Standing surface acoustic waves (SSAW) have been widely used for sorting of cells and particles. However, the major challenges faced with the acoustic driven separation process is the need for an optimized setup to achieve effective separation and the range of particles that can be separated. In this thesis, a custom simulation model is studied to investigate and optimize the separation of varying size particles in a sheathless acoustic separation platform that was developed in our research lab. Insulin-degrading enzyme (IDE) is a zinc metalloprotease that selectively degrades biologically important substrates associated with type 2 diabetes and Alzheimer's disease (AD). As such, IDE is an attractive target for therapeutic innovations. Amajor requirement is an understanding of how other molecules present in cells regulate the activity of the enzyme toward insulin, IDE's most important physiologically relevant substrate. Previous kinetic studies of the IDE-dependent degradation of insulin in the presence of potential regulators have used iodinated insulin, a chemical modification that has been shown to alter the biological and biochemical properties of insulin. Objective: Lynch syndrome (LS) patients are at an elevated risk for early-onset cancers, including endometrial and colorectal (CRC). Prior research has shown a deficit in provider knowledge of LS, which may affect patient satisfaction and adherence to recommended screening and surveillance regimens. Studies suggest patients with LS may educate providers perceived as lacking LS knowledge; however, little is known about these interactions. The goal of this study is to assess patient-reported outcomes from clinical interactions where LS patients educate their providers.
Methods: Participants (n=55) were asked to complete an in-depth telephone interview.

This study examined racial and geographic differences in access to a usual source of care (USC) and it further explored these differences among individuals who had a USC that followed the patient-centered medical home (PCMH) model. Using cross-sectional data from the Household Component of the Medical Expenditure Panel Survey (2008-2013), our sample consisted of non-institutionalized US civilians ages 18-85 (n= 146,233; weighted n = 229,487,016). Our analysis included weighted descriptive statistics and weighted logistic regressions. Although 76% of the respondents had a USC, only 11% of them had a USC that followed the PCMH model. Year-round behavioral observations and surveys were used to investigate the temporal dynamics of life history events and associated variation in behavioral activities of the endangered, island-endemic Hawaiian Duck (Anas wyvilliana) on Kaua‘i, Hawai‘i, USA. Although breeding activity occurred throughout the year, backdated brood observations (n = 67) revealed that birds initiated 94% of nests during the 9-month period from September through May, their peak nesting season. A greater proportion of females (n = 149) were paired during the peak nesting season (75%) than the off-peak season (48%). BACKGROUND: Recent large-scale cancer sequencing studies have discovered many novel cancer driver genes (CDGs) in human cancers. Some studies also suggest that CDG mutations contribute to cancer-associated epigenomic and transcriptomic alterations across many cancer types. Here we aim to improve our understanding of the connections between CDG mutations and altered cancer cell epigenomes and transcriptomes on pan-cancer level and how these connections contribute to the known association between epigenome and transcriptome.

Common practices for invasive species control and management include physical, chemical, and biological approaches. The first two approaches have clear limitations and may lead to unintended (negative) consequences, unless carefully planned and implemented. For example, physical removal rarely completely eradicates the targeted invasive species and can cause disturbances that facilitate new invasions by nonnative species from nearby habitats. Chemical treatments can harm native, and especially rare, species through unanticipated side effects. Biological methods may be classified as biocontrol and the ecological approach. Similar to physical and chemical methods, biocontrol also has limitations and sometimes leads to unintended consequences. Therefore, a …

Environmental gradients constrain physiological performance and thus species’ ranges, suggesting that species occurrence in diverse environments may be associated with local adaptation. Genome–environment association analyses (GEAA) have become central for studies of local adaptation, yet they are sensitive to the spatial orientation of historical range expansions relative to landscape gradients. To test whether potentially adaptive genotypes occur in varied climates in wide‐ranged species, we implemented GEAA on the basis of genomewide data from the anole lizards Anolis ortonii and Anolis punctatus, which expanded from Amazonia, presently dominated by warm and wet settings, into the cooler and less rainy Atlantic …

