The interaction of monoclonal antibodies (mAbs) with air/water interfaces plays a crucial role in their overall stability in solution. We aim to understand this behavior using pendant bubble measurements to track the dynamic tension reduction and x-ray reflectivity to obtain the electron density profiles (EDPs) at the surface. Native immunoglobulin G mAb is a rigid molecule with a flat, “Y” shape, and simulated EDPs are obtained by rotating a homology construct at the surface. Comparing simulations with experimental EDPs, we obtain surface orientation probability maps showing mAbs transition from flat-on Y-shape configurations to side-on or end-on configurations with increasing concentration. …

Many studies have examined the effects of racial and socioeconomic differences on health-related disparities involving obesity but, few have examined the mental and physical well-being intersection. Obesity is a disease that affects many Americans and has become an epidemic affecting the American population. Obesity, especially morbid obesity, is a risk factor for many other obesity-related diseases such as diabetes, cardiovascular diseases, hypertension, and more. Using the 2018 Healthcare Avoidance Study, differences were found between two racial groups, Caucasian and African-American women. This project aimed to examine the effectiveness of a preoperative preparation program (PPP) in decreasing parental anxiety. When children needed anesthesia to get outpatient procedures completed, the accompanying parents received verbal education about the process on the day of the procedure and were assisted by a support person during and after the child's induction anesthesia. Parental anxiety was measured using the State-Trait Anxiety Inventory (STAI) tool before preoperative preparation and after the induction of anesthesia to assess whether the PPP improved parents' anxiety levels. The parents' decreased anxiety score in post-intervention would suggest that the PPP can be effectively and …

Elevated serum levels of trimethylamine N-oxide (TMAO) were first associated with increased risk of cardiovascular disease (CVD) 10 years ago. Research has since defined that serum TMAO accumulation is controlled by the diet-microbiome-liver-kidney axis. Choline related nutrients are consumed in excess during over-nutrition from a Western diet. The resultant elevated serum TMAO is investigated across various chronic metabolic diseases and many tissue types. While TMAO is most clearly linked to CVD mechanisms in vascular tissue, its molecular effects on metabolic tissues are unclear. Background: The p53-up-regulated modulator of apoptosis (PUMA) protein is a pro-apoptotic, BH3-only member of the BCL2 family of effector proteins responsible for promoting organized cell death. PUMA is required for resolution of pneumococcal pneumonia in mice, as mice deficient of PUMA exhibit greater numbers of S. pneumoniae CFU within tissues and higher mortality rates than observed in Puma+/+ mice. Methods: Puma+/+ and Puma-/- mice were intranasally challenged with TIGR4 pneumococcus and sacrificed 24 h post-infection. With the high mortality rate of cardiovascular disease, it is important to study the early signs. The early detection of cardiovascular disease can lead to saved lives. Currently the most prevalent detection methods are the Framingham Risk Score and the carotid intima media thickness, both of which are insufficient. The necessary tool for early detection requires a uniform quantification system. Compound and cascading hydroclimatic extreme events have garnered much attention in recent studies. The combined effects of interconnected extremes can cause widespread damage, with a higher potential impact than individual extremes. Both anthropogenic warming and natural climate variability affect these extremes, which is why detecting past extreme events, understanding the underlying mechanisms, and assessing their future impacts can aid in mitigation efforts to reduce their overall impact. In this work, a pressure sensing system was designed and fabricated by developing a highly sensitive cone-structured pressure sensor with a custom-built software for physiological monitoring applications. A novel highly sensitive cone structured porous polydimethylsiloxane (PDMS) based pressure sensor capable of detecting very low-pressure ranges was developed for respiration monitoring. The pressure sensor was fabricated using a master mold, a hybrid-structured dielectric layer, and fabric-based electrodes. The master mold with inverted cone structures was created using a rapid and precise three-dimensional (3D) printing technique. During the Postmedieval period, children, especially low socioeconomic status (SES) individuals, participated in the workforce. By examining the demographic distributions and paleopathological profiles of juvenile skeletal remains from high SES (St. Bride’s Crypt and Chelsea Old Church) and low SES (St. Bride’s Lower and Crossbones) Postmedieval (1700-1850 CE) London cemeteries, we can better understand how SES impacted the biological health and mortality of children. Unfortunately, model systems that accurately mimic human oncology - a requirement for the development of more effective therapies for these patients - remain elusive. Future human space missions to Mars and beyond may be realized for different research, economic, political or survival reasons. Since space remains a hazardous environment for humans, space exploration and exploitation requires the development and deployment of effective countermeasures. In this paper, we discuss prospects for human enhancement by gene editing, synthetic biology, or implants, for the purposes of future space missions. We argue that there are good reasons to consider such options, and that ethical arguments can be made in favor of human enhancement to enable long-term space exploration.

In this work, particle image velocimetry (PIV) was used for fluid flow visualization in both continuous and segmented-flow microfluidics. Droplet microfluidics is known for its precise and consistent volume dispersion between microliters and picoliters in volume for application ranging from molecular synthesis, drug discover, and diagnostics. Kratom, derived from the plant Mitragyna speciosa, is reported to produce stimulant-like effects at low doses and opiate-like effects at high doses. Mitragynine (MG) and 7-hydroxymitragynine (7-HMG) are the major psychoactive constituents of kratom, but the neural mechanisms of these alkaloids are not clear. As of 2016, an estimated 550,000 new and 200,000 recurrent episodes of myocardial infarction (MI) occur each year in the United States was. Infarction is often distinguished by a considerable decrease in wall thickness and an increase in heart size or left ventricular volume. Lead is a pervasive and ongoing problem in the landscape, especially in urban and suburban areas where historic use of tetraethyl lead gasoline and leaded paint has increased background amounts to dangerous levels. One method to solve this problem is the use of hyperaccumulator plant species, capable of concentrating over 1000 ppm Pb into their tissues. This is a slow process, even with hyperaccumulators, so the use of amendments to improve Pb solubility has gained popularity, though with mixed results. The development of induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs) has been a critical in vitro advance in the study of patient-specific physiology, pathophysiology, and pharmacology. We designed a new deep learning multitask network approach intended to address the low throughput, high variability, and immature phenotype of the iPSC-CM platform. The rationale for combining translation and classification tasks is because the most likely application of the deep learning technology we describe here is to translate iPSC-CMs following application of a perturbation.
