Efforts to enhance the utility of photodynamic therapy as a non-invasive method for treating certain cancers have often involved the design of dye sensitizers with increased singlet oxygen efficiency. More recently, however, sensitizers with greater selectivity for tumor cells than surrounding tissue have been targeted. The present study provides an approach to the modification of the known photosensitizer zinc phthalocyanine (ZnPc), to enhance its solubility and delivery to cancer cells. Targeting a photosensitizer to the site of action improves the efficacy of the sensitizer in photodynamic therapy. In this work we used PLGA-b-PEG to encapsulate a new zinc phthalocyanine derivative, …

Absence Of Endothelial Α5Β1 Integrin Triggers Early Onset Of Experimental Autoimmune Encephalomyelitis Due To Reduced Vascular Remodeling And Compromised Vascular Integrity, Ravi Kant, Sebok K. Halder, Gregory J. Bix, Richard Milner
Sanders-Brown Center on Aging Faculty Publications
Early in the development of multiple sclerosis (MS) and its mouse model experimental autoimmune encephalomyelitis (EAE), vascular integrity is compromised. This is accompanied by a marked vascular remodeling response, though it is currently unclear whether this is an adaptive vascular repair mechanism or is part of the pathogenic process. In light of the well-described angiogenic role for the α5β1 integrin, the goal of this study was to evaluate how genetic deletion of endothelial α5 integrin (α5-EC-KO mice) impacts vascular remodeling and repair following vascular disruption during EAE pathogenesis, and how this subsequently influences clinical progression and inflammatory demyelination. Immunofluorescence staining …

The roles of retinoids in nonalcoholic fatty liver disease (NAFLD) remain unclear and a better understanding may lead to therapies that prevent or limit NAFLD progression. We examined the actions of retinoic acid receptor (RAR) agonists- AM80 for RARαand AC261066 for RARβ2- in a murine model of NAFLD. We fed wild type C57Bl/6 mice a chow or a 45% high fat diet (HFD) for 12 weeks, followed by 4 additional weeks with the HFD+AM80; HFD +AC261066; or HFD. The HFD+AM80 group showed greater hyperglycemia and glucose intolerance compared to other groups. Histopathological evaluation of the livers showed the highest degree …

A Comparison Of Hyperelastic Constitutive Models Applicable To Shear Wave Elastography (Swe) Data In Tissue-Mimicking Materials, David P. Rosen, Jingfeng Jiang
Department of Biomedical Engineering Publications
Shear wave elastography (SWE) techniques have received substantial attention in recent years. Strong experimental data in SWE suggest that shear wave speed changes significantly due to the known acoustoelastic effect (AE). This presents both challenges and opportunities toward in vivo characterization of biological soft tissues. In this work, under the framework of continuum mechanics, we model a tissue-mimicking material as a homogeneous, isotropic, incompressible, hyperelastic material. Our primary objective is to quantitatively and qualitatively compare experimentally measured acoustoelastic data with model-predicted outcomes using multiple strain energy functions. Our analysis indicated that the classic neo-Hookean and Mooney-Rivlin models are inadequate for …
Resveratrol Preconditioning Induces Bioenergetic Efficiency In The Brain Within The Long-Term Window Of Cerebral Ischemic Tolerance, Nathalie Khoury
Resveratrol Preconditioning Induces Bioenergetic Efficiency In The Brain Within The Long-Term Window Of Cerebral Ischemic Tolerance, Nathalie Khoury
Open Access Dissertations
Neuroprotective agents administered post cerebral ischemia have failed so far in the clinic to promote significant recovery. Thus, numerous efforts were redirected towards prophylactic approaches such as preconditioning as an alternative therapeutic strategy. Our lab has revealed a novel long-term window of cerebral ischemic tolerance mediated by resveratrol preconditioning (RPC) that lasts for two weeks in mice. To identify its mediators, we conducted an RNA-seq experiment on the cortex of mice two weeks post RPC, which revealed 136 differentially expressed genes. The majority of genes (116/136) were downregulated upon RPC and clustered into biological processes involved in transcription, synaptic signaling, …

Dataset : Perceived Ability To Regulate Love, Kruti Surti, Sandra Langeslag

Research has shown that romantic love can be regulated. We investigated perceptions about love regulation, because these perceptions may impact mental health and influence love regulation application. Two-hundred eighty-six participants completed a series of items online via Qualtrics that assessed perceived ability to up- and down-regulate, exaggerate and suppress the expression of, and start and stop different love types. We also tested individual differences in perceived love regulation ability. Participants thought that they could up- but not down-regulate love in general and that they could up-regulate love in general more than down-regulate it. Participants thought that they could up-regulate infatuation …

To address the unmet needs for human polyclonal antibodies both as therapeutics and diagnostic reagents, building upon our previously established transchromosomic (Tc) cattle platform, we report herein the development of a Tc goat system expressing human polyclonal antibodies in their sera. In the Tc goat system, a human artificial chromosome (HAC) comprising the entire human immunoglobulin (Ig) gene repertoire in the germline configuration was introduced into the genetic makeup of the domestic goat. We achieved this by transferring the HAC into goat fetal fibroblast cells followed by somatic cell nuclear transfer for Tc goat production. Gene and protein expression analyses …

Most scleractinian corals form obligate symbioses with photosynthetic dinoflagellates (family Symbiodiniaceae), which provide differential tolerances to their host. Previously, research has focused on the influence of symbiont composition and the dynamic processes of symbiont repopulation during single episodes of hyperthermal events, followed by years of less-stressful conditions. In contrast, this study characterized for the first time, the role of Symbiodiniaceae species changes in response to annually recurring hyperthermal events, a scenario soon expected to become the norm. Consecutive hyperthermal events during summer 2014 and 2015 along the Florida Reef Tract offered a unique opportunity to study bleaching susceptibility and recovery …

Effect Of Cooled Perches On Performance, Plumage Condition, And Foot Health Of Caged White Leghorn Hens Exposed To Cyclic Heat, J Y. Hu, P Y. Hester, M M. Makagon, Y Xiong, R S. Gates, H W. Cheng
Department of Animal Science: Faculty Publications
We examined the effects of water-chilled perches as cooling devices on hen performance during 2 summers using daily cyclic heat. White Leghorns, 17 wk of age, were assigned to 36 cages arranged into 6 banks. The banks were assigned to cooled perches, air perches, and no perches resulting in 2 replicate banks and 12 cages per treatment. Chilled water (10◦C) was circulated through the cooled perches during heat episodes. Daily cyclic heat of 35◦C was applied from 0600 to1800 h with a lowering of temperature to 28◦C from 1800 to 0600 h during the …
