PURPOSE: Gliomas, a genetically heterogeneous group of primary central nervous system tumors, continue to pose a significant clinical challenge. Discovery of chromosomal rearrangements involving kinase genes has enabled precision therapy, and improved outcomes in several malignancies.

EXPERIMENTAL DESIGN: Positing that similar benefit could be accomplished for patients with brain cancer, we evaluated The Cancer Genome Atlas (TCGA) glioblastoma dataset. This study explores the palynology of the Cenomanian Woodbine Group in order to answer questions about the paleoecology of North Texas during the Cenomanian and how palynofloras vary at different localities with different facies, as well as the radiation of angiosperms in the Late Cretaceous. This study also helps our understanding of the paleoecology after the Albian Trinity Group in Texas. Three localities in North Texas were sampled and a total of seventeen productive samples were analyzed for palynological data. Pollen was extracted from mudstones from two depositional environments: lower delta plain marshes and a meandering river system. The palynomorphs …

The influence of online education on higher education fundraising practices and giving patterns is not well understood. This qualitative case study explored how fundraising managers perceived the impact of online education on higher education fundraising. To explore this phenomenon, the researcher interviewed fundraising managers at private, nonprofit colleges and universities in the western United States, and reviewed publicly available documents on the participant schools fundraising and financial information. The interviews demonstrated that study participants were concerned about connecting with graduates of online programs, and perceived that online students may have less affinity for their alma mater than on-ground alumni. In 2017, the U.S. Department of Education’s Nations Report Card disclosed performance trends among fourth and eighth grade students in New Mexico. Fourth graders were significantly below the national average in math and reading, while eighth graders were also performing below the national average (U.S. Department of Education). Spatial Exploration Of Trace Metals In Near-Surface Water And Soil In The Bolivian Altiplano, Lizzie Wilson

We analyzed soil and groundwater samples from the Bolivian Altiplano for trace metal concentrations, correlations and partitioning trends. Total digestion, sequential extraction and loss on ignition experiments were performed on soil samples. We ran aqueous samples on an ICP-MS. Results were analyzed to identify trace metal concentrations and find any significant correlations. We generated interpolated maps in ArcGIS to spatially analyze results. Results for elevated concentrations of interest were divided into sub-groups of organophilic metals, oxoanions and divalent metals and include vanadium, arsenic, selenium, zinc, nickel, lead, cobalt, uranium, copper and manganese. Neither the presence of black people in predominantly white spaces nor the appropriation of black culture are indicators of racial justice. Power structures must be integrated even more so than subdivisions, classrooms, breakrooms, church pews, and pop culture. This thesis will explore the absence of African-American leadership from the highest ranks of our power structures that are central to contemporary life and the dynamics within each of these power structures that help to protect white supremacy and therefore maintain segregation.

