and X-ray crystallography studies. Unlike previous reported ligands, 2,4-lutidine binds to two binding pockets that are in close proximity on TTD and …

Manuscripts, Articles, Book Chapters and Other Papers

Bladder and bowel dysfunction (BBD) is a common yet underdiagnosed paediatric entity that describes lower urinary tract symptoms (LUTS) accompanied by abnormal bowel patterns manifested as constipation and/or encopresis. LUTS usually manifest as urgency, urinary frequency, incontinence, and urinary tract infections (UTI). Despite increasing recognition of BBD as a risk factor for long-term urinary tract problems including recurrent UTI, vesicoureteral reflux, and renal scarring, the mechanisms underlying BBD have been unclear, and treatment remains empirical. We investigated how constipation affects the lower urinary tract function using a juvenile murine model of functional constipation. Following four days of functional constipation, animals …

Anorexia Nervosa And The Family: A Look At Well-Siblings, Parents, And Family Dynamics Through A Family Systems Perspective, Martha Avila-Zavala

Modern Psychological Studies

Anorexia nervosa (AN) is one of the main, and one of the most common types of eating disorders among adolescent individuals, especially females. It is characterized by limited eating, increased amounts of physical activity, low self-esteem, and extreme criticism of oneself and one’s body. An individual diagnosed with AN is greatly physically, mentally, and emotionally affected by it. However, an AN diagnosis also has a significant impact on the whole family. In recent years, the family has been incorporated into the treatment of an individual diagnosed with AN, which has shown to be effective in the recovery process and in …

Trimethylornithine Membrane Lipids: Discovered In Planctomycetes And Identified In Diverse Environments, Eli K. Moore Jan 2021

Trimethylornithine Membrane Lipids: Discovered In Planctomycetes And Identified In Diverse Environments, Eli K. Moore

School of Earth & Environment Departmental Research

Intact polar membrane lipids (IPLs) are the building blocks of all cell membranes. There is a wide range of phosphorus-free IPL structures, including amino acid containing IPLs, that can be taxonomically specific. Trimethylornithine membrane lipids (TMOs) were discovered in northern wetland Planctomycete species that were isolated and described in the last decade. The trimethylated terminal nitrogen moiety of the ornithine amino acid in the TMO structure gives the lipid a charged polar head group, similar to certain phospholipids. Since their discovery, TMOs have been identified in various other recently described northern latitude Planctomycete species, and in diverse environments including tundra …

Standard magnetic resonance imaging approaches offer high-resolution but indirect measures of neural activity, limiting understanding of the physiological processes associated with imaging findings. Here, we used calibrated functional magnetic resonance imaging during the resting state to recover low-frequency fluctuations of the cerebral metabolic rate of oxygen (CMRO2). We tested whether functional connections derived from these fluctuations exhibited organization properties similar to those established by previous standard functional and anatomical connectivity studies. Seventeen participants underwent 20 min of resting imaging during dual-echo, pseudocontinuous arterial spin labeling, and blood-oxygen-level dependent (BOLD) signal acquisition. Participants also underwent a 10 min normocapnic and hypercapnic …

Comparison Of Bifurcated Halogen With Hydrogen Bonds, Steve Scheiner Jan 2021

Comparison Of Bifurcated Halogen With Hydrogen Bonds, Steve Scheiner

Chemistry and Biochemistry Faculty Publications

Bifurcated halogen bonds are constructed with FBr and FI as Lewis acids, paired with NH3 and NCH bases. The first type considered places two bases together with a single acid, while the reverse case of two acids sharing a single base constitutes the second type. These bifurcated systems are compared with the analogous H-bonds wherein FH serves as the acid. In most cases, a bifurcated system is energetically inferior to a single linear bond. There is a larger energetic cost to forcing the single σ-hole of an acid to interact with a pair of bases, than the other way …

Faculty Research 2021

Post-transplant lymphoproliferative disorder (PTLD) is one of the most common malignancies after solid organ or allogeneic stem cell transplantation. Most PTLD cases are B cell neoplasias carrying Epstein-Barr virus (EBV). A therapeutic approach is reduction of immunosuppression to allow T cells to develop and combat EBV. If this is not effective, approaches include immunotherapies such as monoclonal antibodies targeting CD20 and adoptive T cells. Immune checkpoint inhibition (ICI) to treat EBV+ PTLD was not established clinically due to the risks of organ rejection and graft-versus-host disease. Previously, blockade of the programmed death receptor (PD)-1 by a monoclonal antibody …

Faculty Publications & Research

To increase student engagement and reduce frustration over grades, data is being collected in my Pathophysiology classes on specific assessment questions which require students to think and apply what they have learned to answer assessment questions (“transfer” questions). These questions are being administered both before and after students have completed specific modules to assess their attitude and understanding of the material. Data is also being collected on student study habits before and after specific assessments for correlation with their scores on “transfer” questions. It is hoped to reduce student frustration over grades by analyzing their behavior and improving their understanding.

