Cyclophilin D (CypD) has been shown to play a critical role in mitochondrial permeability transition pore (mPTP) opening and the subsequent cell death cascade. Studies consistently demonstrate that mitochondrial dysfunction, including mitochondrial calcium overload and mPTP opening, is essential to the pathobiology of cell death after a traumatic brain injury (TBI). CypD inhibitors, such as cyclosporin A (CsA) or NIM811, administered following TBI, are neuroprotective and quell neurological deficits. However, some pharmacological inhibitors of CypD have multiple biological targets and, as such, do not directly implicate a role for CypD in arbitrating cell death after TBI. Here, we reviewed the …

Introduction

This study assessed the hypothesis that circulating human amylin (amyloid‐forming) cross‐seeds with amyloid beta (Aβ) in early Alzheimer's disease (AD).

Methods

Evidence of amylin‐AD pathology interaction was tested in brains of 31 familial AD mutation carriers and 20 cognitively unaffected individuals, in cerebrospinal fluid (CSF) (98 diseased and 117 control samples) and in genetic databases. For functional testing, we genetically manipulated amylin secretion in APP/PS1 and non‐APP/PS1 rats.

Results

Amylin‐Aβ cross‐seeding was identified in AD brains. High CSF amylin levels were associated with decreased CSF Aβ42 concentrations. AD risk and amylin gene are not correlated. Suppressed amylin secretion protected …

Molecular Dynamic Simulation Of Protein Devices And The Parameterization Of Azides And Alkynes For Use In Unnatural Amino Acid Models, Addison Kyle Smith Jan 2021

Molecular Dynamic Simulation Of Protein Devices And The Parameterization Of Azides And Alkynes For Use In Unnatural Amino Acid Models, Addison Kyle Smith

Theses and Dissertations

Proteins that have been modified by attaching them to a surface or to a polyethylene glycol (PEG) molecule can see many uses in therapeutics and diagnostics -- these unique proteins are called protein devices. Current techniques can perform these functionalizations at a specific residue on the protein, but what remains is identifying what happens to protein structure when mutated, and where to perform the attachment. Both of these issues can be examined using molecular dynamic (MD) simulations. Currently, simulations of the unnatural amino acid (uAA) mutations necessary for protein device functionalization cannot be executed, and full-protein screens of all possible …

Experimental Pharmacotherapies In Models Of Alcohol Addiction, Mousumi Akter Sumi Jan 2021

Experimental Pharmacotherapies In Models Of Alcohol Addiction, Mousumi Akter Sumi

Theses and Dissertations

The USA faces $220 billion economic loss and eighty thousand deaths per year due to alcohol abuse while affecting more than 15 million people, making it the third largest life-style related cause of death. The U.S. FDA has approved four medications namely, disulfiram, acamprosate, oral naltrexone, and injectable long-acting naltrexone. These existing drugs are trashed with side effects, have a low success rate, indicating a demand for new potential drugs. We studied the effects of the CB1 negative allosteric modulator, PSNCBAM-1, and the dopamine D4 receptor antagonist, L-745,870 in mouse models of alcohol addiction. PSNCBAM-1 did not significantly reduce CPP …

Department of Biochemistry and Molecular Biology Faculty Papers

The tumor suppressor p53 integrates stress response pathways by selectively engaging one of several potential transcriptomes, thereby triggering cell fate decisions (e.g., cell cycle arrest, apoptosis). Foundational to this process is the binding of tetrameric p53 to 20-bp response elements (REs) in the genome (RRRCWWGYYYN0-13RRRCWWGYYY). In general, REs at cell cycle arrest targets (e.g. p21) are of higher affinity than those at apoptosis targets (e.g., BAX). However, the RE sequence code underlying selectivity remains undeciphered. Here, we identify molecular mechanisms mediating p53 binding to high- and low-affinity REs by showing that key determinants of the code are embedded …

Philosophy And Physics Meet In Quantum World, Liuxiang Hao Jan 2021

Philosophy And Physics Meet In Quantum World, Liuxiang Hao

Bulletin of Chinese Academy of Sciences (Chinese Version)

On the basis of discussion about the ontological status of quantum state in quantum mechanics, the properties of particle in the quantum theory of fields and the problem of time in the canonical quantum gravity, the author argues that the hard core of any physical theory consists of some metaphysical assumptions. The author points out that from the perspective of the history and philosophy of science, the relationship between philosophy and physics is in some way analogous of that between mathematics and physics, so the reunion of philosophy and physics will help us to clarify and even solve those fundamental …

Several Key Points In Territorial Ecological Restoration, Bojie Fu Jan 2021

Several Key Points In Territorial Ecological Restoration, Bojie Fu

Bulletin of Chinese Academy of Sciences (Chinese Version)

The coupled Human-Natural System thinking is considered as an important theoretical basis for territorial ecological restoration. The relationships between ecological space, production space, and living space show three evolution stages in territorial governance: coordinated arrangement, systematic governance, and human-nature harmony. In this regard, coupled Human-Natural System research is necessary to set the perspectives of coupling indicators, coupling hierarchies, coupling effects and coupling models to identify the primary evaluation indicators, delineate hierarchical nested regions, decouple the spatio-temporal relationship of socialecological effects, and develop human-natural coupling models, so as to systematically support the territorial ecological restoration and facilitate the national and regional …

Emerging outbreak of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) infection is a major threat to public health. The morbidity is increasing due to lack of SARS-CoV-2 specific drugs. Herein, we have identified potential drugs that target the 3-chymotrypsin like protease (3CLpro), the main protease that is pivotal for the replication of SARS-CoV-2. Computational molecular modeling was used to screen 3987 FDA approved drugs, and 47 drugs were selected to study their inhibitory effects on SARS-CoV-2 specific 3CLpro enzyme in vitro. Our results indicate that boceprevir, ombitasvir, paritaprevir, tipranavir, ivermectin, and micafungin exhibited inhibitory effect towards 3CLpro enzymatic activity. The 100 …
