© 2020 by the authors. Licensee MDPI, Basel, Switzerland. The growing tips of plants grow sterile; therefore, disease-free plants can be generated from them. How plants safeguard growing apices from pathogen infection is still a mystery. The shoot apical meristem (SAM) is one of the three stem cells niches that give rise to the above ground plant organs. This is very well explored; however, how signaling networks orchestrate immune responses against pathogen infections in the SAM remains unclear. To reconstruct a transcriptional framework of the differentially expressed genes (DEGs) pertaining to various SAM cellular populations, we acquired large-scale transcriptome datasets …

The History Of Early Modern Medicine In New Spain, El Primero Sueño, And Poet Sor Juana Inés De La Cruz, Daniel Hughes

Grand Valley Journal of History

This essay analyzes poetry and other writing by Sor Juana Inés de la Cruz in the context of themes from Greco-Roman antiquity and the historical development of medicine in seventeenth century New Spain, now Mexico. Sor Juana’s El Primero Sueño, a Spanish language poetic silva, exhibits copious references to writers from classical antiquity, including Aristotle and Ovid. Establishing a context steeped in ideas from Greco-Roman antiquity, Sor Juana invokes the medical and philosophical legacy of foundational physician Galen of Pergamon. She also expands upon his ideas into the human anatomical realm, reflecting the increased early modern prominence of …

Simulation And Analysis Of The Carrying Capacity For Road Networks Using A Grid-Based Approach, Junqing Shi, Linwu Chen, Fengxiang Qiao, Lei Yu, Qing Li, Guilian Fan

Faculty Publications

The number of vehicles that a road network can carry is limited. When the limit is exceeded, the network system is not able to function effectively. In this paper, an updated cellular automaton model for grid networks with all-way stop-controlled intersections is proposed to simulate the network level critical density and carrying capacity under different conditions, which essentially indicates the limit number of vehicles that the network can handle before going into gridlock. In the proposed model, two update rules, including lane-changing and the longitudinal location update, are adopted to describe the vehicle's movements on road segments according to the …

Faculty Research 2020

Approximately 30% of older adults exhibit the neuropathological features of Alzheimer's disease without signs of cognitive impairment. Yet, little is known about the genetic factors that allow these potentially resilient individuals to remain cognitively unimpaired in the face of substantial neuropathology. We performed a large, genome-wide association study (GWAS) of two previously validated metrics of cognitive resilience quantified using a latent variable modelling approach and representing better-than-predicted cognitive performance for a given level of neuropathology. Data were harmonized across 5108 participants from a clinical trial of Alzheimer's disease and three longitudinal cohort studies of cognitive ageing. All analyses were run …

Murmurs Of Revolution: Mythical Subversion In Dostoevsky, Connor Guetersloh Aug 2020

Murmurs Of Revolution: Mythical Subversion In Dostoevsky, Connor Guetersloh

English (MA) Theses

Throughout history, revolutions have been plagued by unpredictability; it is all but impossible to know when cultural systems will be turned on their heads. Is there a common motivator, to predict social unrest bubbling beneath the surface of society? I suggest the development of this motivator is detectable by deconstructing Ferdinand de Saussure’s semiotic patterns within the field of rhetorical mythology. “Mythology,” in the rhetorical and linguistic sense developed by Roland Barthes, is the study of a collective system of thinking we subconsciously subscribe to when interpreting meaning, perpetuated by greater society. The struggle for meaning is split into the …

Lowering of prion protein (PrP) expression in the brain is a genetically validated therapeutic hypothesis in prion disease. We recently showed that antisense oligonucleotide (ASO)-mediated PrP suppression extends survival and delays disease onset in intracerebrally prion-infected mice in both prophylactic and delayed dosing paradigms. Here, we examine the efficacy of this therapeutic approach across diverse paradigms, varying the dose and dosing regimen, prion strain, treatment timepoint, and examining symptomatic, survival, and biomarker readouts. We recapitulate our previous findings with additional PrP-targeting ASOs, and demonstrate therapeutic benefit against four additional prion strains. We demonstrate that

Synthesis Of Alpha-Methylselenocysteine, Its Relevant Analogues, And An Unnatural Glutathione Disulfide Core, Robert J. Wehrle Aug 2020

Synthesis Of Alpha-Methylselenocysteine, Its Relevant Analogues, And An Unnatural Glutathione Disulfide Core, Robert J. Wehrle

Dissertations

Selenoproteins, such as glutathione peroxidase, have gained interest for their ability to act as antioxidants, and their potential to act as anti-cancer agents. Synthesizing and studying selenoproteins can be problematic, however, due to their propensity to degrade from over-oxidation. The degradation from over-oxidation can be avoided by the incorporation of the unnatural amino acid, alpha-methylselenocysteine. A synthesis utilizing methyl malonic esters was used to synthesize protected (R)-alpha-methylselenocysteine efficiently (46% over four steps) and in high enantio-purity (88% enantiomeric excess). Using similar procedures, the (S)-enantiomer was also synthesized as well as a beta-analogue.

The use of enzymes …
