Action potentials (APs) are the functional units of fast electrical signaling in excitable cells. The upstroke and downstroke of an AP is generated by the competing and asynchronous action of Na+- and K+-selective voltage-gated conductances. Although a mixture of voltage-gated channels has been long recognized to contribute to the generation and temporal characteristics of the AP, understanding how each of these proteins function and are regulated during electrical signaling remains the subject of intense research. AP properties vary among different cellular types because of the expression diversity, subcellular location, and modulation of ion channels. These complexities, in addition to the …

The Chemotactic Role Of Metallothionein In The Extracellular Environment, Sadikshya Bhandari
Doctoral Dissertations
Metallothionein (MT) is a cysteine rich stress response protein that lacks a signal peptide and thus has historically been considered an intracellular protein. However, the presence of MT in a variety of extracellular fluids and spaces suggests an additional extracellular role for this protein. We have shown that MT released in the extracellular environment can bind to the surface of lymphocytes and is a chemoattractant for Jurkat T cells among other cells. Additionally, when Jurkat T cells are pre-incubated with MT, there is a decline in the subsequent chemotactic response to SDF-1 α. Jurkat T cells pre-incubation with SDF-1 α …

We report field tests of an instrument using multi-wavelength excitation and detection of fluorescence capable of detection and discrimination of viable cells, non-viable cells (not metabolically active but not decomposed), and spores in extreme arid environments where low microbial abundances are expected. These new results are presented for cold and dry volcanic habitats worldwide, e.g., the arid core of the Atacama Desert, Mt. Kilimanjaro glacier and Kibo area, Pali Aike caldera, and the western US volcanic and desert soils in Utah, Idaho, Nevada, and California. Our results are comparable to previous studies reported in the literature for the same environments. …

Climate changes are predicted to drive changes in plant species composition and vegetation cover around the world. Preserved specimens and other botanical information that we gather today—a period future practitioners may look back on as an early stage of modern anthropogenic climate change—will be of value to conservation managers and conservation biologists in the decades and centuries ahead. Here, we present suggestions for the systematic collection, long-term curation (in museums, herbaria, and other research institutions), and maintenance of plant specimens, along with associated data and analyses on the plants and vegetation present today and in the past. The primary aim …

Prostate cancer is a leading cause of cancer death in men over 50 years of age, and there is a characteristic marked decrease in Zn content in the malignant prostate cells. The cause and consequences of this loss have thus far been unknown. We found that in middle-aged rats a Zn-deficient diet reduces prostatic Zn levels (P = 0.025), increases cellular proliferation, and induces an inflammatory phenotype with COX-2 overexpression. This hyperplastic/inflammatory prostate has a human prostate cancer-like microRNA profile, with up-regulation of the Zn-homeostasis-regulating miR-183-96-182 cluster (fold change = 1.41-2.38; P = 0.029-0.0003) and down-regulation of the Zn …

The assessment of individual traits requires that tests are reliable (i.e. consistency over time) and externally valid, meaning that they predict future responses in similar contexts (i.e. convergent validity) but do not predict responses to unrelated situations (i.e. discriminant validity). The aim of this study was to determine if dairy calf personality traits (Fearfulness, Sociability and Pessimism), derived from behaviours expressed in standardized tests, predict individuals’ responses in related situations. The first experiment tested if the trait ‘Sociability’ was related to the expression of social behaviour in the home-pen, with calves assigned individual proximity scores (based on proximity to other …

We are almost oblivious to the presence of microorganisms in our daily lives, but they exist and come into contact with us all the time. While some bacteria are harmless, and even beneficial, other bacteria can cause infections. A common treatment to bacterial infections is antibiotics and the success of an antibiotic depends on the resistance of the bacteria to the antibiotic. We conducted experiments to identify the types of bacteria that can be found on surfaces or within soil environments that come into contact with a vast number of people on a daily basis and how this can affect …

Mathematical models are key tools for the development of surveillance, preparedness and response plans for the potential events of emerging and introduced foreign animal diseases. Creating these types of plans requires data; when data are incomplete, mathematical models can help fill in missing information, provided they are informed by the data that are available. In the United States, the most complete national-scale data available on cattle shipments are based on Interstate Certificates of Veterinary Inspection, which track the shipment of cattle between states; data on intrastate cattle shipments are lacking. Here we develop four new datasets on intrastate cattle shipments …

Diabetes is a common disease affecting majority of populations worldwide. Since 1980, there has been an increase in the number of people diagnosed as prediabetic and diabetic. Diabetes is characterized by high levels of circulating glucose and leads to most microvascular and macrovascular complications such as retinopathy, nephropathy, neuropathy, stroke, and myocardial infarction. Bone marrow vascular disruption and increased adiposity are also linked to various complications in type II diabetes mellitus. In addition to these complications, type 2 diabetic patients also have fragile bones caused by faulty mineralization mainly due to increased adiposity among diabetic patients that affects both osteoblast …

