Molecular Epidemiology, Ecology, And Evolution Of Group A Streptococci, Debra E. Bessen, Pierre R. Smeesters, Bernard W. Beall

NYMC Faculty Publications

The clinico-epidemiological features of diseases caused by group A streptococci (GAS) is presented through the lens of the ecology, population genetics, and evolution of the organism. The serological targets of three typing schemes (M, T, SOF) are themselves GAS cell surface proteins that have a myriad of virulence functions and a diverse array of structural forms. Horizontal gene transfer expands the GAS antigenic cell surface repertoire by generating numerous combinations of M, T, and SOF antigens. However, horizontal gene transfer of the serotype determinant genes is not unconstrained, and therein lies a genetic organization that may signify adaptations to a …

Hypertension results from the interaction of genetic and acquired factors. IgG occurs in the form of different subclasses, of which the effector functions show significant variation. The detailed differences between the glycosylation profiles of the individual IgG subclasses may be lost in a profiling method for total IgG N-glycosylation. In this study, subclass-specific IgG Fc glycosylation profile was investigated in the four northwestern Chinese minority populations, namely, Uygur (UIG), Kazak (KZK), Kirgiz (KGZ), and Tajik (TJK), composed of 274 hypertensive patients and 356 healthy controls. The results showed that ten directly measured IgG N-glycan traits (i.e., IgG1G0F, IgG2G0F, IgG2G1FN, IgG2G1FS, …

Bridging The Gap Between Similarity And Causality: An Integrated Approach To Concepts, Corinne L. Bloch-Mullins Sep 2018

Bridging The Gap Between Similarity And Causality: An Integrated Approach To Concepts, Corinne L. Bloch-Mullins

Philosophy Faculty Research and Publications

A growing consensus in the philosophy and psychology of concepts is that while theories such as the prototype, exemplar, and theory theories successfully account for some instances of concept formation and application, none of them successfully accounts for all such instances. I argue against this ‘new consensus’ and show that the problem is, in fact, more severe: the explanatory force of each of these theories is limited even with respect to the phenomena often cited to support it, as each fails to satisfy an important explanatory desideratum with respect to these phenomena. I argue that these explanatory shortcomings arise from …

Structural Basis Of Mitochondrial Transcription., Hauke S. Hillen, Dmitry Temiakov, Patrick Cramer Sep 2018

Structural Basis Of Mitochondrial Transcription., Hauke S. Hillen, Dmitry Temiakov, Patrick Cramer

Department of Biochemistry and Molecular Biology Faculty Papers

The mitochondrial genome is transcribed by a single-subunit DNA-dependent RNA polymerase (mtRNAP) and its auxiliary factors. Structural studies have elucidated how mtRNAP cooperates with its dedicated transcription factors to direct RNA synthesis: initiation factors TFAM and TFB2M assist in promoter-DNA binding and opening by mtRNAP while the elongation factor TEFM increases polymerase processivity to the levels required for synthesis of long polycistronic mtRNA transcripts. Here, we review the emerging body of structural and functional studies of human mitochondrial transcription, provide a molecular movie that can be used for teaching purposes and discuss the open questions to guide future directions of …

Small Noncoding Rna Profiles Along Alternative Developmental Trajectories In An Annual Killifish, Amie L. Romney, Jason E. Podrabsky Sep 2018

Small Noncoding Rna Profiles Along Alternative Developmental Trajectories In An Annual Killifish, Amie L. Romney, Jason E. Podrabsky

Biology Faculty Publications and Presentations

Embryonic development of Austrofundulus limnaeus can occur along two phenotypic trajectories that are physiologically and biochemically distinct. Phenotype appears to be influenced by maternal provisioning based on the observation that young females produce predominately non-diapausing embryos and older females produce mostly diapausing embryos. Embryonic incubation temperature can override this pattern and alter trajectory. We hypothesized that temperature-induced phenotypic plasticity may be regulated by post-transcriptional modification via noncoding RNAs. As a first step to exploring this possibility, RNA-seq was used to generate transcriptomic profiles of small noncoding RNAs in embryos developing along the two alternative trajectories. We find distinct profiles of …

OBJECTIVE: Unreliable antibodies often hinder the accurate detection of an endogenous protein, and this is particularly true for the cardiac and smooth muscle cofactor, MYOCD (myocardin). Accordingly, the mouse Myocd locus was targeted with 2 independent epitope tags for the unambiguous expression, localization, and activity of MYOCD protein. APPROACH AND RESULTS: 3cCRISPR (3-component clustered regularly interspaced short palindromic repeat) was used to engineer a carboxyl-terminal 3xFLAG or 3xHA epitope tag in mouse embryos. Western blotting with antibodies to each tag revealed a protein product of approximately 150 kDa, a size considerably larger than that reported in virtually all publications. MYOCD …

Application Of Spectral Solution And Neural Network Techniques In Plasma Modeling For Electric Propulsion, Joseph R. Whitman Sep 2018

Application Of Spectral Solution And Neural Network Techniques In Plasma Modeling For Electric Propulsion, Joseph R. Whitman

Theses and Dissertations

A solver for Poisson's equation was developed using the Radix-2 FFT method first invented by Carl Friedrich Gauss. Its performance was characterized using simulated data and identical boundary conditions to those found in a Hall Effect Thruster. The characterization showed errors below machine-zero with noise-free data, and above 20% noise-to-signal strength, the error increased linearly with the noise. This solver can be implemented into AFRL's plasma simulator, the Thermophysics Universal Research Framework (TURF) and used to quickly and accurately compute the electric field based on charge distributions. The validity of a machine learning approach and data-based complex system modeling approach …

