Fusobacterium nucleatum (Fn) is commonly enriched in colorectal cancer (CRC) and associated with poor outcomes, though its mechanisms remain unclear. Our study investigated how Fn affects the tumor microenvironment through single-cell transcriptomic analyses of 42 CRC patient tissues, comparing Fn-positive and Fn-negative tumors. We discovered that Fn impairs IgA plasma cell development and secretory IgA (sIgA) production by disrupting communication with tumor-associated macrophages. Additional experiments in germ-free mice, together with our re-analysis of a publicly available single-cell RNA-seq data set from a CRC mouse model with an intact gut microbiome–both models having been orally gavaged with Fn–jointly validated the causal …

New strategies are urgently needed against antimicrobial resistance, a major health threat, and the different mechanisms regulating the bacterial cell division machinery offer multiple opportunities for developing novel therapeutics. FtsZ, an essential protein of this process, is targeted by multiple small molecules, and benzodioxane-benzamides (BDOBs) are among the most potent inhibitors in several bacterial species. BDOBs mechanisms are however poorly understood, particularly their impact on FtsZ's interplay with partners and ability to assemble phase-separated biomolecular condensates potentially involved in stress sensing. Dysfunction or dysregulation of deubiquitination is closely related to the initiation and development of multiple cancers. Targeted regulation of deubiquitination has been recognized as an important strategy in tumor therapy. However, the mechanism by which drugs regulate deubiquitinase is not clear. Here, we identified ubiquitin-specific peptidase 48 (USP48), a member of the ubiquitin-specific protease family highly expressed in various tumors, as a specific substrate for the activated caspase-3. During drug induced apoptosis of AML cells, activated caspase-3 cleaves USP48 through recognizing the conservative motif DEQD located at 611-614 sites of human USP48. Circadian organization is involved in many gastrointestinal tract (GIT) functions such as the maintenance of intestinal barrier integrity. There is compelling evidence that perturbation of the circadian clock decreases intestinal epithelial cells' resiliency to alcohol-induced injury. One of the most common causes of circadian misalignment is wrong-time eating (largest meal at dinner) in modern societies. Yet, few studies have examined the importance of peripheral circadian rhythms of the GIT to alcohol consumption. Objectives: Onconephrology is an expanding subspecialty focused on the management of cancer patients with renal injury. This study used a comprehensive bibliometric analysis to emphasize the need for cooperation between oncologists and nephrologists, exploring current trends and future research areas in onconephrology.
Methods: Relevant literature on onconephrology published between 1 January 2000 and 27 April 2024 was retrieved from the Science Citation Index Expanded of the Web of Science Core Collection, followed by manual screening. Bibliometric analyses were performed using CiteSpace, VOSviewer, and Bibliometrix software.

Tubulin Regulates Stability And Localization Of Stmn2 By Binding Preferentially To Its Soluble Form, Xiang Deng, Gary A Bradshaw, Marian Kalocsay, Timothy Mitchison

The small, tubulin-binding protein STMN2 is highly expressed in neurons and is implicated in amyotrophic lateral sclerosis. STMN2 degrades rapidly and accumulates at axotomy sites, suggesting fast turnover is crucial for its neuroprotective function. We show that STMN2 was primarily degraded by the ubiquitin-proteasome system. Its membrane-targeting N-terminal domain promoted fast turnover, whereas its tubulin-binding domain promoted stabilization. Proximity labeling and imaging showed that tubulin binding reduced STMN2 targeting to trans-Golgi network membranes. Pull-down assays showed that tubulin binds preferentially to soluble over membrane-bound STMN2. Emotion analysis is a branch of artificial intelligence and natural language processing focused on recognizing emotions hidden throughout various forms of digital data, including text, images, and multi-modal representations. In this dissertation, we present four published and planned works that investigate different methodologies for natural language analysis tasks using deep learning techniques. The first published work we present considers the task of identifying fake news using various text and emotion representations. We demonstrate that emotion representations combined with word embedding techniques can improve the accuracy of fake news classification. The expansion of solar photovoltaic energy infrastructure is transforming landscapes worldwide. Increasingly, facilities are planted with native vegetation, yet how vegetation characteristics and landscape variables shape wildlife communities in solar facilities remains poorly understood. Because avian populations have been in decline and are sensitive to habitat changes, understanding how solar facilities influence avian communities is important. Many opportunities are known to use organic production methods or incorporate native warm-season grasses (NWSG) to accomplish both production goals and environmental or social outcomes. Anthropogenic activities are reshaping the elemental composition of freshwater ecosystems, often creating stoichiometric imbalances that influence the growth, physiology and interactions of aquatic organisms. Ecological stoichiometric theory (EST) provides a framework for examining these changes by linking organismal elemental regulation to broader ecosystem processes. While traditionally focused on elements, such as carbon (C), nitrogen (N), and phosphorus (P), emerging stressors highlight the need to expand this framework to be more elementally inclusive for the examination of novel interactions. One area where they are particularly effective is the treatment of water contaminated with toxic substances. Here, a comprehensive understanding of immune responses triggered by either a multimeric CD40 receptor-agonistic DNA aptamer-based or mannosylated chitosan-based Salmonella Enteritidis inactivated vaccine platforms, along with phytobiotic supplementation as an immune modulatory strategy in response to Salmonella lipopolysaccharides, is presented. This dissertation supports the development of next-generation Salmonella vaccine platforms that simultaneously stimulate both protective cell-mediated and humoral immune responses, while indicating the potential immunomodulatory effects of phytobiotics supplementation to reduce the immunological impact of Salmonella lipopolysaccharides in chickens. Negation is a fundamental linguistic concept used by humans to convey information that they do not desire. Despite this, minimal research has focused on negation within text-guided image editing. This lack of research means that vision-language models (VLMs) for image editing may struggle to understand negation, implying that they struggle to provide accurate results. One barrier to achieving human-level intelligence is the lack of a standard collection by which research into negation can be evaluated. Background: Almost all mammals rely on the thymus and bone marrow to generate and differentiate B and T cells essential for adaptive immunity. A few members of the family Soricidae, or true shrews within Eulipotyphla, have also evolved an enlarged pancreas of Aselli, a kidney-sized organ hypothesized to serve this primary immune role, and whose gene expression profile is unknown. Results: Here we introduce transcriptomes of juvenile Eurasian common shrews (Sorex araneus, family Soricidae) pancreas of Aselli. We compare these to those of the shrew spleen and chick bursa of Fabricius, an analogous, bird-specific organ, and explore differential expression overlaps …

