The nonnative hemlock woolly adelgid is steadily killing eastern hemlock trees in many parts of eastern North America. We summarize impacts of the adelgid on these forest foundation species; review previous models and analyses of adelgid spread dynamics; and examine how previous forecasts of adelgid spread and ecosystem dynamics compare with current conditions. The adelgid has reset successional sequences, homogenized biological diversity at landscape scales, altered hydrological dynamics, and changed forest stands from carbon sinks into carbon sources. A new model better predicts spread of the adelgid in the south and west of the range of hemlock, but still under-predicts …

Mitochondria are bilayer sub-cellular organelles that are an integral part of normal cellular physiology. They are responsible for producing the majority of a cell’s ATP, thus supplying energy for a variety of key cellular processes, especially in the brain. Although energy production is a key aspect of mitochondrial metabolism, its role extends far beyond energy production to cell signaling and epigenetic regulation–functions that contribute to cellular proliferation, differentiation, apoptosis, migration, and autophagy. Recent research on neurological disorders suggest a major metabolic component in disease pathophysiology, and mitochondria have been shown to be in the center of metabolic dysregulation and possibly …

With the absence of effective prophylactic vaccines and drugs against African trypanosomosis, control of this group of zoonotic neglected tropical diseases depends the control of the tsetse fly vector. When applied in an area-wide insect pest management approach, the sterile insect technique (SIT) is effective in eliminating single tsetse species from isolated populations. The need to enhance the effectiveness of SIT led to the concept of investigating tsetse-trypanosome interactions by a consortium of researchers in a five-year (2013-2018) Coordinated Research Project (CRP) organized by the Joint Division of FAO/IAEA. The goal of this CRP was to elucidate tsetse-symbiome-pathogen molecular interactions …

It is well known that young organisms do not maintain memories as long as adults, but the mechanisms for this ontogenetic difference are undetermined. Previous work has revealed that the α-amino-3-hydroxy-5-methyl-4isoxazolepropionic acidreceptor (AMPAr)subunits aretraffickedinto the synaptic membranefollowing memory retrieval in adults. Additionally, phosphorylated PSD-95-pS295 promotes AMPAr stabilization at the synapse. We investigated these plasticity related proteins as potential mediators in the differential contextual stress memory retrieval capabilities observed between adult and juvenile rats. Rats were assigned to either pedestal stress (1h) or no stress control (home cage). Each animal was placed alone in an open field for 5minat the base …

Background: Susceptibility of tsetse flies (Glossina spp.) to trypanosomes of both humans and animals has been associated with the presence of the endosymbiont Sodalis glossinidius. However, intrinsic biological characteristics of the flies and environmental factors can influence the presence of both S. glossinidius and the parasites. It thus remains unclear whether it is the S. glossinidius or other attributes of the flies that explains the apparent association. The objective of this study was to test whether the presence of Trypanosoma vivax, T. congolense and T. brucei are related to the presence of S. glossinidius in tsetse flies when other factors …

Ambergris, which is a coprolith originating from the sperm whale, has been found only rarely, but for centuries, as jetsam on beaches all over the world. There are no reliable data indicating how long such samples may have remained at sea, with unsubstantiated accounts suggesting maybe decades. Here, we obtained over forty jetsam samples, many collected on known dates, from mostly known beach locations across the globe. Such an inventory of verified jetsam ambergris is unprecedented. Each sample was characterised by analytical methods such as gas chromatography-mass spectrometry (GC-MS). We then determined the radiocarbon ages of some of the samples …

Overexpression of the human epidermal growth factor receptor 2 (HER2) is the cause of HER2- positive breast cancer (BC). Although HER2-inactivating therapies have benefited BC patients, development of resistance and disease recurrence have been the major clinical problems, pointing to a need for alternative therapeutic strategies. For that to happen, proteins that play critical roles in the biology of HER2-induced tumorigenesis have to be identified and characterized. Here, we show that the Src homology phosphotyrosyl phosphatase 2 (Shp2) encoded by the Ptpn11 gene is a requisite for ErbB2-induced tumorigenesis. We report that conditional knockout of Shp2 alleles in the ErbB2 …

Background: Cone and rod photoreceptors are two of the primary cell types affected in human retinal disease. Potential strategies to combat these diseases are the use of gene therapy to rescue compromised photoreceptors or to generate new functional photoreceptors to replace those lost in the diseased retina. Cis-regulatory elements specific to cones, rods, or both types of photoreceptors are critical components of successful implementation of these two strategies. The purpose of this study was to identify and characterize the cell type specificity and activity of cis-regulatory elements active in developing photoreceptors.
Methods: Cis-regulatory elements were introduced into the developing chicken …

With the continued adoption of genome‐scale data in evolutionary biology comes the challenge of adequately harnessing the information to make accurate phylogenetic inferences. Coalescent‐based methods of species tree inference have become common, and concatenation has been shown in simulation to perform well, particularly when levels of incomplete lineage sorting are low. However, simulation conditions are often overly simplistic, leaving empiricists with uncertainty regarding analytical tools. We use a large ultraconserved element data set (>3,000 loci) from rattlesnakes of the Crotalus triseriatus group to delimit lineages and estimate species trees using concatenation and several coalescent‐based methods. Unpartitioned and partitioned maximum …