Several grass and broadleaf weed species around the world have evolved multiple-herbicide resistance at alarmingly increasing rates. Research on the biochemical and molecular resistance mechanisms of multiple-resistant weed populations indicate a prevalence of herbicide metabolism catalyzed by enzyme systems such as cytochrome P450 monooxygenases and glutathione S-transferases and, to a lesser extent, by glucosyl transferases. A symposium was conducted to gain an understanding of the current state of research on metabolic resistance mechanisms in weed species that pose major management problems around the world. Optimal treatment of brain metastases is often hindered by limitations in diagnostic capabilities. To meet this challenge, here we profile DNA methylomes of the three most frequent types of brain metastases: melanoma, breast, and lung cancers (n = 96). Using supervised machine learning and integration of DNA methylomes from normal, primary, and metastatic tumor specimens (n = 1860), we unravel epigenetic signatures specific to each type of metastatic brain tumor and constructed a three-step DNA methylation-based classifier (BrainMETH) that categorizes brain metastases according to the tissue of origin and therapeutically relevant subtypes. BrainMETH predictions are supported by routine histopathologic evaluation. …

Brain metastases (BM) are one the most lethal and poorly managed clinical complications in cancer patients. These secondary tumors represent the most common intracranial neoplasm in adults, most frequently originating from lung cancer, breast cancer, and cutaneous melanoma. In primary brain tumors, such as gliomas, recent advances in DNA methylation profiling have allowed for a comprehensive molecular classification. Such data provide prognostic information, in addition to helping predict patient response to specific systemic therapies. Background: Down syndrome (DS) individuals, by the age of 40s, are at increased risk to develop Alzheimer-like dementia, with deposition in brain of senile plaques and neurofibrillary tangles. Our laboratory recently demonstrated the disturbance of PI3K/AKT/mTOR axis in DS brain, prior and after the development of Alzheimer Disease (AD). The aberrant modulation of the mTOR signalling in DS and AD age-related cognitive decline affects crucial neuronal pathways, including insulin signaling and autophagy, involved in pathology onset and progression. Disclosed are monoacylated Toll-like receptor 2 ligands which can be used in both the development of targeted agents for the imaging and treatment of pancreatic cancer as well as other cancers, and as an adjuvant for cancer immunotherapy. The monoacylated compounds disclosed herein have a higher binding affinity for TLR2 relative to a known potent diacylated agonists, but only â½ the bioactivity. Competition of the monoacylated compound with the diacylated compound for binding TLR2 was confirmed. Hence, the reported monoacylated compounds are inhibitors/antagonists of TLR2 activation.

Asymmetric surfaces been shown to inducing unidirectional motion in the Leidenfrost regime; however, very minimal research has been conducted to investigate whether these surface can enhance condensation through the same means. The investigation of heat transfer of ratchets in condensation is a relatively untapped area of study, specifically ratchets with superhydrophobic properties. Anticipated difficulty lies in creating surfaces features or coatings that retain the ratchets and can adequately sustain optimal wetting state of Cassie-Baxter required to improve heat transfer performance during condensation. James Beasley graduated from Eastern Michigan University in 1977 and was an active member of the Student Senate and other associations on campus. This dissertation helped address the commercialization of triploid Crassostrea virginica production and develop tetraploid C. virginica broodstock in Louisiana by addressing triploid commercialization and tetraploid broodstock development. This was done by: 1) developing regional breeding plans for diploid and tetraploid C. virginica targeted at the Gulf Coastal Region, 2) assessing the effect of chromosome set manipulation on gonadal development in diploid, triploid, and tetraploid C. virginica, 3) quantifying the variation of commercially important hatchery traits in triploid C. virginica larvae from different tetraploid parents, 4) developing an up-to-date remote setting manual for the Gulf Coastal Region for industry use, …

Mountain pine beetle (Dendroctonus ponderosae, Hopkins) is a major disturbance agent in pine ecosystems of western North America. Adaptation to local climates has resulted in primarily univoltine generation time across a thermally diverse latitudinal gradient. We hypothesized that voltinism patterns have been shaped by selection for slower developmental rates in southern populations inhabiting warmer climates. To investigate traits responsible for latitudinal differences we measured lifestage-specific development of southern mountain pine beetle eggs, larvae and pupae across a range of temperatures. Developmental rate curves were fit using maximum posterior likelihood estimation with a Bayesian prior to improve fit stability. …

A novel design of a piezoelectric-based energy harvesting pavement system (PZ-EHPS) is introduced in this dissertation work. The design concept of this PZ-EHPS is to transform asphalt layers into a piezoelectric energy harvester to collect dissipated vehicle kinetic energy in a large-scale system, relying on two conductive asphalt layers and one piezoelectric material layer. To verify the feasibility of the above design concept, this dissertation work practically fabricated and tested a series of PZ-EHPS specimens under the loads from a Material Testing System (MTS) in the laboratory, and theoretically analyzed one typical PZ-EHPS specimen via a three-degree-of-freedom electromechanical model and …
