Establishing A Mechanistic Link Between Disturbed Flow And Aneurysm Formation In A 3d Cerebral Bifurcation Model, Nesrine Bouhrira

Theses and Dissertations

The effect of disturbed flow profiles on the endothelium have been studied extensively in systemic vasculature, but less is known about the response of the blood-brain barrier (BBB) to these flow regimes. Here we investigate the effect of steady and pulsatile disturbed flow on the integrity of the BBB using a three-dimensional, perfusable bifurcation model consisting of a co-culture of endothelial cells with mural and glial cells. Experimental flow patterns predicted by computational fluid dynamics mimic in vivo flow regimes, specifically the presence of a recirculation zone immediately downstream of the bifurcation reveal periodic changes in the instantaneous shear stress …

We currently find ourselves in the midst of a global coronavirus disease 2019 (COVID-19) pandemic, caused by the highly infectious novel coronavirus, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Here, we discuss aspects of SARS-CoV-2 biology and pathology and how these might interact with the circadian clock of the host. We further focus on the severe manifestation of the illness, leading to hospitalization in an intensive care unit. The most common severe complications of COVID-19 relate to clock-regulated human physiology. We speculate on how the pandemic might be used to gain insights on the circadian clock but, more importantly, on …

PSNCBAM-1 did not significantly reduce CPP …

Overstraining Human Nature In The Nicomachean Ethics, Douglass Reed Jan 2021

Overstraining Human Nature In The Nicomachean Ethics, Douglass Reed

Philosophy Faculty Publications

In this paper, I investigate Aristotle’s claim in Nicomachean Ethics III.1 about situations that “overstrain human nature.” By setting out and answering several interpretative questions about such situations, I offer a comprehensive interpretation of this passage. I argue that in (at least some of) these cases, the agent voluntarily does something wrong, even though there is a right action available. Furthermore, I argue that Aristotle would think it is possible for a rare agent to perform the right action in (at least some of) these cases, overcoming the limitations of human nature by identifying with the divine part of the …

There has long been an interest in understanding how the hazards from spaceflight may trigger or exacerbate human diseases. With the goal of advancing our knowledge on physiological changes during space travel, NASA GeneLab provides an open-source repository of multi-omics data from real and simulated spaceflight studies. Alone, this data enables identification of biological changes during spaceflight, but cannot infer how that may impact an astronaut at the phenotypic level. To bridge this gap, Scalable Precision Medicine Oriented Knowledge Engine (SPOKE), a heterogeneous knowledge graph connecting biological and clinical data from over 30 databases, was used in combination with GeneLab …

Role Of Na/K-Atpase Non-Enzymatic Signaling In Renal Proximal Tubule Sodium Transport, Shreya Tapan Mukherji Jan 2021

Role Of Na/K-Atpase Non-Enzymatic Signaling In Renal Proximal Tubule Sodium Transport, Shreya Tapan Mukherji

Theses, Dissertations and Capstones

The renal proximal tubule (RPT) is the center stage for renal handling of Na+ , a potent determinant of systemic Na+ and volume homeostasis in health and disease. In the RPT, through its classic ion-pumping function, Na + /K+ -ATPase (NKA) generates the Na+ gradient that drives apical Na+ absorption, mostly through Na+ /H+ exchanger (NHE3). In contrast, pharmacological evidence suggests that the activation of the NKA non-enzymatic signaling function, through Src and other signaling partners, triggers a cellular redistribution of NKA and NHE3 that decreases transcellular Na+ flux in cultured RPT cells. However, the physiological relevance of the nonenzymatic …

Adaptive Two-Stage Edge-Centric Architecture For Deeply-Learned Embedded Real-Time Target Classification In Aerospace Sense-And-Avoidance Applications, Nicholas A. Speranza Jan 2021

Adaptive Two-Stage Edge-Centric Architecture For Deeply-Learned Embedded Real-Time Target Classification In Aerospace Sense-And-Avoidance Applications, Nicholas A. Speranza

Browse all Theses and Dissertations

With the growing number of Unmanned Aircraft Systems, current network-centric architectures present limitations in meeting real-time and time-critical requirements. Current methods utilizing centralized off-platform processing have inherent energy inefficiencies, scalability challenges, performance concerns, and cyber vulnerabilities. In this dissertation, an adaptive, two-stage, energy-efficient, edge-centric architecture is proposed to address these limitations. A novel, edge-centric Sense-and-Avoidance architecture framework is presented, and a corresponding prototype is developed using commercial hardware to validate the proposed architecture. Instead of a network-centric approach, processing is distributed at the logical edge of the sensors, and organized as Detection and Classification Subsystems. Classical machine vision algorithms are …

The Role Of Nutrient Availability In Therapeutic Response Of Leukemia, Rohiverth Guarecuco Jr. Jan 2021

The Role Of Nutrient Availability In Therapeutic Response Of Leukemia, Rohiverth Guarecuco Jr.

Student Theses and Dissertations

Tumor environment influences the response to anti-cancer therapy, but which extracellular nutrients impact drug sensitivity is largely unknown. In this work, we used functional genomics to identify metabolic modifiers of the response to L-asparaginase (ASNase), a therapy that depletes plasma asparagine and targets leukemic cells with insufficient asparagine synthesis. Our approach revealed thiamine pyrophosphate kinase 1 (TPK1), which converts vitamin B1 (thiamine) into the cofactor thiamine pyrophosphate (TPP), as a metabolic dependency under ASNase treatment. In glutamine-anaplerotic leukemia cells, we found that TPP availability enables asparagine synthesis from extracellular glutamine. Mechanistically, TPP is critical for the activity of alpha-ketoglutarate dehydrogenase …

Transcriptional Regulation Of The Metabolic Response To Therapy In Leukemia, Robert Thomas Williams Jan 2021

Transcriptional Regulation Of The Metabolic Response To Therapy In Leukemia, Robert Thomas Williams

Student Theses and Dissertations

Cancer cells are under constant stress due to their uncontrolled growth, oncogenic signaling, and the metabolic insufficiencies of their microenvironments. Under various stresses, cells activate the integrated stress response (ISR), a transcriptional program to restore cellular homeostasis. Activating transcription factor 4 (ATF4) acts as the master transcriptional regulator of the ISR by promoting the transcription of genes that mitigate stress or promote cell death if the stress remains unresolved. Despite being the common mediator of various stress response and metabolic pathways, ATF4 generates tailored transcriptional outputs to distinct cellular stresses by cooperating with other transcriptional machinery. The precise mechanisms by …

The Morass Of Misconceptions: How Unjustified Beliefs Influence Pedagogy And Learning, Morgan Mcafee, Bobby Hoffman

International Journal for the Scholarship of Teaching & Learning

When teachers harbor misconceptions or unjustified beliefs about teaching, learning, and academic motivation, the pedagogical consequences can be severe. It is likely these teachers will unintentionally perpetuate such false beliefs upon students through ineffective teaching strategies or misinterpretations of learning science. Misconceptions among K-12 teachers are particularly deleterious due to the substantial influence teacher beliefs exert upon curriculum development, pedagogy, and the construction of effective learning environments. Prior research has explicated the prevalence of erroneous beliefs about general psychology and neuroscience among various populations but has rarely examined teachers’ misconceptions about pertinent topics in educational psychology. Consequently, this review highlights …
