Background
Metastases play a role in about 90% of cancer deaths. Markers of epithelial-mesenchymal transition (EMT) measured in primary tumor cancer cells might provide diagnostic information about the likelihood that cancer cells have detached from the primary tumor. Used together with established diagnostic tests of detachment—lymph node evaluation and radiologic imaging—EMT marker measurements might improve the ability of clinicians to assess the patient’s risk of metastatic disease. Translation of EMT markers to clinical use has been hampered by a lack of valid analyses of clinically-informative parameters. Here, we demonstrate a rigorous approach to estimating the sensitivity, specificity, and prediction increment …

Daily undulating periodization (DUP), using daily alterations in repetitions, has been advocated as a superior method of resistance training, while traditional forms of programming for periodization (Block) have been questioned. Nineteen Division I track and field athletes were assigned to either a 10-week Block or DUP training group. Year and event were controlled. Over the course of the study, there were four testing sessions, which were used to evaluate a variety of strength characteristics, including maximum isometric strength, rate of force development, and one repetition maximum (1RM). Although, performance trends favored the Block group for strength and rate of force …

Biomedical Applications Of Hydrogels And Gumbos, Kelsey Elizabeth Mcneel
LSU Doctoral Dissertations
Sodium deoxycholate (NaDC) is a small bile salt that was used in this dissertation to produce gelation of tris(hydroxymethyl)amino-methane (TRIS) solutions above, below, and near the pKa of NaDC. These solutions respectively yielded a neutral gelator, a charged gelator, and a mixture of each. Impacts of ionic interactions on gel formation were studied in detail and showed that pH can be used to modify many hydrogel properties including sol-gel temperature, crystallinity, and mechanical strength. It was also observed that pH modification of the hydrogels affected nanoparticle formation. Nanoparticles derived from a Group of Uniform Materials Based on Organic Salts (nanoGUMBOS), …

An Invertebrate Ecosystem Engineer Under The Umbrella Of Sage-Grouse Conservation, Jason D. Carlisle, David R. Stewart, Anna D. Chalfoun
Western North American Naturalist
Conservation practitioners often rely on areas designed to protect species of greatest conservation priority to also conserve co-occurring species (i.e., the umbrella species concept). The extent to which vertebrate species may serve as suitable umbrellas for invertebrate species, however, has rarely been explored. Sage-grouse (Centrocercus spp.) have high conservation priority throughout much of the rangelands of western North America and are considered an umbrella species through which the conservation of entire rangeland ecosystems can be accomplished. Harvester ants are ecosystem engineers and play important roles in the maintenance and function of rangeland ecosystems. We compared indices of the abundance …

Decoherence of quantum systems due to uncontrolled fluctuations of the environment presents fundamental obstacles in quantum science. Clock transitions which are insensitive to such fluctuations are used to improve coherence, however, they are not present in all systems or for arbitrary system parameters. Here we create a trio of synthetic clock transitions using continuous dynamical decoupling in a spin-1 Bose-Einstein condensate in which we observe a reduction of sensitivity to magnetic-field noise of up to four orders of magnitude; this work complements the parallel work by Anderson et al. [R. P. Anderson et al., following paper, Phys. Rev. A 97, …

The Effects Of Tree Plantations And Land Use On Natural Regeneration Of Woody Plants In The Tropics: Diversity, Species Composition, Successional Dynamics, And Functional Traits, John Benjamin Longworth
LSU Doctoral Dissertations
As anthropogenic forests become more common in the tropics, there is a greater need to understand the factors that impact forest succession. I used censuses of woody plants in successional forests to study the effects of prior land use and tree plantations on species composition and richness. First, I tested the hypothesis that communities originating from different land uses were converging in species composition over time. I compared species composition using the Chao-Jaccard similarity index. I observed shifts in the dominant species during the first 30 years of succession, but not convergence of species composition in sites with different land-use …

Stochastic Estimation Of Field Management Zones Using Multi-Year Yield Data And A Hidden Markov Random Field, Alex Layton, James V. Krogmeier, Aaron Ault, Dennis Buckmaster
Department of Electrical and Computer Engineering Technical Reports
Modern precision agriculture equipment enables treating different areas of a field differently, i.e., site specific management. This results in the concept of management zones, which are a compromise between treating a field uniformly and treating every plant individually.
This work presents an algorithm for inferring the management zones for a field based on yield data from multiple years. The algorithm uses a hidden Markov random field model (HMRF) to find regions of the field which likely correspond to the same underlying yield distribution (i.e., “management zones”). These regions are assumed to be the same for each year, but their distributions …

