BACKGROUND: Brain metastases are a significant cause of mortality and morbidity for patients with melanoma. We hypothesize that the development of brain metastases may be explained by molecular heterogeneity between primary cutaneous melanoma (PCM) or extracranial (ECM) and brain (MBM) melanoma metastases.
MATERIALS AND METHODS: We compared next-generation sequencing, tumor mutational burden (TMB), and immunohistochemical staining for PD-L1 expression, among 132 MBM, 745 PCM, and 1190 ECM.
RESULTS: The most common genetic alterations among MBM included: BRAF (52.4%), NRAS (26.6%), CDKN2A (23.3%), NF1 (18.9%), TP53 (18%), ARID2 (13.8%), SETD2 (11.9%), and PBRM1 (7.5%). Four genes were found with higher frequency …

BACKGROUND: Despite the introduction of new insulin analogs, insulin pumps, and continuous glucose monitoring (CGM), young children with type 1 diabetes mellitus (T1D) remain vulnerable to episodes of hypoglycemia because of their unpredictable eating and activity patterns and high degree of insulin sensitivity. Caregivers and young children living with T1D learn to fear hypoglycemia because it is uncomfortable, unpredictable, and dangerous. Up to 60% of caregivers of young children with T1D report moderate to severe levels of fear of hypoglycemia, and caregiver fear of hypoglycemia relates to lower quality of life for families and suboptimal child glycemic control. Thurston County Washington is experiencing a steady increase in the rates of adult obesity within our community. Obesity can have a negative impact on the mental, physical, and financial health of individuals and the county at large (Pul & Suh, 2015). As such, the author proposes addressing this community issue through a social change project that contains both preventative measures as well as postvention efforts to address and decrease rates of obesity in Thurston County Washington.

In this study, the relationships between ethanol yield/concentration and solid loading (6–21%) were investigated to enhance ethanol titer and avoid a random choice of solid loading for simultaneous saccharification and fermentation (SSF). Alkali-pretreated hemp biomass was used for SSF in four scenarios including Case I: 30 filter paper unit (FPU)-cellulase and 140 fungal xylanase unit (FXU)-hemicellulase/g-solid; Case II: 40 FPU-cellulase and 140 FXU-hemicellulase/g-solid; Case III: 30 FPU-cellulase and 140 FXU-hemicellulase/g-solid with 1% Tween80; and Case IV: 30 FPU-cellulase and 140 FXU-hemicellulase/g-solid with particle size reduction (R2 = 0.76–0.93) and quadratic (R2 = 0.96–0.99) correlation with solid loading (6–21%), …

Insects manifest phenotypic plasticity in their development and behavior in response to plant defenses, via molecular mechanisms that produce tissue-specific changes. Phenotypic changes might vary between species that differ in their preferred hosts and these effects could extend beyond larval stages. To test this,we manipulated the diet of southern armyworm(SAW; Spodoptera eridania) and fall armyworm (FAW; Spodoptera frugiperda) using a tomato mutant for jasmonic acid plant defense pathway (def1), and wild-type plants, and then quantified gene expression of Troponin t (Tnt) and flightmusclemetabolismof the adult insects. River otters (Lontra canadensis) encompass a broad geographic range including coastal, riverine, and lacustrine systems. However, knowledge of reproductive behavior and structural den characteristics remain relatively few in the literature, particularly in the Great Plains. Distinctions between the terms “den”, “den site”, “natal den”, and “maternal den” are often ambiguous, obscuring our understanding of river otter’s young-rearing behavior. We used observations and descriptions regarding a single maternal den site and a broad reading of the mustelid literature to propose a more standardized maternal den definition for river otters and specify hypotheses for future research. This paper proposes an interactive approach to aid the compositional process using the brain-computer music interface (BCMI). In particular, the system developed in this paper focuses on creating an interactive compositional process inclusive to non-musicians and those with motor disabilities, in addition to providing a new method of composing. This paper references and compares two BCMI systems that have been developed for compositional purposes and draws upon these ideas to form the proposed system in this paper. Such system would aid the compositional process based on note velocity, chords and key changes.

As Arctic seas rapidly change with increased ocean temperatures and decreased sea ice extent, traditional Arctic marine mammal distributions may be altered, and typically temperate marine mammal species may shift poleward. Extant and seasonal odontocete species on the continental shelves of the Bering and Chukchi Seas include killer whales (Orcinus orca), sperm whales (Physeter microcephalus), beluga whales (Delphiapterus leucas), harbor porpoises (Phocoena phocoena), and Dall’s porpoises (Phocoenoides dalli). Newly documented, typically temperate odontocete species include Risso’s dolphins (Grampus griseus) and Pacific white-sided dolphins (Lagenorhynchus obliquidens). Ischemic heart disease (IHD) is the leading cause of death and chronic disability in the world. IHD affects both the systolic and diastolic function of the heart which progressively leads to heart failure; a structural and functional impairment of filling or ejection of blood from the heart. As frustrations over inequalities in policing and law enforcement continue despite attempted reforms (Beckett, 2016), many are asking for a more effective approach. A 2018 issue statement from the American Public Health Association (2018) highlights that violence is a public health issue that will not go away without the influence of a public health approach. The integrated biological-psychological-social model of health recognizes the complexity in the ways individuals are influenced by their situations, with violence as the unfortunate result of the wrong mix of circumstances. The public health approach to violence focuses on prevention as part of the solution. This …

Life expectancy is a key measure in population health. Although there have been significant gains in the past decades, deep disparities still persist along racial lines. The aim of this paper is to analyze the disparate life chances of racial and ethnic minorities within Shelby County, Tennessee. OLS regression is used to investigate the association between life expectancy and various sociodemographic variables. The overall life expectancy of Shelby County is years below the national average, and the results of this project find that there are stark differences across census tracts.

