Early childhood weight status and its effects on socioemotional development, school readiness, and cognition.Major Professor: Vikki Nolan, DSc, MPHChildhood obesity is associated with negative health and social outcomes in school-age children. These effects range from absenteeism and poor self-esteem to worse test scores, and fewer completed grades. Although previous studies have found these associations in school-age children and adolescents, less is known about this association in young children, who are just beginning school. AbstractThe purpose of this study was to examine the relationships between secondary level teachers perceived involvement with respect to their making pedagogical and administrative decisions at their schools and four measures of school productivity. Grounded in archived accountability information stored on the Tennessee Department of Education website, these four productivity measures were the student attendance, graduation, suspension, and event dropout rates computed for 248 high schools for the 2012-2013 academic year. At these same 248 institutions, teachers perceived involvement was computed from responses to a section on teachers roles in decision making that appeared on the 2013 state-wide administration of …

Gold and hybrid gold nanotechnology are uniquely poised to improve the fields of cancer diagnosis and therapy. Modifying gold nanoparticles with biological proteins has been widely used for decades, however, the common carbodiimide technique requires steps that reduce sensitivity and effectiveness in some spectroscopic techniques. We have developed an antibody conjugation protocol which conducts linking first, and then covalently adheres the antibody-linker complex to the nanoparticle surface. The linking polymer N-hydroxysuccinimide-polyethylene glycol-thiol (NHS-PEG-SH) was incubated with a protein overnight and purified by centrifuge filtration before modifying the surface of gold nanoparticles by slow mixing. The meadow vole (Microtus pennsylvanicus) is a small herbivorous rodent inhabiting grassy fields throughout the Northern United States and Canada. Meadow voles are promiscuous, with males and females taking multiple mates during a single breeding season. Female meadow voles are territorial while males occupy large home ranges that encompass the territories of multiple females. My research focused on how changes in diet or food availability influenced the sexual and exploratory behaviors of meadow voles. In Chapters 1 and 2, I assessed the influence of phytoestrogens on the sexual behavior of meadow voles. Phytoestrogens are nonsteroidal compounds produced by plants that …

SK1 is an intracellular enzyme whose lipid kinase activity regulates the levels of bioactive lipids that are involved in normal homeostasis and disease. SK1 activity is implicated in various diseases including cancer, and inhibition of SK1 activity is a potential treatment for its implicated pathogenesis. However, without a high-throughput SK1 activity assay, screening and identification of SK1 inhibitors is a rigorous process that requires significant time and resources to yield potent lead compounds. Prior research demonstrated intergenerational effects of maternal risk factors on offspring birth weight. However, only a limited number of studies investigated whether grand-maternal risk factors, i.e. body mass index (BMI) and socioeconomic status (SES), affected grandchilds birth weight (GBW). Besides, it is unknown whether such associations are direct or mediated by maternal risk factors. Grand-maternal smoking during pregnancy was additionally included due to many studies addressing the adverse effect of maternal smoking on birth weight. For the past sixty years, acquiring scientific literacy has proven to be a daunting task in education, especially for undergraduate nonscience majors. Although some education scholars have recognized the importance and use of arguments to teach science, these pedagogical practices often are aimed at primary school children rather than college students or involve reporting science experiments rather than actually studying or constructing arguments about issues related to science.However, in this dissertation, I contend that educators often neglect a more available tool that not only examines arguments concerning scientific issues but also demonstrates the very heart of scientific literacy: critical thinking. …

Elucidating how dopamine neurons operate regularly in aspects of neurochemical release and in pathological systems is essential for understanding their role in behavior, drug treatment, and disease. Fixed-potential amperometry (FPA) combined with carbon fiber microelectrodes has proven to be effective at probing these dynamic microsystems and precisely describing the regulatory mechanisms that govern dopamine neurotransmission. Parental posttraumatic stress disorder (PTSD) has been shown to negatively impact children’s socioemotional development (Schwerdtfeger et al., 2014) and increase children’s risk for later psychopathology (Scheeringa & Zeanah, 2008; Yehuda, Halligan, & Bierer, 2001). The gut microbiome may play a role in the development of chronic diseases like obesity, diabetes, and heart disease. Diets including prebiotics or probiotics can alter the abundance of gut bacterial groups and have subsequent health effects. In this study we wanted to establish a method for comparing the benefits of commercial supplements that alter the microbiota by monitoring fecal microbial profiles in male C57BL/6 mice (n = 24) exposed to 15 days of dietary supplementation. Objective: Neurocognitive screening measures reflective of functional status are needed for detection of executive dysfunction. The Frontal Assessment Battery (FAB), a measure designed to assess executive functioning at bedside, lacks sufficient research establishing its ecological validity. The purpose of the study was to evaluate the ecological validity of the FAB by examining its relationship with the Functional Status Interview (FSI), a measure of basic activities (ADL) and instrumental activities (IADL), and a measure of caregiver burden (ZBI-R). The overall goal of this dissertation was to examine the adaptivenss of eight emotion regulation strategies for Chinese middle and high school students. The adaptiveness was evaluated by using an intrapersonal approach that focused on individual students in Study 1 and by using an interpersonal approach that focused on same-sex best friend dyads in Study 2. By using an intrapersonal approach, Study 1 found that students chose to use developing competencies, redirecting attention, expressive suppression, and seeking social support more often than the other four strategies. In response to the growing interest in mind-body practices as preventative, therapeutic and educational interventions, the current work describes the instrument development and validation process to address a gap in yoga research. Yoga is a mind-body practice used in clinical and school settings that uses an integrative curriculum which simultaneously targets skill development/maintenance in physical, cognitive and emotional realms. Yoga is heterogeneous, as there is variability across styles and interventions. Yoga is also educational, as there are integrative skills being learned. This study examines the intersections of place and second language learning. Learner identity has been found to be an important construct in second language learning. In recent years, place and space have become central topics in the study of sociolinguistics and identity. Chytridiomycosis is an infectious amphibian disease caused by the pathogenic fungus Batrachochytrium dendrobatidis (Bd). Bd attaches to keratin on the epidermis of amphibians, invades skin cells, and may lead to pathogenesis in susceptible individuals. However, susceptibility varies within and among species. While this is due to many factors, the skin microbial community is a significant contributor to disease resistance. Amphibians form symbiotic relationships with environmental microbes on the skin surface, some of which produce antifungal agents that inhibit Bd. Interestingly, many agricultural biopesticides utilize the common soil-dwelling bacteria, Bacillus thuringiensis. Through agricultural use, these bacteria likely increase in environmental abundance …

