In archaea, pseudouridine (Ψ) synthase Pus10 modifies uridine (U) to Ψ at positions 54 and 55 of tRNA. In contrast, Pus10 is not found in bacteria, where modifications at those two positions are carried out by TrmA (U54 to m5U54) and TruB (U55 to Ψ55). Many eukaryotes have an apparent redundancy; their genomes contain orthologs of archaeal Pus10 and bacterial TrmA and TruB. Although eukaryal Pus10 genes share a conserved catalytic domain with archaeal Pus10 genes, their biological roles are not clear for the two reasons. Understanding spatial patterns in population characteristics and the principal natal environments supporting riverine fish populations are important for fisheries management. Fin ray microchemistry was used to identify natal environment, and age estimates from sectioned fin rays were used to estimate growth and mortality rates for spotted bass, Micropterus punctulatus (Rafinesque), in a segment of the Ohio River (Smithland Pool) and three tributaries. Differences in water Sr:Ca and Ba:Ca among the Ohio River and tributaries were reflected in fin ray edge Sr:Ca and Ba:Ca. Amyloid-β and α-synuclein are intrinsically disordered proteins (IDPs), which are at the center of Alzheimer’s and Parkinson’s disease pathologies, respectively. These IDPs are extremely flexible and do not adopt stable structures. Furthermore, both amyloid-β and α-synuclein can form toxic oligomers, amyloid fibrils and other type of aggregates in Alzheimer’s and Parkinson’s diseases. Experimentalists face challenges in investigating the structures and thermodynamic properties of these IDPs in their monomeric and oligomeric forms due to the rapid conformational changes, fast aggregation processes and strong solvent effects. Arrestins are soluble relatively small 44–46 kDa proteins that specifically bind hundreds of active phosphorylated GPCRs and dozens of non-receptor partners. There are binding partners that demonstrate preference for each of the known arrestin conformations: free, receptor-bound, and microtubule-bound. Recent evidence suggests that conformational flexibility in every functional state is the defining characteristic of arrestins. Flexibility, or plasticity, of proteins is often described as structural disorder, in contrast to the fixed conformational order observed in high-resolution crystal structures. However, protein-protein interactions often involve highly flexible elements that can assume many distinct conformations upon binding to different partners. Existing evidence suggests …

Butyrophilins (BTNs) are a group of the moonlighting proteins, some members of which are secreted in milk. They constitute a large family of structurally similar type 1 transmembrane proteins from the immunoglobulin superfamily. Although the founding member of this family is related to lactation, participating in the secretion, formation and stabilization of milk fat globules, it may also have a cell surface receptor function. Generally, the BTN family members are known to modulate co-stimulatory responses, T cell selection, differentiation, and cell fate determination. Tick-borne encephalitis (TBE) is a widespread, dangerous infection. Unfortunately, all attempts to create safe anti-TBE subunit vaccines are still unsuccessful due to their low immunogenicity. The goal of the present work was to investigate the immunogenicity of a recombinant chimeric protein created by the fusion of the EIII protein, comprising domain III and a stem region of the tick-borne encephalitis virus (TBEV) E protein, and the OmpF porin of Yersinia pseudotuberculosis (OmpF-EIII). Adjuvanted antigen delivery systems, the tubular immunostimulating complexes (TI-complexes) based on the monogalactosyldiacylglycerol from different marine macrophytes, were used to enhance the immunogenicity of OmpF-EIII. Atp8b1 (ATPase, aminophospholipid transporter, class I, type 8B, member 1) is a cardiolipin transporter in the apical membrane of lung epithelial cells. While the role of Atp8b1 in pneumonia-induced acute lung injury (ALI) has been well studied, its potential role in oxidative stress-induced ALI is poorly understood. We herein show that Atp8b1G308V/G308V mice under hyperoxic conditions display exacerbated cell apoptosis at alveolar epithelium and aberrant proliferation of club cells at bronchiolar epithelium. This hyperoxia-induced ambivalent response in Atp8b1G308V/G308V lungs was followed by patchy distribution of non-uniform interstitial fibrosis at late recovery phase under normoxia. Chikungunya virus (CHIKV) is a mosquito-borne alphavirus. The outbreak of CHIKV infection has been seen in many tropical and subtropical regions of the biosphere. Current reports evidenced that after outbreaks in 2005–06, the