Metabolic Inflexibility Revisited: Muscle Substrate Oxidation Is Dissociated From Muscle Insulin Resistance, Joongyu Daniel Song Jan 2019

Metabolic Inflexibility Revisited: Muscle Substrate Oxidation Is Dissociated From Muscle Insulin Resistance, Joongyu Daniel Song

Yale Medicine Thesis Digital Library

Alterations in basal metabolic substrate preference were proposed as early derangements in the development of skeletal muscle insulin resistance. Here, we report that alterations in muscle substrate oxidation are not associated with muscle insulin resistance in rats or humans. In this study we used a novel stable isotope tracer ([U-13C]glucose) liquid chromatography-tandem mass spectrometry (LCMS/

MS) method to measure the ratio of mitochondrial glucose oxidation by pyruvate dehydrogenase flux (VPDH) relative to rates of citrate synthase flux (VCS) in skeletal muscle. We found that high fat diet (HFD) fed insulin resistant rats did not have altered substrate oxidation in soleus …

In recent decades, computational tools and methods have become pervasive in mathematical and scientific fields (National Research Council, 2010a). Tools such as mathematical and statistical models have expanded the range of phenomena that are explored and have become necessary for analyzing increasingly large data sets across disciplines (National Academy of Sciences, National Academy of Engineering, & Institute of Medicine, 2007). With these advances, entirely new fields such as computational statistics, neuroinformatics, and chemometrics have emerged. The varied applied uses of computational tools across these fields have shown that future scientists will not only need to know how to program, but …

Shattering The Crystal Goblet: Seeking A Pedagogy Of Visuality In Post - Typographic Expository Texts, David Reinking Jan 2019

Shattering The Crystal Goblet: Seeking A Pedagogy Of Visuality In Post - Typographic Expository Texts, David Reinking

Publications

This article synthesizes diverse theoretical perspectives toward developing a pedagogy that addresses the visuality of digital texts. To frame those perspectives and their implications, I use a well - known analogy that Beatrice Warde introduced to typographers in the 1930s: drinking wine from a golden cup or a crystal goblet. I briefly review the theory and research related to visual aspects of texts, generating pedagogical perspectives from several prominent theories and perspectives. I then discuss, illustrated with a few examples, how these pedagogical perspectives might be instantiated in curriculum and instruction and the issues and challenges of doing so. I …

Heuristics For Communicating Science, Risk, And Crisis, Katherine E. Rowan, Andrew S. Pyle Jan 2019

Heuristics For Communicating Science, Risk, And Crisis, Katherine E. Rowan, Andrew S. Pyle

Publications

Scientific information about physical hazards is important and very easy to miscommunicate. Because science uses complex wording and sophisticated reasoning, many assume that sharing science with lay stakeholders is solely a matter of substituting simple words and sentences for unfamiliar terms and complex syntax. An additional frequent assumption is that ‘giving information’ or ‘educating’ are the only important goals when communicating with stakeholders about health and the environment. Indeed, one study shows that, among scientists who share science online with lay stakeholders, their principal goals when communicating with lay audiences are to ‘educate’ or ‘defend science’. Although these goals are …

Phylogenetic Origins For Severe Acetaminophen Toxicity In Snake Species Compared To Other Vertebrate Taxa, Peter Van Den Hurk, Harold M.I. Kerkkamp Jan 2019

Phylogenetic Origins For Severe Acetaminophen Toxicity In Snake Species Compared To Other Vertebrate Taxa, Peter Van Den Hurk, Harold M.I. Kerkkamp

Publications

While it has been known for a while that some snake species are extremely sensitive to acetaminophen, the underlying mechanism for this toxicity has not been reported. To investigate if essential detoxification enzymes are missing in snake species that are responsible for biotransformation of acetaminophen in other vertebrate species, livers were collected from a variety of snake species, together with samples from alligator, snapping turtle, cat, rat, and cattle. Subcellular fractions were analyzed for enzymatic activities of phenol-type sulfotransferase and UDP‑glucuronosyltransferase, total glutathione S‑transferase, and N‑acetyltransferase. The results showed that none of the snake species, together …

Computational Fluid Dynamics Investigation Of A Novel Hybrid Comprehensive Stage Ii Operation For Single Ventricle Palliation, Marwan Hameed Jan 2019

Computational Fluid Dynamics Investigation Of A Novel Hybrid Comprehensive Stage Ii Operation For Single Ventricle Palliation, Marwan Hameed

