Pyruvate kinase M2 (PKM2) is an alternatively spliced variant, which mediates the conversion of glucose to lactate in cancer cells under normoxic conditions, known as the Warburg effect. Previously, we demonstrated that PKM2 is one of 97 genes that are overexpressed in non-small-cell lung cancer (NSCLC) cell lines. Herein, we demonstrate a novel role of subcellular PKM2 expression as a biomarker of therapeutic response after targeting this gene by shRNA or small molecule inhibitor (SMI) of PKM2 enzyme activity in vitro and in vivo. This project examines the cultural differences and similarities between middle class professional fathers in the U.S. and Sweden. These men face extremely different state structured programs, which may or may not provide benefits to ease the pressures of the early years of parenthood. We might expect that differences in welfare state policies result in differences in how people experience parenthood. On the other hand, some welfare state policies of more recent origin may reflect common gendered dynamics. Although experiencing different cultural norms and structures, these fathers’ experiences look relatively similar, their experiences with the family, their descriptions of their role …

Sustenance of the growing world population calls for increased agricultural production. However, this will have to be done while forecasts of water withdrawals on a global scale predict sharp increases in future demand to meet human needs. The inadequacy of irrigation water supplies has led to the need to consider deficit irrigation (DI) as a water saving strategy. DI is a deliberate under-application of water to growing crops.

A patellar-tendon-bearing (PTB) bar is a common design feature used in the socket of trans-tibial prostheses to place load on the pressure-tolerant tissue. As the patellar tendon in the residual limb is subjected to the perpendicular compressive force not commonly experienced in normal tendons, it is possible for tendon degeneration to occur over time. The purpose of this study was to compare patellar tendon morphology and neovascularity between the residual and intact limbs in trans-tibial amputees and healthy controls. Testing — And Mostly Rejecting — The Folk Wisdom Of The Effective Appellate Brief, Steven R. Morrison, Brian Darby

There is a great deal of folk wisdom regarding how to draft an effective appellate brief. Judges and lawyers offer advice that briefs should be short, should present relatively few issues, should always be followed by a reply brief, and so forth. There is little doubt that aspiring appellate advocates, law professors who teach writing, appellate court clerks, and appellate court judges look to this folk wisdom to learn how to write effective appellate briefs, teach the skill, and evaluate, by proxies, which briefs are likely to be the best.

Undergraduate research opportunities have expanded from elite universities in the United States to universities and learning institutions of all ranks and sizes. Research studies have showed some positive outcomes of the research experience for undergraduates (REU), such as enhanced research skills and competencies. However, with the widespread implementation of REU programs across the country, there are some serious and challenging issues, such as fierce competition among students for limited participation opportunities and a overlooking of underrepresented students’ needs. Sensation seeking has been identified as a significant risk factor for adolescent alcohol use. Little is known, however, about the process by which sensation seeking impacts heavy alcohol use. The current study used structural equation modeling (SEM) to examine the relationships among sensation seeking, age of drinking initiation, and heavy alcohol use in a sample of high school seniors (N = 221). Results supporting age of drinking initiation as a mediator of the relationship between sensation seeking and heavy alcohol use. Implications include providing personality-targeted prevention to adolescents who display sensation seeking traits to delay drinking initiation among these …

Steven Yao has called H.D.'s Ion her 'most ambitious feat of translation' (2002a: 83). Two contexts are relevant for thinking about H.D.'s work on this project. One is psychoanalysis, and the other is the scholarship which interprets myth as a narrative reflection of ritual practice. Both contexts are significantly tied to H.D.'s personal life and writing career. Matte Robinson even claims that one of the major characters in the play, Kreousa, 'becomes an extension of H.D.' (2013: 270). This claim may be overstated, but Kreousa's quest for recognition from Apollo does resemble H.D's effort to supply her daughter, Perdita, with …

Mechanical ventilation (MV) is a life-saving intervention for many critically ill patients. Unfortunately, prolonged MV results in rapid diaphragmatic atrophy and contractile dysfunction, collectively termed ventilator-induced diaphragm dysfunction (VIDD). Recent evidence reveals that endurance exercise training, performed prior to MV, protects the diaphragm against VIDD. While the mechanism(s) responsible for this exercise-induced protection against VIDD remain unknown, increased diaphragm antioxidant expression may be required. To investigate the role that increased antioxidants play in this protection, we tested the hypothesis that elevated levels of the mitochondrial antioxidant enzyme superoxide dismutase 2 (SOD2) is required to achieve exercise-induced protection against VIDD. Instructors of postsecondary classes in statistics rely heavily on visuals in their teaching, both within the classroom and in resources like textbooks, handouts, and software, but this information is often inaccessible to students who are blind or visually impaired (BVI). The unique challenges involved in adapting both pedagogy and course materials to accommodate a BVI student may provoke anxiety among instructors teaching a BVI student for the first time, and instructors may end up feeling unprepared or “reinventing the wheel.” We discuss a wide variety of accommodations inside and outside of the classroom grounded in the empirical literature on cognition …

