Urbanization negatively impacts water quality in streams by reducing stream-groundwater interactions, which reduces the stream’s ability to naturally attenuate nitrate. Meadowbrook Creek, a first order urban stream in Syracuse, New York, has a negative urbanization gradient that results in urbanized headwaters that are disconnected from the floodplain, and downstream reaches that have intact riparian floodplains and connection to riparian aquifers. This system allows us to assess how stream-aquifer interactions in urban streams impact the net sources and sinks of nitrate at the reach scale. We used continuous (15-minute) streamflow measurements, along with weekly grab samples at three gauging stations positioned …

G-protein-coupled receptors (GPCRs) are the most diverse membrane proteins in eukaryotes that transmit chemical signals to the cell. GPCRs are considered the largest family of targets for approved drugs on the market. Sphingosine 1-phosphate receptor 1 (S1P1), a widely expressed GPCR protein in many cell types, performs signal transduction function not only in the monomeric state but also in the dimeric or complex oligomeric states. However, the detail of the S1P1 assembly is still not clearly understood. Focal adhesions are the large molecular complexes that anchor cells to the ECM, which is involved in cell spreading, migration, and proliferation. In our previous work, the study of cardiomyocyte morphological changes and myofibril remodeling, responding to the dynamic nano-topographic substrate, have been accomplished. The results showed focal adhesion experienced disassembly and reassembly in the first 12 hours. In this study, the hiPSC-CMs were seeded on the SMP substrates with the flat-to-wrinkle dynamic surface. Paxillin, vinculin, and zyxin from different layers of focal adhesive architectures were co-stained with F-actin. The fluorescent images were analyzed to investigate the details of focal …

Quantum philosopher Ervin Laszlo has taken one step further outside of the contemporary mainstream scientific view, which believes in a reductive “scientific” method primarily of highly controlled experiments to determine truth. Protein prenylation is a posttranslational modification involving the attachment of a C15 or C20 isoprenoid group to a cysteine residue near the C-terminus of the target substrate by protein farnesyltransferase (FTase) or protein geranylgeranyltransferase type I (GGTase-I), respectively. Both of these protein prenyltransferases recognize a C-terminal "CaaX" sequence in their protein substrates, but recent studies in yeast- and mammalian-based systems have demonstrated FTase can also accept sequences that diverge in length from the canonical four-amino acid motif, such as the recently reported five-amino acid C(x)3X motif. Research suggests that exposure to stressors is associated with greater alcohol consumption, more alcohol-related problems, and a greater likelihood of Alcohol Use Disorder. Theory suggests that cognitive processes, particularly unconscious cognitive processes, are key in determining a coping strategy but both alcohol and chronic exposure to stressors may interfere with these processes. The current study tested one such process, attention bias to alcohol cues, as a mediator in the stressor-alcohol relationship. Thirty-nine participants who endorsed hazardous alcohol consumption patterns were recruited from the community; eligible participants were randomly assigned to a stress-exposure or non-stressful control condition. Decades of research into the acceptance of evolutionary biology have revealed a number of factors that are related to an individual’s choice to accept or reject evolutionary biology. This work seeks to extend that work in the following key ways: (1) Use a longitudinal time frame, along with multifactorial linear modeling, to investigate the changes in evolution acceptance and its associated factors across a year of introductory biology education. Medical cannabis is increasingly used as a treatment for chronic pain, and there is initial evidence that medical cannabis may lead to a subsequent decrease in prescription opioid use. The objective of the current study was to conduct a retrospective, naturalistic examination of medical cannabis use (i.e., dose, frequency, type) and subsequent changes in prescription opioid use among a sample of treatment-seeking chronic (non-cancer-related) pain patients (N =277). Data from the electronic medical record (EMR) was paired with information from the State Prescription Drug Monitoring Program and collected at time of initial certification and at six months post-certification. Biofilms are multicellular structures with bacterial cells attached to a surface and embedded in an extracellular matrix. With high-level resistance to antimicrobial agents, biofilms are the cause of chronic infections associated with implanted medical devices such as breast implants, orthopedic devices, pace markers, and many others. Besides the prevalence, biofilm infections are associated with high mortality, presenting an urgent need for more effective controls. The focus of this work is to construct theoretical methods or approaches to tackle computationally challenging problems in quantum chemistry. The primary topic of the work will focus on efficient theories that allow for capturing electron-correlation in semiconductor or metallic nanoparticles. These systems are computationally challenging due to the size of the systems, the number of electrons per atom, and the degeneracy of the particle-hole states. Three methods presented here are aimed at providing novel theoretical methods towards computationally inexpensive electronic excited state calculations. Collective tissue dynamics, more specifically a tissue's ability to fluidize and segregate, is imperative for proper embryonic development and normal physiological functioning. In this thesis, I use vertex models to understand how cell-scale properties govern large-scale collective behavior. I begin with the process of fluidization in ordered monolayers. By perturbing beyond the linear regime, I show that in confluent tissues the linear response does not correctly predict the non-linear behavior, which, in this case, is the ability to exchange neighbors and fluidize. The overall aim is to advance the use of light for noninvasive monitoring of living systems generally defined. We first describe work used to test a methodology for real-time probing of chemical and physical changes in spinal cords in the immediate aftermath of a localized contusive injury by way of calculating pH to monitor physiological changes in the system. Utilizing a previously developed unique algorithm, line scans of injured cords as well as optical profilimetry and scanning NIR autofluorescence images were obtained simultaneously in vivo, on exposed rat spinal cords. There has been an increased participation of female athletes competing in the NCAA for the past thirty years. Amongst these female athletes, there is an increased risk of stress fracture (SFx) injury, which is highly prevalent among female endurance sports. Organization, separation, and cellular regulation are all functions of membraneless organelles (MLOs), which arise by a biophysical phenomenon termed liquid-liquid phase separation (LLPS). By this process, macromolecules in a mixed liquid solution condense together to form liquid droplets within a liquid solution, comparable to oil droplets in water. Some known MLOs formed in cells via LLPS include nucleoli, stress granules, Cajal bodies, and processing-bodies, among other membrane-lacking liquid granules. Previous work has shown that many proteins which compose these liquid compartments also undergo LLPS isolated in vitro, and thus have become model systems to investigate the forces that drive these …

