Mucosal surfaces like those present in the lung, gut, and mouth interface with distinct external environments. These mucosal gateways are not only portals of entry for potential pathogens but also homes to microbial communities that impact host health. Secretory immunoglobulin A (SIgA) is the single most abundant acquired immune component secreted onto mucosal surfaces and, via the process of immune exclusion, shapes the architecture of these microbiomes. Not all microorganisms at mucosal surfaces are targeted by SIgA; therefore, a better understanding of the SIgA-coated fraction may identify the microbial constituents that stimulate host immune responses in the context of health …

Antimicrobial Resistance is a fundamental threat to global health and safety” (WHO, 2016). Nisin, a 34 amino acid lanthipeptide, is currently used as a food preservative worldwide and has been used for decades without significant bacterial resistance having developed. It is an extremely active molecule which kills a range of different bacterial species. However nisin, along with some other similar peptides, suffers from low stability and solubility at physiological pH, which severely restricts its possible use in human and veterinary medicine. This research is concerned with increasing the stability of nisin in physiological pH systems; the stability of the dehydroalanine …

HIV testing among young Black MSM and transwomen (YBMSM/TW) is the gateway to biomedical HIV prevention or treatment. HIV self-testing (HST) is a method that may increase consistent HIV testing. TRUST, a brief, peer-based behavioral intervention, was designed to increase uptake of consistent (every three months) HST among YBMSM/TW in New York City. To test the efficacy of the intervention, we randomized 200 friend pairs into either the intervention condition (TRUST) or a time and attention control condition. A modified intent-to-treat analysis found that self-reported HST at 3-month follow-up was statistically significantly higher (uOR 2.29; 95% CI 1.15, 4.58) and …

Diarrhea is a leading cause of death worldwide because a lack thereof in household sanitization exposes humans to high concentrations of pathogenic Escherichia coli. In 2016, the University of New Mexico’s Nepal Study Center collected cross-sectional survey data using proportional random sampling on three communities in Western Nepal. Structural and parameter learning estimation and approximate inference of Bayesian networks studied diarrheagenic E. coli exposure while incorporating participation in sanitary behaviors, access to sanitary built-in environments, and other human characteristics. Of the reported sickness, hand washing resulted in a 20 percent decrease, water treatment 8 percent, and both 28 percent. Of …

School of Medicine Publications
Background. Mycobacterium tuberculosis remains a global health problem and clinical management is complicated by difficulty in discriminating between latent infection and active disease. While M. tuberculosis-reactive antibody levels are heterogeneous, studies suggest that levels of IgG glycosylation differ between disease states. Here we extend this observation across antibody domains and M. tuberculosis specificities to define changes with the greatest resolving power.
Methods. Capillary electrophoretic glycan analysis was performed on bulk non-antigen–specific IgG, bulk Fc domain, bulk Fab domain, and purified protein derivative (PPD)- and Ag85A-specific IgG from subjects with latent (n = 10) and active (n = 20) tuberculosis. PPD-specific …

BACKGROUND: Although Schistosoma haematobium infection has been reported to be associated with alterations in immune function, in particular immune hyporesponsiveness, there have been only few studies that have used the approach of removing infection by drug treatment to establish this and to understand the underlying molecular mechanisms.
METHODS: Schistosoma haematobium-infected schoolchildren were studied before and after praziquantel treatment and compared with uninfected controls. Cellular responses were characterized by cytokine production and flow cytometry, and in a subset of children RNA sequencing (RNA-Seq) transcriptome profiling was performed.
RESULTS: Removal of S haematobium infection resulted in increased schistosome-specific cytokine responses that were …

Vaccine Delivery To Remote Regions Of The World Using A Novel Delivery Platform, Amelia Bierle
Shared Knowledge Conference
Vaccination has been the most impactful medical intervention ever devised to prevent the global spread of infectious disease. In the past 200 years, vaccination has been credited to have saved more lives from a human intervention only after clean water and improved sanitary conditions. Society has seen this through the eradication of Polio and Smallpox. The current pandemic underscores the need for an effective vaccine ever so. In the last ten months, life has changed drastically around the world. With the pandemic raging on and no cure in sight, it is clear that vaccines are a critical component of public …

Faculty Research 2020
Background: Gut microbial diversity and composition play important roles in health. This cross-sectional study was designed to test the hypothesis that hospitalized children who may be relatively immunocompromised (IC), defined as those with cancer, sickle cell disease (SCD), transplantation, or receiving immunosuppressive therapy) would have decreased microbial diversity, increased Clostridioides difficile colonization and different species composition compared to non-immunocompromised (Non-IC) children admitted to the same pediatric unit. Methods: A stool sample was obtained within 72 h of admission to a single unit at The Children's Hospital at Montefiore, Bronx, NY from March 2016 to February 2017 and the microbiome assessed …

Spatial And Temporal Determinants Associated With Eastern Equine Encephalitis Virus Activity In Florida, Kristi M. Miley
Spatial And Temporal Determinants Associated With Eastern Equine Encephalitis Virus Activity In Florida, Kristi M. Miley
USF Tampa Graduate Theses and Dissertations
Eastern Equine Encephalitis virus (EEEV) is considered the most pathogenic mosquito-borne illness in the United States. Human mortality has been estimated to range between 35 to 75%, with horses experiencing mortality rates greater than 90%. A large number of EEEV cases occur in Florida. Though we have come a long way since the first human infection was identified in the 1930s, there is still much to learn regarding the virus’s ability to maintain transmission year-round in Florida. Phylogenic studies support that Florida may serve as the geographic reservoir for EEEV. This research investigated spatiotemporal and ecological variables associated with risk …

Patient Engagement In Action: Timing And Intensity Of Strategies Used To Engage Low Income Depressed Mothers Of Infants And Toddlers, Maureen J. Baker, Beth Perry Black, Linda S. Beeber
Patient Experience Journal
The purpose of this study was to illuminate the process of patient engagement and to determine how components of patient engagement were operationalized in the nurse-patient interpersonal relationship with low income, depressed mothers, a traditionally underserved population. Using a descriptive quantitative design, we examined how components of patient engagement were executed across three phases of the nurse-patient interpersonal relationship. We assessed for differences in engagement strategies used in different phases of the interpersonal relationship and with mothers with varying levels of engagement. Through this study, we observed that patient engagement has several dynamic components varying in intensity and frequency, depending …
