As critical mediators of cell-to-cell communication, gap junctions (GJs) are comprised of membrane channels that directly link the cytoplasm of adjacent coupled cells thereby allowing for the passage of ions, small metabolites, and secondary messengers. Each channel is formed by the apposition of two connexons from adjacent cells, each composed of six connexin (Cx) proteins. Each GJ channel functions to promote signal propagation and synchronization of cells and tissues in organs. Furthermore, GJs are essential for proper propagation of cardiac action potentials from one cell to the next, leading to the coordinated contraction and relaxation of heart muscle powering circulation. …

Pancreatic Cancer (PC) is one of the vicious cancers as it ranks third in the race of leading cause of cancer-related death. Lack of early diagnostic marker, poor understanding of molecular mechanism of the disease and failure to conventional chemotherapy makes this disease dreadful.
Mucin 4 (MUC4), a high molecular weight glycoprotein is one of the top differentially expressed molecules in PC while not expressed in normal pancreas. How are cholangiocarcinoma cells different from non-malignant cholangiocytes?
All of the hallmarks of cancer apply- those reported in this dissertation include cell proliferation, migration, apoptosis, and evasion of growth suppression. My studies began with testing how apoptosis might be regulated through embelin, a small molecule reported to sensitize cells to apoptosis by blocking XIAP. My data however revealed that embelin reduced the proliferative capacity in cholangiocarcinoma cells, but did not increase cell death. Malignant cells exhibit dysregulation of microRNA processing and expression. Hence, my studies seeking ways that cholangiocarcinoma eludes apoptosis transitioned to the oncomiR miR-106b, which is overexpressed in …

The ethnographic research in this thesis focuses on one group of Afro-Mexicans who migrated to Pasadena, California, from Mexico’s Costa Chica, on the Pacific Coast of Oaxaca and Guerrero, the area where a social movement for Afro-Mexican recognition and collective rights was founded in 1997. This study, using a framework of racial theory, examines the process of racial formation in a transnational setting, which has made this group different in many ways from the Black residents who remain in the Costa Chica. In this work, ferricyanide-doped polyaniline (PANI) is electropolymerized by constant current and cyclic voltammetry (CV) measurements with simultaneous monitoring of mass changes using electrochemical quartz crystal microbalance (eQCM). Raman spectroscopy, Fourier transform infrared spectroscopy and CV measurements confirm the incorporation of the ferricyanide into the film and indicate modified electrical conductivity. UV-Vis absorbance spectroscopy and Raman spectroscopy confirm the conducting ‘emeraldine-salt’ state and confirm the presence of polarons, i.e. charge carriers.
The addition of ferricyanide during electropolymerization of the PANI film results in an increase of polymerization rate as confirmed using eQCM. A decrease in counter ion ingress/egress and an …

In this thesis, we demonstrate an electrochemical approach for monitoring the electric-field induced melting of surface-bound double-stranded DNA. The electrochemical routine involves repeated chronoamperometry pulses to melt the duplex and square wave voltammetry to monitor the extent of melting. We utilize a scanning potential and constant potential technique to generate melting curves and access the stability and kinetics of the DNA duplexes. Our method uses a mixed monolayer of thiol-modified DNA oligomers and mercaptohexanol on gold electrodes, are subsequently incubated with target DNA covalently modified with electroactive methylene blue. This thesis argues that the United States is failing to fulfill its obligations under the Convention on the Rights of the Child (CRC) in its adjudication of the hardship standard in non-LPR cancellation of removal. It is well-documented that the current interpretation of the “exceptional and extremely unusual hardship” standard results in the separation of families and de facto deportation of children, many of whom are U.S. citizens. This thesis contends that this practice is not only unjust, but also unlawful.

Background: Reconstruction of gene regulatory networks (GRNs), also known as reverse engineering of GRNs, aims to infer the potential regulation relationships between genes. With the development of biotechnology, such as gene chip microarray and RNA-sequencing, the high-throughput data generated provide us with more opportunities to infer the gene-gene interaction relationships using gene expression data and hence understand the underlying mechanism of biological processes. Diversification at the transcriptome 3'end is an important and evolutionarily conserved layer of gene regulation associated with differentiation and dedifferentiation processes. Here, we identify extensive transcriptome 3'end-alterations in neuroblastoma, a tumour entity with a paucity of recurrent somatic mutations and an unusually high frequency of spontaneous regression. Utilising extensive RNAi-screening we reveal the landscape and drivers of transcriptome 3'end-diversification, discovering PCF11 as critical regulator, directing alternative polyadenylation (APA) of hundreds of transcripts including a differentiation RNA-operon. PCF11 shapes inputs converging on WNT-signalling, and governs cell cycle, proliferation, apoptosis and neurodifferentiation. Reservoirs throughout the country exhibit degraded shorelines and simplified littoral habitats because of aging. This study evaluated the establishment of agricultural plantings on reservoir mudflats and the effectiveness of supplemental brush pile structures in providing recruitment habitat for juvenile fish. The mudflats of Enid Reservoir, Mississippi were seeded during the winter drawdown in October of 2016 and 2017 with agricultural plantings. Monitoring of plantings found grasses performed best in terms of establishment and providing potential fish habitat. During Summer 2017 brush piles and control sites, without brush, were sampled in Enid Reservoir using rotenone to evaluate juvenile fish use. Juvenile …