The Long Term Effect Of Agricultural, Vadose Zone And Climatic Factors On Nitrate Contamination In Nebraska's Groundwater System, Pongpun Juntakut, Daniel D. Snow, Erin M.K. Haacker, Chittaranjan Ray
Department of Civil and Environmental Engineering: Faculty Publications
A four-decade dataset (1974–2013) of 107,823 nitrate samples in 25,993 wells from western and eastern parts of Nebraska was used to assess long-term trends of groundwater nitrate concentration and decadal changes in the extent of groundwater nitrate-contaminated areas (NO3-N≥10 mg N/L) over the entire state. Spatial statistics and regressions were used to investigate the relationships between groundwater nitrate concentrations and several potential natural and anthropogenic factors, including soil drainage capacities, vadose zone characteristics, crop production areas, and irrigation systems. The results of this study show that there is no statistically significant trend in groundwater nitrate concentrations in western Nebraska, in …

Comparison Of Space-Plant Versus Sward Plot Selection In Thickspike Wheatgrass (Elymus Lanceolatus), Joseph G. Robins, Kevin B. Jensen
Forage and Range Research Laboratory Publications
Thickspike wheatgrass (Elymus lanceolatus [Scribn. & J.G. Sm.] Gould) is an important native perennial grass species used for rangeland revegetation in North America. Plant breeding efforts relying on space‐plant evaluations have resulted in limited improvement in this species. The purpose of this study was to characterize the performance of thickspike wheatgrass half‐sib families under space‐plant and sward plot evaluations, estimate the correlation between measured traits in both evaluation settings, and determine the validity of selecting thickspike wheatgrass for rangeland revegetation in the nontarget environment space‐plant plots. The study included 50 thickspike wheatgrass half‐sib families and five commercial cultivars and experimental …

The Cradle of Humankind (Cradle) in South Africa preserves a rich collection of fossil hominins representing Australopithecus, Paranthropus and Homo1. The ages of these fossils are contentious2,3,4 and have compromised the degree to which the South African hominin record can be used to test hypotheses of human evolution. However, uranium–lead (U–Pb) analyses of horizontally bedded layers of calcium carbonate (flowstone) provide a potential opportunity to obtain a robust chronology5. Flowstones are ubiquitous cave features and provide a palaeoclimatic context, because they grow only during phases of increased effective precipitation6,7, ideally in closed caves. Here we show that flowstones from eight …

‘Matterhorn’ (Reg. No. CV-1151, PI 687896) hard white winter waxy wheat (Triticum aestivum L.) was developed cooperatively by the USDA-ARS and the Nebraska Agricultural Experiment Station and released in 2018. Matterhorn, a sibling of the hard red waxy cultivar Mattern, has white grain color and waxy (amylose-free) endosperm starch. It was released primarily for its unique end-use quality attributes and for grain yield competitiveness with currently grown Nebraskaadapted cultivars. The waxy starch is conditioned by the presence of three naturally occurring mutations that eliminate production of the enzyme granule-bound starch synthase. Granule-bound starch synthase synthesizes amylose in typical wheats and …

Game Play In Education: An Exploration Of The What, How, And Why, Carrin Lynn Blyth
CUP Ed.D. Dissertations
This study explored whether teachers utilize games in the classroom, known as gamification and, if so, whether they used games according to game theory that exists in the field of gaming and game design. In higher education and secondary environments, educators report that problems exist as far as student behavior and engagement. Many students do not want to learn. The literature review conducted as part of this study indicated that when teachers gamified their classrooms few empirical investigations were conducted in the K-12 setting; another shortcoming was a lack of a uniform classification system for game elements among the literature, …

The mechanisms underlying axon regeneration in mature neurons are relevant to the understanding of normal nervous system maintenance and for developing therapeutic strategies for injury. Here, we report novel pathways in axon regeneration, identified by extending our previous function-based screen using the C. elegans mechanosensory neuron axotomy model. We identify an unexpected role of the nicotinamide adenine dinucleotide (NAD+) synthesizing enzyme, NMAT-2/NMNAT, in axon regeneration. NMAT-2 inhibits axon regrowth via cell-autonomous and non-autonomous mechanisms. NMAT-2 enzymatic activity is required to repress regrowth. Further, we find differential requirements for proteins in membrane contact site, components and regulators of the …

© 2018 Field, McVicker and Maruska. Across vertebrates, the use of multimodal (multiple sensory modalities) signals has evolved to convey important information to receivers. Information content of multimodal signals can be the same as or different from information in each unimodal signal, and are classified as redundant or non-redundant, respectively, based on receivers’ behavioral responses. Despite the prevalence and importance of multimodal signaling across taxa, relatively little is known about how and where these signals are processed in the brains of receivers. We used the social African cichlid fish, Astatotilapia burtoni, to investigate how sexually-relevant visual and chemosensory uni- and …
