Using Climate To Explain And Predict West Nile Virus Risk In Nebraska, Kelly Helm Smith, Andrew J. Tyre, Jeff Hamik, Michael J. Hayes, Yuzhen Zhou, Li Dai

National Drought Mitigation Center: Faculty Publications

We used monthly precipitation and temperature data to give early warning of years with higher West Nile Virus (WNV) risk in Nebraska. We used generalized additive models with a negative binomial distribution and smoothing curves to identify combinations of extremes and timing that had the most influence, experimenting with all combinations of temperature and drought data, lagged by 12, 18, 24, 30, and 36 months. We fit models on data from 2002 through 2011, used Akaike's Information Criterion (AIC) to select the best‐fitting model, and used 2012 as out‐of‐sample data for prediction, and repeated this process for each successive year, …

Interactions Of Photosensitizers With Model Membranes And Their Applications In Photodynamic Therapy, Poornima Kalyanram Aug 2020

Interactions Of Photosensitizers With Model Membranes And Their Applications In Photodynamic Therapy, Poornima Kalyanram

Theses

Cancer is caused by the uncontrolled growth and spread of abnormal cells resulting in 1 in 6 deaths every day. The most common treatment methods include surgery, chemotherapy, and radiation. These techniques are invasive, aggressive, and non-specific to cancer cells. Therefore, alternative therapies which are both potent and does not interfere with the quality of living are on a rise. Photodynamic Therapy (PDT) is an approved alternative remedy in the treatment of cancer. The efficacy of PDT depends on the penetration power of the photosensitizer (PS) when injected into to the site of tumor. However, due to complexity involved in …

Valproic Acid Leads To An Increase In Ros Generation By Inhibiting The Deacetylation Of Mitochondrial Sod, Stephen Marc Lucas Aug 2020

Valproic Acid Leads To An Increase In Ros Generation By Inhibiting The Deacetylation Of Mitochondrial Sod, Stephen Marc Lucas

Theses and Dissertations

Valproic Acid Promotes Acetylation of Superoxide Dismutase-2 During Neurogenesis. Valproic acid (VPA) is a known developmental toxicant associated with a high prevalence of neural tube defects (NTD). The mechanism of VPA-induced NTD is unclear, but oxidative stress may be implicated. To understand how embryotoxic oxidative stress may occur, we measured superoxide dismutase (SOD) activity following VPA treatment in the embryonic pluripotent P19 mouse carcinoma cell line. In undifferentiated P19 cultures treated with VPA (5 mM), dichlorofluorescein fluorescence increased 15% compared to untreated controls over 20 min, indicating a modest, yet statistically significant increase in ROS generation. Undifferentiated P19 cells were …

Discovery Of A Novel Regulatory Mechanism Of Tnk1 By 14-3-3 And Its Ubiquitin Association Domain Provides A Potential Therapeutic Targeting Opportunity In Cancer, Tsz Yin Chan Aug 2020

Discovery Of A Novel Regulatory Mechanism Of Tnk1 By 14-3-3 And Its Ubiquitin Association Domain Provides A Potential Therapeutic Targeting Opportunity In Cancer, Tsz Yin Chan

Theses and Dissertations

While a relatively limited number of known oncogenes underlie a large percentage of cancers, a variety of new genes have emerged as low-frequency cancer drivers. Each of these new oncogenes represents a frontier for targeted therapy. However, the discovery of low-frequency targetable oncogenic drivers is challenging. This study focuses on the poorly understood Tyrosine kinase non-receptor-1 (TNK1), which has been reported to have both oncogenic and tumor suppressive functions. TNK1 has been identified to promote cancer cells survival and promote chemoresistance in multiple independent studies. On the other hand, whole-body constitutive deletion of TNK1 in mice caused an increase in …

Imidacloprid Movement Into Fungal Conidia Is Lethal To Mycophagous Beetles, Robin A. Choudhury, Andrew M. Sutherland, Matt J. Hengel, Michael P. Parrella, W. Douglass Gubler

School of Earth, Environmental, & Marine Sciences Faculty Publications

Applications of systemic pesticides can have unexpected direct and indirect effects on nontarget organisms, producing ecosystem-level impacts. We investigated whether a systemic insecticide (imidacloprid) could be absorbed by a plant pathogenic fungus infecting treated plants and whether the absorbed levels were high enough to have detrimental effects on the survival of a mycophagous beetle. Beetle larvae fed on these fungi were used to assess the survival effects of powdery mildew and imidacloprid in a factorial design. Fungal conidia were collected from treated and untreated plants and were tested for the presence and concentration of imidacloprid. The survival of beetles fed …

Aerobic Exercise Alters Opioid Receptors Following Chronic Alcohol Exposure, James N. Brundage Aug 2020

Aerobic Exercise Alters Opioid Receptors Following Chronic Alcohol Exposure, James N. Brundage

