Phox2a and Phox2b are two homeodomain transcription factors playing a pivotal role in the development of noradrenergic neurons during the embryonic period. However, their expression and function in adulthood remain to be elucidated. Using human postmortem brain tissues, rat stress models and cultured cells, this study aimed to examine the alteration of Phox2a and Phox2b expression. The results show that Phox2a and Phox2b are normally expressed in the human locus coeruleus (LC) in adulthood. Furthermore, the levels of Phox2a protein and mRNA and protein levels of Phox2b were significantly elevated in the LC of brain donors that suffered from the …

There is increasing evidence that the hemisphere ipsilateral to a moving limb plays a role in planning and executing movements. However, the exact relationship between cortical activity and ipsilateral limb movements is uncertain. We sought to determine whether 3D arm movement kinematics (speed, velocity, and position) could be decoded from cortical signals recorded from the hemisphere ipsilateral to the moving limb. By having invasively monitored patients perform unilateral reaches with each arm, we also compared the encoding of contralateral and ipsilateral limb kinematics from a single cortical hemisphere. In four motor-intact human patients (three male, one female) implanted with electrocorticography …

Evolution Of Bidirectional Costly Mutualism From Byproduct Consumption, Lon Chubiz, William Harcombe, Jeremy Chacón, Elizabeth Adamowicz, Christopher Marx Nov 2018

Evolution Of Bidirectional Costly Mutualism From Byproduct Consumption, Lon Chubiz, William Harcombe, Jeremy Chacón, Elizabeth Adamowicz, Christopher Marx

Biology Department Faculty Works

Mutualisms are essential for life, yet it is unclear how they arise. A two-stage process has been proposed for the evolution of mutualisms that involve exchanges of two costly resources. First, costly provisioning by one species may be selected for if that species gains a benefit from costless byproducts generated by a second species, and cooperators get disproportionate access to byproducts. Selection could then drive the second species to provide costly resources in return. Previously, a synthetic consortium evolved the first stage of this scenario: Salmonella enterica evolved costly production of methionine in exchange for costless carbon byproducts generated by …

The Relative Effects Of Forest Amount, Forest Configuration, And Urban Matrix Quality On Forest Breeding Birds, Alexandra V. Shoffner, Andrew M. Wilson, Wenwu Tang, Sara A. Gagné Nov 2018

The Relative Effects Of Forest Amount, Forest Configuration, And Urban Matrix Quality On Forest Breeding Birds, Alexandra V. Shoffner, Andrew M. Wilson, Wenwu Tang, Sara A. Gagné

Environmental Studies Faculty Publications

Urbanization modifies landscape structure in three major ways that impact avian diversity in remnant habitat: habitat amount is reduced and habitat configuration and matrix quality are altered. The relative effects of these three components of landscape structure are relatively well-studied in agricultural landscapes, but little is known about the relative effect of urban matrix quality. We addressed this gap by investigating the relative effects of forest amount, forest configuration, and matrix quality, indicated by degree of urbanization and agriculture amount, on the diversity of three guilds of forest birds using data from 13,763 point counts from Pennsylvania, USA. Forest amount …

A combined therapy of human umbilical cord blood cells (hUCB) and G-CSF at the acute stage of TBI was tested as a therapeutic for progressive secondary effects of chronic TBI. Rats were treated with saline carrier, or therapeutic in carrier as follows; G-CSF, hUCB, or hUCB and G-CSF, 7-days after TBI. Eight weeks later, behavioral testing was performed and brains harvested to analyze hippocampal cell loss, neuroinflammatory response, and neurogenesis. Results revealed that the monotherapies partially suppressed neuroinflammation and reduced hippocampal cell loss. However, combined therapy of hUCB and G-CSF robustly dampened neuroinflammation, while enhancing endogenous neurogenesis and reducing hippocampal …

Lcp1 Mutant Zebrafish: A Look At Neutrophils, Cancer, And Gene Compensation, Taylor Mitchell

College of Science and Health Theses and Dissertations

Lymphocyte cytosolic protein 1 (lcp1 or L-plastin) is a small actin-bundling protein that is typically only expressed in motile leukocytes, such as neutrophils and macrophages. However, it is also overexpressed in cancer cells, which may be related to tumor metastasis. Using CRISPR/Cas9 gene editing, our lab has created zebrafish that are genetic knockouts for lcp1 in order to better understand the relationship between L-plastin and cell motility.

Previous studies on L-plastin knockout mice have shown that the mutants have a decreased immune response, and therefore I predicted that our zebrafish mutants might have impaired development or distribution of immune cells. …

Chronic Life Stress And Change In Stress Response Functioning In Urban Youth: The Role Of Social Support And Gender, Heather M. Marshall Nov 2018

Chronic Life Stress And Change In Stress Response Functioning In Urban Youth: The Role Of Social Support And Gender, Heather M. Marshall

College of Science and Health Theses and Dissertations

Salivary alpha-amylase (sAA) has been established as a bio-marker of stress reactivity in acute stressor tasks. Much less research exists exploring whether sAA is bio-marker for chronic stress exposure, and therefore, allostatic load. Extant research is inconsistent with some evidence to support chronic stress exposure having an impact on sAA reactivity and diurnal levels of sAA, while other studies have failed to support this theory. Social support is regarded as a protective factor against stress, mental health, and physical health consequences. It has been suggested that gender may play a role in the utilization and benefits of social support networks. …

