Both virulent and live-attenuated porcine reproductive and respiratory syndrome virus (PRRSV) strains can establish persistent infection in lymphoid tissues of pigs. To investigate the mechanisms of PRRSV persistence, we performed a transcriptional analysis of inguinal lymphoid tissue collected from pigs experimentally infected with an attenuated PRRSV strain at 46 days post infection. A total of 6404 differentially expressed genes (DEGs) were detected of which 3960 DEGs were upregulated and 2444 DEGs were downregulated. Specifically, genes involved in innate immune responses and chemokines and receptors associated with T-cell homing to lymphoid tissues were down regulated. As a result, homing of virus-specific …

Reported cases of Lyme disease in Nebraska have been assumed to be imported from other endemic areas. Previous surveillance efforts provided no evidence of established populations as only individual specimens of Ixodes scapularis (Say) had been collected. In the winter of 2018, adult I. scapularis were found on a dog at Two Rivers State Recreation Area, Douglas County, prompting tick collection at the site and nearby natural areas. In May 2019, all life stages of host-seeking I. scapularis were collected using dragging and flagging techniques in sites located near the Platte River in Douglas, Sarpy, and Saunders counties. This is …

Enterohemorrhagic Escherichia coli (EHEC), a pathogenic subset of Shiga toxin-producing E. coli (STEC), is an important cause of hemorrhagic colitis and hemolytic–uremic syndrome (HUS), and a rare cause of urinary tract infections (UTIs) with associated HUS. EHEC strains attach intimately to intestinal epithelium with formation of actin pedestals (attaching-effacing (A/E) lesions); however, the mechanism of EHEC attachment to the uroepithelium is unknown. We conducted a retrospective study on archived urinary bladder specimens from gnotobiotic piglets that naturally developed cystitis associated with EHEC O157:H7 infection following oral inoculation and fecal shedding. Paraffin-embedded bladder tissues from three piglets with cystitis and immunohistochemical …

School of Veterinary and Biomedical Sciences: Faculty Publications
Inflammatory heart disease (IHD) is a group of diseases that includes pericarditis, myocarditis, and endocarditis. Although males appear to be more commonly affected than females, IHD can be seen in any age group. While the disease can be self-limiting leading to full recovery, affected individuals can develop chronic disease, suggesting that identification of primary triggers is critical for successful therapies. Adding to this complexity, however, is the fact that IHD can be triggered by a variety of infectious and non-infectious causes that can also occur as secondary events to primary insults. In this review, we discuss the immunological insights into …

The recently emerged Zika virus (ZIKV) spread to the Americas, causing a spectrum of congenital diseases including microcephaly in newborn and Guillain-Barré syndrome (GBS) in adults. The unprecedented nature of the epidemic and serious diseases associated with the viral infections prompted the global research community to understand the immunopathogenic mechanisms of the virus and rapidly develop safe and efficacious vaccines. This has led to a number of ZIKV vaccine candidates that have shown significant promise in human clinical trials. These candidates include nucleic acid vaccines, inactivated vaccines, viral-vectored vaccines, and attenuated vaccines. Additionally, a number of vaccine candidates have been …

Moraxella bovoculi is the most frequently isolated bacteria from the eyes of cattle with Infectious Bovine Keratoconjunctivitis (IBK), also known as bovine pinkeye. Two distinct genotypes of M. bovoculi, genotype 1 and genotype 2, were characterized after whole genome sequencing showed a large degree of single nucleotide polymorphism (SNP) diversity within the species. To date, both genotypes have been isolated from the eyes of cattle without clinical signs of IBK while only genotype 1 strains have been isolated from the eyes of cattle with clinical signs of IBK. We used 38 known genotype 1 strains and 26 known genotype …

Evaluation Of A Photograph-Based Citizen Science Program As A Tick Surveillance And Prevention Education Tool, Heather Lynn Kopsco
Open Access Dissertations
The multitude of factors fueling the escalation in tick-borne disease in the United States is further complicated by gaps in public knowledge and inconsistent adherence to tick-bite prevention behaviors. Passive tick surveillance is a growing strategy to monitor tick species trends and associated pathogen prevalence across the country. Most practice involves citizens shipping encountered ticks to a laboratory for identification and pathogen testing, but given potential delays in mailing and processing time, tick bite victims may be placed outside the window of potential prophylactic options, or under unnecessary antibiotic administration. Recently, photographed-based tick surveillance has been shown to be a …