Theses and Dissertations

Opioid receptors have been a target of pharmacological manipulation in alcohol use disorder (AUD) recovery protocols for many years. Aerobic exercise, a common adjunct in AUD recovery, is known to modulate opioid receptors (ORs) both during both acute and long term exposure. The three subtypes of ORs: mu (MOR), delta (DOR), and kappa (KOR) are all expressed on neurons in the mesocorticolimbic circuitry. Kappa-opioid receptors are expressed directly on dopamine (DA) neuron terminals in the nucleus accumbens (NAc). Mu and Delta ORs are expressed on cholinergic interneurons (CINs) and GABA neurons in the NAc. In alcohol dependent rodents, KORs are …

Analyzing Metacommunity Models With Statistical Variance Partitioning: A Review And Meta-Analysis, Kevin Vieira Lamb Aug 2020

Analyzing Metacommunity Models With Statistical Variance Partitioning: A Review And Meta-Analysis, Kevin Vieira Lamb

Theses and Dissertations

The relative importance of deterministic processes versus chance is one of the most important questions in science. We analyze the success of variance partitioning methods used to explain variation in β-diversity and partition it into environmental, spatial, and spatially structured environmental components. We test the hypotheses that 1) the number of environmental descriptors in a study would be positively correlated with the percentage of β-diversity explained by the environment, and that the environment would explain more variation in β-diversity than spatial or shared factors in VP analyses, 2) increasing the complexity of environmental descriptors would help account for more of …

Sarm1 Acts Downstream Of Neuroinflammatory And Necroptotic Signaling To Induce Axon Degeneration, Kwang Woo Ko, Jeffrey Milbrandt, Aaron Diantonio Aug 2020

Sarm1 Acts Downstream Of Neuroinflammatory And Necroptotic Signaling To Induce Axon Degeneration, Kwang Woo Ko, Jeffrey Milbrandt, Aaron Diantonio

Open Access Publications

Neuroinflammation and necroptosis are major contributors to neurodegenerative disease, and axon dysfunction and degeneration is often an initiating event. SARM1 is the central executioner of pathological axon degeneration. Here, we demonstrate functional and mechanistic links among these three pro-degenerative processes. In a neuroinflammatory model of glaucoma, TNF-α induces SARM1-dependent axon degeneration, oligodendrocyte loss, and subsequent retinal ganglion cell death. TNF-α also triggers SARM1-dependent axon degeneration in sensory neurons via a noncanonical necroptotic signaling mechanism. MLKL is the final executioner of canonical necroptosis; however, in axonal necroptosis, MLKL does not directly trigger degeneration. Instead, MLKL induces loss of the axon survival …

Men, Women And Witchcraft: The Feminist Reclamation Of The Witch In The Modern Horror Film, Brian Joseph Hadsell Aug 2020

Men, Women And Witchcraft: The Feminist Reclamation Of The Witch In The Modern Horror Film, Brian Joseph Hadsell

Theses and Dissertations

The witch as a figure possesses a powerful and enduring legacy in Euro-American culture; she is both a victim of patriarchal persecution and the natural enemy of a deeply-gendered society. Recent horror films that employ the witch, however, have generally done so in the form of reclaimed feminist icon: a violently retributive figure avenging the wrongs done to women both past and present. The purpose of this research is to provide insight about our acutely gendered society and culture in the times during and preceding the #MeToo movement through a semiological analysis of three recent horror films that center on …

Faculty, Staff and Students Publications

Lysinuric protein intolerance (LPI) is an inborn error of cationic amino acid (arginine, lysine, ornithine) transport caused by biallelic pathogenic variants in SLC7A7, which encodes the light subunit of the y+LAT1 transporter. Treatments for the complications of LPI, including growth failure, renal disease, pulmonary alveolar proteinosis, autoimmune disorders and osteoporosis, are limited. Given the early lethality of the only published global Slc7a7 knockout mouse model, a viable animal model to investigate global SLC7A7 deficiency is needed. Hence, we generated two mouse models with global Slc7a7 deficiency (Slc7a7em1Lbu/em1Lbu; Slc7a7Lbu/Lbu and Slc7a7em1(IMPC)Bay/em1(IMPC)Bay; Slc7a7Bay/Bay) using CRISPR/Cas9 technology by introducing a deletion of exons …

Assessment Of Agricultural Advisory Messages From Farmer-To-Farmer In Making A Case For Scaling Up Production: A Qualitative Study, Nana Afranaa Kwapong, Daniel Adu Ankrah, Dominic Boateng-Gyambiby, Joseph Asenso-Agyemang, Lydia Oteng Fening

The Qualitative Report

Inadequate access to agricultural extension services often results in poor farm practices, affecting yields and subsequently the income and wellbeing of smallholder farmers. Given the high demand for agricultural information and the limited capacity of extension services, a farmer-to-farmer extension approach has been explored by many underserved farmers. In this study, we use a qualitative case study approach explore how cassava farmers who had limited access to agricultural advisory services from public extension agents managed to up-scale their farming business. Our research question was: what lessons can be learned from the lived experience of these farmers to address current challenges …
