The microbial attachment and colonization on separation membranes lead to biofilm formation. Some selective isolates within the biofilm constitutive microflora acquire resistance and emerge predominant over prolonged use of the membrane. Thus, proving the cleaning and sanitization protocols to be ineffective in adequately removing resilient biofilms. This subsequently leads to selecting microbial resistance within the constitutive microflora to almost all antimicrobial treatments and hence, creates a need to develop novel alternative strategies to control biofilm formation on membrane surfaces. Cells are constantly under threat from both exogenous and endogenous sources of DNA damage. Eukaryotic organisms, however, possess conserved mechanisms that accurately and faithfully respond to DNA damage. The inability to effectively remove DNA lesions can lead to an accumulation of mutations which can compromise cellular viability. The DNA damage response is conserved from bacteria to eukaryotic organisms and have been well characterized, however, how covalently crosslinked proteins are removed from DNA remains enigmatic This thesis provides genetic and biochemical evidence implicating Wss1, a yeast metalloprotease in genome maintenance. The overall objective of this dissertation was to determine the impact of selected post weaning management strategies on beef carcass characteristics and meat quality. The specific objectives were 1) determine the effect of a rumen-protected long chain fatty acids fed during finishing on live performance, serum fatty acid profile, carcass traits, and predicted carcass composition, and 2) determine effects of feeding brassica-based cover crops to cattle during backgrounding on live animal performance, carcass characteristics, tenderness, juiciness, and flavor of strip steaks, and case life of ground beef and strip steaks. In Lake Oahe, a large mainstem Missouri river reservoir that spans 375 river kilometers in central North and South Dakota, naturalized Rainbow Smelt are the primary forage species for a regionally important Walleye Sander vitreus fishery. Walleye condition in Lake Oahe is highly correlated to Rainbow Smelt relative abundance through time, the latter species being typified by erratic abundance trends since accidental introduction in 1971. Cisco Coregonus artedii were introduced in the early 1990s to augment the cold-water prey base. This research aimed to understand the evolutionary mechanism of seed dormancy and flowering time and to provide basic knowledge for the manipulation of germination features in crop breeding. Flowering time and seed dormancy are two adaptive traits for flowering plants. Seed dormancy (SD) helps regulate the timing of germination to promote the survival of seed-bearing plants in adverse environments. Cereal crops and weedy/wild relatives, such as Asian cultivated rice and weedy rice (Oryza sativa L.), have weak and strong seed dormancy, respectively. Historically, pheasants (Phasianus colchicus) have thrived in South Dakota in conjunction with successful land retirement programs or early farming practices through the 1950s, which created interspersions of agriculture and native landscapes that were ideal for pheasants. Recently, the Prairie Pothole ecosystem has undergone rapid agroeconomic expansion, effectively reducing ideal interspersions of native prairie and cropland into agriculturally homogeneous landscapes. Indices of pheasant abundance have suggested persistent population declines since 2008, raising concerns regarding landscape suitability. Biological Nitrogen Fixation is the process of conversion of atmospheric dinitrogen into ammonia which is performed by symbiotic as well as free-living diazotrophic bacteria. A previous project to isolate free-living nitrogen fixing bacteria from a natural prairie yielded Streptomyces which grew on nitrogen free medium. Genome sequencing of three isolates revealed the presence of a second 16S rRNA gene sequence aligning with Bacillus, however repeated streaking only yielded Streptomyces colonies. Mountain Sucker, Pantosteus jordani, is a cold-water species native to the Intermountain West. Fringe populations of Mountain Sucker have experienced declines in recent decades. The population of Mountain Sucker found in the Black Hills of South Dakota represents the eastern fringe of the species’ native range. Higher and stable yield is always a major objective of wheat genetic improvement programs. The SQUAMOSA-promoter binding protein-like (SPL) genes constitute a small family of plant-specific transcription factors with diverse functions in plant development and growth and have great potential in improving yield and other major agronomic traits. However, the functional characterization of the SPL gene family in wheat is far behind other cereal crops such as rice. Using