In a world that is increasingly dominated by advances made in digital systems, this work will explore the exploiting of naturally occurring physical phenomena to pave the way towards a self-powered sensor for Cyber-Physical Systems (CPS). In general, a sensor frontend can be broken up into a handful of basic stages: transduction, filtering, energy conversion, measurement, and interfacing. Membrane proteins are essential in many cellular processes and represent ~60% of all current drug targets. Due to technical limits, membrane proteins of various types were not studied extensively in the past and the biochemistry and functionality of them remain unclear. The structural biology methodologies require pure isolated protein samples for us to resolve their structure and study their biochemical functions. For such in vitro studies, however, membrane proteins often become unstable when isolated from their native lipid bilayer environment. Heart development and the genetic pathways underlying it are highly conserved among vertebrates. During heart development, an embryo must induce mesoderm formation, pattern the mesoderm, specify cardiomyocytes, increase the population of cardiomyocytes through proliferation, and pattern the cardiac chambers. It is becoming increasingly clear that chromatin modifications help mediate the complex processes of heart development by providing spatiotemporal regulation of gene expression. My thesis work focuses on characterizing functions of the chromatin factor Gonad-4-like (Gon4l), encoded by the gene ugly duckling (udu), in zebrafish heart development. Metabolism can convert drugs to harmful reactive metabolites that conjugate to DNA and off-target proteins. Reactive metabolites are a significant driver of both drug candidate attrition and withdrawal from the market of already approved drugs. Unfortunately, reactive metabolites are difficult to study in vivo, because they are transitory and generally do not circulate. Instead, this work computationally models both metabolism and reactivity. Using deep learning, predictive models were developed for the metabolic formation of quinones and epoxides, which together account for about half of known reactive metabolites. The genetic architecture of phenotypic divergence is a central question in evolutionary biology. Genetic architecture is impacted by whether evolution occurs through accumulation of many small-effect or a few large-effect changes, the relative contribution of coding and cis-regulatory changes, and the prevalence of epistatic effects. Our empirical understanding of the genetic basis of evolutionary change remains incomplete, largely because reproductive barriers limit genetic analysis to those phenotypes that distinguish closely related species. Chikungunya virus (CHIKV) is an arthritogenic alphavirus that during acute disease causes fever as well as severe joint and muscle pain. Chronic joint and muscle pain persists in a significant subset of patients, yet we still have a poor understanding of what drives this chronic disease. APOE4 is the strongest genetic risk factor for late-onset Alzheimer's disease (AD). APOE4 increases brain amyloid-β (Aβ) pathology relative to other APOE isoforms. However, whether APOE independently influences tau pathology, the other pathological hallmark of AD and other tauopathies, or tau-mediated neurodegeneration, is not clear. By generating P301S tau transgenic mice on either a human APOE knock in (KI) or APOE knockout (KO) background, we show that the presence of human APOE, regardless of APOE isoforms, leads to various degrees of brain atrophy in 9-month old P301S mice, whereas APOE ablation strongly protects against neurodegeneration. Among optical imaging techniques light sheet fluorescence microscopy stands out as one of the most attractive for capturing high-speed biological dynamics unfolding in three dimensions. The technique is potentially millions of times faster than point-scanning techniques such as two-photon microscopy. This potential is especially poignant for neuroscience applications due to the fact that interactions between neurons transpire over mere milliseconds within tissue volumes spanning hundreds of cubic microns. Animals show circadian rhythms in a variety of physiological functions and behaviors. In Drosophila melanogaster, behavioral rhythms are driven by circadian clock genes that are oscillating in ~150 circadian pacemaker neurons. To explain how circadian neurons encode time and regulate different behavioral rhythms, I performed 24-hour in vivo whole-brain calcium imaging using light-sheet microscopy. First, I found that different groups of circadian neurons show circadian rhythms in spontaneous neural activity with diverse phases. Previous research has found that adding different forms of variability during study can affect later memory at test. For example, having words spoken by different talkers has been shown to improve recall of known and novel words (Goldinger et al., 1999; Barcroft & Sommers, 2005), and varying the cues in cue-target related word pairs has been found to improve recall of the targets (Glenberg, 1979; Bevan et al., 1966). It was unclear, however, whether benefits of variability would extend to more naturalistic stimuli, such as sentences, which have higher working memory demands. This study investigated the relation between rhetorics of transparency and organizational action. Digging into CSR literature from a philosophy of communication perspective, this project seeks to determine if corporate social responsibility delivers on the promises it makes of a better world.

This paper is a reflection on the phenomenon of voluntary isolation in Amazonia, about anthropology’s implication in its formation as a concept, and what anthropologists might profitably say about it as a concrete phenomenon in the world. While knowledge based on ethnographic fieldwork might by minimal or even totally absent for people in voluntary isolation, anthropological research has produced a very impressive understanding of indigenous Amazonian social forms in general, knowledge that can be brought to bear on the question.

With the accelerating deforestation of Amazonia linked with the cultivation of soya used to feed animals for meat, and with calls to reduce or abandon meat consumption as one way of averting catastrophic climate change, it is topical once again. In this capstone project, I advocate for adding makerspaces to art museum education programming. I review the foundational educational concepts of a makerspace which help explain why it is a modern strategy to increasing visitor learning. This includes John Dewey’s theories of experience and education, Howard Gardner’s multiple intelligences theory, George Hein’s studies of museum visitor behavior, and the physiological science of learning. However, in the past seventeen years there have been several new developments in the provenanceresearchfield, and an updated guide has yet to be released. I propose that several changes be made to create an updated version. Drying lakes are a worldwide catastrophe. The goals of this dissertation research were (1) to investigate whether chemical fingerprints of lake sediments, and nearby land soils, could be established by the analysis of 44 chemical elements, moisture content, Eh, pH, and leachable anions and if so, (2) whether the fingerprints could help explain the process of physical and chemical changes in the lake sediments as they became soils when the lake's water level fell and the anoxic sediments were exposed to the air and weather.

This dissertation summarizes over four years of effort towards the completion of a Ph.D. in Biological Sciences. The work presented in this document covers a broad range of topics, but a central unifying theme is prokaryotic life in the continental subsurface. The work presented in each chapter relied heavily on cultivation-independent methods for assessing prokaryotic communities, including prokaryotic community structure reconstruction from high-throughput sequencing of 16S rRNA gene libraries and single-cell genome analysis of novel uncultivated bacteria.