Phenotyping And Sorting Of Synthetic Cells: Building Bridge From Spectroscopy To Genetics, Ma Bo, Xu Jian Nov 2018

Phenotyping And Sorting Of Synthetic Cells: Building Bridge From Spectroscopy To Genetics, Ma Bo, Xu Jian

Bulletin of Chinese Academy of Sciences (Chinese Version)

In synthetic biology, the dazzling methodological innovations in sequencing, editing and synthesis of genomes have resulted in an unprecedented capability in "design and manufacturing of genotype". On the other hand, "testing of cellular phenotype and function" has increasingly become one major bottleneck. Single-cell technologies have tremendous implications and potentials in rapid testing of cellular function. However, ideally, such single-cell methods should allow non-invasive live-cell probing, be label-free, provide landscapelike phenotyping capability, distinguish complex functions, operate with high speed, sufficient throughput and low-cost, and finally, be able to couple with downstream omics analysis via cell sorting. In this perspective article, we …

Nondestructive 3d Imaging And Quantification Of Hydrated Biofilm-Sediment Aggregates Using X-Ray Microcomputed Tomography., N Zhang, Cel Thompson, Ih Townend, Ke Rankin, Dm Paterson, Aj Manning

School of Biological and Marine Sciences

Biofilm-sediment aggregate (BSA) contains a high water content, either within internal pores and channels or bound by extracellular polymeric substances (EPS) forming a highly hydrated biofilm matrix. Desiccation of BSAs alters the biofilm morphology and thus the physical characteristics of porous media, such as the binding matrix within BSA and internal pore geometry. Observing BSAs in their naturally hydrated form is essential but hampered due to the lack of techniques for imaging and discerning hydrated materials. Generally, imagery techniques (scanning electron microscopy (SEM), transmission electron microscopy (TEM), and focused ion beam nanotomography (FIB-nt)) involve the desiccation of BSAs (freeze-drying or …

Metacognitive Reading Strategy And Emerging Reading Comprehension In Students With Intellectual Disabilities, Natasha Cox-Magno, Peter Ross, Kathleen Dimino, Andrea Wilson

Journal of Educational Research and Practice

This article ventures to address the gap in special education practices by providing a metacognitive reading strategy to support the emerging reading comprehension skills of kindergarten students with intellectual disabilities. Historically, students with intellectual disabilities have low reading comprehension skills that can impede their overall academic success. There is a gap in practice regarding the identification and effective use of evidence-based reading comprehension instructional strategies for students with intellectual disabilities. Guided by Piaget’s and Vygotsky’s constructivist theories, the purpose of this study was to test the effectiveness of a metacognitive reading strategy on the emerging reading comprehension (ERC) skills of …

Relating Cyanobacteria And Physicochemical Water-Quality Properties In Willow Creek Lake, Nebraska, 2012–14, David L. Rus, Brent M. Hall, Steven A. Thomas

United States Geological Survey: Water Reports and Publications

Document abstract

Cyanobacteria (also referred to as blue-green algae) are naturally present members of phytoplankton assemblages that may detract from beneficial uses of water because some strains produce cyanotoxins that pose health hazards to people and animals. Cyanobacteria populations observed in Willow Creek Lake during 2012 through 2014 were compared to external nutrient loading from the Willow Creek drainage basin and several other physicochemical properties within the lake, including internal nutrient loading. This report is part of a cooperative study between the United States Geological Survey, the Lower Elkhorn Natural Resources District, the Nebraska Department of Environmental Quality, the Nebraska …

More Than Just Collateral Damage: Pet Shootings By Police, Courtney G. Lee Nov 2018

More Than Just Collateral Damage: Pet Shootings By Police, Courtney G. Lee

The University of New Hampshire Law Review

The Department of Justice estimates that American police officers shoot 10,000 pet dogs in the line of duty each year. It is impossible to ascertain a reliable number, however, because most law enforcement agencies do not maintain accurate records of animal killings. The tally may be substantially higher, and some suggest it could reach six figures.

Deferring to officers’ judgment when they reasonably fear for human safety is sound policy because they regularly must make split-second, life-or-death decisions in highly stressful situations; but many pet shootings occur when officers mistake the behavior of a friendly, curious dog for aggression. Further, …

Don't Go Chasing Waterfalls: The Intrepid, Pioneering, Whitewater Paddler's Right To Stop On Private Land, Jennifer Jolly-Ryan

The University of New Hampshire Law Review

The river roars from the thunder of waterfalls and swift currents. Its water runs wild between large boulders and steep banks. Kayaks and canoes are designed to navigate whitewater rapids that many people believe are unnavigable. Conflicts often arise between private property owners and the recreating public on some of the best whitewater in America. Private property owners along rivers, creeks, and streams have shot at paddlers for entering their private property. Property owners have obstructed navigation, stringing artificial strainers made of barbed wire across creeks and streams.

The sport of running wild water rapids and waterfalls in a kayak …

Professional Practices (ART 399)

My art comprises one unifying idea: my own life experiences, specifically those relating to my family and friends. By depicting highly specific visual cues—such as clothing, crows, and the color blue—I combine multiple facets and memories in an attempt to communicate my feelings and experiences. Sometimes these symbols are universal; a yellow raincoat, for example, calls to mind childhood. Others are encoded within my own memories—a bird skull relates to a dream my friend had one night. My art is about more than myself; it is about finding common ground between me and my audience and exploring that overlap.
