Nationally and internationally, waste generation is increasing and along with it negative environmental impacts. Many people think waste ends at disposal; however, environmental impacts have already begun. Unfortunately, 50% of waste ends up in a landfill which is the third largest source of human related methane emissions and a leading cause of global climate change. An integrated biorefinery approach based on subcritical hydrothermal processes was developed to maximize utilization of biomass components by producing multiple products. Biomass from various sources like agricultural waste (lignocellulose), food waste (spent yeast), and dedicated cultivation (micro-algae) was used in this research.

Corn stover (agricultural waste) was hydrothermally pretreated to remove lignin and xylan while preserving most of the glucan. The pretreated corn stover was utilized to produce levulinic acid (thermochemically) and fermentable sugars (biochemically). Secondary math teachers faced new experiences and struggles this past virtual school year due to the global COVID-19 pandemic. Nine secondary math teachers from a school division in southeastern Virginia participated in this qualitative single case study. Data collection included individual semi-structured interviews and technology integration artifacts. Low molecular weight gelators (LMWGs) are small molecules that self-assemble in appropriate solvents to form three dimensional networks that immobilize the solvent, creating a supramolecular gel. The self-assembly of LMWGs occurs through non-covalent interactions such as hydrogen bonding, aromatic interactions, donor-acceptor interactions, Van der Waals interactions, hydrophobic forces, halogen bonding, etc. Due to self-assembly occurring through reversible non-covalent interactions, supramolecular gels can undergo a gel to solution transformation. Our lab has been studying the synthesis and applications of different classes of glycoconjugates ranging from macrocycles to sugar heterocycle derivatives. Due to the importance of these compounds, our research is based on development of synthetic methods for various carbohydrate derivatives. This dissertation includes introduction and overview of projects, followed by the synthesis and studies of several different series of glycomacrolactones. Emotion regulation is a transdiagnostic factor that functions as a crucial contributor to individual well-being and social functioning. Research seeking to identify adaptive or maladaptive strategies of emotion regulation has often focused on individual strategies, though emerging evidence suggests that what constitutes “adaptive” regulation is often context-dependent. Having a large repertoire of emotion regulation strategies that one is willing to use at different rates has been termed “emotion regulation variability,” and has been linked to positive individual outcomes. Upwelling events are known to support blooms of phytoplankton, important primary producers at the base of the oceanic food web. Phytoplankton community structure changes in response to upwelling support higher trophic level growth and increased efficiency of carbon export from the euphotic zone. While these events occur globally, this study examined upwelling in coastal regions, where alongshore winds can drive Ekman transport and upwelling of deeper waters. The two upwelling regimes examined were the California Current System and the Alaskan Beaufort Sea. The networked society provides an extremely complex environment where we are free to swim, and to sink. In this thesis, I used different materials and forms to visualize this concept. The final work is a series of human faces floating from the water surface with steady gaze. One method of promoting gender equality that has gained popularity in recent years involves revising the fairy tales, primarily the ones compiled and revised by Charles Perrault, Hans Christian Anderson, and the Grimm brothers, to create versions in which the female characters have agency and purpose outside of furthering patriarchal gender ideals. Teachers of the visually impaired (TVIs) regularly present tactile materials (tactile graphics, 3D models, and real objects) to students with vision impairments. Researchers have been increasingly interested in designing tools to support the use of tactile materials, but we still lack an in-depth understanding of how tactile materials are created and used in practice today. To address this gap, we conducted interviews with 21 TVIs and a 3-week diary study with eight of them. There is a growing gap between the racial demographics of public-school teachers and their students. Teachers with demographic backgrounds similar to a school’s student population are often successful, particularly in Title 1 schools with high proportions of minority students; however, teacher turnover is high. The students in Texas public schools are a growing minority-majority population. Changes in the student body racial composition and other school-related variables could influence teacher workforce demographics. To investigate the duration of humoral immune response in convalescent coronavirus disease 2019 (COVID-19) patients, we conduct a 12-month longitudinal study through collecting a total of 1,782 plasma samples from 869 convalescent plasma donors in Wuhan, China and test specific antibody responses. The results show that positive rate of IgG antibody against receptor-binding domain of spike protein (RBD-IgG) to severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in the COVID-19 convalescent plasma donors exceeded 70% for 12 months post diagnosis. The level of RBD-IgG decreases with time, with the titer stabilizing at 64.3% of the initial level by the 9th month. …