fitness of this virus propagating in Aedes albopictus enhanced due to the epistatic mutational changes in its envelope protein. Intrinsically disordered proteins (IDPs) and intrinsically disordered protein regions (IDPRs) are functional proteins and domains that devoid stable secondary and/or tertiary structure. IDPs/IDPRs are abundantly present in various proteomes, where they are involved in regulation, signaling, and control, thereby serving as crucial regulators of various cellular processes. Various mechanisms are utilized to tightly regulate and modulate biological functions, structural properties, cellular levels, and localization of these important controllers. There is growing evidence that parenting stress maintains a strong influence on child externalizing problems; however this link has not been widely studied with adolescent populations. Adolescence can be a period of transitions and is linked to rising levels of parenting stress and negative adolescent outcomes. In this thesis, therefore, in order to properly relate the competition between fractal dimension of a system to its optical scattering properties, and to find the fractal dimension for which maximum scattering occurs, we have performed a systematic characterization of the disorder properties of fractal biological samples with a wide range of fractal dimensions. As cells/tissues are dielectric media, we have indirectly characterize the effective disorder of the fractal media based on their eigenvalues/eigenfunction in a closed-boundary condition using optical waves. We observed a non-monotonous behavior due to the competition between decreases in fractality verses increasing scattering centers.

Biopotential signals such as electroencephalography (EEG), electrocardiography (ECG or EKG), electrooculogram (EOG), and electromyography (EMG) play vital roles in health and clinical diagnoses, monitoring, and therapy. In addition, these signals are required for many nonclinical applications such as Neurofeedback and Brain-Computer Interface (BCI). The quality of the measurement relies on the electrical and mechanical properties of the electrode. AbstractThe purpose of this study was to examine the relationships between secondary level teachers perceived involvement with respect to their making pedagogical and administrative decisions at their schools and four measures of school productivity. Grounded in archived accountability information stored on the Tennessee Department of Education website, these four productivity measures were the student attendance, graduation, suspension, and event dropout rates computed for 248 high schools for the 2012-2013 academic year. Gold and hybrid gold nanotechnology are uniquely poised to improve the fields of cancer diagnosis and therapy. Modifying gold nanoparticles with biological proteins has been widely used for decades, however, the common carbodiimide technique requires steps that reduce sensitivity and effectiveness in some spectroscopic techniques. We have developed an antibody conjugation protocol which conducts linking first, and then covalently adheres the antibody-linker complex to the nanoparticle surface. The meadow vole (Microtus pennsylvanicus) is a small herbivorous rodent inhabiting grassy fields throughout the Northern United States and Canada. Meadow voles are promiscuous, with males and females taking multiple mates during a single breeding season. Female meadow voles are territorial while males occupy large home ranges that encompass the territories of multiple females. My research focused on how changes in diet or food availability influenced the sexual and exploratory behaviors of meadow voles. SK1 is an intracellular enzyme whose lipid kinase activity regulates the levels of bioactive lipids that are involved in normal homeostasis and disease. SK1 activity is implicated in various diseases including cancer, and inhibition of SK1 activity is a potential treatment for its implicated pathogenesis. However, without a high-throughput SK1 activity assay, screening and identification of SK1 inhibitors is a rigorous process that requires significant time and resources to yield potent lead compounds. Prior research demonstrated intergenerational effects of maternal risk factors on offspring birth weight. However, only a limited number of studies investigated whether grand-maternal risk factors, i.e. body mass index (BMI) and socioeconomic status (SES), affected grandchilds birth weight (GBW). Besides, it is unknown whether such associations are direct or mediated by maternal risk factors. For the past sixty years, acquiring scientific literacy has proven to be a daunting task in education, especially for undergraduate nonscience majors.