Mitochondria are critical for energy production and cell signaling, possessing their own DNA (mtDNA) which encodes essential proteins for oxidative phosphorylation (OXPHOS). Lack of mtDNA stability, i.e. diminished mtDNA quality and quantity, is implicated in many diverse diseases and are encompassed under the term mtDNA depletion syndrome (MDS). Two major disease loci are OPA1 and POLG. OPA1 is needed for mitochondrial quality control and mtDNA stability, and POLG is the sole replicative DNA polymerase for mtDNA replication and repair. Current models for pathologies resulting from mutations at these loci do not capture the wholebody aspect of mutations nor the early …

Groundwater below many basins that have been used to store coal combustion residuals (CCRs) may have concentrations of certain inorganic constituents in excess of applicable regulatory standards. Given that many inorganic CCR constituents are also naturally occurring in the aquifer matrix, their detection may not always be indicative of a release from the CCR unit. Inorganics may be present at elevated levels in groundwater due to natural or background conditions, or due to changes to the aquifer geochemistry resulting from the presence of the CCR basin, which then mobilizes these naturally occurring constituents beneath the CCR unit. Manufactured coal ash aggregates (MAs) are a 2:1 (w/w) solidified composite of fly and bottom ash. The current study assessed the feasibility of beneficial utilization of MAs as a subsoil substitute for open-pit restoration to phyto-viable land. A series of indoor and outdoor experiments examined Phaselous vulgaris growth under the influence of MAs in the soil environment. With the MAs layer below the topsoil, P. vulgaris showed enhanced growth with respect to shoot height, leaf number, and leaf chlorophyll intensity. Similar growth enhancement was observed when the MAs were mixed with topsoil or applied as a micronutrient source over the …

The objective of the present study was to determine the effectiveness of the opioid antagonist naltrexone at reducing the consumption of EtOH and nicotine in female alcohol-preferring (P) rats. P rats have been selectively bred to have a genetic predisposition for alcohol abuse, which allows them to be used as an animal model of alcoholism. P rats readily self-administer i.v nicotine (Le et al., 2016), have EtOH consumption during adolescence that is similar to that seen in adulthood, and operantly respond for EtOH until they are impaired/intoxicated (Bell et al., 2006). Background and Objectives: Recognizing the important role that dementia-specific adult day centers have in maintaining persons with a neurocognitive disorder in their home, this article examines three critical indicators at the time when people first enroll in such a center: cognitive and functional impairment of the enrollee, and burden reported by their family caregivers. We also considered variations in these 3 indicators by race/ethnicity and by the relationship of caregiver to the new enrollee.

The process of aging is accompanied by development of chronic disease, reduced physical function, and increased risk of mortality. Timely stalling and resumption of RNA polymerases at damaged chromatin are actively regulated processes. Prior work showed an importance of FACT histone chaperone in such process. Here we provide a new role of OTUD5 deubiquitinase in the FACT-dependent process. Through a DUB RNAi screen, we found OTUD5 as a specific stabilizer of the UBR5 E3 ligase. OTUD5 localizes to DNA double strand breaks (DSBs), interacts with UBR5 and represses the RNA Pol II elongation and RNA synthesis. OTUD5 co-localizes and interacts with the FACT component SPT16 and antagonizes the histone H2A deposition at DSB lesions. OTUD5 interacts with UBR5 and …

- Ecologists use classifications of individuals in categories to understand composition of populations and communities. These categories might be defined by demo- graphics, functional traits, or species. Assignment of categories is often imperfect, but frequently treated as observations without error. When individuals are observed but not classified, these “partial” observations must be modified to include the missing data mechanism to avoid spurious inference.

Microbial pathogens and indicators have contributed to major part of water quality degradation in the United States. Located in the northwestern New Jersey, the Musconetcong River has been included in the New Jersey Impaired Waters List or the 303(d) List due to high concentrations of fecal indicator bacteria. Hence, a Total Maximum Daily Load plan was established to address microbial water quality issues in the watershed.