'Educated' Osteoblasts Reduce Osteoclastogenesis In A Bone-Tumor Mimetic Microenvironment., Alexus D. Kolb, Jinlu Dai, Evan T. Keller, Karen M. Bussard

Department of Cancer Biology Faculty Papers

Breast cancer (BC) metastases to bone disrupt the balance between osteoblasts and osteoclasts, leading to excessive bone resorption. We identified a novel subpopulation of osteoblasts with tumor-inhibitory properties, called educated osteoblasts (EOs). Here we sought to examine the effect of EOs on osteoclastogenesis during tumor progression. We hypothesized that EOs affect osteoclast development in the bone-tumor niche, leading to suppressed pre-osteoclast fusion and bone resorption. Conditioned media (CM) was analyzed for protein expression of osteoclast factors receptor activator of nuclear factor kappa-β ligand (RANKL), osteoprotegerin (OPG), and tumor necrosis factor alpha (TNFα) via ELISA. EOs were co-cultured with pre-osteoclasts on …

A Record Of Vapour Pressure Deficit Preserved In Wood And Soil Across Biomes, Adrian Broz, Gregory J. Retallack, Toby M. Maxwell, Lucas C.R. Silva

Biology Faculty Publications and Presentations

The drying power of air, or vapour pressure deficit (VPD), is an important measurement of potential plant stress and productivity. Estimates of VPD values of the past are integral for understanding the link between rising modern atmospheric carbon dioxide (pCO2) and global water balance. A geological record of VPD is needed for paleoclimate studies of past greenhouse spikes which attempt to constrain future climate, but at present there are few quantitative atmospheric moisture proxies that can be applied to fossil material. Here we show that VPD leaves a permanent record in the slope (S) of least-squares …

Mithramycin And Analogs For Overcoming Cisplatin Resistance In Ovarian Cancer, David Schweer, J. Robert Mccorkle, Jurgen Rohr, Oleg V. Tsodikov, Frederick R. Ueland, Jill M. Kolesar

Obstetrics and Gynecology Faculty Publications

Ovarian cancer is a highly deadly malignancy in which recurrence is considered incurable. Resistance to platinum-based chemotherapy bodes a particularly abysmal prognosis, underscoring the need for novel therapeutic agents and strategies. The use of mithramycin, an antineoplastic antibiotic, has been previously limited by its narrow therapeutic window. Recent advances in semisynthetic methods have led to mithramycin analogs with improved pharmacological profiles. Mithramycin inhibits the activity of the transcription factor Sp1, which is closely linked with ovarian tumorigenesis and platinum-resistance. This article summarizes recent clinical developments related to mithramycin and postulates a role for the use of mithramycin, or its analog, …

A term neonate is born with the ability to suck; this neuronal network is already formed and functional by 28 weeks gestational age and continues to evolve into adulthood. Because of the necessity of acquiring nutrition, the complexity of the neuronal network needed to suck, and neuroplasticity in infancy, the skill of sucking has the unique ability to give insight into areas of the brain that may be damaged either during or before birth. Interpretation of the behaviors during sucking shows promise in guiding therapies and how to potentially repair the damage early in life, when neuroplasticity is high. Sucking …

There has long been an interest in understanding how the hazards from spaceflight may trigger or exacerbate human diseases. With the goal of advancing our knowledge on physiological changes during space travel, NASA GeneLab provides an open-source repository of multi-omics data from real and simulated spaceflight studies. Alone, this data enables identification of biological changes during spaceflight, but cannot infer how that may impact an astronaut at the phenotypic level. To bridge this gap, Scalable Precision Medicine Oriented Knowledge Engine (SPOKE), a heterogeneous knowledge graph connecting biological and clinical data from over 30 databases, was used in combination with GeneLab …

Head and neck squamous cell carcinoma (HNSCC) is a very aggressive disease with a poor prognosis for advanced-stage tumors. Recent clinical, genomic, and cellular studies have revealed the highly heterogeneous and immunosuppressive nature of HNSCC. Despite significant advances in multimodal therapeutic interventions, failure to cure and recurrence are common and account for most deaths. It is becoming increasingly apparent that tumor microenvironment (TME) plays a critical role in HNSCC tumorigenesis, promotes the evolution of aggressive tumors and resistance to therapy, and thereby adversely affects the prognosis. A complete understanding of the TME factors, together with the highly complex tumor-stromal interactions, …

Coral reefs are keystone coastal ecosystems that are at risk of exposure to petroleum from a range of sources, and are one of the highest valued natural resources for protection in Net Environmental Benefit Analysis (NEBA) in oil spill response. Previous research evaluating dissolved hydrocarbon impacts to corals reflected no clear characterization of sensitivity, representing an important knowledge gap in oil spill preparedness related to the potential impact of oil spills to the coral animal and its photosymbiont zooxanthellae. This research addresses this gap, using a standardized toxicity protocol to evaluate effects of a dissolved reference hydrocarbon on scleractinian corals. …