Transcription is regulated through a dynamic interplay of DNA-associated proteins, and the composition of gene-regulatory complexes is subject to continuous adjustments. Protein alterations include post-translational modifications and elimination of individual polypeptides. Spatially and temporally controlled protein removal is, therefore, essential for gene regulation and accounts for the short half-life of many transcription factors. The ubiquitin-proteasome system is responsible for site- and target-specific ubiquitination and protein degradation. Specificity of ubiquitination is conferred by ubiquitin ligases. Cullin-RING complexes, the largest family of ligases, require multi-unit assembly around one of seven cullin proteins. To investigate the direct role of cullins in ubiquitination of …

With the growth of metabolomics research, more and more studies are conducted on large numbers of samples. Due to technical limitations of the Liquid Chromatography-Mass Spectrometry (LC/MS) platform, samples often need to be processed in multiple batches. Across different batches, we often observe differences in data characteristics. In this work, we specifically focus on data generated in multiple batches on the same LC/MS machinery. Traditional preprocessing methods treat all samples as a single group. Such practice can result in errors in the alignment of peaks, which cannot be corrected by post hoc application of batch effect correction methods. In this …

The ability of the F atom of HC≡CF, H2C=CHF and H3CCH2F to serve as an electron donor to the triel (Tr) atom of TrR3 in the context of a triel bond is assessed by ab initio calculations. The triel bond formed by Csp3—F is strongest, as high as 30 kcal/mol, followed by Csp2—F, and then by Csp—F whose triel bonds can be as small as 1 kcal/mol. The noncovalent bond strength diminishes in the order Tr = Al > Ga > B, consistent with the intensity of the π-hole …

Reptiles rely on thermal heat exchange to achieve body temperatures (Tbody) conducive to maintaining homeostasis. Diurnal changes in the thermal environment are therefore liable to influence allostatic mediation of survival processes (e.g., immunity) during environmental challenges or stressors. However, the extent to which Tbody prompts individual variation in physiology remains largely unexplored in reptiles. Our study tested how circulating energy-mobilizing hormone, energy metabolites, and immunity can vary across basal and stress-induced allostatic states for plateau side-blotched lizards (Uta stansburiana uniformis) residing in a heterogeneous thermal environment. Jellyfish are conspicuous yet under-studied components of marine zooplankton communities. Abundance, biomass, size, and distribution of large-jellyfish were measured during July and September of 2018 and 2019 as part of the Northern Gulf of Alaska Long-Term Ecological Research (NGA-LTER) cruises. Nearly 1000 kg dispersed among ~13,800 jellies were collected using a 5 m² Methot net. Catches were dominated by two macro-jellies, the hydrozoan Aequorea sp. and the scyphozoan Chrysaora sp. During 2018, epipelagic macro-jellies biomass averaged 1.46 ± 0.36 g WW m⁻³ for July and 1.14 ± 0.23 g WW m⁻³ for September, while during 2019 they averaged 0.86 ± …

This thesis is a study displaying the connections between female characters and dragons in Old Norse and Middle High Germanic literature. The main associations that I examine are the ways that female characters and dragons share the characteristics of greed or hoarding, prophetic sight or supernatural power, and "monstrosity" or "Otherness." The fundamental argument is that the women and dragons have common characteristics which define them as dangerous and thereby cause the men or heroes of the tale to feel the need to silence or depower them through conquest. Anthropogenic climate change is rapidly altering alpine ecosystems in Alaska. Trees and woody shrubs are expanding upslope and displacing alpine tundra. As alpine tundra habitats shrink and fragment, arthropods and other animals face an increased risk of extirpation due to smaller population sizes and reduced geneflow. Arthropods--insects, spiders, and their relatives--are the most speciose component of the alpine fauna and perform key ecosystem services, such as pollination and nutrient cycling, and are food for vertebrates. Many species have responded by shifting their distribution to higher elevations, but species respond to change idiosyncratically, which could alter species interactions and disrupt communities. …

With increased oil exploration, development, and transport in the Arctic in recent years, the potential for disastrous oil spills is imminent. Biodegradation, the consumption of contaminants by indigenous microorganisms capable of using them as an energy source, can be enhanced using bioremediation treatments and may be a viable spill remediation method when traditional physical recovery techniques are not. The EPA National Contingency Plan (NCP) product schedule lists oil spill response treatments that can be used in the event of a spill, many of which can stimulate intrinsic biodegradation.
