Polymethoxyflavones As Food Factors For The Management Of Inflammatory Diseases, C.-T. Ho, M.-H. Pan, C.-S. Lai, S. Li
Journal of Food and Drug Analysis
Recent researches have expanded the concept that inflammation is a critical component of human diseases. The possible mechanisms by which inflammation contribute to various conditions include increase of reactive species and inflammatory cytokines and growth factors leading to alteration of biomolecules, induction of genetic change, cellular dysfunction and enhancement of transformation or proliferation. Thus, targeting the inflammation is potential strategy for prevention of chronic diseases. Numerous epidemiological and laboratory studies suggest that polymethoxyflavones (PMFs) exhibit a broad spectrum of biological activities. Recently, we isolated and identified hydroxylated PMFs from citrus peels and have investigated their biological activities, including anti-inflammation and …

The Selectivity And Impact Of Polyphenols-Protein Kinases Interactions For Chemoprevention, D.-X. Hou, T. Kumamoto, H. Ishida, A. Hisanaga
Journal of Food and Drug Analysis
Phosphorylation of target proteins by protein kinases leads to the activation of cellular signaling pathways, which induce a great number of biological processes including cell growth, differentiation, development, and death. Although the activities of protein kinases are stringently regulated in homeostasis system, protein kinases can be deregulated under pathological conditions, leading to perturbation of protein kinase-mediated cell signaling pathways and resulting in various disorders including cancer, diabetes, and inflammation. The data from the Human Genome Project has revealed that 518 protein kinases are encoded in the human genome, thereby creating an enormous repertoire of potential targets for chemoprevention. Polyphenols, as …

Journal of Food and Drug Analysis
Resveratrol is one of the best known polyphenol. While its effect on endothelial blood vessel cells, cancer cells, inflammatory processes and neurodegenerative events is well documented, only little is established on the metabolic implication of this phytophenol, particularly on skeletal muscle cells. Here, we report the effect of resveratrol on mouse skeletal muscle cells (C2C12) by measurement of cell proliferation, expression of metabolic genes and their transcription factor dependency, and modulation of non-coding microRNA expression. Resveratrol slightly decreases cell proliferation, while it up regulates PGC-1α a PPAR transcription factor co-activator involved in the control of metabolic genes. It also modulates …

Food for Specified Health Uses (FOSHU) is an official functional food approved by the Consumer Affairs Agency of Japan. The number of FOSHU items reached almost 970 as of October, 2011. It should be emphasized, however, that more than 70% of the current FOSHU products function in the gastrointestinal tract. They improve intestinal microflora or regulate nutrient absorption, thereby reducing the risks of metabolic syndrome. In addition to the gut-modulating FOSHU products already approved, many attempts have been made to develop new functional foods that regulate other intestinal functions. Modulation of the detoxification system is beneficial to reinforce the gut …

Chronic inflammation plays critical roles in tumor initiation and progression. Inflammatory tumor microenvironment populating tumor cells, inflammatory cells, fibroblasts, and adipocytes and a web of signaling molecules are crucial in the promotion and progression of tumors. Numerous studies have identified transcription factors such as nuclear factor-kappa B, hypoxia-inducible factor-1α, and signal transducer and activator of transcription 3 as key modulators in driving inflammation to cancers. Inflammatory cells increase the levels of cytokines that activate these transcription factors to stimulate diverse protumorigenic processes. Additionally, these transcription factors induce production of chemokines that attract more inflammatory cells to maintain tumor-related inflammation. Although …

Journal of Food and Drug Analysis
TGR5 is a member of the G protein-coupled receptor family and is activated by bile acids. TGR5 is thought to be a promising food factor target for preventing metabolic diseases because the activation of TGR5 prevents obesity and hyperglycemia in mice fed a high-fat diet. In the present study, we established an evaluation system for the TGR5 ligand activity, and identified a limonoid nomilin as an activator of TGR5 from among approximate 150 purified food compounds. Unlike bile acids, nomilin did not exhibit the farnesoid X receptor ligand activity. Although the nomilin derivative obacunone was capable of activating TGR5, limonin …

Detection Of Copper Ions In Liquid Foods And Beverages Based On An Enzymatic Method, J.-I. Yang, D.-B. Yeh, J.-M. Kuo, B.S. Pan, G.-C. Lee, Y.-H. Liu, Y.-J. Lai
Journal of Food and Drug Analysis
An economic, simple and sensitive enzymatic method for the determination of copper in aqueous samples was proposed in this study. By removal of copper, superoxide dismutase (SOD) becomes apoenzyme SOD (apo-SOD) that has no SOD activity. The amount of copper added can be estimated by the recovery of SOD activity from reconstituted apo-SOD. The concentration of copper in an aqueous sample can thus be determined. A linear relationship (R2 = 0.998) between the concentrations of copper, ranging from 20 to 80 ppb, and the recovered SOD activities was observed. The detection limit for copper in the proposed assay was 20 …

Journal of Food and Drug Analysis
Photodynamic therapy (PDT) and photodynamic inactivation (PDI) are technologies that utilize visible light and photosensitizers (PS) to inactivate cells. PDT is currently in use for the treatment of several types of tumors. Although cancer has been successfully treated with PS and light, antimicrobial PDI is emerging as a new treatment modality for bacterial infections due to its effectiveness and less likelihood of inducing bacterial resistance. Resistance to therapy is in part due to the ability of the organisms to form a biofilm, which provides a microenvironment that protects the microorganism from antibiotics and attack by the host's immune system. PR is a rich source of isoflavones including puerarin, daidzin and daidzein. This study was aimed to compare the urinary kinetics of isoflavones between traditional decoction (TD) and concentrated powders (CP) of PR in healthy man. Ten male volunteers were given two dosage forms of PR in a crossover design and their urine was collected at specific intervals until 36 h. Urine samples were assayed by an HPLC method before and after hydrolysis with β-glucuronidase and …

Aloe vera whole leaf extracts are incorporated into a wide variety of topically applied commercial products. Aloe vera whole leaf extracts contain anthraquinones, which can generate reactive oxygen species (ROS) upon UVA light irradiation. In this study, the phototoxicity of two Aloe vera whole leaf extracts, Aloe A and Aloe B, were examined. The Aloe B extract was prepared by treating the Aloe A extract with activated carbon (1%, wt/wt) and filtration to remove the Aloe latex components, including the anthraquinones. UVB irradiation of the Aloe vera whole leaf extracts induced lipid peroxidation in the presence of a lipid, methyl …
