Dysregulated Interferon Response Underlying Severe Covid-19, Leann Lopez, Peter C. Sang, Yun Tian, Yongming Sang

Agricultural and Environmental Sciences Faculty Research

Innate immune interferons (IFNs), including type I and III IFNs, constitute critical antiviral mechanisms. Recent studies reveal that IFN dysregulation is key to determine COVID-19 pathogenesis. Effective IFN stimulation or prophylactic administration of IFNs at the early stage prior to severe COVID-19 may elicit an autonomous antiviral state, restrict the virus infection, and prevent COVID-19 progression. Inborn genetic flaws and autoreactive antibodies that block IFN response have been significantly associated with about 14% of patients with life-threatening COVID-19 pneumonia. In most severe COVID-19 patients without genetic errors in IFN-relevant gene loci, IFN dysregulation is progressively worsened and associated with the …

Thermally processed rhizomes of Atractylodes macrocephala (RAM) have a long history of use in traditional Chinese medicine (TCM) for treating various disorders, and have been an integral part of various traditional drugs and healthcare products. Sjögren syndrome (SS) is an immunologically complex, chronic autoimmune disease targeting lacrimal and salivary glands. Nonobese diabetic (NOD) mice spontaneously develop inflammation of lacrimal and salivary glands with histopathological features similar to SS in humans including focal lymphocytic infiltrates in the affected glands. The innate immune signals driving lymphocytic infiltration of these glands are not well-defined. Here we evaluate the role of Toll-like receptor (TLR) 7 in the development of SS-like manifestations in NOD mice. We created a Tlr7 knockout NOD mouse strain and performed histological and gene expression studies to characterize the effects of TLR7 on autoimmunity development. TLR7 …

Angiotensin-converting enzyme 2 (ACE2) is the cellular receptor for the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) that is engendering the severe coronavirus disease 2019 (COVID-19) pandemic. The spike (S) protein receptor-binding domain (RBD) of SARS-CoV-2 binds to the three sub-domains viz. amino acids (aa) 22-42, aa 79-84, and aa 330-393 of ACE2 on human cells to initiate entry. INTRODUCTION: Delirium is a potentially preventable disorder characterised by acute disturbances in attention and cognition with fluctuating severity. Postoperative delirium is associated with prolonged intensive care unit and hospital stay, cognitive decline and mortality. The development of biomarkers for tracking delirium could potentially aid in the early detection, mitigation and assessment of response to interventions. Because sleep disruption has been posited as a contributor to the development of this syndrome, expression of abnormal electroencephalography (EEG) patterns during sleep and wakefulness may be informative. Lymphatic vasculature is an integral part of digestive, immune and circulatory systems. The homeobox transcription factor PROX1 is necessary for the development of lymphatic vessels, lymphatic valves (LVs) and lymphovenous valves (LVVs). We and others previously reported a feedback loop between PROX1 and vascular endothelial growth factor-C (VEGF-C) signaling. PROX1 promotes the expression of the VEGF-C receptor VEGFR3 in lymphatic endothelial cells (LECs). In turn, VEGF-C signaling maintains PROX1 expression in LECs. Kunyit putih (Kaempferia rotunda L.) merupakan tanaman obat yang bermanfaat dan perlu dibudidayakan lebih luas, namun terkendala oleh penyediaan bibit bermutu yang terbatas sehingga menjadi langka. Perbanyakan tanaman dengan cara konvensional memerlukan bibit bermutu dalam jumlah dan waktu yang lama, sedangkan ketersediaan benih masih terbatas. Perbanyakan secara in vitro dapat mengatasi kelangkaan bibit kunyit putih yang bermutu tinggi dalam jumlah besar dan bebas patogen. Abstract

The present study applies the citation analysis method to analyze Library and Information Science (LIS) doctoral theses submitted to Manipur University during 1989-2017. A total of 2,725 citations appended to 27 LIS doctoral theses examined for bibliographic forms, authorship pattern, and their collaboration level, chronological-wise distribution, the applicability of Bradford's law, rank-wise list, core journal list, country-wise list. The study finds that journals are the most cited resources, followed by books. Single-authored works are found dominant in both journal and book citations. The country-wise distribution of journals reveals that 412 citations (38.52%) were from India, followed by the UK …

It was shown that the swelling of the initially hydrophobic Nafion plate in ordinary water (157 ppm deuterium content) and in deuterium-depleted water (1 ppm deuterium content) in a limited volume cell occurs differently. It turned out that small changes in the deuterium content in water lead to significant differences in the dynamics of swelling of the polymer membrane. For a 175-micron thick Nafion plate, this effect is most pronounced when the distance between the windows is L = 200 microns.

The article presents the results of studying the complexity of the Raman spectra of a-alanine in the crystalline state. It has been shown by ab initio calculations that the change in the Raman spectra is explained by the formation of several types of hydrogen bonds. In Raman spectra, the presence of a hydrogen bond between molecules is manifested in the form of asymmetry and splitting of vibrational bands.

The pure-exploration problem in stochastic multi-armed bandits aims to find one or more arms with the largest (or near largest) means. Examples include finding an ∈-good arm, best-arm identification, top-k arm identification, and finding all arms with means above a specified threshold. However, the problem of finding all ∈-good arms has been overlooked in past work, although arguably this may be the most natural objective in many applications. For example, a virologist may conduct preliminary laboratory experiments on a large candidate set of treatments and move all ∈-good treatments into more expensive clinical trials. Cuban Treefrogs (Osteopilus septentrionalis) are established in Florida and Louisiana and have invasive potential further westward, possibly impacting native anurans at multiple life stages. In anurans, competition at the larval stage can decrease adult fitness through slower development and smaller size at metamorphosis, ultimately decreasing recruitment rates. Nitric oxide (NO) is generated by one of the three isoforms of mammalian NO synthases (NOSs), neuronal, endothelial and inducible NOS (nNOS, eNOS, and iNOS, respectively). Dysregulated NOS is implicated in pathologies of numerous severe diseases including Alzheimer and Parkinson’s diseases. Molecular mechanisms of NOS regulation, once fully understood, are potentially key targets for development of selective new pharmaceuticals for treating these diseases that currently lack effective treatments.

Background: Depression and anxiety disorders, believed to be caused by stress-related neuropeptides and pro-inflammatory molecules in the brain, are prevalent comorbidities of fetal alcohol spectrum disorders (FASD).

Methods: Brain tissue was collected following a 4-hour maternal separation stress during the early postnatal period, postnatal days 10 and 12, from mice subjected to 1st and 2nd trimester prenatal alcohol exposure (PAE).

Hypothesis: We hypothesized that PAE leads to glucocorticoid receptor insensitivity, stress molecule overexpression, and potentiated toll-like receptor 4 (TLR4) pathway inflammatory responses in the brain following early life stress.

This dissertation aims to determine if adult hippocampal neurogenesis is a potential therapeutic target in Fetal Alcohol Spectrum Disorders (FASDs). Adult hippocampal neurogenesis plays important and unique roles in certain aspects of learning, memory, and mood (Anacker & Hen, 2017). Individuals with FASDs often demonstrate dysregulation in these arenas (Lewis et al., 2015; Temple, Cook, Unsworth, Rajani, & Mela, 2019). Due to the novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the 2020 coronavirus disease pandemic uncovered health inequities throughout the United States. This paper explores institutional racism as the reasoning behind these health inequities. A discussion of the United States’ failure to implement preventative precautions once notified initially of the coronavirus is featured. Events that took place after the recognition of the coronavirus, such as the economic impacts and passed legislation, are highlighted as well.
