Epithelial oncogenesis is believed to be generally associated with the accumulation over time of an increasing number of mitotic errors until a threshold number of mutations required for the initiation of cancer is achieved. Preemption of cancer through the morphologic detection of dysplastic cells, i.e. cells with a number of mitotic errors that are still below the threshold for cancer, followed by their surgical removal or eradication, has had an enormous impact on reducing the incidence of cancer of the uterine cervix, skin and colon worldwide, but this strategy has been much less successful with cancers in most other body …
Development Of An Spr Method To Monitor The Chemical Interaction Of Dna With A Small Reactive Molecule: Peroxynitrite As An Example, Divya Sharma
ETD Archive
Peroxynitrite is an anion with the formula ONOO-. It is an unstable structural isomer of nitrate, NO3-. Although its conjugate acid (ONOOH) is highly reactive, it is quite relatively stable in basic solutions. Generation of peroxynitrite in-vivo occurs through the diffusion-controlled reaction between superoxide anion and nitric oxide free radical. It is a strong oxidizing and nitrating agent, and its formation has been correlated with many pathological conditions. Because of its oxidizing properties, peroxynitrite can damage a wide array of molecules in cells, including DNA and proteins. Reactions of peroxynitrite with DNA, proteins, and lipids, trigger a wide array of …

Liquid Chromatography-Mass Spectrometry is an advanced analytical technique that offers high sensitivity and specificity and has been increasingly used for analysis of a wide variety of compounds including clinically and pharmacologically relevant molecules. In this dissertation we describe qualitative and quantitative liquid chromatographic mass spectrometric methods to analyze both small molecules and larger macromolecules that provide useful insights into diagnosis and management of several diseases. An LCMS(/MS) analytical method includes extraction of analytes of interest from the matrix followed by liquid chromatographic separation and mass spectrometric detection. Chapter I describes pre-analytical workflows and sample pretreatment techniques and theories underlying LC-MS …
Role Of Tumor Necrosis Factor-Stimulated Gene-6 In Cutaneous Wound Healing And Inflammation, Sajina Shakya
Role Of Tumor Necrosis Factor-Stimulated Gene-6 In Cutaneous Wound Healing And Inflammation, Sajina Shakya
ETD Archive
Controlled inflammation is crucial for normal wound healing. Our main aim in this study was to investigate the effect of loss of tumor necrosis factor-stimulated gene-6 (TSG-6) in cutaneous wound closure and inflammation. TSG-6 by its enzymatic action modifies the extracellular matrix molecule, hyaluronan (HA), through the transfer of heavy chain (HC) proteins from inter-a-trypsin inhibitor to form HC-HA complexes. Both TSG-6 and HC-HA have been associated with inflammation. Here, we showed that loss of endogenous TSG-6 and HC-HA in TSG-6 null mice results in significantly delayed wound closure and differential neutrophil recruitment compared to wildtype mice. Both of these …

It has been examined that poor access to nutritious foods and limited opportunities for physical activity is prevalent among children from underserved families. These deficiencies contribute to increasing childhood obesity rates and emphasize the need for early interventions. Establishing nutritional literacy and understanding the importance of an active lifestyle in early childhood can lead to long-term health benefits and reduction in chronic disease later in life (HHS & USDA, 2015; Tsai et al., 2011; Wang et al., 2011). The InSports Foundation serves as a comprehensive non-profit organization that addresses many of these issues by providing children from underserved families with …

Prevalence Of Mental Health Stigma Among Practicing Pas In Regard To Personal Experience And Patient Exposure, Bailey J. Hanson, Allison Hoy, Madeline Lieberson
All Electronic Theses and Dissertations
Stigmatizing attitudes can be destructive to both the emotional and physical wellbeing of individuals living with mental illness. In fact, individuals with a mental illness are more likely to be seen as responsible for their illness and more likely to be falsely accused of a violent crime (Rüsch, Angermeyer, & Corrigan, 2005). Current research on mental health stigma in healthcare has involved a variety of occupations, but published research has yet to examine stigma held by physician assistants (PAs). This research study attempted to answer the following question: What effect, if any, does personal experience with a family member, friend, …

