Metabolic Engineering Of Cyanobacteria For Production Of Chemicals, Po-Cheng Lin
Metabolic Engineering Of Cyanobacteria For Production Of Chemicals, Po-Cheng Lin
McKelvey School of Engineering Graduate Student Theses & Dissertations
Concerns over the impact of climate change caused by CO2 emission have driven the research and development of renewable energies. Microbial production of chemicals is being viewed as a feasible approach to reduce the use of fossil fuels and minimize the impact of climate change. With recent advances in synthetic biology, microorganisms can be engineered to synthesize petroleum-based chemicals and plant-derived compounds. Cyanobacteria are photosynthetic prokaryotes that use only sunlight, CO2, and trace minerals for growth. Compared to other microbial hosts, cyanobacteria are attractive platforms for sustainable bioproduction, because they can directly convert CO2 into products. However, the major challenge …

Joint Reconstruction For Single-Shot Edge Illumination Phase-Contrast Tomography (Eixpct), Yujia Chen
McKelvey School of Engineering Graduate Student Theses & Dissertations
Edge illumination X-ray phase-contrast tomography (EIXPCT) is an emerging X-ray phasecontrast tomography technique for estimating the complex-valued X-ray refractive index distribution of an object with laboratory-based X-ray sources. Conventional image reconstruction approaches for EIXPCT require multiple images to be acquired at each tomographic view angle. This contributes to prolonged data-acquisition times and elevated radiation doses, which can hinder in vivo applications. In this dissertation, a new “single-shot” method without restrictive assumptions related to the object, imaging geometry or hardware is proposed for joint reconstruction (JR) of the real and imaginary-valued components of the refractive index distribution from a tomographic data …

Mechanosensitive Epithelial Cell Scattering And Migration On Layered Matrices, Christopher Michael Walter
Mechanosensitive Epithelial Cell Scattering And Migration On Layered Matrices, Christopher Michael Walter
McKelvey School of Engineering Graduate Student Theses & Dissertations
Epithelial cells form multi-layered tissue scaffolding that makes up every organ in the body. Along with epithelial cells, the basement membrane (BM) and connective tissue are composed of various proteins that sculpt the organs and protect them from foreign macromolecules. Epithelial cells respond to various cues, both chemical and mechanical, from their surrounding matrices to aid in maintenance and repair of these layers through degradation and deposition of extracellular matrix (ECM) proteins. In cancer progression, epithelial cells lose their normal function of supporting tissue structure and instead adopt more aggressive behaviors through an epithelial-to-mesenchymal transition (EMT) of their cellular traits. …

There are a wealth of tools for fitting linear models at each location in the brain in neuroimaging analysis, and a wealth of genetic tools for estimating heritability for a small number of phenotypes. But there remains a need for computationally efficient neuroimaging genetic tools that can conduct analyses at the brain-wide scale. Here we present a simple method for heritability estimation on twins that replaces a variance component model-which requires iterative optimisation-with a (noniterative) linear regression model, by transforming data to squared twin-pair differences. We demonstrate that the method has comparable bias, mean squared error, false positive risk, and …

The Impact Of Demographics In The Equity Of Global Literacy Education, Saumendra Nath De
The Reading Professor
The right to education has been recognized as a basic human right in a number of international conventions including the 1948 Universal Declaration of Human Rights. The issues of fairness, equal opportunity, and equity in education, however, continue to plague the educational world. According to the UNESCO Institute for Statistics (2017), there are 750 million illiterate adults in the world, six out of ten children and adolescents are not learning globally, and low-income countries are home to a disproportionately large share of children and adolescents not learning. A plethora of demographic factors affects equity in education. This article discusses how …

Hemocompatibility remains a challenge for injectable and/or implantable medical devices, and thromboresistant coatings appear to be one of the most attractive methods to down-regulate the unwanted enzymatic reactions that promote the formation of blood clots. Among all polymeric materials, polyurethanes (PUs) are a class of biomaterials with excellent biocompatibility and bioinertness that are suitable for the use of thromboresistant coatings. In this work, we investigated the thermal and physico-mechanical behaviors of ester-based and ether-based PU films for potential uses in thromboresistant coatings. Our results show that poly(ester urethane) and poly(ether urethane) films exhibited characteristic peaks corresponding to their molecular configurations. …