Preterm birth (PTB) and low birth weight (LBW) are leading causes of neonatal morbidity and mortality. Maternal periodontal disease (PD) has been increasingly linked to these adverse birth outcomes (ABOs), yet questions about causality and treatment effectiveness persist due to study design heterogeneity and potential unmeasured confounding. These gaps hinder the development of clear clinical and public health guidelines. This dissertation addresses these gaps through three interconnected studies assessing the relationship between maternal oral health and ABOs.

Human anatomy stands as a cornerstone prerequisite for admission into graduate-level Physical Therapy Programs. As such, prior anatomy experiences need to prepare students for success in graduate-level coursework. However, studies have demonstrated that undergraduate anatomy courses vary in their ability to predict success in graduate Physical Therapy anatomy coursework. This research aimed to utilize students' perceptions of their undergraduate anatomy experiences to predict grades in graduate-level Physical Therapy anatomy coursework. The Mississippi Alluvial Plain (MAP) of Arkansas is a landscape where many wetlands have been altered for use as aquaculture ponds or agricultural ditches. Commercial harvest of freshwater turtles within the MAP is not restricted or limited, with reported harvest numbers for 2019 alone exceeding 4000 for spiny softshell turtles (Apalone spinifera) and 39,000 for red-eared sliders (Trachemys scripta elegans). Herein, we attempt to provide baseline estimates of freshwater turtle densities and community composition in aquaculture ponds and agricultural ditches of eastern Arkansas, the habitat types most frequently trapped by commercial harvesters. The demand for processed foods has led to increased use of synthetic phenolic antioxidants (SPAs), notably tert-butylhydroquinone (TBHQ), to inhibit lipid oxidation and prolong shelf life. While TBHQ is deemed safe at regulated levels, concerns arise from studies indicating potential adverse effects such as oxidative stress, genotoxicity, and impaired immune function. To investigate TBHQ's immunomodulatory effects, RAW 264.7 murine macrophages were pre-treated with TBHQ and subsequently stimulated with lipopolysaccharide (LPS) or polyinosinic-polycytidylic acid (poly I:C, PIC) to simulate bacterial or viral challenges. Ecological interactions such as predation are vital to shaping the distribution of organisms. For interactions to occur, organisms must undertake movement, such as dispersal, that places them in proximity to the organisms with which they may interact. Geographically isolated wetlands (GIW) are productive and biodiverse ecosystems, but little is known on broad geographic scales about the determinants of their fauna. Fishes have difficulty colonizing GIW, whereas amphibians and macroinvertebrates are adept overland dispersers – though they may be sensitive to top-down control by fishes. Background: Recovering from anterior cruciate ligament (ACL) reconstruction can be challenging. While standard rehabilitation helps restore knee function, many patients still face a slow or incomplete recovery. Extracorporeal shockwave therapy (ESWT) has recently gained attention as a potential way to speed up healing and improve outcomes when added to traditional rehab.
Aim: To explore whether combining ESWT with standard postoperative rehabilitation truly leads to better recovery compared with rehab alone.

The characterization of systems encompasses a variety of modeling frameworks designed to capture specific behaviors and components of various system domains. Whatever the framework, the core elements of a system representation are the information of the system and a description of how that information is related. The relations in deterministic systems are functions, which, when composed to form executable processes, can be used to simulate system data. Trypanosoma brucei is a protozoan parasite which causes human African trypanosomiasis and the livestock wasting disease, nagana. These parasites have highly specialized peroxisomes, called glycosomes, which house a variety of essential metabolic pathways, including glycolysis. Glycolysis is the sole means of energy production in the infective life stage of T. brucei making glycosomes and the glycolytic pathway promising targets for therapeutic strategies. In this work, we have studied the glycosome as a therapeutic target from multiple perspectives. The first portion of this work addresses drug trafficking. Background: Household flooring is increasingly being investigated as a determinant of health, however the pathways through which flooring may impact health and wellbeing are not yet well understood. The SABABU study is a cluster-randomised controlled trial evaluating the impact of an improved flooring intervention on soil-transmitted helminthiasis, tungiasis, and enteric infections in Bungoma and Kwale counties, Kenya. This practice-based case study is designed to offer a tutorial on how to adapt an informal, diagnostic emergent literacy assessment to meet the needs of students like six-year-old Armando, a Latino, multilingual learner with autism spectrum disorder (ASD) and complex communication needs (CCN) in an inclusive kindergarten classroom. Armando’s informal literacy assessment adaptations and outcomes are discussed, including descriptions of his emergent literacy skills observed through his engagement in literacy stations, small group reading instruction in his classroom, and his use of alternative and augmentative communication (AAC) in both instructional and assessment settings.