Electronic Theses and Dissertations

Hypoplastic left heart syndrome (HLHS) is a type of heart defect where the left ventricle is underdeveloped or not developed, resulting in only a single functioning right ventricle. Approximately 7.5% of patients with congenital heart disease are born with a single ventricle (SV) which is accompanied by a spectrum of other malformations such as atrophied ascending aorta, atrial septal defects, and ventricular septal defects (VSD). The existing three-hybrid staged surgical approach serving as a palliative treatment for this anomaly entails multiple complications and achieves a survival rate of only 50%. To reduce the trauma associated with the second stage of …

Parameter Estimation Of Stochastic Models Against Probabilistic Temporal Logic Behavioral Specifications, Arfeen Khalid Jan 2019

Parameter Estimation Of Stochastic Models Against Probabilistic Temporal Logic Behavioral Specifications, Arfeen Khalid

Electronic Theses and Dissertations

The inherent behavioral variability exhibited by stochastic systems makes it a challenging task for human experts to manually analyze them. Computational modeling of such systems helps in investigating and predicting the behaviors of their underlying processes but at the same time introduces the presence of several unknown parameters. A key challenge faced in this scenario is to determine the values of these unknown parameters against known behavioral specifications. The solutions that have been presented so far estimate the parameters of a given model against a single specification whereas a correct model is expected to satisfy all the behavioral specifications when …

Judicial Pragmatism: Strengths And Weaknesses In Common Law Adjudication, Legislative Interpretation, And Constitutional Interpretation, 52 Uic J. Marshall L. Rev. 369 (2019), Doori Song Jan 2019

Judicial Pragmatism: Strengths And Weaknesses In Common Law Adjudication, Legislative Interpretation, And Constitutional Interpretation, 52 Uic J. Marshall L. Rev. 369 (2019), Doori Song

UIC Law Review

Judicial pragmatism is a judicial methodology known for its future-looking mode of analysis, empirically-based decision making, and openness to judicial activism. In terms of strengths, judicial pragmatism helps to (1) maximize wealth and efficiency, (2) resolve truly novel cases, and (3) account for legislative shortcomings. In terms of weaknesses, judicial pragmatism poses the risks of (1) judicial tyranny, (2) overdependence on the social sciences, and (3) marginalization of important moral values. Although judicial pragmatism has generally been accepted as a helpful analytical approach, questions still remain over the extent to which it is helpful to judges in common law adjudication, …

Biomarker-Performance Associations During Nutritional And Exercise Intervention In Air Force Personnel, Jennifer Jurcsisn Jan 2019

Biomarker-Performance Associations During Nutritional And Exercise Intervention In Air Force Personnel, Jennifer Jurcsisn

Browse all Theses and Dissertations

This study evaluated the combined effects of an exercise intervention and nutritional supplement on biomarkers of stress and resilience, and the relationships of those markers with physical and cognitive performance. 130 healthy Active-Duty Air Force (AF) personnel were recruited to participate in a double-blind, placebo controlled 12-week exercise and nutritional intervention. Serum was collected at basal and high stress conditions pre- and post-intervention to track the following biomarkers: cortisol, dehydroepiandrosterone-sulfate (DHEA-S), norepinephrine (NE), neuropeptide Y (NPY), and serotonin. The exercise intervention significantly attenuated the cortisol response and peak stress cortisol levels. The nutritional intervention decreased peak stress NE. The selected …

Move It Or Lose It: Interspecific Variation In Risk Response Of Pond-Breeding Anurans, Philip Matich, Christopher M. Schalk Jan 2019

Move It Or Lose It: Interspecific Variation In Risk Response Of Pond-Breeding Anurans, Philip Matich, Christopher M. Schalk

Faculty Publications

Changes in behavior are often the proximate response of animals to human disturbance, with variability in tolerance levels leading some species to exhibit striking shifts in life history, fitness, and/or survival. Thus, elucidating the effects of disturbance on animal behavior, and how this varies among taxonomically similar species with inherently different behaviors and life histories is of value for management and conservation. We evaluated the risk response of three anuran species—southern leopard frog (Lithobates sphenocephalus), Blanchard’s cricket frog (Acris blanchardi), and green tree frog (Hyla cinerea)—to determine how differences in microhabitat use (arboreal vs …

Correlates Of Snake Entanglement In Erosion Control Blankets, Sarah E. Ebert, Kasey L. Jobe, Christopher M. Schalk, Daniel Saenz, Cory K. Adams, Christopher E. Comer