Obesity is primarily a consequence of consuming calories beyond energetic requirements, but underpinning drivers have not been fully defined. 5-Hydroxytryptamine (5-HT) neurons in the dorsal Raphe nucleus (5-HTDRN) regulate different types of feeding behavior, such as eating to cope with hunger or for pleasure. Here, we observed that activation of 5-HTDRN to hypothalamic arcuate nucleus (5-HTDRN → ARH) projections inhibits food intake driven by hunger via actions at ARH 5-HT2C and 5-HT1B receptors, whereas activation of 5-HTDRN to ventral tegmental area (5-HTDRN → VTA) projections inhibits non-hunger-driven feeding via actions at 5-HT2C receptors. Despite good adherence to supervised endurance exercise training (EET), some individuals experience no or little improvement in peripheral insulin sensitivity. The genetic and molecular mechanisms underlying this phenomenon are currently not understood. By investigating genome-wide variants associated with baseline and exercise-induced changes (∆) in insulin sensitivity index (Si) in healthy volunteers, we have identified novel candidate genes whose mouse knockouts phenotypes were consistent with a causative effect on Si. An integrative analysis of functional genomic and transcriptomic profiles suggests genetic variants have an aggregate effect on baseline Si and ∆Si, focused around cholinergic signalling, including downstream calcium and chemokine signalling. …

This article makes an argument for the value of both replicable research and replication research in writing center studies. In their discussion of replicability, the authors argue that writing about empirical research so that this research can be replicated will improve the quality of communication in writing center studies whether or not replication studies are subsequently undertaken. The authors further provide for researchers specific guidance on how to create replicable studies, focusing on best practices for describing data sets and sampling, sharing surveys and interview protocols, detailing coding efforts, establishing infrastructure to share data sets, and writing about statistics. Background: The Oncotype DX breast recurrence score has been introduced more than a decade ago to aid physicians in determining the need for systemic adjuvant chemotherapy in patients with early-stage, estrogen receptor (ER)+, lymph node-negative breast cancer. Methods: In this study, we utilized data from The Surveillance, Epidemiology, and End Results (SEER) Program to investigate temporal trends in Oncotype DX usage among US breast cancer patients in the first decade after the introduction of the Oncotype DX assay. Riparian ecosystems are vital to the landscape, providing critical services including water filtration and purification, flood and erosion control, carbon sequestration, biodiversity support, and aesthetic value. Bottomland hardwood forests, however, are threatened by invasive species, land loss/conversion, inconsistent or absence of harvesting disturbances, and altered hydrological patterns, leading to reduced success of desired, native species. The Trump Administration’s response to the COVID-19 pandemic is a stark reminder that poorly designed government can be a matter of life and death. This article explains how the Administration’s careless and delayed response to the crisis was made immeasurably worse by its confused and confusing reallocation of authority to perform or supervise tasks essential to reducing the virus’s ravages.

Administrative Agencies often rely on guidance documents to carry out their statutory mandate. Over the past few decades, the Food and Drug Administration (FDA) has been criticized for using soft law guidance documents to exercise powers beyond those authorized by Congress. Since attacks on the use of guidance documents persist and agencies need soft law to respond quickly and flexibly to rapid technological growth, it is essential to develop a solution that preserves this crucial regulatory mechanism and prevents its abuse. Organelle-specific autophagy directs degradation of eukaryotic organelles under certain conditions. Like other organelles, peroxisomes are subject to autophagic turnover at lysosomes. However, peroxisome autophagy (pexophagy) has yet to be analyzed in a live-animal system, limiting knowledge on its regulation during an animal's life. Here, we generated a tandem-fluorophore reporter that enabled real-time tracking of pexophagy in live Caenorhabditis elegans. We observed that pexophagy occurred at a population of non-canonical, tubular lysosomes specifically during starvation and aging. Remarkably, in these contexts, tubular lysosomes were the predominant type of lysosome in the intestine, transforming from vesicles. Though we found that peroxisomes were …