We have shown that epoxyeicosatrienoic acids (EETs), specifically 11,12- and 14,15-EETs, reduce adipogenesis in human mesenchymal stem cells and mouse preadipocytes (3T-3L1). In this study, we explore the effects of soluble epoxide hydrolase (sEH) deletion on various aspects of adipocyte-function, including programing for white vs. beige-like fat, and mitochondrial and thermogenic gene-expressions. We further hypothesize that EETs and heme-oxygenase 1 (HO-1) form a synergistic, functional module whose effects on adipocyte and vascular function is greater than the effects of sEH deletion alone. In in vitro studies, we examined the effect of sEH inhibitors on MSC-derived adipocytes. MSC-derived adipocytes exposed to …

Are Government Incentives Effective For Avoided Deforestation In The Tropical Andean Forest?, Pablo Cuenca, Juan Robalino, Rodrigo A. Arriagada, Cristian M. Echeverria

Forest Collaborative Research

In order to ensure the provision of goods and services from forests, many governments have promoted less-traditional conservation initiatives such as programs of payments for ecosystem services called, more broadly, direct payments for conservation. The Socio Bosque Program (SBP) is a governmental program in Ecuador that directly provides economic incentives to rural families and local and indigenous communities who have voluntarily agreed to comply with some conservation activities. An impact evaluation method (matching) was used to assess the impact of the SBP between 2008 and 2014. This study revealed that on average, the SBP reduced deforestation by 1.5% in those …

This study is among the first to calculate poverty among one of the world’s largest refugee populations, Afghans in Iran. More importantly, it is one of the first to use capability and monetary approaches to provide a comprehensive perspective on Afghan refugees’ poverty. We estimated poverty using data collected from a sample of 2,034 refugee households in 2011 in Iran. We utilized basic needs poverty lines and the World Bank’s absolute international poverty line for our monetary poverty analyses and the global Multidimensional Poverty Index (MPI) for our capability analyses of poverty. Findings show that nearly half of the Afghan …

Flipping The Classroom To Train Citizen Scientists In Invasive Species Detection And Response, Daniel J. Larkin, Megan M. Weber, Susan M. Galatowitsch, Angela S. Gupta

Journal of Extension

Extension educators are increasingly using flipped classrooms, wherein online content delivery precedes in-person learning. We have applied this approach to two Extension programs in which citizen scientists are trained in early detection of invasive species. Our goal in using the tool of flipped classrooms is to accommodate large amounts of content while focusing classroom time on skills development. In 2017, we assessed efficacy of the flipped classroom through knowledge tests and surveys completed by 174 participants and 106 participants, respectively. Results demonstrated large knowledge gains and high participant satisfaction. We encourage Extension professionals to consider whether use of the flipped …

Crispr Knockouts Reveal An Endogenous Role For Ancient Neuropeptides In Regulating Developmental Timing In A Sea Anemone, Nagayasu Nakanishi, Mark Q. Martindale

Biological Sciences Faculty Publications and Presentations

Neuropeptides are evolutionarily ancient peptide hormones of the nervous and neuroendocrine systems, and are thought to have regulated metamorphosis in early animal ancestors. In particular, the deeply conserved Wamide family of neuropeptides—shared across Bilateria (e.g. insects and worms) and its sister group Cnidaria (e.g. jellyfishes and corals)—has been implicated in mediating life-cycle transitions, yet their endogenous roles remain poorly understood. By using CRISPR-Cas9-mediated reverse genetics, we show that cnidarian Wamide—referred to as GLWamide—regulates the timing of life cycle transition in the sea anemone cnidarian Nematostella vectensis. We find that mutant planula larvae lacking GLWamides transform into morphologically normal polyps …

"Misfits" And The Celebration Of Lesbian, Gay, Bisexual, Transgender, And Queer (Lgbtq) Youth At A High School In The United States, Nathan N. Taylor

Journal of Research Initiatives

As part of a six-month case study, this article delineated the relationship between homonormativity and the victimization of students in a high school in the United States by investigating the strategies of policing, resistance, and queering. Not only do these strategies reappropriate practices inherent in homonormative practices, these strategies reconfigure practices associated with heteronormativity and homophobia. The undercurrent of this research highlights how schools marginalize identities in some spaces and elevate identities in other spaces, the socio-political readings of that positioning, and what educators can do to promote an inclusive environment for all students.

Urban Food Entrepreneurship, Governance, And Economic Development In The Post-Industrial Cities Of Newark, New Jersey And Dayton, Ohio, John C. Jones

Dissertations

The local governments of post-industrial cities in the US struggle to foster economic development and to find uses for underutilized or abandoned urban land and buildings. Partly in response to growing consumer interest in locally produced foods, food entrepreneurs are increasingly using these underutilized urban properties for farming or for producing value-added food products. However, sometimes intentionally and sometimes unintentionally, existing policies and regulatory regimes of local and state governments often restrict the ability of urban food entrepreneurs to grow. This dissertation documents urban food entrepreneurship in the post-industrial communities of greater Newark, New Jersey and greater Dayton, Ohio. The …

Temporal And Spatial Effects Of Shock Overpressure On Blood-Brain Barrier Permeability In Blast-Induced Traumatic Brain Injury, Matthew Joseph Kuriakose

Dissertations

Exposure to shock waves is the leading cause of traumatic brain injury (TBI) in military personnel and blast-induced TBI (bTBI) is considered the signature wound in recent conflicts in Iraq and Afghanistan. Many researchers attempt to replicate field-relevant shock waves in laboratory settings through the use of gas-driven shock tubes in order to investigate the generation and propagation of shock waves and also explore possible mechanisms of bTBI. Among several injury mechanisms of bTBI, damage to the blood-brain barrier (BBB) has been identified as a potential candidate and has been the focus of several clinical and experimental investigations aimed to …