Differentially-Dimensioned Furrow Formation By Zygotic Gene Expression And The Mbt, Yi Xie, J Todd Blankenship
Differentially-Dimensioned Furrow Formation By Zygotic Gene Expression And The Mbt, Yi Xie, J Todd Blankenship
Biological Sciences: Faculty Scholarship
Despite extensive work on the mechanisms that generate plasma membrane furrows, understanding how cells are able to dynamically regulate furrow dimensions is an unresolved question. Here, we present an in-depth characterization of furrow behaviors and their regulation in vivo during early Drosophila morphogenesis. We show that the deepening in furrow dimensions with successive nuclear cycles is largely due to the introduction of a new, rapid ingression phase (Ingression II). Blocking the midblastula transition (MBT) by suppressing zygotic transcription through pharmacological or genetic means causes the absence of Ingression II, and consequently reduces furrow dimensions. The analysis of compound chromosomes that …

Effects of curcumin, a biologically active ingredient of turmeric, were tested on the Ca2+transients induced by the activation of α7 subunit of the human nicotinic acetylcholine (α7nACh) receptor expressed in SH-EP1 cells. Curcumin caused a significant potentiation of choline (1 mM)-induced Ca2+ transients with an EC50 value of 133 nM. The potentiating effect of curcumin was not observed in Ca2+ transients induced by high K+ (60 mM) containing solutions or activation of α4β2 nACh receptors and the extent of curcumin potentiation was not altered in the presence of …

Forest biomass is an important resource for producing bioenergy and reducing greenhouse gas (GHG) emissions. The State of Michigan in the United States (U.S.) is one region recognized for its high potential of supplying forest biomass; however, the long-term availability of timber harvests and the associated harvest residues from this area has not been fully explored. In this study time trend analyses was employed for long term timber assessment and developed mathematical models for harvest residue estimation, as well as the implications of use for ethanol. The GHG savings potential of ethanol over gasoline was also modeled. The methods were …

Ovarian cancer (OC) has the highest rate of mortality among gynecological malignancy. Chemokine receptor CXCR2 in OC is associated with poor outcomes. However, the mechanisms by which CXCR2 regulates OC proliferation remain poorly understood. We generated CXCR2-positive cells from parental p53 wild-type (WT), mutant and null OC cells, and assessed the roles of CXCR2 on proliferation of OC cells in p53-dependent and independent manner. CXCR2 promoted cell growth rate: p53WT > mutant = null cells. Nutlin-3, a p53 stabilizer, inhibited cell proliferation in p53WT cells, but had little effect in p53-mutant or null cells, indicating p53-dependence of CXCR2-mediated proliferation. CXCR2 decreased …

Netprophet 2.0: Mapping Transcription Factor Networks By Exploiting Scalable Data Resources, Yiming Kang, Hien-Haw Liow, Ezekiel J Maier, Michael R Brent
2010-2019 OA Pubs
MOTIVATION: Cells process information, in part, through transcription factor (TF) networks, which control the rates at which individual genes produce their products. A TF network map is a graph that indicates which TFs bind and directly regulate each gene. Previous work has described network mapping algorithms that rely exclusively on gene expression data and 'integrative' algorithms that exploit a wide range of data sources including chromatin immunoprecipitation sequencing (ChIP-seq) of many TFs, genome-wide chromatin marks, and binding specificities for many TFs determined in vitro. However, such resources are available only for a few major model systems and cannot be easily …
Is Cancer Progression Caused By Gradual Or Simultaneous Acquisitions Of New Chromosomes?, Mathew Bloomfield, Peter Duesberg
Is Cancer Progression Caused By Gradual Or Simultaneous Acquisitions Of New Chromosomes?, Mathew Bloomfield, Peter Duesberg
Natural Sciences and Mathematics | Student Professional Publications
Background
Foulds defined, “Tumor progression (as a) permanent, irreversible qualitative change in one or more of its characters” (Cancer Res. 1954). Accordingly progressions, such as metastases and acquired drug-resistance, were since found to be subspecies of cancers with conserved and numerous new chromosomes. Here we ask whether cancers acquire numerous new chromosomes gradually or simultaneously in progressions. The currently prevailing theory of Nowell (Science, 1976) holds that unexplained “genetic instability” generates “variant sublines (with) changes in chromosome number” and that “clonal” progressions arise by “stepwise selection of more aggressive sublines”. The literature, however, contains many examples of “immediate” selections of …

Justifying A Standard Of Death, Michael Milhim
Justifying A Standard Of Death, Michael Milhim
Honors Projects
There are three major positions in the legal definition of death debate: the cardio-pulmonary standard, the whole-brain standard, and the higher-brain standard. Prominent arguments for each standard appeal to a theory of human persistence. I’ll contend that these arguments fail for two reasons: the metaphysical underpinnings of the arguments are not decisive, and even if they are decisive, they may not be the right policy to enact. The later of these is more practically important than the former.