phylogenetic analysis, we identified 56 wheat orthologues of rice SPL genes belonging to 19 homoeologous groups. Soybean cyst nematode (SCN) is a microscopic worm that infects and feeds from the soybean roots, hence robbing the plant of nutrients, and entry wounds may facilitate other pathogens to infect the roots. Current SCN management practices include cultural, chemical, and biological methods. Cultural practices such as, crop rotation with non-host and different tillage practices are not efficient due to the persistent nature of SCN in the soil. I compared movement, home range, habitat selection, and mortality among radio-tagged Brown Trout (Salmo trutta) at an American Mink (Neovison vison)-removal site, improved habitat site, and an undisturbed stream site in Rapid Creek, South Dakota. I observed high, post-surgery (> 26 days) survival rate (90%) of radio tagged Brown Trout. Average gross movement of Brown Trout was greater at the improved habitat site (127 m) compared to the mink removal (31 m) site, while average home range size (i.e., stream distance) among all three sites was not significantly different indicating strong site fidelity (125 to 200 m). Transposable elements, also called jumping genes, comprise almost 45% of the human genome. In contrast, only 1% of the human genome is protein-coding sequences. The function and advantages of maintaining such massive copies of transposable elements in the human genome are still unclear. Long interspersed element 1 (L1), the most substantial group and the only active autonomous transposable element in the human genome, has revealed its unique roles in many diseases. The tallgrass prairies of the Northern Great Plains are one example of a heavily imperiled ecosystem. This work combines traditional microbiology with bioinformatic and synthetic biology approaches to study antibiotic tolerance. Antibiotic tolerance is a widespread phenomenon that facilitates antibiotic resistance and decreases the effectiveness of antibiotic treatment. Tolerance is distinct from antibiotic resistance, because tolerance is short term survival and typically results from phenotypic variations rather than genetic variation. The molecular mechanisms underlying tolerance are varied and debated in the literature. I have explored two intracellular processes related to tolerance, toxin-antitoxin (TA) systems (Chapter 2) and proteases (Chapter 4). As newlyweds begin their new adventure as a married couple, the discussion of family expansion is inevitable. This thesis seeks to understand how newlyweds discuss their plans for family expansion among each other and to others by applying the theory of communication privacy management. Ten newlywed couples were interviewed regarding their experiences navigating the topic of family expansion and reproduction. Specifically, the couples were asked about preferences for privacy and control, and what expectations the couples have when disclosing information about their preferences for family expansion. The LEAF (Learning from and Engaging with Assessment and Feedback) project addressed a key issue in third-level teaching and learning: assessment and feedback. The LEAF team comprised 18 academics from across the TU Dublin City Campus and representatives from all Colleges, along with the Director of Student Affairs and the Students’ Union Education Officer. This paper presents the findings of the LEAF project. The purpose of this paper is to describe teachers’ perceptions of experiential learning and its effects on student learning, understanding, and engagement through agriculture education's three-circle model. This paper explores teachers’ perceptions of the value and importance of experiential learning in agriculture education instruction, FFA, and supervised agricultural experience. Further, I explore experiential learning, constructivism, and hands-on learning as the foundational instructional practices used in agricultural education. Agricultural education teachers from southwest Minnesota were surveyed to determine how they are using experiential learning in their agriculture education programs, including classroom instruction, FFA, and supervised agricultural experiences.

We numerically investigated the dynamics of a paramagnetic elliptical particle immersed in a low Reynolds number Poiseuille flow in a curved channel and under a uniform magnetic field by direct numerical simulation. A finite element method, based on an arbitrary Lagrangian-Eulerian approach, analyzed how the channel geometry, the strength and direction of the magnetic field, and the particle shape affected the rotation and radial migration of the particle. The net radial migration of the particle was analyzed after executing a π rotation and at the exit of the curved channel with and without a magnetic field. In the absence of …