School‐Level Body Mass Index Shapes Children's Weight Trajectories, Ashley W. Kranjac
Sociology Faculty Articles and Research
BACKGROUND
Embedded within children's weight trajectories are complex environmental contexts that influence obesity risk. As such, the normative environment of body mass index (BMI) within schools may influence children's weight trajectories as they age from kindergarten to fifth grade.
METHODS
I use 5 waves of the ECLS‐K—Kindergarten Class 1998‐1999 data and a series of multilevel growth models to examine whether attending schools with higher overall BMI influences children's weight status over time.
RESULTS
Results show that, net of child, family, and school sociodemographic characteristics, children who attend schools with higher rates of obesity have increased weight compared to children who …

Early Childhood Stressors and Negative Personality Behaviors
Research is moving away from static to contextual models of understanding personality development, making it imperative to consider daily occurrences, not just biology or significant traumatic events, in personality formation. Temperament has long been considered the innate source of behaviors that later solidify into personality, but behaviors themselves and the conditions that produce and reinforce them are shaped by the daily environment. This behavior-environment interaction is especially important for children experiencing childhood stressors like poverty, food insecurity, and parental aggravation. Non-clinical personality related behaviors may arise from these stressors, which can lead to …

Electrocortical Responses During Script-Driven Emotional Imagery, Nicholas Best, Cassandra Gonzalez, David Herring Ph.D.
Scholars Week
Emotional imagery is a common induction technique used in the laboratory and also employed in various exposure therapy treatments across the anxiety spectrum (e.g., specific and social phobias). Despite its clinical uses, there is a surprising dearth of literature regarding the basic central neural processes underlying emotional imagery, though other peripheral physiological processes have been investigated extensively using heart rate, skin conductance, and startle-blink responses. One imagery study that used a central nervous system psychophysiological measure -event specific brainwave or the event-related potential (ERP) technique- suggests the late positive potential (LPP) of the ERP is larger for unpleasant versus neutral …

Handwriting is a multisensory process known to enhance memory, focus, engagement, and cognition in the learner, thereby making it a valuable study skill. In an attempt to embed handwriting as a study skill in a gateway science course, I used a combination of scaffolding and individual student consultations. Scaffolding emphasized the use of desirable difficulties and retrieval practice through the implementation of techniques such as the flipped learning approach and take home-in class hybrid essay exams. While the former helped to increase overall student success the latter technique was aimed at enhancing written fluency in the subject, curbing procrastination, and …

Ports and shipping play a crucial role in the coastal development, however, it possesses numerous potential risks to the marine environment and the coastal community. Sustainable development is a key concept which integrates the triple bottom line: economic, environmental and social elements into all aspects of decision making. This paper explores and analyses the challenges towards the progression of sustainable development of Indian ports, taking the Ennore port in India as a case study. My objectives in this research were to highlight the lack of consensus on the importance of the two pillars, environmental and social in the concept of …

Drawing on three cases from a larger (N=169) longitudinal study of student writing development, this article shows how STEM students “rewrote” disciplines to suit their writerly purposes as they moved through their undergraduate years. Students made it clear that the institutional dimensions of disciplines, visible in administrative units or departments that control resources and records, remained visible in their mental landscapes, but they had a much more flexible view of the epistemological dimensions of disciplines. Rather than entering a field as novices aiming to emulate the writing of its experts, they drew on the intellectual resources of multiple disciplines in …

Sampling Frequency Influences Sample Entropy Of Kinematics During Walking, Peter C. Raffalt, John D. Mccamley, William Denton, Jennifer M. Yentes
Journal Articles
Sample entropy (SaEn) has been used to assess the regularity of lower limb joint angles during walking. However, changing sampling frequency and the number of included strides can potentially affect the sample entropy. The present study investigated the effect of sample frequency and the number of included strides on the calculations of SaEn in joint angle signals recorded during treadmill walking. Eleven subjects walked at their preferred walking speed for 10 minutes, and SaEn was calculated on sagittal plane hip, knee and ankle angle signals extracted from 50, 100, 200, 300 and 400 strides at sampling frequencies of 60, 120, …

Quantifying The Probability Of Lethal Injury To Florida Manatees Given Characteristics Of Collision Events., B. Lynn Combs
USF Tampa Graduate Theses and Dissertations
Wherever wildlife share space with boaters, collisions are a potential source of mortality. Establishing protection and speed zones are the primary actions taken to mitigate collision risk. However, creation of protection zones may be a point of contention with stakeholders as new zones can have significant socioeconomic impacts. The Florida Manatee is a prime example of a species whose abundance and viability are constrained by this balance between the needs of humans and wildlife on a shared landscape. The goal of this work is to help further understand the risk to manatees by quantifying the probability of lethal collisions. I …

Connectivity is a major contributor to the overall dynamics of marine populations. However, it still remains challenging to describe connectivity on ecologically meaningful scales of time and space. This is a major impediment to evaluating the impacts of marine protected area with respect to fisheries management objectives.
This dissertation brings together a wide array of spatial and connectivity information in the Gulf of Mexico (GOM) with the goal of 1) understanding the spatial distribution of fish populations and source-sink dynamics and 2) evaluating whether this information can be integrated, through a modeling framework, to identify closed areas that could be …