Pharmacokinetics, Pharmacodynamics And Dosing Regimen Of Long-Acting Oxytetracycline In Nile Tilapia (Oreochromis Niloticus), Kananuch Vasuntrarak
Chulalongkorn University Theses and Dissertations (Chula ETD)
Antimicrobial resistance has become a serious global problem and is steadily increasing worldwide. In aquaculture, there are limited antimicrobial options for treatment. Thus, there are growing needs for more specific dosing regimens of existing antimicrobial drugs that are not only to obtain therapeutic efficacy but also to minimize the resistance of pathogens. The purposes of this study were to determine pharmacokinetics (PK) of long-acting oxytetracycline (OTC) after intraperitoneal (IP) administration in Nile tilapia. One hundred and twenty healthy male tilapia (450±37.47 g) were divided into two experimental groups (60 fish/group). Each group received OTC-LA single IP injection at dosage of …

Evaluation Of The Relationship Between Centrality And Individual-Level Characteristics Among Pwid, Benjamin Skov
Open Access Master's Theses
Background: People who inject drugs (PWID) are a well-identified risk population for HIV infection. The risk networks of PWID have been implicated as possible modulators of both HIV risk and educational interventions among this population. In order to further understand the nature of risk networks, we examined how individual characteristics were associated with influential network position based on high closeness, betweenness, or eigenvector network centrality. These centrality measures assess an individual’s importance or potential to influence others based on their connections, closeness is based on proximity to others, betweenness on acting as an intermediary between others, and eigenvector on connection …

Synthesis And The Genetic Encoding Of Novel Lysine Post-Translational Modifications, Sahan A. Galbada Liyanage
Theses and Dissertations
Post-translational modifications (PTMs) play a significant role in regulating cell signaling and regulating the inner cellular environment. The study of PTMs and their primary biological functions is critical for the development of new medicines targeting the pathways and biomolecules responsible for the PTMs.
Previous studies revealed the direct coupling of the β -hydroxybutyryl lysine (KBHB) modification with the gene expression. KBHB in histone proteins links metabolism to epigenetics and gene expression and enables to study in-depth, how cell metabolism controls the gene expression and vice versa. Similar novel histone modification, KLac, attracts a special interest …

Honors Theses and Capstones
The largest recorded cholera epidemic in history is happening right now in Yemen, a country which has reported over 2 million cases of cholera as of 2020. Yemen has a history of endemic cholera, but prolonged conflict in the country has led to deteriorating conditions that have triggered massive outbreaks of the disease. The purpose of this study is to investigate the direct and indirect causes of the cholera epidemic in Yemen by proposing factors that may confer vulnerability and resilience in Yemeni governorates. Case studies were constructed for three governorates: Sana’a and the inner municipality of Amanat Al Asimah; …

Pre-exposure prophylaxis (PrEP) for HIV prevention may not only benefit the individual who uses it, but also their uninfected sexual risk contacts. We developed an agent-based model using a novel trial emulation approach to quantify disseminated effects of PrEP use among men who have sex with men in Atlanta, USA from 2015 to 2017. Components (subsets of agents connected through partnerships in a sexual network, but not sharing partnerships with any other agents) were first randomized to an intervention coverage level or control, then within intervention components, eligible agents were randomized to PrEP. We estimated direct and disseminated (indirect) effects …

Few studies have prospectively compared the relative transmissibility and propensity to cause disease of Mycobacterium tuberculosis Beijing strains with other human-adapted strains of the M. tuberculosis complex. We assessed the effect of Beijing strains on the risk for M. tuberculosis infection and disease progression in 9,151 household contacts of 2,223 culture-positive pulmonary tuberculosis (TB) patients in Lima, Peru. Child contacts exposed to Beijing strains were more likely than child contacts exposed to non-Beijing strains to be infected at baseline, by 12 months of follow-up, and during follow-up. We noted an increased but nonsignificant tendency for child contacts to develop TB. …