The first epithelial to mesenchymal transition (EMT) occurs in trophoblast stem (TS) cells during implantation. Inactivation of the serine/threonine kinase MAP3K4 in TS cells (TSKI4 cells) induces an intermediate state of EMT, where cells retain stemness, lose epithelial markers, and gain mesenchymal characteristics. Investigation of relationships among MAP3K4 activity, stemness, and EMT in TS cells may reveal key regulators of EMT. Here, we show that MAP3K4 activity controls EMT through the ubiquitination and degradation of HDAC6. Loss of MAP3K4 activity in TSKI4 cells results in elevated HDAC6 expression and the deacetylation of cytoplasmic and nuclear targets. In the nucleus, HDAC6 …

Prior studies linking breastfeeding duration and maternal gestational smoking, respectively, to offspring eczema report contradictory findings. However, it is not known whether maternal gestational smoking and breastfeeding duration can jointly influence eczema and whether these exposures mediate their effect on eczema via DNA Methylation (DNA-M) of CpG (cytosine-phosphate-guanine) sites. With data from the Isle of Wight (IOW) birth cohort, UK, we first evaluated the combined effect of maternal gestational smoking and breastfeeding duration on eczema at ages 1-to-2 (n=980), 4 (n=902), 10 (n=966), and 18 years (n=929) using Generalized estimating equations (GEE). Circadian clocks are oscillator systems that drive rhythms in behavior and physiology in an organism to coordinate with the light-dark cycle. The molecular clock is made up of core clock genes acting in a transcriptional negative feedback loop. Transcriptional activators BMAL1 and CLOCK form a heterodimer which binds to cis E/E-box elements present in the promoter region of target genes including Per1,2,3 and Cry1,2. PER and CRY proteins then translocate to the nucleus to repress BMAL1/CLOCK-mediated transcription. This transcriptional negative feedback mechanism underlies circadian oscillations. Direct drug administration has several shortcomings such as low circulation concentration due to rapid flushing by the immune system, non-specificity to target tissue and inefficient delivery to avascular sites. A widely adopted solution to these limitations is to sterically shield the drug from a harsh in vivo environment by encapsulation in a biocompatible, bio-degradable substrate. Hip-hop has been shown to be a significant force of identity, knowledge, and cultural development, particularly for Black youth (Brown, 2009; Emdin, 2010; Hill, 2009; Love, 2012). Building on research surrounding identity and knowledge development within hip-hop, this Black/hip-hop feminist research study seeks to understand the role that hip-hop plays in the lives, identity and personal epistemology of Black adolescent girls. Urbanization of the southeastern region of the United States is an ongoing process with wide ranging environmental implications. Tennessee is among southern states expected to develop at higher than average rates. Therefore, the purpose of this doctoral research is twofold; to holistically examine urbanization in Jackson, Madison County, TN and the environmental impact of urbanization on the region. Adults with disabilities face multiple internal, social, and systemic barriers that hamperefforts to attain and maintain meaningful employment. In order to achieve success in vocational pursuits, individuals with disabilities must demonstrate perseverance in the face of probable adversity; this construct is career resilience. No scale has been developed to measure a persons career resilience that is specifically tailored to people with disabilities. The purpose of this study was to develop an instrument to measure the career resilience of adults with disabilities. This study determines the content for a supplemental curriculum designed to improve musical literacy and musical expression skills using the Arkansas School Band and Orchestra (ASBOA) all-state junior high flute etudes. It addresses the first two steps (analysis of practical problems and development of possible solutions) of a Design-Based Research (DBR) project. The author intends to address the third and fourth steps (iterative cycles of testing and refinement and reflection and enhancement) in future research studies. Identifying Pathways To Smoking And Alcohol Consumption After A Natural Disaster: Findings From Hurricane Katrina, Adam Alexander

Survivors of disasters often experience severe psychological symptoms and disorders, including depression, anxiety, post-traumatic stress disorder (PTSD), and report using psychoactive substances, such as alcohol, marijuana, and cigarettes. This dissertation examined whether depression severity and PTSD were pathways to postdisaster smoking and alcohol consumption using a representative cohort of adult cigarette smokers from Memphis, Tennessee and New Orleans, Louisiana nine and eighteen months after Hurricane Katrina (N=2004). Results from path analyses showed that disaster exposure increased depression and posttraumatic stress nine months after Hurricane Katrina, and the effect persisted across time for depression.