Introduction: Anatomic morphology commonly depicted in atlases or textbooks is often emphasized in gross anatomy classrooms; however, considerable variation may be observed in cadavers during dissection, particularly in the vascular system. This study statistically assesses the frequency of the classic versus variant axillary artery branching pattern and compares these observations to the incidence of variation described in the literature. Material and Methods: Axillary artery branching pattern was studied in 62 cadaver limbs. A Chi square goodness of fit test with post hoc analyses on the adjusted standardized residuals was used to evaluate branching patterns. Results: A statistically significant difference existed …

Previous research has found dualistic effects on alcohol consumption with low doses of buprenorphine increasing alcohol and higher doses of buprenorphine reducing alcohol consumption in rats (Ciccocioppo et al., 2006). Other existing research on naloxone treatment and alcohol consumption in opioid use has demonstrated that alcohol consumption decreases after naloxone treatment in rats (Hyytia & Sinclair, 1993). Yet, no research has been conducted on either rats or humans on the effects of buprenorphine and naloxone medication combined. Ghrelin is a peptide hormone primarily secreted by the stomach that acts as the endogenous ligand for the GHSR1a G-protein-coupled receptor. This 28 amino acid peptide stimulates appetite and has been associated with the cardiovascular, gastrointestinal, neuroendocrine, and immune systems. Ghrelin requires a unique posttranslational modification to be biological active. Ghrelin O-acyltransferase (GOAT) is responsible for catalyzing octanoylation of the 3rd serine side chain of ghrelin to generate the active form of this hormone. Dopant ions have been found to diffuse/migrate throughout solid materials when the thermal energy of the system is elevated above a temperature boundary allowing the cations and anions within a system to move freely. The first act encapsulates a study of the rigidity transition in frictional particle packings, or how a packing goes from floppy-to-rigid as its particle density increases, for example. While consensus has emerged regarding the nature of the rigidity transition in frictionless packings, there is much less consensus with frictional packings. Therefore, I introduce two new complementary concepts, frictional rigidity percolation and minimal rigidity proliferation, to help identify the nature of the frictional rigidity transition. In this thesis, we investigate the relationships between the disorder, structure, and deformation in amorphous materials. First, to understand the surprising low-frequency vibrational modes in structural glasses, and how it arises from the microscopic disorder in the system, we study the spectra of a large ensemble of sparse random matrices where disorder is controlled by the distribution of bond weights and network coordination. When there is a finite probability density of infinitesimal bond weights, we find a region in the vibrational density of states that is consistent with the low-frequency behavior in structural glasses.