Technological advancements enable collecting vast data, i.e., Big Data, in science and industry including biomedical field. Increased computational power allows expedient analysis of collected data using statistical and machine-learning approaches. Historical data incompleteness problem and curse of dimensionality diminish practical value of pure data-driven approaches, especially in biomedicine. Advancements in deep learning (DL) frameworks based on deep neural networks (DNN) improved accuracy in image recognition, natural language processing, and other applications yet severe data limitations and/or absence of transfer-learning-relevant problems drastically reduce advantages of DNN-based DL. Our earlier works demonstrate that hierarchical data representation can be alternatively implemented without NN, …
On The Interpretation Of The Normalization Constant In The Scaling Equation, Karl J. Niklas, Sean T. Hammond
On The Interpretation Of The Normalization Constant In The Scaling Equation, Karl J. Niklas, Sean T. Hammond
Earth System Science and Policy Faculty Publications
The scaling equation, Y1 = βYα2, has been used empirically and explored theoretically primarily to determine the numerical value and meaning of the scaling exponent, α. The mathematical interpretation of α is clear—it is the quotient of the relative rate of change of Y1 with respect to the rate of change of Y2. In contrast, the interpretation of the normalization constant, β, is obscure, so much so that some workers have rejected the idea that it has any biological importance. With the notable exception of Steven J. Gould's early work, Huxley's dismissal of β largely relegated the study of its …

Rapid, efficient and accurate nucleic acid molecule detection is important in the screening of diseases and pathogens, yet remains a limiting factor at point of care (POC) treatment. Microfluidic systems are characterized by fast, integrated, miniaturized features which provide an effective platform for qualitative and quantitative detection of nucleic acid molecules. The nucleic acid detection process mainly includes sample preparation and target molecule amplification. Given the advancements in theoretical research and technological innovations to date, nucleic acid extraction and amplification integrated with microfluidic systems has advanced rapidly. The primary goal of this review is to outline current approaches used for …

On many public lands in the Great Plains region of the USA and Canada, cattail (Typha spp.) growth has far exceeded the 50:50 distribution recommended for optimum wetland wildlife habitat. Excessive cattail growth is the primary concern of wetland managers and its integrated management is reviewed here. The coverage of this mostly hybrid cattail (T. latifolia × T. angustifolia) is often over 90 % and if partially removed for habitat enhancement represents a substantial biomass resource in sites such as conservation wetlands, water retention basins and roadside drainage ditches. Available biomass is estimated to be 3,000 kg/ha assuming a 50 …

Implications Of Channel Catfish Movement In An Internationally Managed System, Henry Hansen
School of Natural Resources: Dissertations, Theses, and Student Research
The large-scale movements and spatial behavior of channel catfish in the Red River of the North, have direct interactions with geopolitics, anthropogenic structures, and ecosystems. Investigating the spatial dynamics and exploitation of this mobile and internationally managed fish species provides opportunities for resource managers to design evidence-based policy for the diverse interest groups that utilize the fishery. My thesis comprised two parts: 1) characterize systemwide movement and survival patterns using mark-recapture methods and acoustic telemetry and 2) project the interaction of hypothetical exploitation scenarios and alternative movement methodologies to assess the fishery from an ecosystem service flow perspective. Channel catfish …

From the earliest days of the web, users have been aware of the fickleness of linking to content. In some ways, 1998 was a simpler time for the Internet. In other ways, like basic website design principles, everything old is new again. Jakob Nielson, writing “Fighting Linkrot” in 1998, reported on a then-recent survey that suggested 6% of links on the web were broken. The advice then hasn’t changed: run a link validator on your site regularly, and update or remove broken links. Also set up redirects for links that do change. The mantra for Nielson was “you are not …

In the U.S. Great Plains (GP), diagnosing precipitation variability is key in developing an understanding of the present and future availability of water in the region. Building on previous work investigating U.S. GP pluvial years, this study usesERAtwentieth century (ERA-20C) reanalysis data to investigate key circulation anomalies driving GP precipitation anomalies during a subset of GP pluvial years (called in this paper Pattern pluvial years). With previous research showing links between tropical Pacific sea surface temperature (SST) anomalies and GP climate variability, this study diagnoses the key circulation anomalies through an analysis of SSTs and their influence on the atmosphere. …

Bacterial resistance to antibiotics is a growing public health threat. Each year, ~2 million infections and 23,000 deaths are caused by antibiotic resistance in the US. 1 Agents targeting the essential bacterial type II topoisomerases DNA gyrase and topoisomerase IV, as exemplified by the fluoroquinolones (FQs), have been shown to be useful antibiotics. FQs such as ciprofloxacin, moxifloxacin, and levofloxacin, to name a few, are broadspectrum antibacterial agents that have been in clinical use since the 1980's. Unfortunately, heavy use of the FQs has inevitably led to the development of FQresistant pathogens. 2 A gyrase inhibitor lead compound called QPT-1 …

In 2013, Plaster Creek Stewards built Kreiser Pond, a green infrastructure bioswale in East Grand Rapids that flows into Plaster Creek. It has reduced the creek’s water flow, making the creek less prone to flooding. A similar green infrastructure project was completed at Shadyside Park in Dutton in 2019. At Shadyside Park, a floodplain was built to slow the flow of the creek when there is heavy rainfall and prevent erosion caused by flooding.1 Plaster Creek’s flow rate isn’t the only concern. There is also significant microbial (bacterial and viral) contamination in Plaster Creek.2 This project investigated whether the green …
