Pandemic Vibrio Cholerae Shuts Down Site-Specific Recombination To Retain An Interbacterial Defence Mechanism, Francis J. Santoriello, Lina Michel, Daniel Unterweger, Stefan Pukatzki

Vibrio cholerae is an aquatic microbe that can be divided into three subtypes: harmless environmental strains, localised pathogenic strains, and pandemic strains causing global cholera outbreaks. Each type has a contact-dependent type VI secretion system (T6SS) that kills neighbouring competitors by translocating unique toxic effector proteins. Pandemic isolates possess identical effectors, indicating that T6SS effectors may affect pandemicity. Here, we show that one of the T6SS gene clusters (Aux3) exists in two states: a mobile, prophage-like element in a small subset of environmental strains, and a truncated Aux3 unique to and conserved in pandemic isolates. This dissertation report covers Yan Ma’s Ph.D. research with applicational studies of machine learning in manufacturing and biological systems. The research work mainly focuses on reaction modeling, optimization, and control using a deep learning-based approaches, and the work mainly concentrates on deep reinforcement learning (DRL). Yan Ma’s research also involves with data mining with bioinformatics. Large-scale data obtained in RNA-seq is analyzed using non-linear dimensionality reduction with Principal Component Analysis (PCA), t-Distributed Stochastic Neighbor Embedding (t-SNE), and Uniform Manifold Approximation and Projection (UMAP), followed by clustering analysis using k-Means and Hierarchical Density-Based Spatial Clustering with Noise (HDBSCAN). Background Carbon nanotubes and nanofibers (CNT/F) have known toxicity but simultaneous comparative studies of the broad material class, especially those with a larger diameter, with computational analyses linking toxicity to their fundamental material characteristics was lacking. It was unclear if all CNT/F confer similar toxicity, in particular, genotoxicity. Nine CNT/F (MW #1-7 and CNF #1-2), commonly found in exposure assessment studies of U.S. facilities, were evaluated with reported diameters ranging from 6 to 150 nm. 5 fluorouracil (5FU) has been a first-choice chemotherapy drug for several cancer types (e.g., colon, breast, head, and neck); however, its efficacy is diminished by patient acquired resistance and pervasive side effects. Leukopenia is a hallmark of 5FU; however, the impact of 5FU-induced leukopenia on healthy tissue is only becoming unearthed. Recently, skeletal muscle has been shown to be impacted by 5FU in clinical and preclinical settings and weakness and fatigue remain among the most consistent complaints in cancer patients undergoing chemotherapy. A benchmark project was conducted regarding the use of a CPM machine postoperatively to increase standardization of care for total knee replacement patients while also increasing the patient’s range of motion of the knee and decreasing the post-surgical pain. Over a six week period of time, each patient was given a CPM machine no later than 2 hours post op after the total knee replacement surgery and each patient was discharged after a one night stay in the hospital under observation instead of inpatient. The Cretaceous Almond Formation, located in the Greater Green River Basin, records deposition of coastal plain fluvial sandstones and shallow marginal-marine sandstones in a net-transgressive sequence along the western margin of the Cretaceous Interior Seaway (CIS) from the late Campanian to early Maastrichtian. The Almond Formation is an important hydrocarbon reservoir, with development mainly along the Wamsutter Arch and the northeast margins of the Washakie Basin. Previous studies have primarily focused on outcrops along the eastern flank of the Rock Springs Uplift and subsurface data targeting the Wamsutter Arch. Further development of the Almond petroleum system requires extending our understanding …

Modern plant breeding increasingly relies on genomic information to guide crop improvement. Although some genes are characterized, additional tools are needed to effectively identify and characterize genes associated with crop traits. To address this need, the mPing element from rice was modified to serve as an activation tag to induce expression of nearby genes. Embedding promoter sequences in mPing resulted in a decrease in overall transposition rate; however, this effect was negated by using a hyperactive version of mPing called mmPing20. Transgenic soybean events carrying mPing-based activation tags and the appropriate transposase expression cassettes showed evidence of transposition. Expression analysis …

