A Pan-Cancer Analysis Of Driver Gene Mutations, Dna Methylation And Gene Expressions Reveals That Chromatin Remodeling Is A Major Mechanism Inducing Global Changes In Cancer Epigenomes., Ahrim Youn, Kyung In Kim, Raul Rabadan, Benjamin Tycko, Yufeng Shen, Shuang Wang

Faculty Research 2018

BACKGROUND: Recent large-scale cancer sequencing studies have discovered many novel cancer driver genes (CDGs) in human cancers. Some studies also suggest that CDG mutations contribute to cancer-associated epigenomic and transcriptomic alterations across many cancer types. Here we aim to improve our understanding of the connections between CDG mutations and altered cancer cell epigenomes and transcriptomes on pan-cancer level and how these connections contribute to the known association between epigenome and transcriptome.

METHOD: Using multi-omics data including somatic mutation, DNA methylation, and gene expression data of 20 cancer types from The Cancer Genome Atlas (TCGA) project, we conducted a pan-cancer analysis …

Improving Ecological Restoration To Curb Biotic Invasion—A Practical Guide, Qinfeng Guo, Dale G. Brockway, Diane L. Larson, Deli Wang, Hai Ren Nov 2018

Improving Ecological Restoration To Curb Biotic Invasion—A Practical Guide, Qinfeng Guo, Dale G. Brockway, Diane L. Larson, Deli Wang, Hai Ren

United States Department of Agriculture, Forest Service / University of Nebraska-Lincoln: Faculty Publications

Common practices for invasive species control and management include physical, chemical, and biological approaches. The first two approaches have clear limitations and may lead to unintended (negative) consequences, unless carefully planned and implemented. For example, physical removal rarely completely eradicates the targeted invasive species and can cause disturbances that facilitate new invasions by nonnative species from nearby habitats. Chemical treatments can harm native, and especially rare, species through unanticipated side effects. Biological methods may be classified as biocontrol and the ecological approach. Similar to physical and chemical methods, biocontrol also has limitations and sometimes leads to unintended consequences. Therefore, a …

Articles, Abstracts, and Reports

Optimal treatment of brain metastases is often hindered by limitations in diagnostic capabilities. To meet this challenge, here we profile DNA methylomes of the three most frequent types of brain metastases: melanoma, breast, and lung cancers (n = 96). Using supervised machine learning and integration of DNA methylomes from normal, primary, and metastatic tumor specimens (n = 1860), we unravel epigenetic signatures specific to each type of metastatic brain tumor and constructed a three-step DNA methylation-based classifier (BrainMETH) that categorizes brain metastases according to the tissue of origin and therapeutically relevant subtypes. BrainMETH predictions are supported by routine histopathologic evaluation. …

Articles, Abstracts, and Reports

Brain metastases (BM) are one the most lethal and poorly managed clinical complications in cancer patients. These secondary tumors represent the most common intracranial neoplasm in adults, most frequently originating from lung cancer, breast cancer, and cutaneous melanoma. In primary brain tumors, such as gliomas, recent advances in DNA methylation profiling have allowed for a comprehensive molecular classification. Such data provide prognostic information, in addition to helping predict patient response to specific systemic therapies. However, epigenetic alterations of metastatic brain tumors with specific biological and translational relevance still require much further exploration. Using the widely employed Illumina Infinium HumanMethylation 450K …

Background: Down syndrome (DS) individuals, by the age of 40s, are at increased risk to develop Alzheimer-like dementia, with deposition in brain of senile plaques and neurofibrillary tangles. Our laboratory recently demonstrated the disturbance of PI3K/AKT/mTOR axis in DS brain, prior and after the development of Alzheimer Disease (AD). The aberrant modulation of the mTOR signalling in DS and AD age-related cognitive decline affects crucial neuronal pathways, including insulin signaling and autophagy, involved in pathology onset and progression. Within this context, the therapeutic use of mTOR-inhibitors may prevent/attenuate the neurodegenerative phenomena. By our work we aimed to rescue mTOR signalling …

Blockchain Demystified: A Technical And Legal Introduction To Distributed And Centralised Ledgers, Jean Bacon, Johan David Michels, Christopher Millard, Jatinder Singh

Richmond Journal of Law & Technology

This paper provides an introduction to blockchain technology and its legal implications. The paper consists of two parts. The first part looks at the technology behind the hype. It explains how blockchain technology works and can be deployed in various ways to create applications with different features, including open, distributed and closed, and centralised platforms. The second part analyses the technology’s implications for several areas of law that will be relevant to companies and other organisations that seek to use blockchain technology, namely: contract law, data protection law, securities law, property law, intellectual property, and company law. The purpose of …

Gdpr: The End Of Google And Facebook Or A New Paradigm In Data Privacy?, Kimberly A. Houser, W. Gregory Voss

Richmond Journal of Law & Technology

EU Data Protection Agencies have been vigorously enforcing violations of regional and national data protection law in recent years against U.S. tech companies, but few changes have been made to their business model of exchanging free services for personal data. With the Cambridge Analytica debacle revealing how insufficient American privacy law is, we now find ourselves questioning whether the General Data Protection Regulation (GDPR) is not the onerous 99 article regulation to be feared, but rather a creation years ahead of its time. This paper will explain how the differences in U.S. and EU privacy and data protection law and …

Toll-Like Receptor 2 Ligands And Methods Of Making And Using Thereof, David L. Morse, Josef Vagner, Mark Mclaughlin, Robert Gillies, Amanda Huynh, Michael Doligalski

USF Patents

Disclosed are monoacylated Toll-like receptor 2 ligands which can be used in both the development of targeted agents for the imaging and treatment of pancreatic cancer as well as other cancers, and as an adjuvant for cancer immunotherapy. The monoacylated compounds disclosed herein have a higher binding affinity for TLR2 relative to a known potent diacylated agonists, but only −½ the bioactivity. Competition of the monoacylated compound with the diacylated compound for binding TLR2 was confirmed. Hence, the reported monoacylated compounds are inhibitors/antagonists of TLR2 activation.

Effects Of A Community Population Health Initiative On Blood Pressure Control In Latinos, James R Langabeer, Timothy D Henry, Carlos Perez Aldana, Larissa Deluna, Nora Silva, Tiffany Champagne-Langabeer

Faculty, Staff and Student Publications

Background Hypertension remains one of the most important, modifiable cardiovascular risk factors. Yet, the largest minority ethnic group (Hispanics/Latinos) often have different health outcomes and behavior, making hypertension management more difficult. We explored the effects of an American Heart Association-sponsored population health intervention aimed at modifying behavior of Latinos living in Texas. Methods and Results We enrolled 8071 patients, and 5714 (65.7%) completed the 90-day program (58.5 years ±11.7; 59% female) from July 2016 to June 2018. Navigators identified patients with risk factors; initial and final blood pressure ( BP ) readings were performed in the physician's office; and interim …