Faculty Publications

In road construction projects across the United States, erosion control methods (e.g., erosion control blankets [ECBs]), are mandated to stimulate seedbed regeneration and prevent soil loss. Previous reports have suggested that snakes are vulnerable to entanglement in ECBs. We conducted a literature review, field surveys, and an entanglement experiment to examine what factors increase a snake’s risk of ECB entanglement. Our literature review produced reports of 175 reptiles entangled in mesh products, 89.1% of which were snakes, with 43.6% of snake entanglements occurring in erosion control products. During our field surveys, we found 10 entangled snakes (n = 2 alive; …

The Loss Of Atrx Increases Susceptibility To Pancreatic Injury And Oncogenic Kras In Female But Not Male Mice., Claire C Young, Ryan M Baker, Christopher J Howlett, Todd Hryciw, Joshua E Herman, Douglas Higgs, Richard Gibbons, Howard Crawford, Arthur Brown, Christopher L Pin

Paediatrics Publications

Background

Pancreatic ductal adenocarcinoma (PDAC) is the third leading cause of cancer death in North America, accounting for >30,000 deaths annually. Although somatic activating mutations in KRAS appear in 97% of PDAC patients, additional factors are required to initiate PDAC. Because mutations in genes encoding chromatin remodelling proteins have been implicated in KRAS-mediated PDAC, we investigated whether loss of chromatin remodeler ɑ-thalassemia, mental-retardation, X-linked (ATRX) affects oncogenic KRAS’s ability to promote PDAC. ATRX affects DNA replication, repair, and gene expression and is implicated in other cancers including glioblastomas and pancreatic neuroendocrine tumors. The hypothesis was that deletion of Atrx …

Surgical Technique For Developing A Rabbit Model Of Congenital Diaphragmatic Hernia And Tracheal Occlusion, M. Mudri, S. A. Smith, C. Vander Tuin, J. Davidson, T. R.H. Regnault, A. Bütter

Paediatrics Publications

The surgical model of congenital diaphragmatic hernia (CDH) has been utilized in exploring treatments and innovative therapies, such as tracheal occlusion (TO). The rabbit is an excellent surgical model compared to others due to lower cost, ease of care, short gestational period, and large litter size. This model is also ideal in studying lung hypoplasia of CDH because rabbit lung development is most similar to humans as alveolarization begins prior to birth and continues post-natally. However, the surgical technique in creating a rabbit model of CDH is quite difficult and information is lacking on how to establish this model. Therefore, …

Probiotics, Health and Nutrition

The study of traditional populations provides a view of human-associated microbes unperturbed by industrialization, as well as a window into the microbiota that co-evolved with humans. Here we discuss our recent work characterizing the microbiota from the Hadza hunter-gatherers of Tanzania. We found seasonal shifts in bacterial taxa, diversity, and carbohydrate utilization by the microbiota. When compared to the microbiota composition from other populations around the world, the Hadza microbiota shares bacterial families with other traditional societies that are rare or absent from microbiotas of industrialized nations. We present additional observations from the Hadza microbiota and their lifestyle and environment, …

The Effects Of Methylphenidate (Ritalin) On The Neurophysiology Of The Monkey Caudal Prefrontal Cortex, Sébastien Tremblay, Florian Pieper, Adam Sachs, Ridha Joober, Julio Martinez-Trujillo

Physiology and Pharmacology Publications

© 2019 Tremblay et al. Methylphenidate (MPH), commonly known as Ritalin, is the most widely prescribed drug worldwide to treat patients with attention deficit disorders. Although MPH is thought to modulate catecholamine neurotransmission in the brain, it remains unclear how these neurochemical effects influence neuronal activity and lead to attentional enhancements. Studies in rodents overwhelmingly point to the lateral prefrontal cortex (LPFC) as a main site of action of MPH. To understand the mechanism of action of MPH in a primate brain, we recorded the responses of neuronal populations using chronic multielectrode arrays implanted in the caudal LPFC of two …

A Mouse Model Of Gestational Glucose Intolerance Through Exposure To A Low Protein Diet During Fetal And Neonatal Development, Sandra K. Szlapinski, Renee T. King, Gabrielle Retta, Erica Yeo, Brenda J. Strutt, David J. Hill

Paediatrics Publications

Key points: Pancreatic β-cell dysfunction is hypothesized to be the significant determinant of gestational diabetes pathogenesis, however pancreatic samples from patients are scarce. This study reports a novel mouse model of gestational glucose intolerance in pregnancy, originating from previous nutrition restriction in utero, in which glucose intolerance was restricted to late gestation as is seen in human gestational diabetes. Glucose intolerance was attributed to reduced β-cell proliferation, leading to impaired gestational β-cell mass expansion in maternal endocrine pancreas, in addition to reduced glucose-stimulated insulin secretion. This model reproduces some of the features of gestational diabetes and is suitable for testing …