Background: Epigenetic clocks are mathematical models that predict the biological age of an individual using DNA methylation data and have emerged in the last few years as the most accurate biomarkers of the aging process. However, little is known about the molecular mechanisms that control the rate of such clocks. Here, we have examined the human epigenetic clock in patients with a variety of developmental disorders, harboring mutations in proteins of the epigenetic machinery. Results: Using the Horvath epigenetic clock, we perform an unbiased screen for epigenetic age acceleration in the blood of these patients. We demonstrate that loss-of-function mutations …

Social learning is ubiquitous across the animal kingdom, where animals learn from group members about predators, foraging strategies, and so on. Despite its prevalence and adaptive benefits, our understanding of social learning is far from complete. Here, we study observational learning in zebrafish, a popular animal model in neuroscience. Toward fine control of experimental variables and high consistency across trials, we developed a novel robotics-based experimental test paradigm, in which a robotic replica demonstrated to live subjects the correct door to join a group of conspecifics. We performed two experimental conditions. In the individual training condition, subjects learned the correct …

Type 2 diabetes mellitus (T2DM) is a nation-wide prevalent chronic condition, which includes direct and indirect healthcare costs. T2DM, however, is a preventable chronic condition based on previous clinical research. Many prediction models were based on the risk factors identified by clinical trials. One of the major tasks of the T2DM prediction models is to estimate the risks for further testing by HbA1c or fasting plasma glucose to determine whether the patient has or does not have T2DM because nation-wide screening is not cost-effective.
Those models had substantial limitations on data quality, such as missing values. In this dissertation, I …

Background: Calls for science education reform have been made for decades in the USA. The recent call to produce one million new science, technology, engineering, and math (STEM) graduates over 10 years highlights the need to employ evidence-based instructional practices (EBIPs) in undergraduate STEM classes to create engaging and effective learning environments. EBIPs are teaching strategies that have been empirically demonstrated to positively impact student learning, attitudes, and achievement in STEM disciplines. However, the mechanisms and processes by which faculty learn about and choose to implement EBIPs remain unclear. To explore this problem area, we used social network analysis to …

Schooling fishes, like flocking birds and swarming insects, display remarkable behavioral coordination. While over 25% of fish species exhibit schooling behavior, nighttime schooling has rarely been observed or reported. This is due to vision being the primary modality for schooling, which is corroborated by the fact that most fish schools disperse at critically low light levels. Here we report on a large aggregation of the bioluminescent flashlight fish Anomalops katoptron that exhibited nighttime schooling behavior during multiple moon phases, including the new moon. Data were recorded with a suite of low-light imaging devices, including a high-speed, high-resolution scientific complementary metal-oxide-semiconductor …

Introduction
Laboratory rat welfare is critically influenced by laboratory animal personnel through their implementation, or lack of implementation, of various enrichment techniques. One such promising technique is heterospecific play, or “rat tickling”, which mimics aspects of rat rough-and-tumble play and can contribute to improving welfare, but may be infrequently implemented. The theory of planned behavior can be used to study implementation by measuring intentions and beliefs about rat tickling, including behavioral attitudes (whether it is good or bad), subjective norms (whether there is social/professional pressure to provide it), and control beliefs (whether they feel in control of providing it). Therefore, …

Adventures In Structureland: Exploring Protein Conformational Plasticity By Nmr, Anne R. Kaplan
Doctoral Dissertations
Proteins are complex macromolecules that play a critical role in all biological processes. Their ability to change shape, or conformation, in response to environmental factors is intimately linked with function. Thus, understanding the mechanisms underlying conformational changes is imperative for complete understanding of protein function(s), and an overall comprehension of biological systems. This work explores the structural and dynamic attributes of conformational exchange in the C-terminal domain of pore-forming toxin Hemolysin II (HlyIIC), and the coiled-coil leucine zipper, GCN4p, using nuclear magnetic resonance (NMR). The HlyIIC domain contains a sole proline at position 405 that leads to equal populations of …