Clapper Rails possess the potential to indicate changes to marsh ecological state due to population variation related to habitat features exhibited in previous study. Estimates for Clapper Rail survival are among the key missing parameters to create predictive models for Clapper Rail populations. (1998), individuals of all ages have been exposed to a variety of traumatic events that had the capacity to alter overall functioning in a variety of ways. Despite budding research on parent-child discrepant perceptions of a child’s psychosocial functioning, the potential difference in individual perception of coping socialization is unclear. Further, literature suggests the onset of various impulse-control disorders occurs around middle childhood. Thus, the current study used a phenomenological approach and thematic analysis to examine perspectives of coping socialization, identify intended take away messages, and examine the perceived impact of inconsistent understanding of the interaction. Results indicated both parents and children use verbal and physical cues to understand one another during coping socialization and that their lab discussion was representative of their typical interactions. An increasing trend in mass mortality events (MMEs) has been observed in recent years, leading to an increased study of these events and their causes. Still to be investigated are the immediate and long-term effects of these environmental disturbances. Microbial communities found on and within the carcass are a major contributor to decomposition. Prominent Feature Analysis (PFA) is a reliable and valid writing assessment tool, derived from the writing it is used to assess. PFA, used to assess on-demand expository essays in Grades 3-12, uncovers positive and negative characteristics of a sample. To extend PFA to a new academic level and genre, I assessed scientific writing of 208 undergraduates, identifying 35 linguistic and 20 scientific prominent features. An essay could earn up to 28 positive (24 linguistic and four scientific), and up to 27 negative marks (11 linguistic and 16 scientific). The cultural significance of effigy pipes among southeastern groups during the Mississippian period (A.D. 1000-1600) has yet to be fully understood. Recent studies, however, have provided new archaeological contexts for framing explanations of their possible use and distribution among such groups. Apart from conjectures about their use as ceremonial objects, selection for effigy pipes in the Mississippian Southeast was directly related to fluctuating environmental and demographic conditions under which such objects were manufactured and distributed. Bab edh-Dhra is the most extensively excavated cemetery from Early Bronze Age, Jordan. Despite thorough study, the social structure and existence of social inequality remain unclear. This was addressed using osteological evidence of physiological stress to compare between family tombs. In societies exhibiting social inequality, individuals of lower status experience higher levels of stress. Evidence of physiological stress (femoral length, LEH, metabolic disorders, periosteal reactions, cribra orbitalia, and porotic hyperostosis) was recorded using standard methods for 250 adults. The impact of parental depressive problems on children’s depressive symptoms has been widely studied. The Stress Buffering Hypothesis states that social support acts as a protective factor between the impacts of stress from negative life events on physical and psychological health. The current study examined the stress buffering hypothesis in terms of the relationship between parental depressive problems and emerging adult depressive problems. Necrotic enteritis (NE) caused by Clostridium perfringens type A is an important bacterial enteric disease of global broiler production. However, the dynamic interactions of NE and its predisposing factors are not fully presented by current studies. An impact of nitrogen, phosphorus and potassium fertilizers on the concentration changes of carotenoid pigments in the Indigofera tinctoria plant by using Raman spectroscopic techniques is studied. Three different concentration levels of the fertilizers with a normal supply as control were added to the plants at two stages. The Raman spectra were taken to determine the carotenoid concentration level changes in the plant leaves in vivo. The ring-stretching mode are the Raman spectroscopic signatures for the carotenoid pigment and its magnitude increased significantly (over 170%) for the case of phosphorus and potassium fertilizers. The world’s oceans absorb a significant percentage of anthropogenic carbon emissions, and CO2 levels have profound effects on the marine environment. Of primary concern is the acidification of the oceans due to dissolved CO2. The goal of this research is to design new sensing technologies for deployment in the marine environment to detect CO2 pollutant. This dissertation contains two secondary quantitative data analyses studies. In the first, implementation of the WannaBee Healthy? Be Smart! Be Active! Be a Leader! health science curriculum was examined to expand understanding about teacher usage of an integrated health curriculum. Specifically, researchers assessed the amount of activities within each curriculum domain (i.e., books, creative expressions, language/literacy, math, science) and the number of activities within each theme of the lessons (i.e., nutrition, physical activity, sleep) utilized by participating teachers. For this study, in-depth, semi-structured interviews were conducted with 16 transgender adults who lived, or had previously lived, in Mississippi and sought healthcare services in the state. The questions asked focused on identifying the barriers to care that these individuals faced as well as the strategies for navigating the barriers they employed. Recommendations for providers were also considered.

Removal Of Dietary Antimicrobials And Effects Of Their Replacement With Bacillus Subtilis On The Growth And Intestinal Health Of Male Broilers, Kacey O'Donnell

The effects of dietary antimicrobial removal and Bacillus subtilis supplementation on the growth and intestinal health of male broilers were investigated. Birds were fed either a control, antimicrobial, or a B. subtilis probiotic diet at different feeding phases. Birds were challenged with a 10 × dose of a coccidiosis vaccine. Supplementation of B. subtilis in for antimicrobials in the late grower and early finisher phases improved growth similar to birds fed antimicrobials until withdrawal, while antimicrobial removal without B. subtilis supplementation in those periods hindered growth. The improved growth suggests that the probiotic was able to alleviate the stress of …

Corn (Zea mays) is frequently infected by a soil fungal pathogen Aspergillus flavus. The fungus produces aflatoxins, which cause liver cancer. Maize inbred lines that are resistant to infection by A. flavus have been developed, and these inbred lines provide excellent models for studying molecular mechanisms of maize resistance to the fungus. MicroRNA-like RNAs (milRNAs) recently identified in A. flavus had been found to be correlated with aflatoxin production conditions, suggesting that the milRNAs might play a role in the regulation of aflatoxin production.
