Department of Nutrition and Health Sciences: Faculty Publications

Emerging evidence suggests intestinal microbiota as a central contributing factor to the pathogenesis of Relapsing-Remitting-Multiple-Sclerosis (RRMS). This novel RRMS study evaluated the impact of fecal-microbiota-transplantation (FMT) on a broad array of physiological/clinical outcomes using deep metagenome sequencing of fecal microbiome. FMT interventions were associated with increased abundances of putative beneficial stool bacteria and short-chain-fatty-acid metabolites, which were associated with increased/improved serum brain-derived-neurotrophic-factor levels and gait/walking metrics. This proof-of-concept single-subject longitudinal study provides evidence of potential importance of intestinal microbiota in the pathogenesis of MS, and scientific rationale to help design future randomized controlled trials assessing FMT in RRMS patients.

Time-Restricted Feeding Extends Longevity In Drosophila Melanogaster, Daniel Cabrera Jan 2020

Time-Restricted Feeding Extends Longevity In Drosophila Melanogaster, Daniel Cabrera

Student Theses and Dissertations

Time-restricted feeding (TRF) is a dietary intervention in which daily feeding patterns align with behavioral patterns, synchronizing feeding times with periods of higher activity, e.g. humans eating only during the day or rodents (nocturnal animals) at night. TRF has been shown to improve cardiac health in Drosophila melanogaster, reduce metabolic markers in rodent models, and reduce glycemic indices in prediabetic men. However, the mechanism and long-term effects of this intervention remain elusive. To understand the effect of TRF on longevity we used the fruit fly Drosophila melanogaster, which is a useful model for longevity, sleep and circadian studies because of …

Evaluating The Utility Of Diagnostic Workups For Biliary Atresia In Neonates With Cholestatic Jaundice Following Prolonged Tpn, Melissa Austin, Zubair H. Aghai

Phase 1

Introduction: Parenteral nutrition associated cholestasis (PNAC) develops in 40-60% of premature infants following TPN for 2-4+ weeks. The incidence of biliary atresia is low and there is a 60-day, post-term window for corrective surgery. There is limited data on both the natural history of PNAC in premature infants following prolonged TPN, as well as the utility of diagnostic tools investigating biliary atresia in these patients.

Methods: A retrospective chart review using EMR data from the Intensive Care Nursey was performed with the following criteria: premature babies diagnosed with cholestasis, born at <1500 grams, and received TPN for 14 days. Ultimately 61 babies met criteria and data was collected and pooled to produce descriptive statistics and graphs describing laboratory trends.

Results: Median gestational age was 26 [IQR 25, …

Assessment Of Dobhoff Tube Malposition On Radiographs Using Deep Learning, Kevin George, Paras Lakhani, Md

Phase 1

Introduction: Dobhoff tubes (DHT) are narrow-bore flexible devices that deliver enteral nutrition for critically ill patients. Tracheobronchial insertion of DHTs presents a significant risk for pulmonary complications. Thus, DHT insertion requires radiologist confirmation of correct placement with chest x-ray (CXR), increasing clinical delays. To address this, we demonstrate the novel application of Deep Convolutional Neural Networks (DCNNs) to automatically and accurately identify DHTs in CXRs in real time.

Methods: 141 de-identified HIPAA compliant frontal view chest radiographs containing DHTs in various positions were obtained. The DHTs were first manually segmented and verified by a board certified radiologist. Images were split …

The Effect Of Advanced Glycation End Products (Ages) On Macrophages, And Their Implications For Prostate Cancer Progression, Callan C. Frye Jan 2020

The Effect Of Advanced Glycation End Products (Ages) On Macrophages, And Their Implications For Prostate Cancer Progression, Callan C. Frye

MUSC Theses and Dissertations

Advanced glycation end products (AGEs) are sugar-protein adducts that may worsen cancer severity and may contribute to health disparity outcomes among prostate cancer (PCa) patients. AGEs are formed endogenously via a slow, nonenzymic reaction; however, AGEs are also formed in foods as a result of subjecting meats and sugars to high-heat conditions. AGEs are therefore prolific in the Western Pattern Diet (WPD), which has become increasingly common among industrialized nations. Previous work has identified dietary AGE as contributing to MYC-driven PCa progression, and the dependence of this process upon stromal receptor for advanced glycation end products (RAGE). This study therefore …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Previously, we have shown that RNA interference (RNAi) can prevent aflatoxin accumulation in transformed peanuts. To explore aflatoxin control by exogenous delivery of double-strand RNA (dsRNA) it is necessary to understand the generation of small RNA (sRNA) populations. We sequenced 12 duplicate sRNA libraries of in-vitro-grown peanut plants, 24 and 48 h after exogenous application of five gene fragments (RNAi-5x) related to aflatoxin biosynthesis in Aspergillus flavus. RNAi-5x was applied either as double-stranded RNA (dsRNA) or RNAi plasmid DNA (dsDNA). Small interfering RNAs (siRNAs) derived from RNAi-5x were significantly more abundant at 48 h than at 24 h, and the …

Association Of Social Support And Depressive Symptoms Among African Americans, Jacketta Renee Cobbs Jan 2020

Association Of Social Support And Depressive Symptoms Among African Americans, Jacketta Renee Cobbs

Walden Dissertations and Doctoral Studies

Depression affects over 120 million individuals worldwide; in the United States, depression is a leading cause of disability for individuals’ ages 15-44 years. Social support can affect both physical and depressive symptoms; therefore, most patients with heart failure (HF) need support from family and/or friends to effectively manage their health condition. This indicates family and/or friends are expected to be the core support system for long term care of those with HF. The purpose of this study was to determine whether social support contributes to depressive symptoms among African Americans with HF. The research questions examined the experience of different …