Computer users are often the last line of defense in computer security. However, with repeated exposures to system messages and computer security warnings, neural and behavioral responses show evidence of habituation. Habituation has been demonstrated at a neural level as repetition suppression where responses are attenuated with subsequent repetitions. In the brain, repetition suppression to visual stimuli has been demonstrated in multiple cortical areas, including the occipital lobe and medial temporal lobe. Prior research into the repetition suppression effect has generally focused on a single repetition and has not examined the pattern of signal suppression with repeated exposures. We used …

Endocrine disrupting chemicals (EDCs) pose a threat to biodiversity at the individual, population, and ecosystem level, as they can interfere with processes that are responsible for regulating metabolism, development, behavior, and reproduction in living organisms. 17α-ethynylestradiol (EE2) is a synthetic estrogen and EDC utilized in pharmaceutical and livestock industries, and contaminates waterways worldwide. This research explores the effects of dose-dependent and multi-generational exposure of EE2 in three strains of zebrafish. High dose (10-25 ng/L EE2) exposure led to complete reproductive failure, as well as significantly decreased survival and growth. Implications of religious practices in Islam go far beyond religiosity, and this paper analyzed the relationship between the COVID-19 pandemic and religious practices of Muslim students in higher education. The analyzed data is from the survey of the Muslim Student Life at Syracuse University and the Center for Islam in Contemporary World at Shenandoah University. The survey was conducted through a non-random convenience sampling from March 30th through April 10th of 2020 and had 498 responders. Teh merupakan salah satu komoditas perkebunan yang memegang peranan cukup penting dalam perekonomian Indonesia, yaitu sebagai sumber pendapatan dan devisa serta penyedia lapangan kerja bagi masyarakat. Teh di Indonesia sebagian besar berasal dari Jawa Barat dengan kontribusi produksi (rata-rata lima tahun terakhir) sebesar 66,93%, sedangkan provinsi lainnya hanya berkontribusi kurang dari 10%. Produksi teh di Indonesia padatahun 2017 sebesar 146,17 ton, selalu berfluktuasi dari tahun ke tahun, hingga diperkirakan tahun 2021 menurun dengan produksi sebesar 141,63 ton. Perbanyakan tanaman stroberi secara konvensional dilakukan dengan menggunakan stolon, tetapi kurang efektif serta kualitas bibit yang dihasilkan kurang baik akibat adanya akumulasi penyakit. Budidaya stroberi memerlukan adanya perbanyakan bibit secara massal, tetapi tidak mengubah kualitasnya. Multiplikasi in vitro menjadi solusi untuk penyediaan bibit berkualitas dalam jumlah besar. Upaya untuk mendapatkan tunas in vitro dalam jumlah banyak yakni perlu adanya penambahan zat pengatur tumbuh golongan sitokinin seperti Benzylaminopurine (BAP) atau Thidiazuron (TDZ). Awareness of environmental issues has led to increasing interest from composite researchers in using “greener” materials to replace synthetic fiber reinforcements and petrochemical polymer matrices. Natural fiber bio-based thermoplastic composites could be an appropriate choice with advantages including reducing environmental impacts, using renewable resources and being recyclable. The choice of polymer matrix will significantly affect the cost, manufacturing process, mechanical properties and durability of the composite system. In deserts, global environmental changes, plant community structure, and soil water availability form a complex relationship characterized by direct and indirect relationships and feedbacks. Plant community structure is affected by the establishment and spread of invasive grasses, which form a positive feedback with wildfire. Plant community structure is also controlled by top-down trophic interactions by small-mammals and climate change, which alters the timing and amount of soil water. In deserts, soil water availability mediates the effects of global environmental changes and trophic interactions on plant community structure because it is the limiting resource for plant growth. Nomadic pastoralism persists at a substantial scale in Tuva and neighboring regions of Inner Asia. Tuvan pastoral lifeways reflect adaptations to both local environments and current economic realities. Much of our quantitative understanding of the economics of Tuvan nomads is derived from data collected in the first half of the 20th century. Accordingly, this paper provides an updated picture of the inner workings of nomadic households using data collected in Barun-Khemchik and Bai-Taiga provinces in 2013–2015. It analyzes herd composition and size, and compares the frequency of different animals kept today with values recorded in Tuva in 1916 and 1931. …
