Dual-Display Laparoscopic Laser Speckle Contrast Imaging For Real-Time Surgical Assistance, Jaepyeong Cha, Corey Zheng, Lung Wai Lau
Pediatrics Faculty Publications
Laser speckle contrast imaging (LSCI) utilizes the speckle pattern of a laser to determine the blood flow in tissues. The current approaches for its use in a clinical setting require a camera system with a laser source on a separate optical axis making it unsuitable for minimally invasive surgery (MIS). With blood flow visualization, bowel viability, for example, can be determined. Thus, LSCI can be a valuable tool in gastrointestinal surgery. In this work, we develop the first-of-its-kind dual-display laparoscopic vision system integrating LSCI with a commercially available 10mm rigid laparoscope where the laser has the same optical axis as …

Influence Of Crossbreeding On Meat Goat Doe Fitness When Comparing Boer F1 With Base Breeds In The Southeastern United States, Piush Khanal, Maria L. Leite-Browning, Richard Browning Jr.
Agricultural and Environmental Sciences Faculty Research
Understanding fitness level among various breeds is essential for sustainable meat goat production. Research on the relative fitness of Boer F1 does and straightbred base breed has been limited. Meat goat does of various genotypes (Boer, Kiko, Spanish, Boer × Kiko reciprocal F1 crosses, and Boer × Spanish reciprocal F1 crosses) were studied to evaluate breed effects on doe fitness traits and the expression of heterosis over 7 production years. The herd was semi-intensively managed under humid subtropical pasture. Doe age affected (P < 0.05) various traits. Boer × Kiko does were heavier (P < 0.05) than Boer does at fall breeding, but Boer × Spanish does did not differ (P > 0.05) from Boer does for breeding weight. The body weights of …

Kinetic Characterization Of The Shigella Type Three Secretion System Atpase Spa47 Using Α-32p Atp, Heather B. Case, Nicholas E. Dickenson
Chemistry and Biochemistry Faculty Publications
ATPases represent a diverse class of enzymes that utilize ATP hydrolysis to support critical biological functions such as driving ion pumps, providing mechanical work, unfolding/folding proteins, and supporting otherwise thermodynamically unfavorable chemical reactions. We have recently shown that the Shigella protein Spa47 is an ATPase that supports protein secretion through its specialized type three secretion apparatus (T3SA), supporting infection of human host cells. Characterizing ATPases, such as Spa47, requires a means to accurately determine enzyme activity (ATP hydrolysis) as a function of time, reaction conditions, and potential cofactors, regulators, inhibitors, etc. Here, we describe a detailed protocol for characterizing …

Survivors of childhood brain tumors (SCBT) face a higher risk of cardiometabolic disorders and premature mortality compared to the general population. Excess adiposity is a known risk factor for these comorbidities. However, while SCBT have higher adiposity compared to healthy controls, measuring adiposity in clinical practice involves access to specialized equipment and may impact busy clinical services. Application Of Click Chemistry In The Development Of Peptide Based Hiv Fusion Inhibitors, Mukesh Madan Mudgal, Nagaraju Birudukota
Chemistry and Physics Faculty Articles
Human Immunodeficiency Virus (HIV) continues to be a major global public health issue. Inhibition of HIV envelope fusion with the CD4 cell membrane prevents the entry of HIV into the CD4 cells providing a novel approach to the treatment of HIV infection. Thus, interference in the fusion of the virus with the co-receptor substrate appears to be a specific and potential way to fight HIV infection and replication. Applications of click chemistry are spreading in the field of drug discovery and it became a powerful tool for the synthesis of medicinally important compounds.
Remarkably, the Cu (I)-Catalyzed Azide-Alkyne Cycloaddition (CuAAC) …

Impact Of Precision Medicine On Drug Repositioning And Pricing: A Too Small To Thrive Crisis, Venkataswarup Tiriveedhi
Biology Faculty Research
The pricing of targeted medicines continues to be a major area of contention in healthcare economics. This issue is further complicated by redefining the role of molecular testing in precision medicine. Currently, whilst pricing of clinical laboratory diagnostics is cost-based, drug pricing is value-based. The pricing for molecular testing is under pressure to change the traditional business model, for it has a critical subsidiary role in determining the final value of targeted medicines. The market size for drugs is reduced by molecular testing when patients with the same disease are stratified based on their genetics, it is critical to determine …

Role Of Noise And Parametric Variation In The Dynamics Of Gene Regulatory Circuits., Vivek Kohar, Mingyang Lu
Role Of Noise And Parametric Variation In The Dynamics Of Gene Regulatory Circuits., Vivek Kohar, Mingyang Lu
Faculty Research 2018
Stochasticity in gene expression impacts the dynamics and functions of gene regulatory circuits. Intrinsic noises, including those that are caused by low copy number of molecules and transcriptional bursting, are usually studied by stochastic simulations. However, the role of extrinsic factors, such as cell-to-cell variability and heterogeneity in the microenvironment, is still elusive. To evaluate the effects of both the intrinsic and extrinsic noises, we develop a method, named sRACIPE, by integrating stochastic analysis with random circuit perturbation (RACIPE) method. RACIPE uniquely generates and analyzes an ensemble of models with random kinetic parameters. Previously, we have shown that the gene …

Articles, Abstracts, and Reports
The present study seeks to present a single-blind, randomized control trial of a hypothermic anti-inflammatory device, the endorectal cooling balloon (ECB), to assess whether regional hypothermia could improve 90-day and time to pad-free continence following robot-assisted radical prostatectomy (RARP). Five high-volume surgeons at three institutions had patients randomized (1:1) to regional hypothermia with ECB versus control. Patients were blinded to device use, as it was inserted and removed intraoperatively. Knowledge of device use was restricted to the operating room personnel only; recovery room and ward nursing staff were not informed of device use and instructed to indicate such if a …

Kinetics Of Dextromethorphan-O-Demethylase Activity And Distribution Of Cyp2d In Four Commonly-Used Subcellular Fractions Of Rat Brain, Barent N. Dubois, Farideh Amirrad, Reza Mehvar
Pharmacy Faculty Articles and Research
The purpose of this study was to compare the enzymatic kinetics and distribution of cytochrome P450 2D (CYP2D) among different rat brain subcellular fractions.
Rat brains were used to prepare total membrane, crude mitochondrial, purified mitochondrial, and microsomal fractions, in addition to total homogenate. Michaelis–Menten kinetics of the brain CYP2D activity was estimated based on the conversion of dextromethorphan (DXM) to dextrorphan using UPLC-MS/MS. Protein levels of CYP2D and subcellular markers were determined by Western blot.
Microsomal CYP2D exhibited high affinity and low capacity, compared with the mitochondrial CYP2D that had a much lower (∼50-fold) affinity but a higher (∼six-fold) …

What Conditions Enable Decentralization To Improve The Health System? Qualitative Analysis Of Perspectives On Decision Space After 25 Years Of Devolution In The Philippines, Harvy Joy Liwanag, Kaspar Wyss
Ateneo School of Medicine and Public Health Publications
Background
Decentralization is promoted as a strategy to improve health system performance by bringing decision-making closer to service delivery. Some studies have investigated if decentralization actually improves the health system. However, few have explored the conditions that enable it to be effective. To determine these conditions, we have analyzed the perspectives of decision-makers in the Philippines where devolution, one form of decentralization, was introduced 25 years ago.
Methods
Drawing from the “decision space” approach, we interviewed 27 decision-makers with an average of 23.6 years of working across different levels of the Philippine government health sector and representing various local settings. …
